Abstract

Background: Acanthus ilicifolius L. has antifungal potential and makes it possible to be a candidate for denture cleanser. This study aims to evaluate the effect of effervescent Acanthus ilicifolius L. extract on antifungal properties and flexural strength acrylic after immersion for three duration times.Method: Extracts were made by maceration method with ethanol. For the antifungal test, effervescent Acanthus ilicifolius L extract was prepared by combining the Acanthus ilicifolius L extract 4% and sodium perborate with two ratios, 1:7 and 1:14. The microdilution method was carried out. Effervescent Acanthus ilicifolius L extract 4% with ratio 1:7 was prepared for the flexural strength test. The fifty-four heat-cured acrylic resin (20x10x2.5mm) were fabricated and divided into 3 groups. Group 1 was immersed in aquadest (control); Group 2 was immersed in sodium perborate; Group 3 was immersed in effervescent Acanthous ilicifolius L. extract. Each group was immersed for 15 min, 8, and 56 hours (n=6). The flexural strength test was performed using Universal Testing Machine.Result: Effervescent Acanthus ilicifolius L. extract exhibited antifungal activity which increased in an effervescent Acanthus ilicifolius L extract-dependent manner significantly (0.57 to 0.50 OD). There were no significant differences between sodium perborate, effervescent_AI1 (1:7) and effervescent_AI2 (1:14). The flexural strength was found to be within the range of 66.1 - 68.3 MPa. There were no significant differences between groups (p < 0.05).Conclusion: Effervescent Acanthus ilicifolius L extract with the ratios 1:7 had a good antifungal effect and appropriate flexural strength after immersion for 15 min, 8, and 56 hours.

Abstract

Introduction: Cross-infection is the transfer of microorganisms that can occur from individual to individual or to object. Candida albicans is one of the infectious pathogens that can cause cross-infection in dentistry. One example is the manufacture of working models of gypsum material, which can cause cross-infection from patients to dentists or technicians, so it is necessary to control infection by means of    disinfection    of    type    III    gypsum     material     work models. Objective: To determine the effectiveness of soaking type III Gypsum in a 0.5% Sodium hypochlorite and 2% Povidone Iodine solution against the growth of Candida albicans. Methods: A laboratory experiment with a total of 21 samples, consisting of 7 samples for each group. The sample is made of a type III gypsum plate measuring 2 x 1 x 0.5 cm. The sample consisted of 3 groups: treatment 1, treatment 2, and treatment 3. The study used the diffusion method. Gypsum plate type III was contaminated with Candida albicans and then immersed in a solution of sterile distilled water (C), 0.5% sodium hypochlorite (P1), and 2% povidone-iodine (P2). The soaking results were planted in SDA media, incubated for 2x24 hours, and then counted using a colony counter. The data were analyzed using Kruskal- Wallis and Mann-Whitney post hoc analyses. Results: In the Kruskal- Wallis analysis of significance (p), between each group, there was a significant difference (p<0.05). The Mann-Whitney post hoc analysis test showed a correlation between group K and P1 and P2 (p<0.05). Conclusion: 0.5% Sodium Hypochlorite and 2% Povidone Iodine solutions are effective as disinfectants using the immersion method in Type III Gypsum against the growth of Candida albicans.

Abstract

Introduction. Training of Trainer (ToT) program for teachers and parents for a 21-day morning and evening toothbrush activity teaches children and adults about the benefits of good oral hygiene and shows them how to brush their teeth twice a day with fluoride toothpaste to realizing caries-free Indonesia in 2030 Method. This activity used a digital / online system via the zoom application. Participants' knowledge of dental health was measured through pre-test and post-test questionnaires. Results. The results of the statistical analysis used a paired sample t-test to compare the mean pre-test and post-test scores of the ToT participants showed a significance value (p=0.000). Discussion. There was a significant difference in the level of knowledge of maintaining dental and oral health in teachers and parents of students at Vita School before being given DHE material and after being given DHE through the "Training of Trainer" Conclusion. ToT activities were useful for increasing participants' knowledge about dental health

Abstract

ABSTRAKPendahuluan: Kandidiasis oral adalah infeksi jamur di mukosa mulut yang sebagian besar disebabkan oleh Candida albicans (C.albicans). Kondisi imunosupresi dapat menyebabkan penurunan sistem imun tubuh, sehingga menurunkan kemampuan melawan C. albicans. Pemberian obat anti jamur nystatin telah diketahui dapat menurunkan infeksi kandidiasis oral, selain itu terapi adjuvant menggunakan oksigen hiperbarik dapat memicu aktivasi makrofag dan menurunkan sitokin proinflamatori. Penelitian ini bertujuan untuk menganalisis pengaruh nystatin topikal dan terapi oksigen hiperbarik terhadap jumlah makrofag pada tikus kandidiasis oral dengan kondisi imunosupresi. Metode: Jenis penelitian eksperimental laboratoris. Tikus Wistar jantan sejumlah 20 dibagi menjadi 4 kelompok: P1 (normal/sehat), P2 (tikus kandidiasis oral kondisi imunosupresi), P3 (tikus kandidiasis oral kondisi imunosupresi diberi nystatin topikal), P4 (tikus kandidiasis oral kondisi imunosupresi diberi terapi oksigen hiperbarik). Imunosupresi dilakukan dengan memberikan deksametason secara peroral selama 14 hari dengan dosis 0,5 mg/hari/tikus, ditambahkan tetrasiklin 1 mg/hari/tikus. Induksi C. albicans dilakukan pada dorsum lidah, dua hari sekali selama 12 hari. Nystatin topikal dan terapi oksigen hiperbarik 2,4 Atmosfer Absolut (ATA) diberikan selama 5 hari. Jumlah makrofag pada sediaan lidah tikus diamati menggunakan mikroskop cahaya dengan perbesaran 1000x. Analisis data menggunakan uji Anova dan Least Significance Different (LSD). Hasil: Uji Anova menunjukkan terdapat perbedaan signifikan (p < 0,05) antara kelompok penelitian. Analisis statistik LSD menunjukkan terdapat perbedaan signifikan (p < 0,05) antara kelompok P1 dengan P2 dan P3, serta kelompok P2 dengan P3 dan P4, sedangkan antara kelompok P1 dan P4, serta P3 dan P4, tidak terdapat perbedaan yang bermakna (p > 0,05). Simpulan: Pemberian nystatin topikal dan terapi oksigen hiperbarik berpengaruh pada jumlah makrofag tikus kandidiasi oral dengan kondisi imunosupresi.Kata kunciimunosupresi, kandidiasis oral, makrofag, nystatin topikal, terapi oksigen hiperbarik  The effect of topical nystatin and hyperbaric oxygen therapy towards number of macrophages in oral candidiasis rats with immunosuppressed condition: experimental laboratory studyABSTRACT Introduction: Oral candidiasis is a fungal infection in oral mucosa, mostly caused by Candida albicans (C. albicans). Immunosuppressive conditions can cause a decrease in the immune system, thereby reducing the ability to fight C. albicans. Nystatin has known can decrease oral candidiasis infection, furthermore an adjuvant therapy using hyperbaric oxygen can trigger the activation of macrophages and decrease proinflammatory cytokines. The aim of this study was to analyze the effect of topical nystatin and hyperbaric oxygen therapy towards the number of macrophages in oral candidiasis immunosuppressed rats. Methods: 20 male Wistar rats were divided into 4 groups: P1 (normal/ healthy), P2 (Oral candidiasis immunosuppressed rats,) P3 (Oral candidiasis immunosuppressed rats with topical nystatin), and P4 (Oral candidiasis immunosuppressed rats with hyperbaric oxygen therapy). Immunosuppressed condition was made by giving dexamethasone 0.5 mg/day orally, added with tetracycline 1 mg/day for 14 days. After 3 days, rats were induced with C. albicans once in two days for 12 days. Topical nystatin and hyperbaric oxygen therapy 2,4 ATA were given for 5 days. The number of macrophages were observed using microscope at 1000x magnification. The data were analyzed using Anova and LSD tests. Results: Anova test showed a significant difference (p < 0.05) between groups. LSD analysis showed a significant difference (p < 0.05) between P1, P2, and P3, also P2 with P3 and P4, while there was no significant difference (p > 0.05) between P1 and P4, also P3 and P4. Conclusion: Topical nystatin and hyperbaric oxygen therapy have effect into the number of macrophages in oral candidiasis immunosuppressed rats.Key wordsimmunosuppression, oral candidiasis, macrophages, topical nystatin, oral candidiasis, hyperbaric oxygen therapy
