Abstract

Cryptosporidiosis is a disease caused by Cryptosporidium spp. protozoan parasites and are zoonotic. Cryptosporidium canis is the main species that infects dogs. Transmission of C. canis in dogs to humans is possible. This study aims to detect microscopic C. canis infection based on morphology and molecularity using the Polymerase Chain Reaction (PCR) in dogs in Surabaya City. A total of 80 diarrhea dog feces samples were taken from Animal Hospitals and animal clinics in several areas in the Surabaya City, then added potassium dichromate and stored at 4OC. Detection was made of the presence of Cryptosporidium spp. oocysts microscopically which is then confirmed by molecular examination using the PCR method. The results showed 40 positive samples containing Cryptosporidium spp., oocysts, with a size of 2-6 ?m. Ten samples from the total positive sample of Cryptosporidium spp. oocysts by microscopic examination, with the PCR test there were seven positive samples of C. canis. Based on the results of the study it can be concluded that the species that causes Cryptosporidiosis in dogs in Surabaya City is C. canis. The high cases of Cryptosporidiosis in dogs can be a warning to be able to prevent Cryptosporidium spp. infections, especially in pets that have the potential as a reservoir in spreading disease.

Abstract

The purpose of this study was to determine the analysis of twin dairy cattle business in Lembang District, Bandung Regency, West Java. The study used a metoda survey of 25 farmers in the Corner Village and 25 in Sunterjaya Village, Lembang District, Bandung Regency by using interviews and filling in the questionnaire. Primary data and secondary data are then analyzed. The results of the study show that, the average ownership of dairy cows is 4.40 head/farmers. Net income of farmers is IDR 45,591 million/year, equivalent to 3.79 million/ month, B/C 1.78. The While in Sunterjaya Village has 5.28 head/farmer. Net income of farmers is IDE. 38,808 million/year,white to 3.32 million/year, B/C 1.77. Dairy business with a scale of 5-10 head/ farmer, economic value will be proportional to the cost and labor spent for one year. The difference in calf sale value of IB-TE and IB results in B/C ratio > 1, economically the twin calves of IB-TE results are feasible to be maintained and maintained.

Abstract

This study aims to identify morphology and molecular of Blastocystis infection in pigs in the Tabanan and Badung District, Bali Province, Indonesia. 100 samples of pig stool were taken, fifty samples from the Tabanan and fifty samples from Badung District. Blastocystis sp. isolated and identified microscopically and confirmed by polymerase chain reaction (PCR) as a molecular identification method. 63% (63/100) Blastocystis is found in pig feces from these two District by microscopic detection. Positive samples were found 27 samples from Badung and 36 samples from Tabanan. Five samples were taken from each District conducted a confirmation test using the PCR method and found 60% (6/10) positive samples, two samples from Badung and four samples from Tabanan. The results of this study indicate that in healthy pigs, they also contain parasites that have the potential to spread disease to humans. Therefore, the high incidence of Blastocystis in pigs can be an alarm or warning to prevent Blastocystis infection from livestock that can potentially to be a reservoir in the spread of disease.

Abstract

Toxoplasmosis is a zoonotic disease and known as Food borne diseases as main transmission pathway in Indonesia. Regular monitoring of animal health with surveillance is very important and requires a diagnostic devices that have good accuracy and continuous availability at affordable costs. Serological testing techniques with agglutination method are suitable and appropriate choices because they are quite accurate and simple in their application and does not require skilled human resources. Aims of study aims are to develops an agglutination test kit and evaluate its performance comparatively with several commercial kits. 98 samples of positive and negative standard serum that have been confirmed by using 4 commercial serological kits were used to evaluate the inhouse-toxoplasma modified agglutination kit (ToMAT). Furthermore 186 field serum samples were used in inter reliability agreement assay for comparing the ToMAT kit with four commercial kits available in Indonesian market’s and immunoblotting. Performance evaluation of the ToMAT shows the sensitivity, specificity and diagnostic accuracy, were 98.55%; 86.21% and 94.9% respectively. The value of PPV and NPV is 94.4%; 96.2%. The ToMAT test kit had a good agreement with immunoblotting (76%) and the Toxotest ELISA kit (74.1%) was based on the value of Gwet’s AC1.

Abstract

Ovarian cysts are a disorder characterized by the growth of smooth muscle cells in the ovaries forming like fluid-filled balloons. Oviductal Glycoprotein (OVGP1) is secreted in the oviduct and plays a role in early embryonic development. The incidence of ovarian cysts is thought to affect the level of expression of the OVGP1 gene. The purpose of this study was to determine the potent role of OVGP1 gene as biomarker for detection of ovarian cyst on goats by Polymerase Chain Reaction (PCR) method. The samples used were DNA isolated from normal female PE goat oviduct and goat with follicular ovarian cyst obtained from RPH. The OVGP1 gene identification was performed by PCR method using 5'GACTGCAACCCACACAAGGA-3 forward primer (OVN1 _F) and reverse primer (OVN1_R) 5'GAGAGCAGCACCCACTTTTC-3 ‘. The sequencing of PCR results is done by Sanger method. The gene sequence analysis used the Bioedit and NCBI BLAST programs by comparing the PCR sequence on the follicular ovarian cyst and the normal ovary with the OVGP1 gene of the gene bank NM_001285584.1. The results showed that two normal samples and two samples of follicular cysts had similarities above 90% with OVGP1 gene bank NM_001285584.1. Samples with follicular ovarian cysts indicate a change in nucleotide base sequence due to deletions, insertions, transitions and transversions that affect the amino acid sequence. The arrangement of amino acids on samples with follicular ovarian cysts found an increase in the number of proline (P), threonine (T) and serine (S). In conclusion there is a difference in OVGP1 gene sequences in sample follicular ovarian cysts compared with sample normal ovaries . So the OVGP1 gene potentially can be used as a marker of follicular ovarian cysts.

Abstract

This study aims to describe the anatomical differences in the gills of the thai mahseer’s (Tor tambroides), carp (Cyprinus carpio) and tilapia (Oreochromis niloticus). The stages of this study include the preparation of test fish, preparation of gill, documentation, identification, and analysis. This study used five fish for each type of fish with total weight ranged from 500-1000 g and length 28 to 40 cm. Analysis of each gill section was carried out morphology and morphometric. Morphometric data on the arcus branchialis section observed included the ratio of branchial arcus length to total length, on the part of the branchial filament including the average number of branchialis filaments per branchial arcus, density of branchialis filaments (filaments/cm), and length ratio of branchial filaments with the length of the branchial arcus, while in the branchiospinal part includes the average number of branchiospinalis per branchialis arcus and density of branchiospinalis. Result showed that morphologically, the anatomical differences in keureling gills, carp and tilapia are located in the form of branchiostegal and branchiospinalis membranes. Anatomical differences were seen in relative gill weight, branchial arcus length ratio, branchiospinal density, average number of branchial filaments, average density of branchialis filaments and average number of branchiospinalis. Carp have higher relative weight values of gills compared to tilapia and keureling fish which are 3.73 ± 0.43%, 2.82 ± 0.64% and 1.92 ± 0.55%, respectively. Tilapia has the shape and size of gills that are more developed in supporting the performance of the respiratory system than carp and keureling fish.

Abstract

Staphylococcus aureus is one of the normal flora that can cause infection in injured skin. Resistance to antibiotics has an impact on the difficulty of therapeutic treatment so that other alternatives are needed. The purpose of this study was to observe the effectiveness of apple vinegar as an antimicrobial against Multidrug Resistant Staphylococcus aureus isolated from infection wounds in dogs in Surabaya. The methods in this study were the isolation of bacteria from 30 samples of dog festering wounds on Manitol Salt Agar (MSA) media and identification through macroscopic, microscopic, catalase tests, coagulase tests, hemolysis tests on Blood Agar media, and Voges–Proskauer (VP) tests. Bacteria that have included the S. aureus criteria were followed by sensitivity tests to the antibiotics Amoxycillin, Ampicillin, Gentamicin, Chloramphenichol, and Ciprofloxacin. Apple vinegar activity test was carried out using disk diffusion method against Multidrug Resistant Staphylococcus aureus. The results showed that of the seven S. aureus isolates, there were two isolates belonging to the Multidrug Resistant S. aureus. The results of the apple vinegar activity test showed the presence of antimicrobial activity shown by the formation of a clear zone around the paper disk with an average diameter of 24.06 mm at a concentration of 90%. The conclusion shows that apple vinegar has antimicrobial activity against Multidrug Resistant S. aureus which is isolated from dog festering wounds in Surabaya.
