Abstract

Indonesia merupakan negara dengan angka kelahiran yang relatif tinggi, sehingga dibutuhkan alternatif dalam menurunkan jumlah penduduk seperti penggunaan alat kontrasepsi berbahan herbal. Penelitian bertujuan mengetahui pengaruh ekstrak etanol daun pepaya Calina terhadap kualitas sperma dan morfologi epididimis tikus wistar. Penelitian menggunakan 24 tikus wistar, dibagi menjadi 4 kelompok terdiri dari Kontrol (akuades), ekstrak dosis 100 mg/Kg BB, 200 mg/Kg BB, 300 mg/Kg BB yang diberikan menggunakan sonde lambung (1ml) selama 30 hari. Pada hari ke-31 tikus dibedah dan diambil cauda epididymis untuk dilakukan pengamatan kualitas sperma dan pembuatan preparat histologis dengan metode parafin (pewarnaan HE). Parameter kualitas sperma terdiri dari motilitas, viabilitas, jumlah, dan morfologi sperma. Parameter morfologi epididimis terdiri dari tebal sel epitel, diameter dan luas epididimis. Seluruh parameter dianalisis menggunakan uji one way Anova dan dilanjutkan uji Duncan dengan selang kepercayaan 95%. Hasil menunjukan pemberian ekstrak dosis 100-300 mg/Kg BB berbeda secara signifikan dibandingkan kontrol dengan menurunkan parameter motilitas, jumlah, viabilitas dan morfologi sperma (P<0,05). Morfologi epididimis menunjukan penurunan tebal epitel, diameter dan luas epididymis pada dosis 100-300 mg/kgBB dibandingkan dengan kontrol (P<0,05). Kesimpulan menunjukan ekstrak etanol daun pepaya Calina dosis 100 mg/kgBB memiliki potensi sebagai agen antispermatogenik ditandai dengan penurunan kualitas sperma dan morfologi epididymis tikus wistar.

Abstract

An in silico study was conducted to analyze the potential of Human Immunodeficiency Virus (HIV) glycoprotein 160 as a candidate source of B cell subunit (epitope) vaccine for Acquired Immunodeficiency Syndrome (AIDS). The method used was immunoinformatics. The tools used were a set of laptop device, uniprot application, IEDB AR application and ToxinPred application. The materials used were glycoprotein sequences. The analysis results showed that there were 4 candidate peptides for B cell subunit (epitope) vaccines for AIDS, namely MRVMGIQ (Methionine, Arginine, Valine, Methionine, Glycine, Isoleucine, Glutamine, NMWKNNM (Asparagine, Methionine, Tryptophan, Lysine, Asparagine, Asparagine, Methionine), EIMGDIR (Glutamate, Isoleucine, Methionine, Glycine, Aspartic Acid, Isoleucine, and Arginine) and MWQRVGQ (Methionine, Tryptophan, Glutamine, Arginine, Valine, Glycine and Glutamine). All peptides are non-toxic so they are safe for the body

Abstract

Industrialisasi produk dengan bahan nanoperak semakin meningkat. Salah satu metode sintesis nanoperak (AgNPs) yang ramah lingkungan adalah radiosintesis menggunakan sinar gamma. Tantangan lingkungan terkait industrialisasi AgNPs tidak hanya terkait sintesis dan produksi tetapi juga terkait penggunaan produk. Peningkatan penggunaan nanoperak juga berarti peningkatan limbah pemakaian produk dengan nanoperak yang terpapar ke ekosistem perairan. Tujuan penelitian ini adalah untuk mengetahui bagaimana pengaruh koloid nanoperak terstabilisasi alginat terhadap daya tetas telur dan struktur tulang ikan wader pari (Rasbora lateristriata, Bleeker, 1854). Koloid AgNPs disintesis menggunakan sinar gamma pada dosis 5 KGy, 10 KGy, 15 KGy, dan 20 KGy kemudian dikarakterisasi dengan UV-Vis. Telur ikan didedahkan dengan koloid AgNPs selama 24 jam. Embrio diamati pada umur 2 hari menggunakan mikroskop leica. Pewarnaan tulang dilakukan dengan pewarna Alizarin Red dan Alcian Blue pada ikan berusia 5 pekan. Terdapat penurunan daya tetas embrio ikan wader pari pada perlakuan AgNPs yang disitesis pada 5 KGy. Morfologi jantung pada larva megalami edema. Osifikasi pada bagian kranial ikan dengan perlakuan terlihat mengalami keterlambatan jika dibandingkan dengan normal. Terdapat penurunan laju jantung pada ikan perlakuan. Pendedahan AgNPs pada fase embrio ikan wader dapat memengaruhi sistem sirkulasi, struktur tulang dan daya tetas embrio ikan wader pari.

Abstract

Xylanases are hydrolytic enzymes which randomly cleave the β-1,4 backbone of the complex plant cell wall polysaccharide xylan. In this study, we investigate the extracellular enzyme activity of three strains of Bacillus altitudinis which produced with various production times in neutral (pH 7) and alkaline (pH 9) conditions. Crude enzyme was obtained from inoculated production medium containing 2% w/v xylan after six different time of incubations, including 0, 24, 48, 72, 96 and 120 hours.  The stationary phase of those three strains was observed after 72 hours of incubation, in accordance with significant increase of xylanase activity both in neutral and alkaline assay conditions. However, the highest of xylanase activity was obtained after 92 hours of incubation, in all three bacterial strains tested. A thin layer chromatography (TLC) was performed, and it confirmed that the crudes enzymes was able to breakdown the xylan into oligosaccharides. A higher activity of xylanase was obtained in alkaline condition but not significantly different between those two conditions. B. altitudinis KBX08 has the highest xylanase activity (46.9U/ml) which produced after 96 hours of incubation, indicating their potential for further development as xylanase producer.

Abstract

The immune response is divided into innate immune response and adaptive immune response. Macrophages are one of the immunity cells that work both in innate and adaptive immune response. The activities of secondary metabolites present in agarwood-producing plants allow the potential for natural immunomodulators so that the purpose of this study was to determine the potential G. versteegii extract leaves as immunomodulator and to examine the effect of G. versteegii leaves extract toward macrophage cells of mice (Mus musculus L.). The steps to be carried out in this study were the preparation of G. versteegii leaves extract with multi-level extraction, collection of M. musculus, cytotoxicity test with MTT assay, phagocytosis activity test of macrophages calculated by light microscopy (400X), and Griess assay for NO production of macrophages. The cytotoxicity test resulted an IC50 value of G. versteegii leaves extract was 183.45 µg/mL and the Imboost® as comparison solution is 20.12 µg/mL. This value used to determine the concentration of extract for phagocytosis activity test and NO production of macrophages test. The results showed that G. versteegii leaves extract has the potential as an immunomodulator and extract with 20 µg/mL concentration has the highest effect on phagocytosis activity and NO production.

Abstract

Andaliman (Zanthoxylum acanthopodium) plant, a unique spice in the Batak tribe limited only to the fruit. The leaves available most of the time and consisting of essential oils, there are no research has been conducted on molecular docking to determine the potential of the drug. The Covid-19 pandemic is still ongoing, so information as Corona antivirus is needed. This study aims to find the differences of chemical components from Andaliman leaves essential oil, classify the varieties, and get potential components as Corona antivirals based on molecular docking. The Andaliman leaves were collected from North Tapanuli District, than isolated using water distillation and Gas chromatography-mass spectrometry (GC-MS). Data analysis using Principal Component Analysis (PCA) with the XL stat 2020. The results showed that three varieties of Andaliman, Siholpu, Siganjangpat, and Variety three are not grouped in one quadrant. There are 53 compounds were detected. The dominant chemical compounds in all varieties are D-Limonene (Siganjangpat: 87.7%, Siholpu: 50.4%, and Variety three: 42.6%). Each variety is characterized by specific compounds, Siholpu by caryophyllene and sabinene; Singanjangpat by D-Limonene; Variety three by neryl acetate and 1,8-cinneol. The chemical component in Andaliman leaves that most potent as Corona antiviral is beta-caryophyllene in the Siholpu variety.

Abstract

Jahe merupakan tanaman rempah-rempah yang berpotensi sebagai imunomodulator untuk meningkatkan sistem kekebalan tubuh manusia. Senyawa gingerol dilaporkan memiliki aktivitas imunomodulator dan imunosupresan. Informasi mengenai keragaman stomata genotipe lokal jahe merah diperlukan untuk melengkapi deskripsi genotipe jahe merah yang digunakan sebagai dasar kegiatan pemuliaan tanaman jahe merah. Observasi morfologi daun dilakukan dengan mengamati jumlah, panjang, dan lebar stomata, serta jumlah kloroplas. Data yang diperoleh dianalisis dengan menggunakan peranti lunak IBM SPSS versi 25 dan PAST 3.25. Hasil penelitian menunjukkan bahwa dari 14 genotipe jahe lokal koleksi pada 4 karakter pengamatan sitologi menunjukkan bahwa rerata panjang stomata, lebar stomata, dan jumlah kloroplas berbeda nyata.  Sementara karakter rerata jumlah stomata tidak berbeda nyata antar genotipe jahe merah. Analisis lanjut dilakukan dengan metode DMRT, terlihat bahwa panjang stomata pada genotipe Ziof-110 (60,1µm) berbeda nyata dengan Ziof-68 (66,3 µm). Pada karakter lebar stomata secara statistik Ziof-108 (66,7 µm) berbeda nyata dengan Ziof-69 (70,3 µm) dan Ziof-70 (69 µm). Karakter jumlah kloroplas Ziof-72 (30) berbeda nyata dengan Ziof-13 (46) dan Ziof-115 (43). Dendogram kekerabatan dan analisis komponen utama membagi 14 genotipe jahe merah lokal menjadi 2 kelompok. lebar stomata dan jumlah kloroplas merupakan karakter yang berkontribusi terhadap keragaman jahe merah dengan nilai PC1 0,70 dan PC2 0,73. Komponen utama 1 dan 2 memiliki nilai eigenvalue > 1 dan dapat menjelaskan keragaman total jahe merah sebesar 65,24%.

Abstract

Daphnia sp. is one of the most important natural feeds in freshwater fish farming sector. Daphnia contain considerably high protein, easy to be cultivated, short harvest time, the size is appropriate to the fish's mouth size, its movement can stimulate fish larvae to eat it, does not pollute the aquatic environment or larval rearing media and can be enriched with certain ingredients. Carbon is a source of energy for the growth of Daphnia directly or through microorganisms that become feed for Daphnia. This study aimed to analyze the effect of providing vegetable waste as a carbon source on the abundance of Daphnia and determine the best type of vegetable waste for the cultivation of Daphnia. The research method was an experimental method using 3 (three) different vegetables, namely spinach, mustard greens, and cabbage. The study was conducted using a completely randomized design with 4 (four) treatments in the form of different vegetables and control and the test sample was repeated 3 (three) times. The growth of Daphnia was observed for 16 days of cultivation time. The results showed that the provision of vegetables affected the population growth of Daphnia. Spinach (P3) was the treatment with the highest growth value, which average growth was sebanyak 96,67 individual (ind) of Daphnia with a specific growth rate of 0.055%. The temperature in Daphnia culture media generally ranged from 25-27℃, while the pH in Daphnia culture media showed a value of 7.4-9.5. The results of PCA analysis showed that water temperature was a variable that was strongly correlated with the growth of Daphnia.

Abstract

The Paradisaea minor jobiensis, Rothcshild 1897 is one of the most beautiful Cenderawasih birds in Papua, especially in the Yapen Islands and is a species currently threatened with extinction. The purpose of this study is to find out the population of P. m. jobiensis in the forest area of Imbowiari Barawai. Observation of population P. m. jobiensis is done with Demographic Parameters through direct observation using IPA method and then analyzed quantitatively parametrically to see population size, density and sex ratio. The population of Cenderawasih bird in the forest area of Imbowiari Barawai is 188 individuals, of which males are 136 individuals and females are 82 individuals. The population of Cenderawasih Birds in the Imbowiari forest is still very abundant, where the males are moro dominant than females.

Abstract

Pandanus tectorius Park., tree of life, which almost all of its parts are used by humans. Industrial fiber made from screw pine leaves has been successfully exported to several countries, while the fruit is consumed as an alternative staple food. Fruit is only produced by female plants, because screw pine is dioecious plant. Farmers obtain screw pine seedling using stem cuttings from female trees, but the number of seedling produced is limited and this technique can damage the plant. Alternative seedling production using seeds has constraints in the form of uncertainty over the sex of the seedling. One of the best ways to provide screw pine seedling is to use a shoot-tip culture technique. Shoot-tip culture is able to reproduce plants efficiently and quickly and does not damage the plant. Seedling will also have the same genetic traits as the selected plant. However, efforts to develop an efficient and fast screw pine shoot-tip culture protocol have not been carried out massively. The induction and shoot multiplication stages still had a low success rate. Another problem in seedling production through tissue culture is the certainty seedlings are true-to-type. This review article aims to reveal the potential of shoot-tip culture for in vitro screw pine seedling production and to test the genetic stability of the screw pine seedling.
