Abstract

Contamination of processed beef foods such as abon can be caused by various types of microbes, one of which is Staphylococcus aureus. Staphylococcus aureus can cause various infections, both on the skin, gastrointestinal tract, or endocarditis. The objective of this research was to determine the presence of Staphylococcus aureus in beef abon sold in Pahing Market, Kediri. Abon used is non-branded beef abon which is as many as 10 samples obtained by total sampling technique. Samples were tested by observation of colony morphology through Gram staining, mannitol fermentation test, catalase and coagulase test, and acetoin test. The samples were inoculated on Broth NaCl (ink. 24 hour-37°C), then inoculated on MSA (ink. 24 hour-37°C), and VP (ink. 2x24 hours-37°C). Catalase and coagulase tests were carried out by taking colonies on MSA media. The results showed that there were 9 abon samples contaminated with Staphylococcus aureus as indicated by Gram positive staining results, positive (perfect) mannitol fermentation, and positive acetoin, catalase, and coagulase test. The causes of contamination are contaminated abon ingredients, the manufacturing process using less sterile tools, poor handling and processing, processing food with dirty hands, food stored without cover, sick food processors, and dirty markets

Abstract

Azotobacter spp. merupakan salah satu genus bakteri nonsimbiotik yang berperan penting dalam bidang agrikultur terkait kemampuannya memfiksasi nitrogen. Penelitian ini bertujuan untuk mengetahui potensi isolat Azotobacter dari lahan Eco Urban Farming ITS dalam mendegradasi lipid dan protein. Kemampuan isolat dalam mendegradasi lipid dan protein ditentukan dengan pengukuran Indeks Biodegradasi (IB). Isolat yang digunakan adalah A1a, A1b, A2, A3, A5, A6, A7, A8, A9, dan A10. Semua isolat mampu mendegradasi lipid kecuali A5, dengan IB tertinggi pada isolat A1b (1,00). Semua isolat dapat mendegradasi protein kecuali A7, dengan IB tertinggi terdapat pada isolat A8 (3,00).

Abstract

Studi perilaku populasi monyet ekor panjang dilakukan di Taman Wisata Alam (TWA) Grojogan Sewu Tawangmangu Karanganyar. Penelitian ini bertujuan untuk mempelajari aktifitas harian populasi monyet ekor panjang dengan menggunakan metode scan sampling. Penelitian ini dilaksanakan pada bulan November 2014. Pengamatan dilakukan selama 150 menit dengan interval waktu 5 menit. Hasil penelitian menunjukkan bahwa persentase perilaku harian populasi monyet ekor panjang yang diperoleh yaitu: bergerak (35%), grooming (25%), bermain (15%), inaktif (10%), makan (6.8%), agonistik (3.6%), tidur (2.3%), kawin (0.9%), dan bersuara (0.8%).

Abstract

Dengue Hemorrhagic Fever (DHF) is one of the environmental health problems that several case increasing the number of patients and the wider area of distribution. The spread of dengue is influenced by several factors such as the vector disease, the behavior of people and the environment. In some sub-tropical countries is like  in the winter season , a number of   Aedes albopictus is found the eggs can still hatch at temperatures 0,5?C. Eggs that will be tested for each treatment amounted to 100 eggs and had been through the process selected.  The research carried in Parasitology and Entomology Labolatories, South Sumatra. The research was conducted from March to December 2014. In a multivariate test showed that the interaction of temperature and storage time affect the hatchability of eggs of Aedes aegypti strain Japan. Humidity and temperature can influence one of the insects are mosquitoes. At a certain temperature and humidity mosquitoes can not do the lifecycle and inhibite the morfology. The conclusion of this research is the cold storage and extreme temperature influence to eggs hacthingof Strain Japan the Aedes aegypti. Suggestions in this research is the public should continue to implement programs 3M plus, due to the Aedes eggs can survive in cold weather.

Abstract

Chromium metal is one of the heavy metal wastes from various industries and is persistent for the agricultural environment, especially in rice fields. Chromium metal can change biodiversity and ecosystem function in paddy soil. Chromium metal phytoremediation that pollutes paddy soils with hemp (Boehmeria nivea) is important because paddy soils play a role as a living medium for food crops, especially rice (Oryza sativa). One indicator of the success of phytoremediation is the reduction of chromium metal content in the soil, so it requires a policy system to maintain a healthy environmentally friendly agriculture. This study aimed to determine the ability of Boehmeria nivea to reduce levels of chromium metal in the soil and provide policy solutions to keep environmentally healthy agriculture. This study used a complete randomized block design, random sampling of chromium metal data. The results showed the initial concentration of chromium metal in the soil was 2.36 ppm, after treatment with the interaction between Agrobacterium sp. I3 with Boehmeria nivea (P0B1T1) and interaction of organic matter (compost) with Boehmeria nivea (P0B2T1) obtained Cr 1.37 ppm metal content with a decrease of 42.01%. The resulting policy solution is the Environment Health Agriculture System (EHAS). The conclusion of this study was phytoremediation of chromium metal using Boehmeria nivea combined with the Environment Health Agriculture System can create a healthy environmentally friendly agricultural system.

Abstract

Tujuan penelitian ini adalah mengetahui aktivitas antibakteri 10 isolat Actinomycetes dari sampel pasir Gunung Merapi menggunakan metode sumuran dan fermentasi terhadap bakteri E.coli multiresisten antibiotik dengan lama waktu fermentasi yang berbeda. Penelitian ini merupakan penelitian eksperimen menggunakan Rancangan Acak Lengkap (RAL) dengan 2 faktor perlakuan yaitu lama waktu fermentasi (L) dan jenis isolat Actinomycetes (S). Masing-masing perlakuan dengan 2 kali ulangan. Isolat Actinomycetes tersebut difermentasi dalam kultur cair yang mengandung 2% manitol, 2% pepton, dan 1% glukosa selama 6, 7, dan 8 hari, pada suhu 280C menggunakan shaker 50 rpm, selanjutnya diuji menggunakan metode sumuran terhadap E.coli multiresisten. Hasilnya ke 10 isolat mempunyai aktivitas antibakteri terhadap E.coli dengan diameter zona hambat bervariasi. Aktivitas antibakteri terkuat pada hari ke-6 pada isolat D (S4) dengan diameter zona hambat iradikal 17,25 mm, pada fermentasi hari ke-7 pada isolat G (S8) dengan diameter zona hambat radikal 7 mm, dan pada hari ke-8 pada  isolat A (S1) dengan diameter zona hambat radikal 10 mm.

Abstract

Minuman instan yang dikemas dalam kantong plastik dapat dijual dan ditemukan pada toko-toko, warung kecil, dan pedagang kaki lima dengan bebas. Masyarakat tidak menyadari bahwa mengkonsumsi minuman instan berlebihan dapat mengganggu kesehatan, karena dalam komposisi minuman instan tersebut terdapat bahan pemanis buatan. Penelitian ini bertujuan untuk mengetahui pengaruh konsumsi minuman instan dengan frekuensi berbeda terhadap kadar ureum darah mencit (Mus musculus). Penelitian dilakukan dengan metode eksperimen, yang mengguna-kan sampel 20 ekor mencit jantan, berumur 2-3 bulan dan berat 20-40 g yang terbagi ke dalam 4 kelompok, yaitu Placebo, kelompok 1, kelompok 2, dan kelompok 3. Bahwa pemberian minuman instan dengan volume 0,5 ml/20g BB selama 10 hari ada pengaruh terhadap kadar ureum darah mencit (Mus musculus). Kemudian dilakukan uji lanjut Duncan antara kelompok perlakuan placebo, perlakuan 1, dan perlakuan 2 tidak menunjukkan hasil yang bermakna, sedangkan ke-lompok perlakuan 3 yang menunjukkan perlakuan yang paling berpengaruh terhadap kadar ureum darah mencit (Mus musculus). Sehingga dari hasil penelitian dan analisa data dapat disimpulkan bahwa pemberian minuman instan merk Marimas rasa jeruk dengan volume 0,5ml/20 g BB berpengaruh terhadap kadar ureum darah mencit (Mus musculus), dan perlakuan yang paling berpengaruh yaitu kelompok perlakuan 3 dengan pemberian minuman sachet merk Marimas rasa jeruk tiga kali sehari diwaktu pagi, siang dan sore selama 10 hari dibandingkan dengan kelompok placebo, perlakuan 1 dan perlakuan 2.

Abstract

The use of medicinal plants is increasing due to the lack of side effects caused and the number of bioactive compounds that cannot be represented by synthetic chemical synthesis compounds. However, the management and use of natural medicines for the main handling of diseases are often hampered by the quality of the ingredients which are low and unstable. The standardized quality control system of OAI (Indonesian Natural Medicine) is the main key to improve clinical assurance and safety of the use of herbal medicines in Indonesia. One of the medicinal plants known to the public is Curcuma longa L. (turmeric). The main active components contained in turmeric are curcumin, demetoksikurkumin, bis-demetoksikurkumin, and ar-turmeron. Information about the quality of turmeric is needed in its use as a raw material for drugs so we need an analytical technique that is able to identify the diversity of metabolite profiles of active compounds. In this research, an optimization method is used to improve efficiency in the extraction of turmeric rhizome metabolites so that the best solvent concentration is known for the analysis of fingerprinting secondary metabolites with 1H-NMR 500 MHz spectroscopy in turmeric rhizomes. The results were analyzed with MNOVA software and chemical shift obtained compared with the reference. From the results obtained a concentration of methanol-d4 (CD3OD) 100% able to extract curcumin better than other solvents. The solvent is able to extract saccharide (sugar) compounds in the form of sucrose, amino acids and fatty acids in the form of methionine, glutamine, acetate, and glycero phospho choline.

Abstract

Rambutan seed flour could be utilized as a food especially as source of carbohydrate, fat, and protein. Red cabbage could be used as natural food color because of their anthocyanin content. The aims of the research were to investigate the effect of red cabbage to the antioxidant activity and consumer preference of rambutan seed flour biscuit. This research was use completed randomized design with 9 combinations and 2 replications. The results showed that the highest antioxidant activity was biscuit made by 50 g of rambutan seed flour addition and 20% of red cabbage extract, e.i 19.48 % radical scavenging activity (RSA) of DPPH. But biscuit which was most prefere by panel test was only 25 g of rambutan seed flour without any addition of red cabbage extract. Rambutan seed flour and red cabbage addition to the biscuit did affect the antioxidant activity and the consumer preference of the biscuit.

Abstract

Lipase memiliki manfaat penting di bidang industri. Tujuan penelitian ini adalah untuk mendapatkan kapang lipolitik yang mampu tumbuh dan menghasilkan aktivitas lipase tinggi pada limbah ampas kelapa menggunakan metode solid state fermentation. Isolat kapang uji dipurifikasi kemudian dilakukan skrining dan seleksi kapang lipolitik dan dilanjutkan dengan produksi lipase menggunakan substrat ampas kelapa yang sebelumnya diukur kandungan biokimia. Hasil menunjukkan bahwa 8 isolat kapang lipolitik mampu tumbuh baik pada substrat ampas kelapa yang ditunjukkan dengan adanya sporulasi dan perubahan pH medium selama reaksi. Diantara kapang lipolitik tersebut, isolat kapang KLC-333 diketahui menghasilkan aktivitas hidrolisis lipase terbesar yaitu 13,33 U/ml dan volume produksi 46 ml. Biosintesis dan peningkatan produksi lipase dipengaruhi oleh kandungan nutrien di dalam substrat ampas kelapa.
