Abstract

Rice is a principal food source for most of the world's population. Rice has many nutrients including thiamin hydrocloride (THC) and pyrodoxine hydrocloride (PHC). Before consumption, rice is usually processed such as milling, washing, and cooking. Determination of THC on IR-64 and Rojolele rice with washing three times has been conducted. Also determined of PHC on the milled IR-64 rice and the pounded IR-64 rice. The THC was determined by spectrophotometric method at 246 nm. Determination of PHC based on the coupling reaction with diazo reagent in alkaline solution to form azo dye then the absorption was measured at 440 nm. The result of determination of THC on IR-64 and Rojolele rice after washing three times can decrease THC level 12,93% and 12,88% respectively. Determination of PHC on the milled IR-64 rice obtained 1.54 ± 0.027 mg/g and on the pounded IR-64 rice 1.80 ± 0.048 mg/g. The result of statistical test with independent T test showed there were significant difference (α = 0,05) PHC level on both treatments.

Abstract

Meloxicam (MLX) is a nonsteroidal anti-inflammatory drugs (NSAID) which belong to Biopharmaceutical Classification System (BCS) class II, which have a low solubility level with a high permeability. Therefore, to enchance is solubility level, physical modification of the meloxicam is required. It  can be done by the cocrystal formation. Cocrystal contained active ingredients and coformer which bind through the hydrogen bond. This study used malonic acid (MA), since it contained carboxylic group which expected to form hydrogen bonds. Slurry is a method of cocrystal formation by mixing two ingredients, i.e. active ingredients of the drugs and its cofomer, dissolved in a solvent and may be formed due to the heat energy released by the friction between particles and their crusher. This study aimed to determine the characteristic of meloxicam-malonid acid cocrystal by 1:1 mol ratio using PXRD, DSC and FTIR. The result of PXRD charaterization indicated a new peak at angle 9,4° and 18,5°. The result of DSC characterization indicated an endothermic peak with a low melting point at 97,64°C ; 152,62°C ; 176,87°C temperature. The result of FTIR characterization indicated a shirt of the O-H uptake band from at wave number 3126-2980 cm-1 to 3292-2951 cm-1, the N-H uptake band of meloxicam at wave number 3492 cm-1 to 3525 cm-1

Abstract

The root of Calotropis gigantea (thistle) is an Indonesian herb to treat cancer based on empirical and scientific evidences. The study aims to find out the infuence of ethanol extract of Calotropis gigantea root in inhibiting cancer cell growth of Mus musculus fibrosarcoma in vivo and to find out metabolite compound in the Calotropis gigantea root extract. The effect of cancer cell growth inhibition was tested on Mus musculus inducted with 7.12-dimetilbenz (α) antrasena (DMBA) and dose treatment of 50, 100 and 150 mg/Kg body weight. The metabolomic analysis on the root employs UPLC-QToF-MS/MS as a positive ESI ion source, the movement phase of water/acid mixture is 99.9/0.1 [v/v] and acetonitrile/formic acid was 99.9/0.1 [v/v] with gradient elution system and stationary phase of C18. The chromatogram was analyzed using Masslynx 4.1. The component identification was based on the m/z ratio measured in Masslynx and m/z counted in chemdraw. The result of the study showed that the Calotropis gigantea root extract with the dose of 50, 100 dan 150 mg/Kg body weight were able to increase the weight of fibrosarcoma mice and have a significant influence on caspase-3 expression with cell apoptosis index 24.3 %; 13.3 % and 12.3 % respectively. The result of metabolomic analysis showed 14 compounds found in the root extract. There are two major compounds: 4-Chlorobenzenethiol with the area 32.51% and N-[1-(Adamantan-1-yl) ethyl]-2-(1-piperidinyl)-4-quinazolinamine with the area 37.20%. They are indicated responsible for the anticancer activity of Calotropis gigantea root extract. Keywords: Calotropis gigantea, caspase-3, fibrosarcoma, metabolomic, UPLC-QToF-MS/MS

Abstract

Chrysanthemum cinerariifolium (C. cinerariifolium) is an empirically proven medicinal plant that has anticancer activities. This study aimed to profile metabolites, and cytotoxic activity of root, leaf, stem, and root extracts C. cinerariifolium on T47D cells and to determine the correlation of metabolite content with cytotoxic activity. The metabolite profile was carried out using UPLC-QToF-MS / MS, and cytotoxic activity was carried out using the MTT method. The results obtained in the form of a chromatogram were processed with the application Masslynk so that a metabolite profile data obtained. The data is then analyzed statistically using Principal Component Analysis (PCA). The results obtained on metabolite profiling showed that there were differences in metabolite profiles in the roots, stems, leaves, and chrysanthemums. The characteristic compounds in the flower section are D - (-) - Morphine and in the leaf part of genistein  and N - [(5-Chloro-1,2,3-thiadiazol-4-yl) methyl] -1- (2-isopropyl-4 -methyl-1,3-thiazol-5-yl) -N-methyl ethanolamine. The major compounds in the roots, stems, and leaves are Orphenadrine with successive percentages of 9.11%, 10.16%, and 3.24%, and the major compounds in the flower section are D - (-) - Morphine with a percentage of 10, 86%. Furthermore, the results of the cytotoxic activity test showed differences in anticancer cytotoxic activity in the parts of the flower, leaf, stem, and root of C. cinerariifolium. Besides, there is a relationship between the metabolite content and anticancer cytotoxic activity of each part of C. cinerariifolium. The higher the level of orphenadrine compounds in the plant, the higher the potential for anticancer. Keywords: C. cinerariifolium, UPLC-QToF-MS/MS, T47D cells.

Abstract

Indonesia has so many kinds of a parasite (mistletoe), one of them which has potential anticancer activity is mango parasite (Dendrophthoe pentandra). This research aimed at determining the cytotoxic activity profile of mango parasite leaves extracts from several locations in Indonesia against T47D and Vero cell lines. Mango parasite leaves obtained from 4 locations in Indonesia: Kediri, Pekalongan, Sumatra, and Kalimantan. Three samples was obtained from 3 different sub-districts in each locations. Cytotoxic activities of samples were performed by MTT assay method. The results of this study showed that there were differences between samples obtained from different locations, especially in cytotoxic activity and selectivity indexes of mango parasite leaves (D. pentandra) ethanol extract. D. pentandra obtained from Kalimantan has the most potential cytotoxic activity and could be potentially developed into a T47D breast anticancer agent. Extracts from the Kalimantan have an IC50 values of287.39 μg/ml, LC50 798.28 μg / ml and selectivity index 2.77, which means it was toxic to cancer cells but safe for Vero cell lines, and its selectively kills cancer cells rather than Vero cell lines. The result of this study could support D. pentandra to be developed as a new potential Fitofarmaka.

Abstract

Corn stalk has a high cellulose content, so that it is potential to be used as a composition for making alginate-carboxymethyl cellulose beads. Alginate and cellulose are biodegradable, renewable and non-meltable polymers that have wide applications in various industrial sectors. The purpose of this study was to determine the effect of crosslinking agent C4H6O4Zn on the adsorption and shape of beads. The varied concentrations of C4H6O4Zn are 3%; 5% and 10%. Characterization of alginate-carboxymethyl cellulose beads composites using Fourier Transform InfraRed (FTIR), and Scanning Electron Microscope-Energy Dispersive X-Ray (SEM-EDX). Based on research, the highest swelling value is obtained at 5% C4H6O4Zn crosslink which is 59.68%. FTIR data shows the appearance of wave numbers at 1413 cm-1 which indicates the presence of C-O Na groups, while at wave number 458 cm-1 indicates the presence of Zn-O groups. SEM-EDX data with a 5% C4H6O4Zn crosslink has a round shape with a wrinkled surface, multiple grooves causing a non-homogeneous surface. Whereas in C4H6O4Zn 10% the surface is almost smooth

Abstract

Low Back Pain is one of the disease that causing global disability in the worldwide. In the past 10 years, prevalence of LBP has not decreased, it makes global burden increase substantially. Chronic low back pain(CLBP) is pain that is almost experienced daily by patients in the lower back region in a period of more than 3 months. Chronic low back pain is a mixture of pain that arises through nociceptive and neuropathic mechanisms. Over the past decade, numerous  guidelines have been used  in the management of CLBP. Problems that are still a controversy include the management of pain. The mechanism of many treatment measures is still in the low to moderate effectiveness stage, so that the pain management target is not optimal. Substance P is a neuropeptide that release  both in acute and chronic pain. Therefore study about mechanisms that affect SP release may show important information regarding pathological alterations in pain perception.

Abstract

Chronic low back pain (LBP) is the frequent chronic pain and notable health problem which is the main reason of disabling conditions, individual suffering, sick leave, and costly treatment.  Unfortunately, available therapy especially oral medicines only beneficial in no more than 50% of patients. Tricyclic antidepressants (TCAs) are commonly used for neuropathic pain but the data for neuropathic LBP is limited. The purpose of this review is to evaluate the efficacy and adverse drug reaction of TCAs in chronic LBP. We searched scientific search engines including Scopus and PubMed and collected original articles including Randomized Clinical Trial (RCT), comparative, and observational study about Tricyclic Antidepressants treatment in chronic LBP from 1998 to 2019. In this review, 5 articles and 543 patients were evaluated. All recruited articles are randomized clinical trials. Among all five relevant articles, three of them evaluate amitriptyline treatment in chronic LBP while the rest articles evaluate imipramine and nortriptyline. Amitriptyline is the most well-studied TCA for chronic LBP. We found that both amitriptyline and nortriptyline can reduce pain severity significantly when the dose is increased gradually depending on patient tolerance. TCAs can also improve disability significantly. However, single-dose imipramine is not superior than placebo. We conclude that TCAs can be effective for treating chronic LBP when used routinely for several weeks and the dose should be increased gradually depending on patient tolerance although further studies are required to confirm the optimal dose for analgesia. The most prevalence Adverse Drug Reactions (ADRs) of TCA including dry mouth, somnolence, insomnia, sedation, and constipation.

Abstract

Ibuprofen is an anti-inflammatory class of non-steroidal propionic acid used in the treatment of osteoarthritis, analgesics, and antipyretics. Ibuprofen made in a transdermal patch to avoid side effects of irritation to the stomach, first pass effect on application oral. This aims of the research to determine the physical characteristics of the patch, the effect of propylene glycol concentration (150 mg, 200 mg, 250 mg) on the thickness patch, pH, and the rate of penetration as increase in penetration of the ibuprofen transdermal patch. Patch preparations were made in 3 draft formulas with varying concentrations of propylene glycol 150 mg, 200 mg, and 250 mg. The method used for penetration testing is Franz diffusion cells. The results of physical characteristics are clear, odorless, dry, not cracked, 300 folds, thickness ranges from 0.142 to 0.184. The concentration of propylene glycol effects (p-value 0.05) the penetration and no effect (p-value 0.05) the pH test. Increasing the concentration of propylene glycol causes an increase medicine that penetrate the skin.Keywords: Ibuprofen, transdermal patch, propylene glycol

Abstract

World demand for halal pharmaceutical products, which include food, cosmetics, drugs, and other products will continue to grow with the increasing of global Moslems population in the world. Milk is one of the pharmaceutical products produced from livestock commodities. It is processed into 2 types of products namely ESL and UHT milk. In Indonesia, the halal status of a product is a crucial point for consumers who are predominantly by Moslems. In its processing, Indonesian Ulema Council (MUI) has a rule called HAS 23101, a halal certification requirement as an implementation of Halal Guarantee System. This study examines the importance of the Halal Assurance System criteria in milk production in the Indonesian food industry. The purpose of this study is to analyse the suitability of the halal guarantee system criteria for milk production in PT. Greenfields Indonesia based on HAS 23101 standards. This research is a descriptive observational study. The data collection process is carried out by providing a checklist to the halal management team in the food industry. The results obtained are the percentage of conformity to the 100% Halal Policy, 100% Halal Management Team, 100% Training and Education, 100% Material, 100% Product, 91.6% Product Facility, Written Procedure for Critical Activity 100%, 100% Search Capability, Products that do not meet 77.8% criteria, 100% internal audit, and 100% management review. The percentage of conformity results obtained as a whole was 97.2%.
