Abstract

Thalassemia is an autosomal recessive genetic mutation disorder with symptoms similliar to anemia that causes deficiency synthesis of the globin chains (hemoglobin component inside erythrocytes). Thalassemia is classified based on secondary protein structure abnormalities in α globin protein or β globin protein. Based on data from Indonesian Thalassemia Foundation (YTI) in Indonesia, people with thalassemia, especialy β thalassemia is constantly increasing 8 to 10% per year, so it is very impotant to have a strategy that reduce the increasing number in the population. Genetic examination on the individual is very effective to detect quickly the person with thalassemia trait carrier, to reduce the number of thalassemia carrier population. Identification of 1st exon β globin gene mutations with sequencing method is one of the way to know the specific mutation in thalassemia carriers. Identification results can be used as a reference for the rapid detection of thalassemia trait carrier. This study aims to determine the type of mutation and location of nucleotide mutations in 1st exon β globin gene on carrier of thalassemia and the changes of amino acid translated by the mutant gene. The study was conducted by isolating the genome from the β thalassemia carrier blood, amplifying and sequencing the 1st exon β globin gene. The location of point mutation analysis and the changes of amino acid, was analyzed using computational method by comparative alignment using normal β globin gene as a comparison. The results of the research showed there is a point mutation in the 59th nucleotide caused by transition (T to C) and 147th nucleotide caused by transversion (G to C). The mutation type found on this study was a silent mutation because there is no change in the translated amino acid.Keywords: β globin, exon 1, mutation, thalassemia

Abstract

Central Sulawesi is a province produce eels (Anguilla marmorata) in Indonesia. Eels is the one commodity that has a high economic value. In the last years, eels trading traffic through the airport had increased. Fish health status to be one of the important issues in trade of eels. Genus Aeromonas is disease-causing pathogen on eels and as the etiologic agent responsible for a variety of infectious complications pathogenic bacteria. The purpose of this study is identify the various species of the genus Aeromonas on the eels which are traffic through Mutiara Sis Al jufri Airport Palu. This observation have been from April to December 2014. Specimens Anguilla sp. was obtained from supplier in the Palu city. Laboratory examination performed use conventional methods (Biochemistry) and performed in Microbiology Laboratory of Fish Quarantine Palu. The result showed from 396 eels have been identified, 128 of them are of the genus Aeromonas (32.32%). Aeromonas were identified some species among other: Aeromonas sobria, Aeromonas hydrophilla, and Aeromonas caviae. A.sobria is higher percentage in April and August 2014 (57,14%), A. hydrophilla in December 2014 (92,85%), and A. caviae August 2014 (21,43%). The existence of pathogenic bacteria in eels this should be a concern to prevent widespread dissemination through trade traffic.

Abstract

In Indonesia, the increasing prevalence of diabetes mellitus is accompanied by the increasing financial burden to the patients. It is necessary to find alternative herbal treatment for diabetes, which is effective, cheap, and easy to find: the mahkota dewa. This research aims to find out the histological regeneration of Langerhans islet cells after given by flesh extract of mahkota dewa.This experimental research used complete randomized design (CRD). There were 5 treatments given to the twenty five mice; the negative control group (the normal one), the positive control (male white mice (Rattus norvegicus which were made diabetic by administering injection of 70 mg/kg of body mass of diabetogenic alloxan), the group of diabetic male white mice given 3, 6 and 9 grams/day/200 bodyweights), then were measured glucose in blood at 0, 1st and 4th week and counted Langerhans islet cells every 5 overviews.Statistically results showed that flesh extracts of mahkota dewa can decreased glucose in diabetic mice (r=0,75) with contribute 56,89% in decreasing, regenerated Langerhans islet cells of pankreas {Fcount(17,558)>Ftab(2,870)} with the highest increasing is 3,84 amount and started with 3grams/day/200 bodyweights doses, and the best effectiveness in regenerating was 11,81% consentration or 0,1181grams/day (6.56 grams/day in human).

Abstract

This research is about the isolation and characterization Vibrio sp. from grouper fish (Plectropomus sp.). This research aimed to observe the amount of vibrio bacteria species based on colony morphology and characterization morphological colony of vibrio isolated from grouper fish(Plectropomus sp.). Bacteria isolation was conducted by isolate some external organs (lesion on its body) and internal organ (jejenum) from grouper fish (Plectropomus sp.). Bacteria was grown on selective medium for vibrio, TCBS (Thiosulfate Citrate Bile Sucrose) agar. The incubation time was 24 hours at 370C. The result showed there are 3 isolate vibrio bacteria that was coded by V1, V2, and V3. V1 morphological characteristis was rounded, edge is entire, convex elevation, green colony colour with blue in the center of colony and smooth tekstur. V2 morphological characteristic was rounded, edge is entire, tortous elevation, colony colour yellow, and smooth tekstur. Morphologicalcharacteristic of V3 was Irregular, egde is Undulate, elevation hill, colony colour yellow and own the smooth tekstur.Keywords: characterization, grouper fish (Plectropomus sp.), isolation, Vibrio

Abstract

Ornithoptera croesus lydius is one of endemic butterlfy in Halmahera island North Maluku. It is macrolepidotera tipe, with interesting colour body at male and female butterfly. At Ornithoptera croesus lydius male and female butterfly have variation morphology size and colour body. The genetic variability on the  O.c lydius  butterflies is level intraspecies variability. The objective of the research is to know the genetic variability on the O.c lydius based on PCR-RAPD marker. The most value of genetic distance  is 0,783 on O.c lydius male and female from hotspot at 780 masl. The percentage of polimorphic  is 71,95%. Base on similarity value and polymorphic percentage show genetic variability on intraspecies level of the Ornithoptera croesus lydius.

Abstract

Among the industries in Lampung, PT. Perkebunan Nusantara VII (Persero) Bekri Business Unit is one of the Palm Oil Processing Plant (POPP) industry who spend a lot of waste water in the form of residual organic liquid materials. The rest of the liquid material can cause problems such as pollution so it needed special treatment before the waste water is discharged into the environment. This study was conducted to determine the quality of liquid waste in an anaerobic pool IV on Wastewater Treatment Plant (WWTP) PT. Plantation Nusantara VII (Persero) Business Unit Bekri so it can be determined whether the waste water treatment process can improve the quality of waste water for the better. Waste water quality measurement is done using the parameters of physics,chemistry, and heavy metals, namely pH, COD, BOD5, TSS, Cd, Cu, Pb, and Zn. The results showed that the quality of the wastewater in an anaerobic pool IV was good and meets the quality standards of palm oil mill effluent by the State Minister of Environment Resolution No. 03 / MENKLH /11/1991 and the Indonesian Government Regulation No. 20 of 1990 on Water Pollution Control. It also indicates that the effluent treatment system at the Wastewater Treatment Plant (WWTP) PT. Perkebunan Nusantara VII (Persero) Bekri Business Unit is very effective.Keywords: anaerobic pool, liquid waste, PTPN VII

Abstract

Candida sp. are dimorphic fungi that grow as the yeast cells. These fungi are commonly found on stagnant water, for example is toilet water tub. Several factors that affect the growth of Candida sp. on the toilet water tub are the source of water, toilet users, and the draining. The purpose of this research is to find out the effect of draining frequency on the number of Candida sp. in women’s toilet water tub at the gas station in Surakarta. This study is an observational analytical research with cross sectional approach. The examination was showed that the average number of Candida sp. in the regularly drained toilet water tubs was 66,67 colonies/10ml sample and the average number ofCandida sp. in the irregularly drained toilet water tubs was 240,3 colonies/10ml sample. The datawere tested by using independent t-tes and the significant number was 0,003 (P < 0,05). It could beconcluded that there was a distinctive effect of draining frequency on the number of Candida sp. inwomen’s toilet water tub at the gas station in Surakarta.Keywords: Candida sp., draining, water tub

Abstract

The research aims to study the effect of rice storage period against the growth of Sitophilus oryzae population and weight loss characteristics of rice.  The rice storage period consists of five levels: rice storage for 40, 60, 80, 100, and 120 days.  The results showed that rice storage period affects the growth of S. oryzae population, weight loss characteristics of rice, and the percentage of rice powder. The 120-day storage period may increase S. oryzae populations, weight loss characteristics of rice, and the percentage of rice powder. The relationship between the storage period and the growth of S. oryzae population, weight loss characteristics of rice, and the percentage of rice powder increased linearly. There was a positive and very significant correlation between the rice storage period and the imago of S. oryzae population (r = 0.996**; P <0.01), weight loss percentage (r = 0.990**; P <0.01), perforated grain percentage (R = 0.994**; P <0.01), and the percentage of rice powder (r = 0.971**; P <0.01).

Abstract

Gecko is one of the reptiles are used as traditional Chinese medicine which has been reported as antitumor capabilities. The purpose of this study was to determine the cytotoxic effect of rat serum that treated with gecko powder and its ability to inhibit proliferation and stimulate apoptosis of WiDr cells. Gecko powder  the treated in vivo in the rat (Rattus novergicus) SD female. Twelve mice treated with powder gecko at a dose of 9 g/kg body weight per day. Then the rat blood serum on the in vitro treated in WiDr cells. Cytotoxicity test using the MTT method. Observation of the proliferation kinetics using doubling time test. Then apoptosis test performed by the doublestaining method. Cytotoxicity assay results showed that the powder is toxic to the gecko cell WiDr. In the cytotoxicity test showed IC50rat serum with gecko powder body parts obtained at a concentration of 7,914%. The observation of cell proliferation kinetics demonstrated  rat serum with gecko powder body parts most effective in inhibiting the proliferation of WiDr cells Data apoptotic WiDr cells  showed the rat serum with gecko powder effective agency in promoting apoptosis. Rat serum with treat gecko powders bodies part are toxic and can inhibit proliferation and stimulate apoptosis of WiDr cells.

Abstract

This study aims to determine the concentration of NaCl which can be used as maize resistance limit to salinity and to obtain varieties that are tolerant to salinity. This research was conducted in the form of an experiment with randomized design factorial using two factors in the group. As the first factor is the variety of Arjuna, Sukmaraga, Bhishma, Gumarang, Heroine Yellow, Pacakka, Anoman-1, and LAMURU. The second factor is the concentration of NaCl with 0 g L-1, 3 g L-1, 4 g L-1, and 5 g L-1. The results showed that the level of resistance of some maize varieties to salinity using NaCl produce varieties that have different resistance levels to high levels of salinity, there are varieties that can withstand the high NaCl concentration and some are only able to survive on a low NaCl concentration. The test results obtained sequentially showed varieties that are resistant to salinity is Pacakka. The concentration of 5 g L-1 NaCl can not be used as a concentration limit for the grouping of maize resistance to salinity.Keywords: Maize (Zea mays L.), NaCl, Salinity
