Abstract

Availability of moleculer marker to identify the important agronomic character of plant is needed to accelerate selection activity of plant. Particularly for the complex quantitatively inherited traits - like e.g. shading tolerance of soybean - the use of such a technique will speed up the process to produce adapted genotypes.The objective of this research is to identify the linkage of molecular marker RAPD with character of shading tolerance. The morphological specific characters wich is corelated to shade tolerance is the number of productive branchs. The intensity 75% of artificial shading is optimal level for doing selection of soybean genotypes. The inheritance of shading tolerance of soybean was controlled by gene with full dominant or by two genes pairs with dominant and ressesive epistasis. There was no maternal effect in the inheritance to shade tolerance. Heritability value (0.45 – 0.54) indicated that the proportion variation caused by the genetic factors was moderate. Molecular analysis by using RAPD technique showed that UBC153, ROTH 480.01, and ROTH 480.03 primer have polymorphic band that can be used for inheritance study and linkage analysis. All polymorphisms segregated independently of each ather. Interval mapping with Mapmaker/QTL indicated that the location of the three QTLs on linkage group were at marker tolerance locus of Roth 480.01-8125 , Roth 480.03-1125, and UBC 153-19125 .

Abstract

The objectives of the research were to make land suitability map for sugarcane plant (Saccharum officinarum), to give recommendation of location including area for sugarcane plant cultivation and to increase sugarcane plant productivity. The research used maps overlay and Geographical Information System (GIS) which used Arch-View Spatial Analysis version 2,0 A in Remote Sensing Laboratory, Agency for the Assessment and Application of Technology (BPPT), Jakarta. The research was carried out in Tegal Regency starting from June to October 2004.The results of the research showed that the suitable, conditionally suitable, and not suitable land for sugarcane cultivation in Tegal Regency reached to a high of 20,227 ha, 144 ha, and 81,599 ha respectively. There were six most dominant kind of soil: alluvial (32,735 ha), grumosol 5,760 ha), mediteran (17,067 ha), latosol (18,595 ha), glei humus (596 ha), and regosol (22,721 ha).

Abstract

Medium and small industry sector has an important role, not only as a job provider for the number of job seekers that increases from year to year, but also for production and distribution of goods and services or society. The expectationtowards the contribution of this sector in creating employment, generating regional income and national income from foreign exchange has been increasing especially because of the industry's capacity to compete in the global market due to its use of local content in production. In the Gunung Kidul District there are some various products that have potential and prospect to be developed. Some of them even has penetrated the export market. The Agency of District Development Planning of Gunung Kidul District has identified and analyzed some products and has therefore determined 5 (five) products that have potential to become the region's competitiveproducts. These are (based on rank) : 1. Wood Mask Handicraft, 2. Bamboo Handicraft, 3. Stone Handicraft, 4. Dried Cassava (Gaplek), 5. Cattle. In this article, the writer provides suggestions to the Gunung Kidul District Government for theirpolicy making for the development of wood mask handicraft small industry.

Abstract

The transportation sector is the second largest final energy consumer after the industry sector. The main energy issue in the transportation sector in Indonesia is the dominant use of oil fuels. Moreover, almost 60% of total oil fuels are being subsidized. On the contrary, the development and application of New and Renwable Energy (NRE) in the transportation sector has not been very successful. This paper applies the Analytical Hirarchy Process (AHP) in conjuction with optimization of the energy system using MARKAL model to select the best strategy for increasing the role of New and Renewable Energy (NRE) in the transportation sector. Three case studies are applied to evaluate the intervention by policy as an effort to increase the role of NRE in the transportation sector.

Abstract

Most of Country Border Area in Indonesia are underdeveloped area with limited social and economic infrastructure. On the adjacent area, the neighbor country such as Malaysia that adjacent with Sumatera already developed growth centersand border corridors with so many economic activities and trade. The disparity among country can not be avoided, mostly in economic aspect. To develop Country Border Area as equal with neighbor country and surrounding area, it is needed policy to establish Economic Development Area (KPE) in Country BorderArea. KPE is an area consist of some districts in border area that functionally bond to develop integrated superior economic. One of KPE developed is KPE Bagansiapiapi in Riau Province that adjacent with Malaysia. It has strategic location and abundant natural resources, but limited infrastructure as a constrainon economic development. For that reason, it is needed to formulate KPE Bagansiapiapi Strategic Plan based on agropolitan-marine integrated development area (AMIDA) concept.

Abstract

Perencanaan suatu proses produksi sederhana yang mudah diaplikasikan merupakan latar belakang dari riset ini. Pada riset ini dipelajari sejauh mana mekanisme pembentukan bahan dengan proses hot press metalurgi serbuk di lingkungan udara yang tidak dikondisikan berpengaruh terhadap karakteristik Aluminium Matrix Composites (AMCs). Komposit terbuat dari serpihan AC8A cor yang di haluskan sebagai matrik dan 20% Vf partikel SiC sebagai penguat. Komposit AC8A/SiCp dibuat dengan tekanan hot-press sebesar 425 MPa pada beberapa variasi temperatur yaitu 360ËšC, 380ËšC, 400ËšC dan 430ËšC di lingkungan udara yang tidak dikondisikan. Dilakukan pengukuran densitas dan uji kekerasan Vickers untuk melihat pengaruh temperatur terhadap karakteristik komposit AC8A/SiCp. Nilai kekerasan matrik meningkat seiring dengan peningkatan temperatur hingga 400ËšC dan menurun pada 430ËšC, sedangkan densitas dan nilai kekerasan komposit meningkat seiring dengan peningkatan temperatur

Abstract

Suatu penelitian telah dilakukan untuk mengevaluasi general combining ability (GCA) dan specific cobining ability (SCA) untuk sifat produksi dan reproduksi pada babi. Materi yang digunakan adalah catatan keturunan hasil persilangan tiga bangsa babi (Duroc, Landrace, Yorkshire) sebanyak 573 anak babi yang berasal dari 13 pejantan dan 65 induk. Formula yang digunakan untuk menganalisis parameter adalah Partial Diallel Cross Metode II dari Griffings. Parameters yang diamati pada anak adalah litter size, bobobt lahir, jumlah puting, pertambahan bobot badan (PBB) sebelum dan sesudah sapih dan bobot sapih. Hasil penelitian menunjukkan bahwa GCA Duroc lebih tinggi dibandingkan Yorksire and Landrace, yang nilai GCA untuk litter size, bobot lahir, jumlah puting, jumlah anak pada saat sapih, bobot sapih, PBB sebelum sapih, PBB setelah sapih dan bobot badan pada umur 42 hari masing-masing -0,18; -0,02; -0,52; -0,13; -0,21; -0,01; 0,01 dan 0,03. Persiilangan antara Duroc x Yorkshire menunjukkan SCA paling tinggi untuk bobot lahir (0.30), jumlah puting (3.60), bobot sapih (1.10) dan PBB sebelum sapih (0.04). Persilangan antara Yorkshire x Landrace menghasilkan SCA paling tinggi untuk litter size (2.47), jumlah anak waktu sapih (2.23), PBB sebelum sapih (0.04), PBB setelah sapih (0.06) and bobot badan pada umur 42 hari (2.30). Kesimpulan penelitian ini adalah nilai paling baik untuk GCA ditunjukkan oleh Duroc, sementara itu SCA oleh persilangan antara Yorkshire x Landrace.

Abstract

Kebun Agromedika Hamboro (Kamaro) is cultivation of medicinal plants demonstration farm (demfarm), especially rhizome plants. The purpose of this research is to identify the internal conditions (strengths and weaknesses) and external (opportunities and threats) Kamaro and to provide alternative strategies that can be applied in the development of educational tourism Kamaro as a garden of medicinal plants. Theresults showed that the IFE matrix that became a major force Kamaro factors include the product being offered is a useful medicinal plants for health, while its main weakness is a condition of the garden is not good. The results showed that EFE matrix is a major opportunity Kamaro factor is the development of science and technology so as to incrase business transactions, while the threat factor is climate and weather nature conditions are unpredictable. Based on the IE matrix results, Kamaro puts on depicting V cells in the internal conditions of the average and Kamaro response to external factors faced by relatively moderate. The strategy can be implemented is to preserve and maintain, an alternative strategy that many do is market penetration and product development. Results of the SWOT matrix are obtained seven alternative strategies. The results matrix QSP is obtained main strategic priority to improve the management system such as increase farm productivity, post-harvest handling, gardensanitation and magnificence, administration and finance and others.

Abstract

Demo–plant CFB boiler 30 ton steam/hour is a granted equipments of cooperation program between BPPT and NEDO Japan in technological study area of CFB Boiler to utilize low–rank coal which functioned as steam consumption supply as well as electricity at production process system in Paper Mill of Basuki Rachmat – Banyuwangi. But outside calculation in the year of 1998, Indonesia occured economic crisis which result activity of construction which have 90% perforced to be discontinued, because PKBR as area owner and construction funder expressed have national debt, so that the asset taken over by BPPN. Since the project activities discontinued, various effort of project solving have been conducted, one of them, BPPT propose study of CFB Boiler displace function as steam and electricity generation (co–generation). In this paper, writer try to elaborate result of study giving 5 option, that is (1) Stand–alone power station, (2) Co–generation 1 (new steam turbine), (3) Co–generation 2 (new steam turbine+cooling tower), (4) Co–generation 3 (existing steam turbine) and (5) Co–generation 4 (existing steam turbine+cooling tower).

Abstract

Aspergillus terreus is a Saprophyte fungus that produces several secondary metabolites as lovastatin (anti-cholesterol drug) and itaconic acid (a polymer material). Lovastatin is one of the statin class of drugs that have efficacy as antihypercholesterolemic. Plasmid transformation is the introduction and incorporation of exogenous plasmid into cells orprotoplast. In this study, pTRLI plasmid (pTRI inserts containing lovE gene as a regulator gene in the biosynthesis of lovastatin) will be transformed by electroporation transformation. The purpose of this research is transformation of pTRLI plasmid into protoplasts of Aspergillus terreus by electroporation and obtain stable transformants. The research was initiated by isolation of pTRLI plasmid. Then pTRLI plasmid was determined purity and concentration by nanodrop. Furthermore, Protoplasts of Aspergillus terreus were isolated enzymatically by adding an enzyme which can degrade the cell wall of Aspergillus terreus which contains chitin and cellulose. PTRLI plasmid were transformed into protoplasts of Aspergillus terreus by electroporation. These transformants were grown in Czapek-Dox medium containing pyrithiamine agar and the number of transformants mg-1 of pTRLI plasmid was calculated. Transformants were selected to grow in Czapek-Dox medium containing piritiamin 1 mg l-1. The number of transformants produced 187 transformants mg-1 of PTRLI plasmid. Transformants are stable up to five generations by growing the transformants in Czapek-Dox medium agar containing piritiamin 1 mg l-1. The success of the transformation indicated by ptrA gene in transformants that can be amplified by PCR. The size of fragment DNA is 801 bp.
