Abstract

Pain is a sensory and emotional feelings associated with tissue damage. Leaves kerehau recognized by people Dayak and Kutai as postpartum pain medication. Kerehau leaf has long been known by the public because many have efficacy in medicine. This research to determine the analgesic activity of ethanolic extract of leaves kerehau (Callicarpa longifolia Lamk.) on male white mice (Mus musculus). This is experimental research, randomized complete unidirectional pattern. The animals test were divided into 5 groups with each treatment of 5 mice. Group I (Na CMC 0.5% negative control), Group II (ibuprofen as a positive control), group III - V the ethanol extract of leaves kerehau dose of 150 mg , 300 mg and 600 mg/ Kg BW). Inductor pain acetic acid 0.5 % (ip). Observed number of writhing in mice for 1 hour and calculated percent analgesic . The results obtained in this study has efficacy as an analgesic with Strength of power dose I: 51.65 %, dose II: 65.40 %, and dose III: 75.98 %.

Abstract

Shiitake mushroom (Lentinula edodes (Berk.) Pegler) is one of the fungi that has potentially been used as food and medicinal mushroom. Cultivation in liquid medium by the fermentation process has been developed over recent years. Four isolates of L.edodes (isolates from Malang, Cianjur, Lembang and Yogyakarta) cultured on three types of fermentation media (KM: Medium Kauffman; GYMT: Glucose, Yeast Extract, Malt Extract, Thiamin; YEMR: Yeast extract, Malt Extract, Rice bran). Bioactive metabolites  extracted from mycelium biomass and fermentation culture filtrates were tested using pathogenic microbial Staphylococcus aureus ATCC 25923, Escherichia coli ATCC 35218, Candida albicans ATCC 10231, and Trichophyton mentagrophytes. Inhibition zone diameter data were analyzed using analysis of variance followed by Duncan test at 5% error rate. The results showed that bioactive compounds in the culture filtrates and mycelium biomass of L. edodes extracted using chloroform, ethyl acetate and water, were able to inhibit S. aureus, E. coli and C. albicans,  but were not able to inhibit T.mentagrophytes. Isolates of L.edodes from Lembang and Yogyakarta cultured on Kauffman Medium gave  the best results with inhibitory zones of 24.97 to 31.14 mm. Microbes which gave the most sensitive of bioactive compounds test from L. edodes were C. albicans followed by E. coli, S. aureus and T. mentagrophytes. Bioactive metabolites from L. edodes inhibited potentially pathogenic microbes  from the group of  bacteria and  yeasts but did not inhibit the fungus.

Abstract

Benalu belimbing (Macrosolen cochinensis) and onion sabrang / onion dayak (Eleutherine palmifolia (L) Merr.) is a traditional medicinal plant that has been used by the community as a cancer drug from generation to generation. This study aims to prove scientifically about the anticancer potential from combination of both plants. Anticancer potential of the plant has been proven by testing the extract both in single form and in combination with Hela cells. While to know the selectivity of the extract to HeLa cell then extract is tested on normal cell Vero cell line. Furthermore, a combination test of both extracts was done using variation dose under IC50. The method used to determine anticancer activity was the MTT method. The results obtained in this study showed that ethanolic extract onion sabrang selectively able to inhibit the growth of cervical cancer cells HeLa on IC50: 40.36 μg / ml with 4.06 for the SI (selectivity index) value. While ethanolic extract of benalu belimbing bselectively can inhibit the growth of cervical cancer cells HeLa on IC50: 217.72 μg / ml with 5.77 for the SI (selectivity Index) value. The combination test of sabrang onion extract and extract of benalu belimbing showed that the combination was highly synergistic in combination dose of EBS 22.62 μg / ml and EBB 23.3 ug / ml with combination index 0.15. Whereas the combination doses having synergistic effects on combination doses 23.3 μg / ml for EBB and 11.31 μg / ml for EBS; 23.3 μg / ml EBB and 16.96 μg / ml EBS with a consecutive combination index; 0.51 and 0.64. From these results it can be concluded that the onion sabrang extract has anticancer effect on HeLa cells and selectively killing HeLa cells because the value of selectivity index is greater than 3. The result of the combination test of both extracts shows that the combination dose having strong synergistic effect, so that combination can be recommended for further research.

Abstract

Essential oil of Curcuma heyneana Val & Zijp has antifungal activity against Candida albicans with minimum inhibitory concentration (MIC) 0.25% v/v. Characteristic of essential oil is easy to evaporate from the skin surface. Therefore, it should be formulated in cream. The research aims to know influence of cream formulations of Curcuma heyneana oil toward physical characteristics and antifungal activity against Candida albicans. Essential oil was obtained by water and steam destillation. Cream were made into oil in water base (O/W) and water in oil base (W/O) containing 10% of Curcuma heyneana oil. Cream were tested for physical characteristics (such as viscosity, spreadability, protection ability, adhesive time, pH) and also in vitro antifungal activity against Candida albicans. Observation for antifungal activity was done after incubation for 48 hours. Data for physical characteristics and antifungal activity were analyzed using one way anava and continued with t-LSD test (p=0,95). The research indicated that W/O cream containing Curcuma heyneana oil had higher viscosity than O/W cream. Spreadability and protection ability O/W cream was better than W/O cream. Adhesive time of W/O cream was longer than O/W cream. Both of O/W and W/O cream had pH level of 6-7. O/W cream containing Curcuma heyneana oil had better inhibition zone against Candida albicans than cream W/O, they were 1.84±0.071 cm and  1.70±0.074 cm, respectively.

Abstract

Some compounds were potentially as a radical catcher, it was also found in the papaya fruit (Carica papaya L. (orentang)). This study aimed to determine 2,2-diphenyl-1-pycrylhydrazyl (DPPH) radical capture of  ripe and raw papaya fruits with vitamin C synthetic as a benchmark. Mashed papaya pulp was dried by the freeze-drying method. The dryed mass was dissolved in ethanol and reacted with DPPH. Vitamin C was also treated by the same method, then made a standard and curve of percent DPPH radical arrest by vitamin C. Percent DPPH radical capture by the ripe and raw papaya fruit powder extracts was plotted on a standard curve. Ripe papaya fruit had the ability to capture the highest DPPH radical (1.340,931 mg of fresh ripe papaya proportional to 1 mg of vitamin C). The raw papaya had the capability of 3.557,814 mg of fresh raw papaya proportional to 1 mg of vitamin C.

Abstract

Bulls fertility has an important role to conception, so bulls should have a high libido, high quality semen, and good performance. This study aimed to improve the quality of semen bulls using traditional herbal medicine by utilizing a combination of medicinal plants. The study used 20 heads of Peranakan Ongole (PO) bulls, is divided into four trials traditional herbs, namely Curcuma xhanthorriza, Curcuma eoriginosa, Alpinia galanga, Andrographis paniculata, Piper retrofractum, Eurycom longifolia, Pimpenella purwatjan, each of which is F1 (15%, 15%, 20%, 5%, 30%, 15% and 0%); F2 (15%, 15%, 20%, 5%, 30%, 10% and 5%); F3 (15%, 15%, 20%, 5%, 25%, 20% and 0%); and F4 (15%, 15%, 20%, 5%, 25%, 15% and 0%). Giving the bull 50 g dose of herbal formula every 300 kg of body weight bull mixed with 200 ml honey and 5 eggs. Bulls were given traditional herbal formula once a week orally. The dependent variable of sperm volume, quality of sperm and strength libido were assessed with randomized block design one way. The results showed that all four traditional herbal formulas that are given to PO bulls did not appear to significantly affect the volume of semen and libido for a month with an average volume of 3.8 ± 2.8 cc / ejaculate and libido 159.9 ± 20.2 seconds. Sperm motility, sperm concentration, percentage of live sperm, and sperm abnormalities in PO bulls were given traditional herbal medicine with four different formulas also not significant (P> 0.05); with as well as the color, pH and consistency is still within the normal range. It was concluded that the four herbal formula derived from a mixture of traditional medicine can be given to bulls in order to increase libido and semen quality. It is suggested for granting herbal medicine ingredient formula of traditional medicine that are cheap and easily obtained to bulls.

Abstract

Antioxidant is a substance which in small concentrations can significantly inhibit or prevent the oxidation of the substrate. One plant is efficacious as an antioxidant is the persimmon (Diospyros kakiThunb.) where is cultivated widely in East Asia, Spain and Indonesia. In the search for natural antioxidant compounds, have been studied isolation and identification of antioxidant compound of persimmon leaves (DiospyroskakiThunb.) using DPPH (2,2-diphenyl-1-pikrilhidrazil) method. The extraction of persimmon leaves was carried out by maceration method with wasbenzen. Extracts obtained by evaporating the solvent wasbenzenon a rotary evaporator and were then re-macerated with methanol. Antioxidant activity of extracts was evaluated with DPPH 0.2% method. Active extract was partitioned with chloroform, methanol, distilled water and tested for antioxidant activity by DPPH 0.2% method. The active compound was purified byPTLC and the purified active isolates was then confirmed by TLC. The antioxidant activity (IC50) active isolates were analyzed using spectrophotometry. Analysisof active isolates resulted the antioxidant activity (IC50) of 100.00 ug / ml. UV-Vis spectrum of the active isolates showed absorption at λmax 285 and 401 nm. Infrared spectra (KBR) showed absorption at 3446 cm-1 (OH), 2926 cm-1 (CHaliphatic), 1456 cm-1 (CH2), 1384 cm-1 (CH3), 1255 cm-1, and 1115 cm-1 (C-0-C), 1631 cm-1 (C=Caromatic). GC-MS spectra gave two peaks with retention time 11.408 minutes with a similarity index of 79.243% with the molecular ion (M•)+ appeared at m/z 178, 163, 147. These spectra are expected to be methyl eugenol. Furthermore, the peak with retention time 12.982 minutes with a similarity index of 20.757%, producing a mass spectrum with molecular ion peaks (M•)+appeared at m/z 192, 177, 161. These spectra are expected to be myristicin compound.

Abstract

Essential oil is a natural substance that has been known as antibacterial. This research aimed to examine the effectiveness of antiseptic gel from essential oil which taken from rind Pontianak orange. Essential oil from rind Pontianak orange are formulated into a gel and it tested its effectiveness as an antibacterial to Escherichia coli and Staphylococcus aureus. Based on the result of the study, the effective concentration of essential oil to inhibit Escherichia coli is 0,5%, while Staphylococcus aureus is inhibited at concentration 0,1%, 0,3% and 0,5%. The results of the gel test stabilization showed that the homogenity of gel, pH value and stable adhesion are good. Coverage gel increased during 30 days of storage. One Way Anova test analysis showed that the dispersive power of each gel formula does not differ significantly.

Abstract

Inflammation is a protective action that role on defending mechanism to defeat agent causing cell injury. Glucoside of skunkvine (Paederia scandens) has anti-inflammation effect. The aim of this study was to know the anti anti-inflammatory effect of skunkvine (Paederia scandens) extract on artificial edema in Wistar rat. Thirty-five Wistar rats were divided into 7 groups, each group consisted 5 rats. In the control and treatment groups, aquadest (negative control), fenilbutazon (positive control), skunkvine extracts in dose 10mg/kgBW, 20mg/kgBW 30mg/kgBW, 40mg/kgBW, and 50mg/kgBW were administered per orally. In order to induce the edema, one hour after administration, in each group, right hind paw of the rats was injected subcutaneously with by 1% carrageenin. The volume measurement performed with one hour interval for 5 hours assessing with pletysmometer. The data were analyzed by ANOVA. The result of this study showed there was significant difference on group with skunkvine extract 20mg/kg BW to negative control and positive control. In conclusion, that the skunkvine extract 20 mg/kgBW has potency as anti-inflammation substance.

Abstract

Jatropha multifida L. (jarak tintir) was a shrub, annual, and had ± 2 m high. Empirically jarak tintir sap was used as traditional medicine by Indonesian people for a long time. Only limited studies were conducted regarding its chemical compound. It was reported that multifidone (diterpenoid compound from the stem) had an activity against cancer cells in vitro. This study aimed to test the toxicity of various extracts (n-hexane, ethyl acetate, and methanol) with Brine Shrimp Lethality Test (BSLT) of Jatropha multifida L. stem. A Fraxidin (8-hidroxy-6,7-dimethoxy coumarin) has been isolated from ethyl acetate fraction based on highest cytotoxic with LC50 value 3.69 µg/mL. The isolated compound was elucidated to gain chemical structure base on spectroscopic data (UV-Vis Spectrofotometric, IR Spectrofotometric, and NMR). Toxicity of fraxidin was tested on BSLT and showed no potential activity with LC50 value > 500 μg/mL.
