Abstract

 Diabetes mellitus is a metabolic disorder characterized by hyperglycemia. “Jamu gendong kunyit asam” (Curcuma domestica Val.; Tamarindus indica L.) is a traditional medicine that has antioxidant activity which can contribute to diabetic because of the phenolic content. This aim of this study is to determine the effect of “jamu gendong kunyit asam” on lowering blood glucose levels and repairing damage of the islet cells. 25 male wistar rats (Rattus norvegicus) were divided into five groups. Group I as a normal group without any treatment, group II, III, IV and V were induced with streptozotocin (7mg/ 200gBB) then treated with  CMC 1%, glibenclamide (0.27mg / 200gBB), “jamu gendong kunyit asam” 1.90mL/ 200gBB and 3.80mL/ 200gBB. The treatment was done for 28 days. Glucometer was used to measure the gloucose blood level on day 4, 8, 12, 16, 20, 24 and 28. The damages on the islet cells were examined under the microscope on the histology sample of pancreas prepared with Hematoxyllin eosin stain. Datas were analyzed statistically with One Way Anova test, T-Test, and the Kruskal-Wallis test using SPSS 17.0 for Windows. Results showed there is a significant difference (p> 0.05) in blood glucose levels and damage to the islets of Langerhans of the pancreas between the glibenclamide group and treatment of “jamu gendong kunyit asam”. From the research it can be concluded that the “jamu gendong kunyit asam” have antidiabetic activity which is  showed by a decrease in blood glucose levels and an improvement of pancreatic islets of Langerhans in streptozotocin-induced diabetic rats with an effective dose of 1.90mL/ 200gBB.

Abstract

Muna tribe in Kota Wuna Settlement has utilized various types of medicinal herbs. However, until nowthere has been no documented information about the scientific names and their utilization of the medicinal plant species in the area. The objective of this study was to determine the number and types of medicinal herbs used by the society in the study area, efficacy and organs of the plants are utilized. The research was conducted from January until November 2013, in Wuna City Settlement Tongkuno districts, Muna regency, Southeast Sulawesi Province. This research method used was survey and interview. Identification of the types of medicinal herbs was done by matching the characteristics that exist with images referring to the plant identification book. The results showed that there were 34 collections of plants that have medicinal properties traditionally utilized by the Muna tribes at Wuna City Settlement. Based on 34 types of plants that used, Based on 34 types of plants used, 31 species have been identified and 3 plants species have not been identified.

Abstract

Daun legundi (Vitex trifolia) has been used by Indonesian people as antiasthma. This research aims to produced the good quality syrup of extract of daun legundi and also to evaluate the relative level change of viteksikarpin in syrup after formulation. Syrup of extract of daun legundi was made for 4 formulas of variation of propylene glycol (PG) level. Formula 1 (PG 11%), formula 2(PG 12%), formula 3 (PG 13%) and control (without PG). Each formula was tested for physical characteristic (organoleptic, viscosity, time of flow, flavor test), relative level of viteksikarpin against extract. The result showed that increase of propylene glycol level was increased the viscosity syrup and time of flow while the percentage of relative level of viteksikarpin against extract was decreased. In this research, formula 1 (PG 11%) was the selected formula because it has a good flavor and appearance, less viscosity, least precipitate, highest relative level of viteksikarpin and acceptable in the market.

Abstract

Keloid is a skin lesion caused by an abnormal proliferation of fibroblasts and the deposition of collagen in the wound healing process. Therapy for keloids is relatively limited and mostly has side effects. Recent studies show that 5α-oleandrin from N. indicum Mill. (N. indicum) has anti-keloid effect by inhibiting (proliferation, migration activity and TGF-β1 expression) keloid fibroblast. Further studies with N. indicum extract standardized 5α-oleandrin should be conducted to develop it as an anti-keloid agent by performing at proliferative effects, collagen accumulation, and keloid fibroblast migration. The antifibrotic activity of N. indicum extract on the proliferation of keloid fibroblasts was measured by MTT assay, whereas collagen deposits were observed by Sirius Red method. Observation of keloid fibroblast migration by scratch assay is according to Liang et al. (2009). Ethanol extract of N. indicum standardized 5α-oleandrin can inhibit the proliferation of keloid fibroblas with IC50 0.458 μg/mL, also inhibited collagen deposits with IC50 0.055 μg/mL at incubation for 72 hours. In incubation 48 hours after treatment with ethanol extract of 1.0 μg/mL occurred inhibition of migration significantly compared with control.

Abstract

Immunomodulator has been used as one of the attention to develop herbal medicine. Jahe is one of herbal medicine which is usually used to heal various diseases. Jahe Emprit  is one of local name of Jahe in Indonesia. The aim of this research was to investigate the effect of pungent principle containing extract of Jahe emprit rhizome on the macrophage activity compared by levamisol and Echinacea extract. Extraction was done by 70%v/v ethanol. The concentrations of extract were 5 mg/kg B.W; 25 mg/kg B.W; and 100 mg/kg B.W. CMC Na 1,5%w/v was used as solvent control. Levamisol 2,5 mg/kg B.W and Echinacea extract 10 mg/kg B.W were used as positive control. The macrophage ability  were measured according to Leijh et al., (1986) that was basically on the amount of latex which was captured  by macrophage. The extracts were orally administrated to the Swiss male mice at the dose of 0,2 ml/20 g B.W. The Macrophage  activity was analyzed by one way ANOVA at 95% of confidence level. The extract which had specification of 35% relative concentration of pungent principles; 3,554% ± 0,145 % W/W total phenolic substances; 13,70 mg/ml  antioxydative value  (IC50) at the concentrations of  5 mg/kg B.W and 25 mg/kg B.W could increase phagocytosis  ability. The ability of the extract at the concentration 25 mg/kg.B.W had no different with either the Levamizol 2,5 mg/kg. B.W., and Echinacea extract 2,5 mg/kg. B.W.

Abstract

Peptic ulcer were been caused by an imbalance between the aggressive factors and a number of defense mechanisms. Pepino (Solanum muricatum Aiton) fruit predicted had antiulcer activity. This study is aimed to test the antiulcer activity of pepino fruit juice in Swiss-Webster mice model which induced by 50 µL concentrated acetic acid. The steps in this study consist of making of fruit juice, phytochemical screening, testing of antiulcer activity at 300, 500, dan 800 µL/20 g BW in doses, and data analyzed using ANOVA. The concentration of pepino fruit juice was 2.5 g/mL. Phytochemical screening showed that pepino fruit juice contain alkaloids, flavonoids, and tannins with protection ratio were 6.02, 23.15, and 63.29% at 300, 500, dan 800 µL/20 g BW, respectively.

Abstract

Our susceptibility to various infectious diseases can be avoided by increasing the specific immune responses either by the proliferation of lymphocytes. Immunomodulator effect of Andrographis paniculata (Burm.f.) Nees) and Curcuma xanthorrhiza Roxb have been well evaluated. Empirically, the East Borneo’s tribe combined both herbs to treat jaundice disease. However, scientific evidence is still needed in their use as a drug candidate. This study explores further effect of the immunomodulator from 5 ethanol extract groups. Quantitative analysis was also completed using a densitometer to obtain andrographolide and curcumin levels of ethanol extract. Lymphocyte proliferation was tested by using MTT colorimetric method, the results was read by an ELISA reader and statistically analyzed with One-Way ANOVA test with 95 % of confidence level. The maceration yield of sambiloto ethanol extract (EES) and temulawak ethanol extract (EET) were respectively 5.78 % w / w and 3.92 % w / w. The results of quantitative analysis in 28.6 mg EES contained 11.43 % andrographolide and from the 251.8 mg EET contained 28.79 % curcumin. From the MTT test, all treatment groups were significantly different from the control group. Based on the OD values, the best combination to increase lymphocyte cell proliferation ETS was a group contained 56.25 mg of temulawak and 18.75 mg of sambiloto in 1 ml of solvent. Nonetheless, the value was not higher than the ability of ET in increasing cell proliferation.

Abstract

Oxidative stress induced by the radicals have been known to affect the occurrence of various degenerative disease. In order to search natural antioxidant compounds, it had been studied the isolation and identification of antioxidant compounds in fern stems (Alsophila glauca J.Sm) using DPPH method (1,1-diphenyl-2-picrylhydrazyl). Fern stem extraction was done by maceration with wasbenzen and metanol. Extracts was obtained by evaporating the wasbenzen solvent with a rotavapor. Material was re-macerated with methanol after wasbenzen evaporation. The same way was done to obtain the metanol extract. These extracts were tested for antioxidant activity using DPPH method with TLC. Active extract was partitioned in 80% methanol. Both 80% methanol soluble and insoluble extract were tested their antioxidant activity using DPPH method (TLC). Active extract was fractionated in vacuum liquid chromatography using a mobile phase with different polarity gradient and different concentrations (wasbenzen:chloroform). Active fraction was isolated by preparative TLC method and purity of obtained compounds was tested by TLC. Antioxidant activity of obtained isolates was tested by DPPH method using spectrophotometry. Compounds known to had antioxidant activity as radical catcher with IC50 178.4µg/mL.

Abstract

Tawas ut tuber (Ampelocissus rubiginosa Lauterb.) is empirically used in wound healing by Dayak community. The present study was undertaken to assess wound healing activity of gel from ethanol extract of A. rubiginosa tuber using parameters of the closure of wound length, tensile strength, and histopathological observations. This study used twenty-five male Wistar rats divided into five groups: positive control (Bioplacenton®), negative control (placebo gel), and three of testing groups with gel extract (concentrations of 1.5%, 2.0%, and 2.5%). Wound healing activity was evaluated to 4 cm length and ± 2 mm depth incision wound model on the back skin of rats. The measurement of wound length was observed on the day of 4th, 8th, 12th, 16th, and 20th used quantitative analyze. On the day of 20th, animal was anaesthetized and the skin has been taken for tensile strength evaluation and histopathological observations. This study showed that group treated with gel extract at the concentration of 2.5% experienced higher wound healing activity with average percentage of wound closure of 99.00% ± 0.16, tensile strength 3.8541 gram/mm2, and proved with the re-epithelization, neocapillarization, and increase collagen density appeared in histopathological observations. Based on this study, gel from ethanol extract of A. rubiginosa tuber possesses wound healing activity.

Abstract

Medicinal plants by the community is still in traditional form, boiling or brewed, so it requires innovation in their use more practical, comfortable consumed, and nutritious with the right dose. Eleuterine palmifolia (L) Merr  which are traditionally used for breast cancer drug, facilitating breastfeeding, and antioxidants developed in effervescent powder preparation. This study uses a randomized block design (RBD) consists of 3 groups with different formulations to mix effervescent compositions containing tartaric acid, citric acid, and sodium bicarbonate on the basis of the rules of stoichiometry, namely : Formula A (50%), Formula B (55 %), and Formula C (60 %). Tests include the physical and chemical properties of powders and hedonic test. Formula B is the best, with a moisture content (0.6%), flow-ability (11.36 g/sec), compressibility (12.5 %), solubility rate (1.52 min), and pH (5). These results meet the criteria of a good powder. Hedonic test results have shown that Formula B as the most preferred product.
