Abstract

The process of freezing and thawing on semen can lead to physical stress, often called cold shock, and couses the structural and biochemical damage that affecting cell function and ultimately lead to the death of the cell The aim of this study was to know the effect of the addition of various concentrations of dimethylsulfoxide (DMSO) as the intracellular cryoprotectant in phosphate yolk diluent on the post thowing quality of the green jungle fowl semen. The study used eight green jungle fowl semens which were collected with massage techniques. Semen was evaluated macroscopically and microscopically. Good quality semen was diluted with phosphate yolk which was added four different concentration of DMSO, namely 4%, 6%, 8%, and 10%. Semen was then filled and sealed in a mini straw (0.25 mL) with the concentration of 150.106 cells, and equilibrated at 4oC for 4 hours. The semen freezing was processed using conventional method. Evaluation was performed on post thawing semen. The evaluation of semen quality included the progressive motility and plasma membrane intact. Data were analyzed by analysis of variance. If there were any significant differences, the data were futher analyzed by Duncan test. Abstract

The purpose of this study was to examine the effect of the extract of Moringa oleifera leave on Argulus sp infestation in comet fish, and determine an optimal concentration and time. Moringa oleifera leave was extracted using ethanol 96%. In total, 25 comet fishes (Carassius auratus auratus) were used in this research, in which infested by one to five individuals of Argulus sp per fish. This research used pretest and posttest design with four treatments of extract concentration of Moringa oleifera leave, namely, TI 25 mg/l, TII 37.5 mg/l, TIII 50 mg/l, TIV 62.5 mg/l, and T0 without extract as a control. Each treatment was imposed on five comet fishes. The parameter of this study was the amaount of Argulus sp released from the body of the fish. The results showed that the extract of Moringa oleifera leave had an effect on ectoparasite on comet fish. There was an indication that the more concentration of the extract of Moringa oleifera leave, the more Argulus sp released, although statistically, it was not significant (P>0.05). The optimal time of giving the extract of Moringa oleifera leave to Argulus sp infestation was 12-h.

Abstract

This study aimed to determine the effort of fermented purple sweet potato in the rations to improve carcass, antioxidant profile, meat and eggs lipid profile of bali ducks. Completely randomized design with three treatments was used in this study. Sixty baliducks were divided into 3 treatments that provision by rations; A: without purple sweet potato, B: with 10% purple sweet potato, C: with 10% fermented purple sweet potato.The experiment used four replications to each treatment which 5 layer ducks were used as a repetition in each unit of experiment. The experiment was done for 12 weeks. Antioxidant consumption was counted during the experiment. In the end of experiment the ducks were killed and meat antioxidant profile namely antioxidant capacity, malondialdehida (MDA) and superoxide dismutase (SOD), carcass profile including slaughter weight, carcass weight, carcass percentage, and physical carcass composition (meat, bone, fat including skin) were recorded. Meat and egg lipid profile (total cholesterol, highdensity lipoprotein (HDL), low density lipoprotein (LDL) and triglycerides) were also counted in the end of experiment.The results showed fermented purple sweet potato in the rations significantly increased the consumption of antioxidants, accompanied by increasing in slaughter weight, carcass weight, carcass percentage, and meat percentage. Fermented purple sweet potato in the rations also significantly reduced the percentage of sub-cutaneous fat and skin. Fermented purple sweet potato improved antioxidant profile significantly, increased antioxidant capacity, and superoxide dismutase (SOD), and decreased the levels of malondialdehide (MDA). It also decreased significantly the total cholesterol, HDL, LDL and triglycerides in meat and eggs. The conclusion of the research is a 10 % fermented purple sweet potato in the rations be able to improve the antioxidant consumption, carcass, antioxidant profile, meat and eggs lipid profile of first laying phase of bali ducks.

Abstract

Genomic selection has a high potency to improve the genetic quality of Bali cattle. In effort to find out a molecular marker or DNA segment which is linked with a certain phenotype, we examined the association between BoLa-DRB3 microsatellite polymorphism and dam body weight and birth weight of calves in Bali cattle. A total of 40 dams from Abuan Village, the District of Susut was investigated. The total DNA was extracted with QIAamp DNA blood mini kit. The locus was amplified using PCR technique with a 54OC annealing temperature.. Alleles were separated through PAGE 6%, and visualized with silver staining. We found 10 alleles which their length varied from 145 bp to 233 bp. Allele 195 had the highest frequency that was 35.0%. The HE, HO, PIC were 0.821, 0.875, and 0.791 consecutively. Statistical analysis showed that the polymorphism of DRB3 microsatellite had a marginally significant association (p < 0.1) to the dam body weight. However, it had no significant association to the birth weight of the calves. The results of the research reflect that the DRB3-BoLA gene has a weak potency in controlling the body weight.

Abstract

Newcastle disease virus (NDV) is very contagious disease agent, and causes many outbreaks trough out thecountry of Indonesia. This study was conducted to determine the virulence of Bali field isolate of NDVbased on haemaglutinin-neuraminidase (HN) protein genes. Four field isolates from different locationswere propagated at fertilized chicken eggs of 9 days old. Allantoic fluid was harvested and NDV wasconfirmed using standard haemaglutination (HA) and haemaglutination inhibition test (HI) methods. Thefragment of HN protein gene was amplified using RT-PCR. The product was sequenced using Big-Dyetermination method. All four isolates grew well with the titer of 2[5]-2[9] HA unit and could be confirmedusing HI test. The HN genes, however, exhibited variations at its 3-D structure and hydrophobicity betweenthe virus that previously circulated in Indonesia and vaccine virus. It is concluded that all four Bali’sisolates under this study are virulent VND of the genotype VII. Further testing is needed to justify the bestformula of NDV vaccine to be used trough out Indonesia.

Abstract

Recent years the infection of Neospora caninum parasite has been a significant reproductive disease on cattle. The purpose of this study was to determine the seroprevalence of Neospora caninum in Bali cattle that are slaughtered in Denpasar’s slaughter house.A total of 184 serums was collected and tested using enzyme linked immunosorbent assay (ELISA). The results showed that there were 2 serum samples (1.08%) contain Neospora caninum antibodies. While the remaining samples, 182 samples (98.92%), were negative. Though the neosporosis seroprevalence rate in Bali is very low, a sustainable surveylance must be carried out to mitigate the broadness of the disease distribution.

Abstract

The aims of this research were to overview the effect of maternal antibodies on the histopathological changes and viral antigen distribution of the broiler chickens challenged with ND APMV-1 virus. A total of 100 chicken were allotted into 3 treatment groups consisting of group I (titer antibodies< 23 HI Unit), group II (titer antibodies 23 – 24 HI Unit) and group III (titer antibodies> 24 HI Unit). All group I, II and III were inoculated with ND virus isolates of type viscerotropic velogenic at the dose of 1000 TCID50. The histopathological changes observed in nervous system were endotheliosis and perivascular cuffing. Immunohistochemistry staining showed that NDV infected cells were found in most organs both in inflammatory cells and in epithelial cell of many organs mainly in nervous, respiratory and digestive systems. Neurological symptoms and neural lesions were highest in group II (titer antibodies 23 – 24 HI Unit)

Abstract

Live Bird Market (LBM) has a high potential for spreading Avian Influenza Virus (AIV) between fowls or from fowl to human. Up to now, a dinamic of avian flue incidents at many LBMs in Bali has not been reported. This research aimed to reveal a dynamic of seroprevalences of avian influenza in ducks at Beringkit (Badung) and Galiran (Kelungkung) LBMs. A total of 35 duck blood samples was collected from each of LBMs. Sampling was conducted monthly from March to August, 2012 . AIV antibody of duck serum was measured using Rapid Hemagglutination Inhibition (Rapid HI) test. Seroprevalence differences were analyzes with Chi-square (?2) Nonparametric statistical test. The results showed that seroprevalences of AIV H5 in ducks at Beringkit and Galiran LBMs were very high, ranged from 68.6% to 100% and 65.7% to 97.1% respectively. A Dynamic of AIV H5 seroprevalences in ducks at Beringkit and Galiran LBM had a similar pattern, except in July 2012. This indicates that VAI H5 has been circulating for a long time and has been to be an endemic virus infection in ducks at LBMs in Bali. It can be suggested that an Avian Influenza Virus monitoring should be done continuously over a long period.

Abstract

A study to determine the presence and prevalence of bovine tuberculosis and Klebsiella pneumoniae atcattle slaughterhouse in Bali was carried out in Pesanggaran and Mambal abattoirs from January to March2015.The Lungs and lymph nodes were inspected for lesions and then examined through DNA analysis forMycobacterium bovis and culture for isolation of Klebsiella pneumoniae. Of the samples examined, no onehad lesions suggestive of tuberculosis in two abattoirs.However, Klebsiella pneumoniae was observed1/4513 sample (0.02%) only in Mambal abattoir. This study is the first report of the presence of Klebsiellapneumoniae in Bali. Therefore, a aware of this zoonotic Klebsiellosis and its control managementformulation are need.

Abstract

This retrospective study was conducted to know the relation between time and frequency of mating on litter size of Golden Retriver  in Denpasar Region. Mating incident records of 19 Golden Retrievers from 8 kennels were compiled to be analyzed. These data can be use as a reference in effort to maximize the amount of  Golden Retriever clans. The result showed that the three times of mating on the 10th , 12th and 14th day after onset of proestrus produced the most litter size 10 ± 2.82. However, mating twice on wice on 9th and 11th after onset of proestrus produced a litter size of 8.50 ± 0.70. The statistic analysis depicted that there was no significant relationship between mating frequencies and mating time on litter size.
