Abstract

Ginger is one of some medicinal plants that has been used to make someone still in the comfortable condition e.g. headache, diarrhea, flatulent, cold, cough, and masuk angin. The purpose of this study was to determine effects of pungent principle extract of the “jahe merah” rhizome in the relation of increasing macrophage phagocytic activation. This method was done by using latex beads.( Leijh et al., (1986) The effects of 10 mg/kgBW, 25 mg/kgBW, and 100 mg/kgBW of pungent principle extract of “jahe merah” rhizome ethanolic extract, that extracted by 70%v/v Ethanol  was decided according to the number of latex which was phagocyted  by macrophages. The conditioning time was done in 20 days. At 15th day, mice were infected by Listeria monocytogenes. The data was analyzed by Kolmogorov-Smirnov, and one way ANOVA with by 95% confidence level. As the specific parameter of the pungent principle extract were: pungent principle spot 1 : 2  = 6.8:1; 3.27%w/w GAE (Gallic Acid Equivalent) as total phenolic concentration, and IC50 14.57 mg/mL. The result was shown that 100 mg/kgBW of 70% ethanolic extract of “jahe merah” rhizome, was capable to increase macrophage activity. The effect of increasing macrophage phagocytosis of 100 mg/kgBW of 70% ethanolic extract of jahe merah was the same with Echinacea extract and Levamisol.

Abstract

One way to prevent the spread of Haemorrhage Dengue Fever is the use of abate. The use of abate as larvicides often complained causing an unpleasant smell, and can cause resistance. Lemongrass oil is reported to have activity as larvicides, and this study aims to make granules of lemongrass oil preparation, as well as determining the value of LC50, LC90 against larvae of Ae. aegypti instar III. The granules of lemongrass oil preparation are made with lactose filler and binder CMC-Na. Larvicidal activity test using 20 larvae, 5 concentration solution to each granules, and contacted for 24 hours. Measurement of total larvae deaths, and data were analyzed using probit analysis from Finney modification, to determine the LC50 and LC90 values. Results shown that lemongrass oil can be made as granules with lactose filler and CMC-NA binder. The LC50 and LC90 values of granules from lemongrass oil are 38,30ppm and 51,57ppm.

Abstract

Antioxidant is known to inhibit free radical reaction. Tyrosine photodegradation can be caused by radical reaction. Nowadays, plant with antioxidants are widely used to inhibit free radical reaction. Study of inhibition of photodegradation used four groups. Those groups are: P1 consisted of 2mL tyrosine 0,05 %; P2 consisted of 2 mL tyrosine 0,05 %, and 600 μL Rhetoflam (topical ketoprofen) 1 %; P3 consisted of 2 mL tyrosine 0,05 %, 60μL Rhetoflam 1 %, and 100 μL tea leaf water ekstract 0,15 %; P4 consisted of 2 mL tyrosine 0,05 %, 600 μL Rhetoflam 1 %, and 100 μL mahkota dewa fruit water ekstract 0,15 %; P5 consisted of 2 mL tyrosine 0,05 %, 600 μL Rhetoflam 1 %, and 100 μL finger root etanolic ekstract 0,15 %; P6 consisted of 2 mL tyrosine 0,05 %, 600 μL Rhetoflam 1 %, and 100 μL vitamin C 0,15 %; each group is added with aquadest up to 5,0 mL and illuminated with mercuric lamp for four hours. Level of remaining tyrosine was measured with visible spectrophotometric method. We used ANOVA to analyse the data with convidence level of 0,95 and then continued by Tukey (HSD). Follin-Ciocalteu method with galic acid calibration curve was used to determine total phenolic level. The level of total phenolic of tea leaf aquoeus extract, mahkota dewa fruit aquoeus extract, fingerroot ethanolic extract were 29.64±0.86 %; 8.29 % 0.27 %; and 7.11 %, 0.15 %, respectively. Our investigation also found gallic acid equivalent (GAE) with the inhibition activity of 4.03; 1.58; and 2.09 and they were bigger than Vitamin C with the same concentration of 0.15 %.

Abstract

Amilum bengkuang (Pachyrhizus erosus (L.) Urb.) dengan ukuran mikropartikel dan mempunyai sifat yang opaque dapat dimanfaatkan sebagai tabir surya fisik. Amilum bengkuang dibuat dalam sediaan krim dan losion dan digunakan secara topikal. Penelitian ini bertujuan untuk mengetahui pengaruh kenaikan konsentrasi amilum bengkuang terhadap viskositas sediaan krim dan losion serta aktivitasnya sebagai tabir surya pada mencit yang dilihat dari nilai Sun Protecting Factor (SPF). Amilum bengkuang diperoleh dengan metode pengendapan, selanjutnya dilakukan karakterisasi dan pengukuran diameter partikel. Pembuatan sediaan krim dan losion masing-masing dalam 3 formula dengan konsentrasi amilum bengkuang 15%, 20%, dan 25%, selanjutnya diamati viskositas dan stabilitas fisik dalam kondisi ekstrim panas dan dingin. Pengamatan selama penyimpanan meliputi warna, bau, homogenitas, dan viskositas. Aktivitas tabir surya ditentukan secara in vivo menggunakan mencit betina galur Swiss Webster yang diinduksi dengan 8-MOP selanjutnya dilihat nilai SPF. Data viskositas pada kondisi ekstrim panas dan dingin dianalisis dengan statistik pada taraf kepercayaan 95%. Hasilnya menunjukkan bahwa kenaikan konsentrasi amilum bengkuang menaikkan viskositas, sedangkan warna, bau, dan homogenitas sediaan tidak berubah selama penyimpanan. Sediaan krim dan losion dengan konsentrasi amilum 15%, 20%, dan 25%, memiliki aktivitas tabir surya secara in vivo pada mencit, dengan nilai SPF berturut-turut sebesar 1,22; 1,52; dan 2,38.

Abstract

The main therapy of wounds infection using antibiotic such as gentamicin sulfate can be applied topically. Increasing incidence of antibiotic resistance forces new strategy to combine plant extract and antibiotic. These two combinations are expected to reduce the incidence of microbial resistance. Ethanolic extract of Aloe vera skin leaves contain anthraquinone that has antimicrobial activity. The aim of this study was to determine the effect and Fractional Inhibitory Concentration Index (FICI) from combination of ethanolic extract of Aloe vera skin leaves and gentamicin sulphate which can inhibit the growth of Staphylococcus aureus. Minimum Inhibitory Concentration (MIC) determination was used Kirby-Bauer disc diffusion method. The concentration of extract solution used were 1,25; 2,5; 5; 10 mg/mL while the solution concentration of gentamicin sulfate used were 2,5; 5; 15; 25 µg/mL. Solution DMSO was used as negative control. Combination solution was made with volume ratio 1:1 from MIC of extract and gentamicin shulphate. The result showed that the MIC of the of ethanolic extract of Aloe vera skin leaves was about 2.5 mg/mL and MIC gentamicin sulphate was about 5 µg/mL. Combination of ethanolic extract of Aloe vera skin leaves and gentamicin sulfate can inhibit the growth of Staphylococcus aureus with zone of inhibition 7,63 mm. FICI of combination was 2 and its antibacterial activity was indifferent compared to single extract and single gentamicin sulphate.

Abstract

Kasturi (Mangifera casturi) is a plant originally from Kalimantan, mainly in South Kalimantan. Although there is no report regarding the usage of the fruit in traditional healings, current research demonstrate that methanolic extract of Mangifera casturi fruit demonstrated high antioxidant activity. As oxidant plays crucial role in inflammatory related diseases, we investigate the antiinflammatory effect of Mangifera casturi fruit methanolic extract. To assess the antiinflammatory activity, we used thioglycollate-induced leukocyte migration in mice.  Male balb/c mice were pretreated (i.p) with Mangifera casturi fruit methanolic extract followed by thioglikolat (i.p) to induce leukocyte migration. After 4,5 hours, the mice were sacrificed and the number of leukocytes were counted using hemocytometer under light microscope.   Our experiment indicates that Mangifera casturi fruit methanolic extract significantly exhibited antiinflammatory activity by inhibiting the migration of leukocytes induced by thioglycollate. However, the activity of the extracts was lower than the positive control, indomethacin. Further purification was required to obtain the active compound with the activity comparable to indomethacin. Thin layer chromatography (TLC) analysis demonstrates that the methanolic extract of Mangifera casturi fruit contains terpenoid and phenolic compounds.

Abstract

Antidiabetic activities of Phaleria macrocarpa fruits and Annona muricata leaves through α-glucosidase inhibition have been reported. Each extract of the two medicinal plants showed weak α-glucosidase inhibitory activities. The effect of synergies is expected from combining the two extracts. The purpose of this research was to investigate α-glucosidase inhibitory activity of the combination of ethanolic extract of P. macrocarpa fruits and A. muricata leaves and to confirm the presence of different chemical constituents of the most active individual and extracts combination by thin layer chromatography (TLC). Extracts were obtained by maceration method using 96% ethanol. Various concentration of extracts combination was analyzed based on IC50 of the enzyme inhibition of each individual extract. The result showed that α-glucosidase inhibitory activity of each extracts was concentration dependent, with IC50 values of individual extracts of P. macrocarpa fruits and A. muricata leaves were 261.34 and 428.79 μg/mL, respectively. However, the extracts combination with the highest inhibition activity (84.51±0.79%) was obtained at its IC50 of P. macrocarpa fruits and two-third IC50 concentration of A. muricata leaves. The TLC profile of the most active extract combination showed a phytochemical constituent that is also detected in the profile of each individual extract. This can be assumed as a synergistic effect due to increased phytochemical contents of the combined extract.

Abstract

Free radical induced oxidative stress that influences the occurrence of various degenerative diseases such as cancer, coronary heart disease and premature aging. In the case that body's antioxidant defense system does not have excessive antioxidants, additional natural antioxidant via food or other nutrients intake is needed. Stems of ferns are known to contain phenolic compounds that are known to have antioxidant activity. A study has been carried out to determine antioxidant potential of stems of ferns of western Borneo origin compared to quercetin as positive control. Initially, material was macerated gradually with wasbenzen followed by methanol. The extract obtained was filtered and evaporated to dryness to give dried wasbenzen extract and methanol extract respectively. Antioxidant properties of extracts were performed by DPPH on TLC. Active (wasbenzen) extract was then partitioned with 80% methanol to give soluble and insoluble fractions. Active (insoluble) fraction was further fractionated by vacuum liquid chromatography using a gradient mobile phase with increasing polarity (wasbezen: chloroform). Fractions obtained were tested antioxidant activity, and isolation was done by preparative TLC. The purity of isolated antioxidant compound was confirmed by TLC, and antioxidant potential was measured spectroscopically with DPPH method. This compound displayed antioxidant activity, but its value was lower than that of quercetin used as a positive control (IC50 178.4 v / s 2.17 microg / mL)

Abstract

Essential oil of clove (Syzygium aromaticum) containing eugenol has an anti-inflammatory activity. The study was aimed to develop the formulation of lotion by adding of oleic acid and propylene glycol as penetration enhancer. The effect of enhancer composition was also studied. Lotion was prepared with the composition of oleic acid (AO) and propylene glycol (PG) as follow: 1:0 (FI), 0,5:0,5 (FII), 0:1 (FIII). Capacity an anti-inflammatory of formulation based on parameters of the amount of cells with COX-2 expression, the number of inflammatory cells and the epidermis thickness was evaluated using male mouse strain BALb/C induced by crotton oil as inflammatory agents. The results showed that the increasing composition propylene glycol caused the decreasing of the amount of cells with COX-2 expression (p <0.05), the inflammatory cells (P <0.05) and the epidermis thickness (p <0.05)

Abstract

Squamous cell carcinoma is one of the most common malignancy in the oral cavity.  Currently, cancer treatment using surgery, radiotherapy, chemotherapy and its combination.  New strategy for cancer therapy is by searching herbal which can induce apoptosis. Bromelain is one of a potential anticancer agent that can induce apoptosis. Pineapple stem is one of a potential herbal medicine because of its bromelain enzyme content. The aim of this study was to investigate the relation between pineapple stem ethanol extract and apoptosis of oral tongue squamous cell carcinoma cell line. Oral tongue  squamous  cell  carcinoma  cell  line was  treated  with  three concentrations  under  IC50  of  pineapple  stem  ethanol  extract,  including  5.000, 5.500,  6.000  µg/ml.  In  apoptosis  test,  cell  stained  with  fluorochrome  ethidium bromide and acridine orange after 24 hours incubation. Fluorescence microscope was used for counting the cell. Viable cell would be stained green and apoptotic cell would be stained yellow to orange. Probit analysis was used to determine IC50 of pineapple stem ethanol extract on Oral tongue squamous cell carcinoma cell line. Pearson analysis used to know the correlation between pineapple stem ethanol extract concentration and Oral tongue squamous  cell  carcinoma  cell  line apoptosis. The  result  showed  that  there  was  a  positive  correlation  between concentration  of  pineapple  stem  ethanol extract  and  apoptotic  cell  (r=0,999, p<0,05). In conclusion, pineapple stem ethanol extract treatment induced apoptosis of  oral  tongue  squamous  cell  carcinoma  and  the  increases  of  pineapple  stem ethanol extract concentration followed by the increase of apoptosis induction.
