Biodiversitas 21: 1060-1064. Leucaena leucocephala (Lam.) de Wit (petai Cina), Parkia speciosa Hassk. (petai), and Archidendron jiringa (Jack) I.C. Nielsen (jengkol) seeds extracts contain sulfhydryl compounds and have various therapeutic properties. The main objective of this research was to compare the effect of various ethanol concentration on the yield of extract and levels of sulfhydryl compounds from leguminous seeds. Dried seeds were macerated at room temperature (25°C) with a ratio of solids and solvents was 1:10. The solvent concentrations used were 30%, 50%, 70% and 96% ethanol. The yield of extract was expressed as the ratio of the weight of extract to the weight of dried seeds, the level of sulfhydryl compound was expressed as reduced L-glutathione (GSH) equivalent. The level of sulfhydryl compounds was determined by Ellman reagent and further analyzed using a spectrophotometer at 411 nm wavelength. Stink bean produced the highest yield of extract, while the highest level of sulfhydryl compounds was obtained from petai cina seed extract. The concentration of ethanol gave a significant difference to the yield of extract and the level of sulfhydryl compounds. The results showed that ethanol concentration affected the efficiency of the extraction of total sulfhydryl compounds and the yield of the extract on leguminous seeds.

Effects of supplementation with phosphorus, calcium and manganese during oil palm frond fermentation by Phanerochaete chrysosporium on ligninase enzyme activity. Biodiversitas 21: 1833-1838. The objective of this study was to evaluate the effects of supplementation with phosphorus (P) in combination with calcium (Ca) and manganese (Mn) during oil palm frond (OPF) fermentation by Phanerochaete chrysosporium on ligninase enzyme activity and lignin degradation. This study was carried out using a randomized complete design with 3 treatments (addition of P, Ca and Mn) and 5 replicates. The following treatments were performed: T1 (P 1000 + Ca 2000 + Mn 150 ppm), T2 (P 1500 + Ca 2000 + Mn 150 ppm), and T3  (P 2000 + Ca 2000 +Mn 150 ppm). The data were subjected to an analysis of variance (ANOVA), and differences between treatment means were tested using Duncan's multiple range test (DMRT). The parameters measured were as follows: lignin peroxidase (LiP) activity (U/mL), manganese peroxidase (MnP) activity (U/mL), crude protein (CP) content (%), crude fiber (CF) content (%) and the decrease in lignin (%). The results revealed a significant increase in LiP activity and CP content and a decrease in the lignin content (p<0.05) by the addition of P in the T3 treatment. However, the treatment nonsignificantly increased (p>0.05) MnP activity and significantly decreased (P<0.05) the CF content. In conclusion, supplementation of the OPF fermentation process with P 2000, Ca 2000, and Mn 150 ppm resulted in the highest ligninase enzyme activity and in decreased lignin content.

Development strategy for ecotourism management based on feasibility analysis of tourist attraction objects and perception of visitors and local communities. Biodiversitas 21: 689-698. Liwa Botanic Gardens (LBG), Lampung, Indonesia ecotourism development must be carried out as an effort to develop nature tourism with the most careful consideration in its management, without temporary benefits orientation, and the doers must have benchmarks in the process of sustainable development so that the next generation can utilize it in their time. This study aims to determine the management conditions and potential of Tourist Attraction Object (TAO) that have been operating — knowing the perceptions of tourists and local people. The method used is (i) scoring; (ii) assessing the perception of satisfaction of tourists and local people; (iii) doing intensive interviews with area managers and community leaders; (iv) identifying problems using the SWOT matrix; (v) constructing a development strategy. Data collection techniques with accidental sampling method with a total of 130 respondents based on tourist visits at the time of the study. The management of the LBG was previously managed by the Forestry Service until 2016, and in 2017 the management was handed over to the West Lampung Research and Development Agency (Balitbang). The results showed that overall, TAO gets a feasibility rating to be developed. The perception of tourists is quite satisfied with the attractions offered and the services of the manager. The community is satisfied with the ongoing management. The community wants are always to be involved in the development process. Researchers give advice on the examination of the management and marketing system in introducing the LBG to increase local and foreign tourist visitors.

Xylan-degrading ability of thermophilic Actinobacteria isolated from soil in a geothermal area. Biodiversitas 21: 144-154. This study was conducted to obtain the potential isolates of xylan-degrading thermophilic Actinobacteria. Seventeen isolates were obtained from the soil samples collected from Cisolok geysers in West Java, Indonesia. These isolates were screened for their xylan-degrading ability on minimal medium with the addition of 0.5% xylan as substrate, and were incubated at various temperatures for 7 days. A total of 15, 14, 4, and 3 isolates showed a xylan-degrading ability at 45°C, 50°C, 55°C, and 60°C, respectively. The three isolates (SL1-2-R-2, SL1-2-R-3, and SL1-2-R-4) that showed xylan-degrading ability at 60°C on 0.5% xylan were further tested by using minimal medium with the addition of 0.1% RBB-xylan as a substrate. The results showed that these isolates were also able to utilized RBB-xylan at 45 to 60°C. The xylanolytic activity of the crude enzymes from the three isolates on minimal medium containing 0.5% xylan or 0.1% RBB-xylan as substrates, indicated that these isolates were able to produce extracellular xylanase. This study revealed that the soil of Cisolok geysers served as an ideal source for finding the thermophilic Actinobacteria which produced xylanase at high temperatures.

Antibiotics resistant Escherichia coli isolated from aquatic ecosystems in Palembang, South Sumatra, Indonesia. Biodiversitas 21: 86-97. Antibiotics-resistant bacteria (ARB) are one of the emerging water contaminants currently gaining serious global concern due to their adverse risk to ecosystems, wildlife, and public health. This study determined antibiotics resistant Escherichia coli isolated from aquatic ecosystems in Palembang. Most probable number (MPN) was used for bacterial estimation and the Kirby-Bauer method was used for susceptibility test against antimicrobial agents. The results indicated that 82% of E. coli isolates from 28 sampling sites were resistant to ampicillin, 57% to tobramycin, and 71% to tetracycline. The isolates showed intermediate profile to kanamycin (50%), 57% to cotrimoxazole, 50% to cefixime, and 54% to gentamycin. These isolates still showed sensitivity towards ciprofloxacin (86%) and chloramphenicol (61%). Total coliform (TC) numbers ranged from 0 to >1600 MPN/100 mL. Sampling sites with high MPN values of ?1600 MPN/100 mL were Sekanak watersheds (SW1, SW8, SW11, SW12, SW13, and SW14) followed by SW2 and SW9 with a value of 1600 MPN/100 mL, while samples from cattle and fish farms (CW) varied from 0 to 170.000 MPN/100 mL. TC of samples collected from retention ponds (RP) ranged from 0 (RPJSC) to 1.600.000 MPN/100 mL (RPSH3). The Escherichia coli (EC) counts varied from 1.7 x 103 (RPSH2) to ?1.6 × 104 MPN/100 mL (SW11, SW12, SW13, and SW14). Several samples (SW3, RPPI, RPTS, RPSB, RPIBA, and RPOPI) have no E. coli. The results indicated some of the sampling locations that exceeds the quality standard of water have been regulated by the Governor of South Sumatra and the Indonesian Government.

Biodiversitas 21: 211-218. Proteolytic and chitinolytic bacteria are needed to degrade protein-chitin fibers in crab shell waste. These bacteria are important for enhancing the degradation rate of crab shell waste in the environment. This study aims to study the enzymatic activity and identify potent proteolytic and chitinolytic bacteria based on 16S rDNA sequences. The bacteria were isolated from crab shell waste using skim milk agar and colloidal chitin agar for proteolytic and chitinolytic bacteria, respectively. The bacterial isolates were screened based on their protease and chitinase activity using casein and chitin as substrates, respectively. The selected isolates were identified based on 16S rDNA sequences similarity. A total of 20 proteolytic and 22 chitinolytic bacteria were successfully isolated from the samples. The isolate of AP9, AP5, and BP14 had a high protease activity. However, only BP14 and CK20 showed a high chitinase activity. The isolate of AP9 was identified as Bacillus subtilis, AP5, and CK20 were identified as Bacillus licheniformis, and BP14 was identified as Bacillus cereus. AP5 dan CK20 were classified as non-pathogenic isolates.

Biodiversitas 21: 98-105. Farmers have traditional ecological knowledge related to the management of their fields, including knowledge of climate, soil types, soil fertility, various plants and animals, plant pests and irrigation. This knowledge is based on customs, beliefs and cosmos. Over time, government policies in agriculture such as the green revolution were implemented with the aim of increasing productivity of rice in wet-rice field farming. This initiated a change in the community's farming system from subsistence farming to commercial farming. Not to mention the existence of climate change that requires people to adapt. This change has an impact on ecological, economic and social aspects which are interesting to study. This study conducted in Sindang Hamlet, Rancakalong Village,  Sumedang District, West Java, which is known to have local rice cultivars and still practices rituals related to rice management. This study aims to analyze the changes in paddy management system in Sindang Hamlet due to green revolution and climate change, using qualitative data collection methods such as semi-structured interviews (deep interviews) with competent locals and participatory observations. Result of study showed that presently there are 3 groups of farmers in Sindang Hamlet, farmers who plant only local rice cultivars, farmers who plant only superior rice cultivars, and farmers who plant both local rice and superior rice cultivars in their land. Most farmers in Sindang Hamlet planted local rice but the diversity of local rice has decreased leaving only six cultivars, namely rayot, mesir, omas, torondol, mataram and randa kaya. There are still farmers in Sindang Hamlet who practice traditional farming, but many are starting to use modern tools and synthetic chemicals. The rice field management ritual called ngalaksa is still observed by the community, although there are some changes over time. The changes in rice field management have a negative impact on the local environment, economy and social aspects.

Ethnobotany of suweg (Amorphophallus paeoniifolius): Folk classification, habitat, and traditional conservation in Cisoka Village, Majalengka District, Cimanuk Watershed Region, Indonesia. Biodiversitas 21: 546-555. Amorphophallus paeoniifolius (Dennst.) Nicolson or suweg is commonly called in Sunadese of West Java as suweg that is a species of tuber growing in many regions of the world, including in the Cimanuk Watershed, West Java Province, Indonesia. A. paeoniifolius usually grows in rural areas of West Java in some agroecosystem types, including the homegarden, garden, and mixed garden system. This plant species, in contrast to other tuber species, such as taro, has not been intensively cultivated by rural community in West Java. Even though A. paeoniifolius has not been intensively cultivated, this plant species in the future have the potential to be used for various purposes for human being. Because A. paeoniifolius, one of plant tubers, has a high resistance to unpredictable environmental changes, such as climate change, pest disruption, and easy to maintain. This study aimed to elucidate the traditional ecological knowledge (TEK) of rural people of Cisoka Village, Cikijing Sub-district, Majalengka District of Cimanuk Watershed, West Java on folk classification, habitat, and traditional conservation of A. paeoniifolius. The method used in this study was qualitative. Field data collection was undertaken by observation and semi-structured interviews or deep interviews with competent informants. The results of study showed that the rural population of Cisoka Village, Majalengka District, has recognized two kinds of Amorphophallus. Firstly, Amorphophallus is a commonly consumed that is called suweg which has scientific name as Amorphophallus paeoniifolius (Dennst.) Nicolson. Secondly, Amorphophallus is recognized as not commonly consumed which is called 'iles-iles' (Amorphophallus variabilis Blume). Both species can be traditionally classified based on morphology (tubers, leaf blade and petioles, and flowers), habitat, utilization, cultivation, and suitability of growing places. Specifically, A. paeoniifolius, grows in various conditions, growing at non-cultivated lands, such as forests and cultivated land, such as homegarden, garden, mixed-garden, and rice fields, or pond edges, but predominantly grows in homegardens and gardens. Some of these lands are hieum or shaded areas and negrak or open areas. A. paeoniifolius starts to grow and develop at the beginning of the rainy season until approaching the dry season, which is characterized by yellowing and falling of petiole. Conservation of A. paeoniifolius has been based on tradition is carried out by the rural community through storage/ preparation of seeds, which does not destroy A. paeoniifolius that grows in several types of agroecosystems, and is managed semi-intensively in several types of agroecosystems such as homegardens and gardens.

Abstract. Muhakka, Suwignyo RA, Budianta D, Yakup. 2020. Nutritional values of swamp grasses as feed for Pampangan Buffaloes in South Sumatra, Indonesia. Biodiversitas 21: 953-961. This study was aimed at determining the types and quality of vegetation in the swamp area, which could be used as sources of roughage for Pampangan buffaloes in South Sumatra. In this descriptive study, Results showed that the highest crude protein and ether extract contents, 20.56 and 2.92%, respectively, were found in (Neptunia oleracea). The highest ash content of 25.19% was found in Cyperus digitatus. The lowest fiber content (11.01%) was found in Ludwigia hyssopifolia, which also had the lowest content of Neutral Detergent Fiber (NDF) (40.64%). Hemicellulose content was found to be highest (37.14%) in Leersia hexandra. The lowest ADF (23.66%) and lignin (14,84%) contents were found in Nymphaea lotus. N. oleracea was found to have the lowest content (27.08%) of cellulose. It was concluded that the swamp vegetation found in this study consisted of 17 species of grasses and herbs (89.47%) and two species of legumes (10.53%). C. digitatus and L. hyssopifolia were found to have the best nutritional values. Dominant and palatable vegetation with good nutritional values, which were potential to be used as sources of roughage for Pampangan buffaloes, were Hymenachne acutigluma, Oryza rufipogon, Hymenachne amplexicaulis, Polygonum barbatum, and Eleocharis dulcis.

Abstract. Fatimah IN, Iskandar J, Partasasmita R. 2020. Ethnoecology of paddy-fish integrative farming (minapadi) in Lampegan Village, West Java, Indonesia. Biodiversitas 21: 4419-4432. In the past, the traditional paddy-fish integrative farming (minapadi) was predominantly undertaken by rice farmers in West Java, particularly in the water abundant areas. Currently, however, the practice of the rice-fish system has been dramatically reduced due to the Green Revolution programs, including water pollution that may have affected fish farming in paddy fields. Although the rice-fish systems have rarely been practiced in West Java villages, some farmer groups of Lampegan Village, Ibun Sub-district, Bandung District are still practicing the rice-fish farming system. The purpose of this study is to collect and document the traditional ecological knowledge (TEK) of Lampegan Village about the rice-fish system farming managements, including data on various fish species that are widely cultivated in the community, cultural practices of the rice-fish farming management, and benefits of the rice-fish farming system. A combination of qualitative and quantitative methods was used, with an ethnoecological approach. Techniques of data collection, such as observation, semi-structured interviews, or deep interviews, and structured interviews were employed in this study. The results of the study show that the farmers of the rice-fish farming in Lampegan Village, Bandung District own rich and deep knowledge of fish species and practice of the minapadi system. The TEK of the rice-fish farming management system has been inherited from ancestors and is mixed with western scientific knowledge. The rice-fish farming systems have provided some ecological and socio-economic benefits for village people.
