Abstract

Konsumsi ikan di Indonesia terus mengalami peningkatan seiring dengan target nasional mencapai 59 kg/kapita, menjadikan ikan gurami sebagai salah satu komoditas budidaya penting. Namun, produksi gurami menghadapi tantangan serius akibat tingginya kerentanan terhadap penyakit, khususnya Motile Aeromonas Septicemia (MAS) yang disebabkan oleh Aeromonas hydrophila. Bakteri ini memiliki tingkat virulensi tinggi yang dipengaruhi oleh berbagai faktor patogenik, salah satunya enzim dan toksin hemolisin (hlyA) yang berperan dalam melisis sel darah merah, memicu nekrosis jaringan, dan memperparah infeksi. Upaya pengendalian penyakit dapat dilakukan secara organik maupun anorganik, namun pengembangan vaksin berbasis bioinformatika menjadi pendekatan yang menjanjikan dengan menargetkan gen virulensi seperti hemolisin. Penelitian ini merupakan penelitian laboratorium dengan pendekatan observasional deskriptif. Sampel diperoleh dari ikan gurami yang diduga terinfeksi MAS dan dianalisis melalui tahapan isolasi bakteri, pemurnian, serta uji biokimia. DNA bakteri kemudian diekstraksi dan gen hemolisin diamplifikasi menggunakan metode PCR dengan primer spesifik. Produk PCR dianalisis melalui elektroforesis dan disekuensing menggunakan metode Sanger. Analisis homologi dilakukan menggunakan BLAST, sementara hubungan filogenetik dianalisis dengan perangkat lunak MEGA11. Selain itu, prediksi epitop imunogenik dilakukan menggunakan perangkat bioinformatika berbasis IEDB. Hasil penelitian menunjukkan keberadaan gen hlyA dengan ukuran 592 bp pada isolat A. hydrophila. Analisis homologi menunjukkan tingkat kesamaan 98–100% dengan tujuh strain dari GenBank, serta hubungan filogenetik yang erat dengan strain asal India dan Cina. Prediksi epitop menghasilkan tujuh kandidat terbaik untuk sel B, serta 30 epitop MHC II dan 10 epitop MHC I yang bersifat antigenik, non-alergenik, tidak toksik, dan larut air. Temuan ini menunjukkan potensi gen hemolisin sebagai kandidat vaksin berbasis bioinformatika untuk pengendalian MAS pada ikan gurami.

Abstract

Coastal areas are dynamic ecosystems that hold strategic value both ecologically and economically. The presence of infrastructure, such as jetties, has the potential to cause a decline in seawater quality in coastal regions. This study aims to determine the seawater quality and the Pollution Index based on physical, chemical, and microbiological parameters over a two-year period around a jetty used for fuel termina operations. The research was conducted in the sea water of Ulakan Village, Manggis District, Bali Province. The research method involved in-situ measurements of several parameters and the collection of seawater samples for laboratory analysis. The laboratory test results were then used to determine the seawater quality status using the Pollution Index (PI), based on the quality standards for marine life. The results showed that the seawater quality around the jetty for fuel storage operations exhibited significant dynamics in physical, chemical, and microbiological parameters throughout the period from 2024 to early 2025. Parameters that did not meet the established quality standards were water clarity in July 2024, as well as Copper (Cu) at locations AL2 and AL3 in January 2025. The Pollution Index analysis concluded that while all locations were initially in 'Good' condition in January 2024 with a value of 0.62, a degradation in quality occurred, moving into the 'Slightly Polluted' category in July 2024 with values ranging from 1.02 to 1.03. By January 2025, only location AL1 showed recovery in quality with an index of 0.59, demonstrating a better environmental recovery capacity in that area. Meanwhile, AL2 and AL3 experienced an increase in the index to 2.15 and 1.98, respectively, remaining in the 'Slightly Polluted' category.

Abstract

The fishing centers that can be utilized in Lampung Province is the TPI Gudang Lelang Market. The catch from fishermen around TPI Gudang Lelang consists of economically valuable fish such as hairtail, scads, and mackerel fish. Fishermen at TPI Gudang Lelang can produce catches of approximately 90-100 tons per month. The objective of this independent project is to determine the weight and length of the mackerel fish (Rastrelliger sp) caught at TPI Gudang Lelang and to analyze the relationship between the length and weight of the Kembung fish at this location. This independent project was conducted from September to December 2022, located at TPI Gudang Lelang, Teluk Betung, Bandar Lampung. The method employed was direct observation in the field. The subjects of the study were the length and weight of the Kembung fish. The mackerel fish (Rastrelliger sp) found at TPI Gudang Lelang have an average body length of 19.6 cm and an average weight of 0.116 grams. The relationship between the length and weight of the mackerel fish, analyzed using regression analysis, yielded a value of (b) 2.84, indicating that the growth in length does not correspond with the growth in weight (with length growth being more dominant than weight growth). Additionally, the value of (R²) is 0.607, suggesting a weak relationship between the length and weight of the mackerel fish.

Abstract

This study developed a Slim Fit Wall (SFW) Aquarium as a medium for ornamental fish conservation and a decorative element for minimalist buildings. The aquariums were designed to be slim with dimensions of 80 × 10 × 40 cm and 100 × 10 × 60 cm, using glass materials and lightweight steel brackets. This design offers a space-saving solution without sacrificing visual and functional aspects. Testing involved strength simulations using Autodesk Inventor software, showing Von Mises stresses below the material's safe limit, ensuring structural strength. Visual and functional tests were conducted with respondents from the KP Kupang Polytechnic. The results showed an average visual satisfaction level of 83.9%, with advantages in space efficiency and visual comfort. Functionality was tested for one month, demonstrating stable performance without component damage. The study's conclusion confirms that the SFW Aquarium is suitable for minimalist buildings, fulfilling aesthetic needs, and supporting conservation awareness. Further research is recommended to explore variations in size and design.

Abstract

Frozen tuna loin is a leading product of the fisheries industry for export. To ensure food safety and high quality, consistent cold chain implementation needs to be strictly implemented, as well as product quality analysis before distribution. The quality of frozen tuna loin is greatly influenced by microbiological conditions, histamine levels, organoleptic quality, and yield efficiency during the processing process. This study aims to analyze the microbiological quality, histamine levels, yield, and organoleptic quality of frozen tuna loin products produced by PT. XYZ, Bitung City. The parameters tested include Total Plate Count (TPC), E. coli, Salmonella, histamine levels, yield, and organoleptic tests. The results showed that the temperature during the processing process was within the safe range according to the Company's standards and SNI. The ALT, E. coli, and Salmonella values were below the SNI 4104:2015 threshold, as was the histamine level below 50 ppm. The organoleptic value ranged from 7.9 to 8.5, categorized as very good, and the final cumulative yield reached ±58%. These results indicate that the implementation of a cold chain-based processing system effectively maintains the microbiological, physical, and chemical stability of frozen tuna loin products, thereby meeting export safety and quality standards.

Abstract

This study aimed to isolate psychrotrophic histamine-forming bacteria (BPH) from bigeye tuna using Niven and EMB (eosine methylene blue) pre-screening media, and to identify the obtained BPH based on their biochemical and molecular characteristics. BPH were isolated from bigeye tuna that had been incubated at 16 oC for 5 days. Positive BPH isolates from Niven and EMB media were then grown in TSBH (trypticase soya broth histidine) medium at 5, 16, and 30 oC to test their activity in forming histamine using TLC (thin layer chromatography). Selected BPH were then identified biochemically and molecularly. BPH with high histamine-forming ability were obtained from Niven medium as many as 16 isolates (39.02%) and from EMB medium as many as 3 isolates (5.35%). The highest histamine-forming activity was obtained at 30 oC and then biochemical testing was carried out for all isolates at that temperature. Presumptive biochemical characterization of 19 isolates obtained 6 isolates that have different biochemical characteristics, namely oxidase, motility, ability to hydrolyze gelatin, and the ability to grow in 10% NaCl. The isolates came from Niven medium with codes TMB-N4, TMB-N9, TMB-N25, and TMB-N26 and from EMB medium with codes TMB-E38 and TMB-E40. The results of molecular testing for 6 isolates obtained the genus Morganella with a similarity percentage of >99% and biochemical characterization that leads to the species Morganella morganii subsp. sibonii. The acquisition of Morganella morganii subsp. sibonii bacteria from both EMB and Niven medium indicates that these bacteria are important organisms in the formation of histamine in fish that experience temperature abuse conditions during handling.

Abstract

The productivity of whiteleg shrimp (Litopenaeus vannamei) cultivation is influenced by technical and non-technical factors. Technical factors include water quality, growth and survival, and feed efficiency. Non-technical factors, such as business analysis and feasibility, are related to cultivation productivity. The study was conducted in Pandeglang, Banten, Indonesia, for three months. Six ponds were observed, each with an area of 2.500 m2, 2.000 m2 and 1.500 m2, with a stocking density of 125 shrimp/m2 . The results showed that water quality was mostly within the optimal range, except for parameters such as brightness, nitrite, ammonium, ammonia, phosphate, Total Organic Matter (TOM), and magnesium. The growth values of Average Body Weight (ABW) and Average Daily Growth (ADG) were very good, with a low mortality rate or high SR, and optimal feed efficiency throughout the pond. The results of the business analysis show a positive value on the productivity of L. vannamei. In general, productivity factors such as water quality, growth and survival, feed efficiency and business analysis are related to biomass production with R2 values, namely: 0.9942 (pond 1), 0.9922 (pond 2), 0.9956 (pond 3), 0.9598 (pond 4), 0.9903 (pond 5) and 0.9844 (pond 6).

Abstract

This study aimed to evaluate the sensory quality, consumer acceptance, and nutritional composition of several commercial nori products available on the market. Three commercial nori samples coded as A, B, and C were tested using an organoleptic method by assessing color, aroma, taste, and texture attributes. In addition, the nutritional composition was analyzed based on the nutrition facts listed on the product packaging, including energy, fat, carbohydrates, protein, fiber, and sodium. The results of the organoleptic test showed that product B obtained the highest average score of 4.56, followed by product A with 3.84 and product C with 3.54. Sensory evaluation indicated that product B had a dark glossy color, distinctive roasted aroma, sweet savory taste, and stable crispy texture, making it the most preferred by panelists. Nutritional composition analysis showed that products B and C had the highest energy content of 25 kcal per serving, while product A had a lower energy content of 15 kcal. Product C contained the highest fat content (2.5 g), whereas product A had the highest carbohydrate and protein contents. Differences in sensory characteristics and nutritional composition among the products influenced consumer acceptance of commercial nori products.

Abstract

Demang Gedi Mangrove Education Park (TEM) in Purworejo Regency holds significant natural resource potential; however, it faces challenges such as declining visitor numbers and management constraints. This study aims to analyze the ecological condition of the mangrove forest by examining mangrove density and to formulate appropriate ecotourism development strategies using the IFAS (Internal Factor Analysis Summary) and EFAS (External Factor Analysis Summary) matrix approach. The research methodology involved field surveys to collect vegetation density data, alongside interviews and questionnaires for SWOT analysis. The results indicate that the ecological condition of the mangrove forest is classified as 'very good,' with vegetation density ranging from 3,466 to 4,366 ind/ha. Based on the strategic analysis, the development position of TEM Demang Gedi is located in Quadrant I (Growth-Oriented Strategy). This position indicates that the area possesses strong internal strengths to capitalize on existing external opportunities. The recommended priority strategy is a progressive strategy, specifically maximizing natural potential and government support to develop sustainable conservation-based educational tourism.
