Abstract

The objective of this study was to determine the anatomical morphology and histology of the intestineof water monitor (Varanussalvator). Intestine organ from an adult male water monito was used in thisstudy. Perfusion was performed to flow the 4% paraformaldehyde fixative solution through the heart.Intestine was collected, then divided into six sections, followed with dipping them into 4% paraformaldehydesolution for four days, continued histological preparations and stained with haematoxylin eosin (HE). Theanatomical morphology observation was performed before the intestine was processed into histologicalpreparations by observing the outer shape of the intestine and measuring it’slength. Morphologicalobservation was made †after processing theintestinal tissue by observing each portion of the intestinalwall structure. The results showed that transverse fold were found on the outer and inner surfaces of theintestine section I-IV, while in the V-VI part of the intestine, the folds were not observed anymore. Theintestine wall consisted of four layers, such as mucosa, submucosa, tunica muscularis, and serosa. Thevilli on intestine section I-IV were relatively longer and higher, while the intestine vili in section V-VI wererelatively short. The entire mucosa in water monitor was coated with absorptive columnar epitheliumwith goblet cells. Based on the result study, it can be concluded that the water monitor has more efficientdigestive process which can be seen morphologically from the relatively short intestinal, has transversefolds on the surface of the small intestine, and does not have cecum. The intestinal wall consists of layersi.e : mucosa, submucosa, tunica muscularis, and serosa.

Abstract

The research was aimed to evaluate the combination effect of inulin dahlia tuber and Lactobacillus spdevelopment micro flora on intestinal and productivity of crossbred local chicken. The present researchwas assigned in a completely randomized design with factorial pattern, 2x3. The first factor was two levelsof dahlia tuber powder as inulin source 0.8% (A1) and 1.2% (A2), and, the second factor was three levels ofLactobacillus sp namely, without Lactobacillus sp (B0), 1.2 mL (108 cfu/mL) (B1) and 2.4 mL (108 cfu/mL)(B2). Each treatment combination was replicated four times with seven birds each. Parameters observedwere total of lactic acid bacteria (BAL), Escherichia coli, pH and body weight gain. The results showed thatfeeding inulin dahlia tubers at 1.2% and Lactobacillus sp at 1.2 mL (108 cfu/mL) sp either in partial orinteraction significantly (P<0.05) affected the total LAB, E. coli, pH and body weight gain. In conclusion,combination of inulin from dahlia tubers and Lactobacillus can increase total LAB and body weight gain,decrease total E. coli and pH.

Abstract

The aim of this research is to observe blood profile of domestic cats which includes red blood cell, white blood cell, haemoglobin, haematocrit, and trombocyte during skin graft recovery period via autograft. A total of nine male domestic cats (Felis catus) aged 1-2 years weighting 3-4 kg were separated into three treatment groups. Hair shaving and disinfectant application were done on lateral area of front leg, and then 2x2 cm incision was made. The wound was wrapped by sterile gauze dampened by poviderm iodine and left for different period of days per treatment group; where Group I (two days), Group II (four days), and Group III (six days). Transplantation was done by taking the skin on thorax area and placing it on the recipient after first cleaning subcutaneous tissue from the skin surface of donor’s wound and the base of recipient’s wound. Blood sample was taken from vena cephalica antebrachii anterior on day 0 before skin graft, on day three, six, nine, 12, and 18 after skin graft. Based on the result, significant difference waso bserved from red blood cell count, white blood cell count, haemoglobin count, and hematocrit, whereas trombocyte count did not show any significant difference between treatment groups. The cats on Group II and Group III were systemic good condition compared to Group I. ABSTRAK Penelitian ini bertujuan melihat profil darah kucing lokal, meliputi sel darah merah, sel darah putih, hemoglobin, hematokrit, dan trombosit selama kesembuhan tranplantasi kulit secara autograft. Sebanyak sembilan ekor kucing lokal (Felix catus) jantan umur 1-2 tahun dengan bobot badan 3-4 kg dibagi menjadi tiga kelompok perlakuan. Pada kucing tersebut dilakukan pencukuran rambut dan desinfeksi pada area kaki depan sisi lateral, selanjutnya dibuat luka sayat/incisi 2x2 cm. Luka dibalut dengan kasa steril yang dibasahi povidone iodine dan dibiarkan selama beberapa hari. Kucing dengan sayatan yang dibiarkan tersebut dikelompokkan menjadi kelompok I (dua hari), kelompok II (empat hari), dan kelompok III (enam hari). Transplantasi dilakukan dengan mengambil kulit dari area thorak dan ditempatkan pada resipien dengan terlebih dahulu permukaan luka kulit donor dan dasar luka resipien dibersihkan dari jaringan subkutis. Pengambilan sampel darah dilakukan melalui vena cephalica antebrachii anterior pada hari ke-0, 3, 6, 9, 12, dan 18. Berdasarkan hasil penelitian terjadi perbedaan yang signifikan (P<0,05) terhadap jumlah sel darah merah, sel darah putih, hemoglobin, dan hematokrit, sedangkan jumlah trombosit tidak menunjukkan perbedaan yang signifikan (P>0,05) di antara kelompok perlakuan. Kondisi sistemik tubuh kucing kelompok II dan III lebih baik dibandingan kelompok I.

Abstract

This study was conducted to determineSDS concentration also to compare Tris egg yolk and citrateegg yolk on the quality of ettawa grade chilled semen. The study consist of two experiments. The firstexperiment was to determine the best SDS concentration in Tris egg yolk diluents and the second experimentwas to compare the SDS suplementation in tris and citrate egg yolk in the quality of ettawa grade chilledsemen. The semen were collected from three bucks, immediately after collection the semen evalutedmacro and microscopycally. Semen demonstrate more than 70% motility and less than 10% spermatozoaabnormality, individually divided into four tube, each tube was diluted in egg yolk supplemented with 0;0,025; 0,05; and 0.075% SDS then stored at 5°C for 72 hours. The quality of chilled semen was observefor motility and viability every 12 hours. Result demonstrated that 0.05% SDS was the best concentrationcompared to others. In the second experiment,the semen were diluted in four different diluents, it wereTris egg yolk (TEY), Tris egg yolk with 0.05% SDS (TEYS), citrate egg yolk (CEY) and citrate egg yolk with0.05% SDS (CEYS). Result demonstrated that the mean value of motility and viability of spermatozoa inTEYSdiluents (67.08 ± 6.43% and77.07 ± 6.78%) was higher (p<0.05) than CEYS (60.42 ± 9.05% and72.31± 7.45%), TEY (59.23 ± 9.41% and71.21 ± 8.56%) and CEY (53.45 ± 11.33% and67.74 ± 8.90%). In conclusion,supplementation of 0.05% SDS in TEY was maintained best sperm quality compared to other diluentsduring preservation.

Abstract

Hot water extract of Spirulina platensis which is contain of lipopolysaccharide can be used as animmunostimulant. This study aimed to determine the effect of hot water extract of S. platensis throughinjection on the non-specific immune response of Osphronemusgouramy. The treatment was given throughintraperitoneal injection of hot water extract of S. platensis with doses of 1%, 10%, 20% and PBS, PhosphateBuffer Saline (control) 0.1 ml/fish. Fish reared in the aquarium (67 l volune) with density 12 fishes/aquarium with average weight of 7.5 – 8.5 g/fish. Blood samples of fish taken from the caudal vein on day0 (before administration of the extract), 3 and 5 days after administration of the extract for the measurementof total leucocytes, differential leucocytes and phagocytic activity. Results showed an increased in totalleucocytes, phagocytic activity, monocytes and neutrophils of fish that given hot water extract of S. platensis.The addition of hot water extract of S. platensis at 1% through injection gave total leucocytes, phagocyticactivity, monocytes and neutrophils on days 3 were 543.73 x 103sel/mm3,69.30%, 34.33% and 14%respectively higher and significantly different to control.

Abstract

Knowledge and information about the age of puberty in muntjac (Muntiacus muntjac muntjac) isindispensable to the interests of females breeding in conservation efforts. The aims of this study were todetermine the age of puberty and age at first mated females muntjac kept in captivity through the analysisof estrogen and progesterone metabolites in feces. This study used 155 fecal samples that were collectedfrom three female muntjacs. Sample collection was began when muntjac aged three months, four monthsand six months. Total of 10-20 g fecal samples were collected every 2-4 days. Analysis of steroid hormonemetabolites was performed by using enzyme immunoassay (EIA) method with specific antibodies.Determination of the age of puberty was based on the appearance of the first time estrus and ovulation,which was indicated by the appearance of the highest estrogens secretion, on hormone metabolites profile.Hormone metabolites data then were tabulated in the average and standard deviations were presentedwith graphs and analyzed descriptively. The results showed that the age of puberty detection based onanalysis of the estrogens and progesterone metabolite in the feces can be applied in muntjac. Muntjacfemales kept in captivity flats reached puberty at age 5±1 month or 4-6 months range. It is recommendedthe first mated in the muntjac is at least after the female experienced two period of oestrous or has reachedat age of six months.

Abstract

Tartrate resistant alkaline phosphatase 5b (TRACP5b) is secreted by osteoclasts during bonedifferentiation and resorption. The objective of the research was to study TRAP5b expression inovariectomized Wistar rat consuming the combinations of calcitriol and raloxifene supplementation foreight weeks. Twenty five female Wistar rats aged eight weeks were randomly divided into five groups:normal control (NK), ovariectomy control (OVK), ovariectomy+calcitriol supplementation (OVD),ovariectomy+ raloxifene supplementation (OVR), and ovariectomy+calcitriol+ raloxifene supplementation(OVDR). At the end of the treatment, blood samples were taken from plexus orbitalis medialis forestrogen analysis. All rats were euthanized, the uteri were taken and weighed. Left femur was taken forhistopatological examination and immunohistochemistry TRAP5b using monoclonal antibody anti TRAP5bwhich was detected with streptavidin-biotin. The results showed that estrogen levels of the rats in OVKgroup were significantly decreased compared to the rats in NK group, meanwhile estrogen levels in the OVDR rat group were significantly decreased compared to the NK and OVK rat group. Histopathologicalobservation of distal femur epiphysis in group NK showed normal structure, meanwhile, distal femurepiphysis in OVK group was found osteoporosis, with some abnormalities, such as: increased of bonemarrow space, domination of adipocytes in the bone marrow, and decrease of trabecular bone speculum inepiphysis. Histopathological findings of distal femur epiphysis in OVDR group were increased of trabecularbone speculum in epiphysis and the domination of adipocytes in the bone marrow of epiphysis.Immunohistochemistry of distal femur epiphysis in OVDR group revealed increasing tartrate resistantalkaline phosphatase 5b (TRAP5b) expression in trabecular bone, which was located in bone marrow spaceand trabecular speculum surface as well. It can be concluded that the combination of calcitriol and raloxifenesupplementation in ovariectomized rats aged 8 weeks increases TRACP5b expression as a marker for theincrease of osteoclast acitivity in bone resorption, which may lead to the formation of the bone.

Abstract

This research objective was to analyze the effect on stocking density on performance, blood profile’s and meat quality of crossed kampung broiler chicken. Ninety day old chick of crossed kampung broiler chickens were maintained in three cages that had same size of 1x1 m2. Commercial feed was given for the chickens’ feeding. Productivity parameter was recorded since day old chick to 12 week. As many as 30% of the total chickens were taken randomly and used as samples to analyze the blood profile in 10 week old and another 30% of the chickens aged 12 weeks were also taken randomly and used to analyze the meat quality. This study used Randomized Complete Design with different cage density treatments: 8 heads/m2, 10 heads/m2, and 12 heads/m2. Each treatment was repeated three times. Result showed that different cage density (8 heads, 10 heads and 12 heads/cage) had no significant affect on performance (temperature humidity index, feed intake, water consumption, body weight, feed conversion, and mortality), blood profile’s (erythrocyte, leukocyte, hemoglobin, hematocrit, difenrensiasi leukocyte, heterofil/limfosit and glucose) and meat quality (physical, chemical and sensory). In conclusion, the stocking density does not effect on the performance, blood profile’s and meat quality of crossed kampung broiler chicken. The chickens can be maintained with density up to 12 heads/cage. Kampung broiler chicken aged nine weeks may reach higher growth than kampung chicken. Cholesterol meat of kampung broiler chicken is lower than the broiler and kampung chickens. ABSTRAK Penelitian ini bertujuan untuk menganalisis tingkat kepadatan kandang terhadap performans, profil darah dan kualitas daging pada ayam persilangan kampung broiler. Sebanyak 90 day old chick hasil persilangan ayam kampung dengan ayam broiler dipelihara di dalam petak kandang yang berukuran 1 x 1 m2. Ayam diberikan pakan ayam pedaging komersial. Pengambilan data performans dimulai pada umur satu hari sampai 12 minggu. Sebanyak 30% ayam diambil secara acak pada minggu ke-10 dari setiap petak kandang kemudian dilakukan pengujian profil darah. Sebanyak 30% ayam diambil secara acak pada minggu ke-12 untuk dikorbankan nyawanya dengan cara disembelih, kemudian dilakukan pengujian kualitas daging. Data dianalisis dengan sidik ragam. Perlakuan yang diberikan ialah kepadatan kandang, yaitu 8 ekor/m2, 10 ekor/m2 dan 12 ekor/m2. Hasil penelitian menunjukkan perbedaan tingkat kepadatan ayam kampung broiler dalam kandang tidak berpengaruh nyata terhadap performans (temperature humidity index, konsumsi pakan, konsumsi air minum, bobot badan, pertambahan bobot badan, konversi pakan dan mortalitas), profil darah (eritrosit, leukosit, hemoglobin, hematokrit, diferensiasi leukosit, heterofil/limfosit dan glukosa) dan kualitas daging (kimia, fisik dan organoleptik). Simpulan penelitian adalah kepadatan kandang tidak memengaruhi performans, profil darah, dan kualitas daging ayam silangan kampung broiler sehingga dapat dipelihara sampai pada kepadatan 12 ekor/m2 dan ayam tersebut tumbuh optimal sampai umur sembilan minggu dengan kadar kolesterol daging ayam kampung broiler lebih rendah dibandingkan ayam broiler dan ayam kampung.

Abstract

The objectives of this study to evaluate the effect of b-mercaptoethanol addition in maturation mediumon maturation and fertilization rate of sheep oocytes in vitro. Collected oocytes were then matured inmaturation medium without (control group) or with b-mercaptoethanol in concentration of 50 ?M or 100?M. There was a significant increase in the percentage of oocytes that reached the metaphase II (MII) stageafter cultured in maturation medium with b-mercaptoethanol either in concentration of 50 ?M or 100 ?M(p<0.05) (62.67%; 77.78%; 82.05% for control, 50 ?M, and 100 ?M group respectively). However no differencewas found in maturation rate of those oocytes in 50 ?M and 100 ?M groups (p>0.05). When maturedoocytes were fertilized, no difference was found on fertilization rate among the groups (P>0.05) (70.93%;77.50%; 69.14% for control, 50 ?M and 100 ?M group respectively). These data indicated that the additionof b-mercaptoethanol increased the percentage of sheep oocytes that reached the MII stage in vitro, but hadno effect on the fertilization rate.

Abstract

Bovine viral diarrhea virus (BVDV) is an important viral disease, which a ubiquitous pathogen ofcattle with worldwide economic importance and due to its misdiagnose with other viruses. The goal of thecurrent study was to identify and characterize of BVDV by reverse transcriptase polymerase chainreaction (RT-PCR) and followed by sequence genome analyses. Blood, feces, and semen samples werecollected from 588 selected cattle from animals suffering from diarrhea and respiratory manifestation. RTPCRresults showed that the 69 (11.74%) samples were positive to BVDV. Further molecularcharacterization was conducted only with 17 PCR positive samples. The results indicated the 17 IndonesianBVD virus isolates were belonging to the genotype-1 of BVDV (BVDV-1) based on sequence analysis anda phylogenetic relationship between Indonesian BVDV isolates and BVDV in the world. This finding is thefirst report of BVD-1 circulated in Indonesian cattle.
