Abstract

Multiplication of Aquilaria malaccensis with naa and yeast growth regulators on in vitro culture. This study aims to obtain the best concentration of NAA and yeast extract for multiplication of agarwood on in vitro culture. This research was conducted from January to October 2020 at the Laboratory of Plant Physiology and Biotechnology, Faculty of Agriculture, Universitas Sebelas Maret, Surakarta. The experimental design used was a factorial completely randomized design (CRD) with two factors, namely NAA (0 ppm; 0.1 ppm; 0.2 ppm; and 0.3 ppm) and yeast extract (0 mg/l, 700 mg/l, 800 mg/l, and 900 mg/l).(+) The results showed that the combination of 0 ppm NAA and 900 mg/l yeast increased the number of shoots of A. malaccensis explants with the highest average number of 3.67 shoots. A single NAA concentration of 0 ppm was able to increase the number of leaves of explants of A. malaccensis with the highest average leaf rate of 24.5 leaves. A single yeast concentration of 0 mg/l was able to increase the number of leaves of A. malaccensis explants with an average of 22 leaves.

Abstract

Nyamplung (Calophyllum inophyllum) is a potential type of non-timber forest product (NTFP) with high oil yield which is useful as an alternative renewable energy sources and herbal medicine-cosmetics. Result of population selection level in Java Island have been used to build Provenan Seed Stand (PSS) nyamplung in KHDTK Wonogiri (Central Java) as an improved seed source with high oil yield and fruit productivity. Seed become nyamplung part that is used as oil raw material which is strongly influenced by flowering and fruiting period. Information on flowering fruiting ability and period is needed to support the availability of raw materials in a sustainable supply for the nyamplung oil industry. Observation of flowering and fruiting period focused on 465 individual trees in nyamplung PSS at KHDTK Wonogiri through monthly monitoring by calculated flowering fruiting individualtree during 5 years (2014-2018). The results showed that the flowering period of nyamplung in TBP Wonogiri varied during 2014-2018. Flowering and fruiting occurs almost throughout the year. However, the peak of flowering and fruiting occurs mostly in March-August (end of the rainy season to the middle of dry season). In general, the trend of flowering and fertilization increased in 2014-2016, then decreased in 2017-2018, which is thought to be influenced by the higher canopy density. The environmental factor that most influences flowering is number of rainy days, while fertilization of TBP was strongly influenced by temperature. The management of TBP require future action such: second thinning, top pruning, application of fertilizers and hormones to stimulate flowering, and further detailed observations of flowering fruiting stage and its interaction with microclimate.

Abstract

Identification of insect species using molecular approach is one of the first steps in managing Ceratocystis. Proper species identification of the vectorsof Ceratocystiswill provide an effective way in limiting the distribution of this pathogenic fungi. The isolation of insect DNA is a crucial step in species identification using molecular characters. The aim of this research was to obtain the most effective method for isolating the COI gene and to confirm the insect DNA using amplification of the DNA through polymerase chain reaction (PCR). Twelve unidentified Coleoptera specimen collected from Acacia spp. plantation in Pelalawan Riau were randomly selected for this study. The insect DNA were isolated using the CTAB buffer with four different preincubation treatments, involved a non destructive method, i.e. soaking (R), destructive methods, i.e. crushing (H), soaking-crushing (RH) and freezing-crushing (FH). Two sets of primer, LCO1490/HCO2196 and LCO1490/HCO2198 were used to amplify the DNA of COI gene. The results shows, the COI gene was isolated from all pre-incubation treatments, except in the non-destructive treatment. The isolated DNA of COI gene was successfully amplified using both primer sets used in this study.

Abstract

Aquilaria malaccensis (agarwood) is a native species to Indonesia, it produces fragrance. Aim in this study was to assess genetic diversity and structure of A. malaccensis in order to manage seed stand to produce high quality and quantity seeds/seedlings. Leaf samples for DNA template were collected from  populations at western Bangka i.e. Air Gantang, Pelangas, and Simpang Gong. Amplifications of 14 RAPD markers were confirmed; 42 DNA samples of A. malaccensis were analysed using RAPD markers. Parameters of genetic diversity within population i.e. private alleles (PA) and expected heterozygosity (H) were calculated using GeneAlex software. Genetic structure among populations were determined as genetic distance (Da), Analysis Molecular of Variance (AMOVA), a neighbor-joining tree, were calculated using GeneAlex and PopTrew software. Eleven out of 14 RAPD markers were produced 104 stable and polymorphic alleles; PE were found at Air Gantang and Pelangas, 1 and 2 alleles, respectively. The H ranged between 0.133 (Simpang Gong) and 0.328 (Pelangas). The Da was moderate (0.099). AMOVA showed significant genetic diversity among populations (14%). A neighbor-joining tree showed the populations divided into two clusters with high boostrap value, and concordance with their geographical positions. Restricted gene flow might cause a high genetic relationship among populations. Low HEE in Simpang Gong caused this population was priored to being conserved; Air Gantang and Pelangas should be designed as different conservation units due to the population contained different private alleles.

Abstract

Thinning is one of silvicultural management actions in maintaining and increasing growth and quality of standing stands. This study aims to determine growth response of selected teak clone to several intensities of thinning and intercropping. The method was a factorial completely randomized block design (RCBD) with thinning intensity (4 levels): 0% (control = A1), 25% (A2), 50% (A3) and 75% (A4) as the main plot, while intercropping (2 levels), as the sub plot. The results showed that the Current Annual Increment (CAI) of diameter at breast height (CAI DBH), volume/tree (CAI vol/tree) and volume/hectare (CAI vol/ha) of stands gave a significant positive response to thinning intensity treatment. CAI DBH one year after thinning were 0.79 cm, 1.47 cm, and 2.46 cm; increased by 32%, 145% and 310% at the thinning intensity of 25%, 50% and 75% when they compared with the control (0.59 cm). CAI vol/ha of stand in the first year, namely 11.88 m/ha, 14.04m3/ha, and 16.78 m3/ha or with an increase of 9%, 29% and 54% respectively at an intensity of 25 %, 50% and 75% compared to the control.

Abstract

Development of Acacia hybrid is important to be done due to their superiority such as high productivity, good wood quality, more tolerance to pest/disease and adaptive to marginal sites, although its wood durability to wood-destroyer organism attacks is still unknown yet. This study aimed to observe the wood natural durability of Acacia hybrid to drywood termites (Cryptotermes cynocephalus Light.) attack. The wood samples used were taken from 6 years old trees of Acacia hybrid in Wonogiri, Central Java. The experimental design used is completely randomized design using two factors, axial and radial of wood section with the number of trees as replication. Parameters measured were extractive content, termite mortality and wood mass loss. The results showed that the axial section of wood significantly affected the wood extractive content at the level of 0.01, while termite mortality and the mass loss of wood were not significantly affected by the wood section. The largest average percentages of termite mortality and mass loss of wood found on the middle of wood section (96.67%) and near the bark (1.14%) respectively. Based on this result, the Acacia hybrid wood is categorized as very durable wood and potential to be developed as raw material for solid wood-based industries.

Abstract

Conservation management strategies for native Sumatran species such as andalas (Morus macroura) are currently being developed through genetic diversity studies. This genetic diversity information will be used to enhance ongoing conservation and regeneration programs and to control logging of this high value plant species. The aims of this initial study were to develop a method for extracting DNA from andalas cambium, to select polymorphic RAPD primers and to study genetic diversity using selected RAPD primers. DNA was successfully extracted using the CTAB method from all 32 cambium samples collected from Jambi and West Sumatra. Eighteen RAPD primers were screened and six polymorphic primers selected (OPO-6, OPY-6, OPY15, OPW-1, OPW-3 and OPW-4). RAPD analysis using the six selected primers resulted in 52 polymorphic loci and the value of genetic diversity within the population (Hs) of 0.308 and the value of genetic diversity among populations (DST) of 0.091. The highest genetic diversity was observed in Batipuah, Tanah Datar (0.351) and the lowest was observed in Halaban, Lima Puluh Kota (0.205). A test with AMOVA shows that the genetic diversity between individuals is greater than the diversity between populations. The dendogram based on the genetic distance between populations shows that the andalas population in West Sumatra is separated into two groups and both are separated from the Jambi population. The results of this study can be used as a basis for further research on several other andalas populations as well as the basic information for conservation strategies of andalas.

Abstract

The progeny trial of Shorea leprosula was established in Central Kalimantan under the IUPHHK-HA management of PT. Erna Djuliawati by planting 30 families to examine the genetic superiority of  trees as plywood raw material. The trial was laid-out using  8 blocks, 4 tree-plots and a spacing of 6 x 3 m. The aim of the study was to practice the plus trees selection in the progeny trial. The study was conducted using a survey method on phenotypically superior trees based on a diameter at breast height (dbh) baseline of ≥ 30 cm. The data collected and analyzed for the plus trees and comparison trees were total tree height (m), dbh (cm), and dbh increment (cm/yr). The results of the initial selection showed that 110 trees from 29 families were selected as candidate plus trees in the progeny trial. Futhermore, the honest significant difference test (BNJ) and scoring showed that there were 15 trees from 9 families that can be designated as plus trees.

Abstract

Bitti (Vitex cofassus Reinw.) is a native plant species in Sulawesi which belongs to Verbenaceae family. This species has great potential to be developed as a house construction material, boat base material, and household utensils. Development of bitti silviculture techniques is very necessary in supporting the efforts to develop these plants. One of problems in this cultivation is disease attack. This study aimed to determine symptoms of disease attack, level of severity, determine kind of pathogen that cause disease, and analyze some factors that cause pathogen attack on bitti seedlings. The results showed that leaf spot disease was found on bitti seedlings with level of severity 33.22%. The identification results showed that the pathogen causing leaf spot disease is the fungus, namely Curvularia sp. Microclimate conditions such as temperature and humidity assumed to affect the emergence of pathogen on bitti seedlings. The attack of Curvularia sp tends to affect the growth of bitti seedlings.

Abstract

Dryobalanops aromatica is a highly economic value resin-producing tree and has been known as an important international trade commodity that is widely used in the perfume, cosmetic, medicine, and wood industries. In natural habitats, this species has been found to grow well on peatland and mineral soils. The information regarding adaptive genes in Indonesian native trees to abiotic stress is still very limited. The research was conducted to analyse the differentially expressed genes (DGEs) that can elucidate the role of several upregulated and down-regulated genes under peat media and mineral soil treatments. DGE analysis was carried out using R software, Bioconductor package 'edgeR'. Using the Benjamini and Hochberg approach to control FDR (FDR 0.05), with a Log2FC 2 and p-value of 0.05, showed 320 contigs were up-regulated and 439 contigs were down-regulated, while 58129 contigs were not significantly expressed. Furthermore, this study also presents an overview of the genes involved in different pathways, such as photosynthesis, carbon and energy metabolism, hormone-related genes, nitrogen metabolism, reactive oxygen species, and transcription factor. This information will be useful in understanding D. aromatica molecular responses to stress condition that may be of use for selecting genotypes in the breeding programs or peatlands restoration
