Abstract

Common palm civets (Paradoxurushermaphroditus) have a potential to be commercial commodityanimal which produce best coffee, parfume, but also as reservoir potential infection desease such as SevereAcute Respiratory Syndrome (SARS). Therefore, their health status is very important. This study wasaimed to discribe the leukocyte profile, and stress index of 4 males Java common pal civets and 4 females.Blood sampling was taken from vena femoralis and done at weeks 1st, 5th, 6th, and 7th. Leukocyte profilewere examined using haemocytometerand blood smear. The average leukocyte count of males and femalescommon palm civets per mm3 were (3.33±0.86)x103 and (2.83±0.70)x103, neutrophil were (1.01±0.47)x103and (0.68±0.30)x103, eosinophil were (0.16±0.18)x103 and (0.04±0.05)x103, lymphocyte were (2.06±0.42)x103and (2.05±0.59)x103, monocyte were (0.09±0.06)x103 and (0.06±0.06)x103. Stress index were (0.49±0.18)for males and (0.37±0.22) for females. Generally, leukocyte count of males Java common palm civet werehigher than those of females.

Abstract

Formaldehyde is one of the exogenous free radicals. During formaldehyde exposure, there will be morefree radical or reactive oxygen compound formed through electron transport chain. Excessive reactiveoxygen compound triggers the process of lipid peroxide reaction in the membrane of spermatozoa cell. Theaims of this study were to find out that formaldehyde exposure caused abnormalities in the number of thenumber of spermatogonium type A, spermatogonium type Pachytene, 7th spermatid and 16th spermatid inspermatogenesis process.This study was experimentally and randomly pretested-posttested- with controlgroup design. The samples of this study were adult male mice strain Balb-C (age 2-3 months) with thefollowing criteria: body weight between 22-25 grams and healthy. Randomly, 48 mice were divided intothree groups, were control group, the first treatment group and the second treatment group. Prior to thetreatment, were taken a half from each group for the pre-test, by preparing microscopic preparation testicleand examination was performed to the total spermatogenic cells. The rests of the mice were used as posttestexamination after 35 days treatment. On the 36th day, all the rest mice were necropsied for microscopictesticle preparation. The result of this study showed that the formaldehyde exposure caused significantlydecrease in the number of spermatogenic cells (p<0,05), the average of type A spermatogonium cells atcontrol group, 1st treatment group and 2nd treatment group were 39,90±0,51; 20,42±0,72; 15,65±0,88respectively; spermatogonium type Pachytene were 48,47±1,28; 32,60±3,06; 23,14±3,16 respectively; 7thspermatid were 97,47±5,28; 39,98±4,28; 30,36±2,96 respectively and16th spermatid were 73,08±4,05;21,70±1,70; 16,38±1 respectively.It can be concluded that the formaldehyde exposure decreased the amountof spermatogenic cells. The result of this study is expected to be used as the baseline for further study inorder to measure formaldehyde content as a free radical in testicle .

Abstract

As chromosome of different species differs in the size, shape, and number, their caryotpes areexpected to provide a distinctive and characteristic feature of the organism. The caryotypes ofseven amphibian species : Limnonectes gruniens, L modestus, Rana (Hylarana) chalconota,Fajervarya cancrivora, Polypedates celebensis, Bufo celebensis and B biporcatus of South EastSulawesi-origin were investigated. The result showed that the number of diploid chromosomes inthe seven anuran varied between 22-26. The highest diploid chromosomes were observed in Rchalconota, F cancrivora (26), followed by L gruniens, L modestus (24) and the lowest were observed22 in P celebensis, B celebensis and B biporcatus (22). In general, all the the seven anuran sharesfour metacentric chromosomes pairs which is in pairs No 5, 6, 9, and 10 respectively. Howeverdifferences were observed in other chromosomes pairs numbers among the seven species.

Abstract

The purpose of this study  was to find out the effect of lime total acids  on the characteristics ofintestine broiler base on its pH, digestive  viscosity, and the number and hight of small intestineof villi. Atotal of120 male Lohmann broiler chicks with the average body weight of 45.29 ± 1.76g, were used in thisstudy which were then allotted into six groups. The six treatment groups consist of : animals feed with noadditional acid (PO) as negative control, animals feed with addition of  0.4% (w/w) commercial citric acid(P1) as positive control, and animals feed with additional of 0.2% (P2), 0.4% (P3), 0.6% (P4), and 0.8% (P5)total acids extracted from lime, respectively.   This study was carried out using Completely RandomizedDesign with four replications.  The results showed that lime total acids had a very significant effect(P<0.01) on the  pH and digestive viscosity.  However, no significant effect (P>0.05) was found on thenumber and height of the villi. The addition of 0.8% total acid on the animals fed give the best result on theintestinal characteristics compared to other treatments.

Abstract

The use of appropriate diluent is intended to ensure the physical and chemical requirements ofspermatozoa so that the quality, particularly capacitation can be maintained. This study aimed todetermine the quality of Limousin cattle sperm after sexing with density percoll centrifugation methodusing Andromed diluent and Cauda epididymal Plasma 2 (CEP-2) plus 10% egg yolk. Observations ofspermatozoa quality including : motility, viability, percentage of abnormal, concentrations, membraneintegrity and state of capacitation. The semen used were obtained from Limousin cattle at the Center forArtificial Insemination(BBIB) Singosari Malang, with motility criteria e” 70%. Data were analysed usinga paired T-test in order to determine differences in the use of thinning treatments at each layer. Theresults showed that both diluent, andromed and CEP-2 diluent plus 10% egg yolk were able to maintainthe quality of spermatozoa either in the upper and lower layers

Abstract

Serological test is one of diagnostic method to detect pathogenicity brucella. Several methods are being improving such as Rose Bengal Test (RBT), Complement Fixation Test (CFT) dan Enzyme-linked Immunosorbent Assay (ELISA). Immunostick has an accuracy equivalent to ELISA and is easy to apply in the field so it is possible to be applied as a rapid test for brucellosis detection. The study aim was to know sensitivity and specificity of immunostick that were used to detecte antibody Brucella abortus using commercial antigens of B. abortus Strain 19 (S19) and B. abortus Strain 99 (S99). The test have compared with ELISA. The tests were conducted in two stages, namely (i) immunostick ability to detect antibodies in seropositive and seronegative serum, and (ii) the immunostick result were compared with ELISA result in serum grup that were be know and unknown status. A total of 250 serums were examined and result indicated that immunostick can be detect B. abortus antibodies in cattle serum with sensitivity 100%. Immunostick specifity were 45,45% for B. abortus S99(1) antigen; 78,79% for B. abortus S99(2) antigen and 51,52% for B. abortus S19 antigen. When the test compared with ELISA, the sensitivity 82,86% and the spesifity were 52,31% for B. abortus S99(1) antigen; 93,54% and 79,71 for B. abortus S99(2) antigen and 82,86% and 58,46% for B. abortus S19 antigen.

Abstract

This research aimed to study the effect of curcumin supplementation and red light exposure to the growth of magelang ducklings. The research design used complete randomized design that divided into four groups and each group consists of five ducklings, they were ducklings from ducks which was not given curcumin and exposed to white light (control), ducklings from ducks which was not given curcumin and exposed to red light (P1), ducklings from ducks was given curcumin 18 mg/head/day and white light (P2) and ducklings from ducks was given curcumin 18 mg/head/ day and red light (P3). The curcumin that given to the duck were mixed with feed as a supplement. The variables of this research were the body weight and morphometry that consisted of the chest perimeter, abdominal perimeter, and fur necklace (the white fur that surounded the neck) width . The collected data were analized with Kruskal Wallis non parametric test and would be further tested with Mann Whitney-U test and linear regression. The result of this research showed that curcumin supplementation and red light exposure to ducks affected the body weight, chest perimeter, abdominal perimeter, and fur necklace width of magelang ducklings. We concluded that 18 mg curcumin supplementation and red light exposure increased ducklings quality, we hope 18 mg curcumin supplementation and red light exposure can be applied to increase ducklings quality, futhermore the ducklings have better productivity.

Abstract

Brucellosis is one of the constraints in development of animal husbandry in Indonesia and is thought to have a huge economic impact. This study aimed to calculate the national economic losses cause by brucellosis in livestock in Indonesia. The method of calculating the estimated economic losses used two ways, by calculating the direct losses and by calculating the price of economic externalities. Source of secondary data for the calculation derived from scientific publications, government data, expert opinions, market data, and assumptions that are based on expert discussions. The results showed that by the direct calculation method it is estimated the economic losses at approximately Rp. 3,516,401,986,082 per year whilst by the second method the losses is estimated at Rp. 3,637,773,925,000 per year. Both methods of calculation used give relative similar economic losses estimation. The total loss due to brucellosis is estimated to reach 1.8% of the total value of livestock assets in Indonesia.

Abstract

The objective of this study was to investigate the effect of transport 12 hours on haematological and some biochemical responses of local Indonesian sheeps. Ten adults (2.0-2.5 years) male local Indonesian sheeps BW adult 30-40 kg were transported by L-300 pickup truck starting at 3:00 pm to 3:00 am on the d 2 without stopping. Jugular blood were collecte-d at 7 am (-8 h pre- transportation as baseline), 15:00 immadiately after (0 h), 19:00 (4 h), 23:00 (8 h) , 03.00 (12 h ), and at 11.00 d 2 (+8 h) post- transportation as recovery period). Under the conditions of the present study, transport of local Indonesian sheeps significantly increased glucose during 4 h transport (P <0.05), increased eosinophil at 0 h (P <0.05) and no significant changed in erythrocytes, haemoglobin , monocytes, neutrophils, lymphocytes, leukocytes, packed cell volume (PCV), and total plasma protein (TPP) (P> 0.05). Our findings showed that transportation 12 h affected eosinophil during 0 h and glucose metabolism during 4 h of transportation, but did not any affect on some haematology and dehydration responses in local Indonesian sheeps.

Abstract

This study was carried out to determine the use of bird flu polyvalent vaccines containing two or threeor more virus isolates representating of circulating viruses in the region. Three seed isolates of avianinfluenza H5N1 virus were used in this experiment. The isolates were Chicken/Denpasar/Unud-01/2004,Chicken/Klungkung/Unud-12/2006, and Chicken/Jembrana/Unud-17/2006. The seeds were inactivatedusing 0.01% formaldehide than mixed (AI3G) alumunium hidroxide adjuvant and then injectedintramuscularly to Isa Brown layer chicken at 3 weeks of age and repeated at the age of 5 weeks. The doseof each seed virus was 27 HA units. Sera were collected at one and two weeks after the second vaccination.The result showed that the arithmetic meant titer (AMT) of sera that tested with homologous isolate washigher than the test using a heterologous isolates, in the standard haemaglutination inhibition (HI) assay.The mixed AI3G vaccine produced a uniform AMT against the constituent isolates, while vaccines withindividual isolate yielded a lower and more variation in AMT. Further experiments using a commercialhomologous H5N1 and heterologous H5N2 commercial vaccines has resulted AMT that 1-4 log lower thanAI3G vaccine. It is concluded that polyvalent vaccine with field seed isolates is recommended to be appliedin the poultry farm in Indonesia.
