Porphyromonas gingivalis and Candida albicans, which are found in dental plaque and can form a biofilm, have virulence factor that facilitates the bacterial colonisation and proliferation in periodontal pockets. Chitosan extract of flower crab (Portunus pelagicus) has antimicrobial potential which can be used as an alternative therapy. The objective of this research was to analyse the potential of flower crab (Portunus pelagicus) chitosan in the inhibition of Porphyromonas gingivalis and Candida albicans biofilms. Methods: This research was a pure experimental laboratory. This study used flower crab (Portunus pelagicus) chitosan to inhibit the biofilm formation of Porphyromonas gingivalis and Candida albicans. The subjects were divided into four groups, where each group consisted of 4 samples. The K+ (positive control group), P1 (0.25% chitosan), P2 (0.5% chitosan), and P3 (1% chitosan). The biofilm inhibition was determined using the microtiter plate methods, which results in the value of optical density, then calculated using the inhibition formula percentage. Data analysis was conducted using the one-way ANOVA followed by the LSD test. Results: There were significant differences in the Porphyromonas gingivalis biofilm inhibition between groups (p < 0.05), except in group K+ with P3. Whereas for Candida albicans biofilm inhibition showed no significant difference (p < 0.05) between group K+ with P2 and P3; group P1 with P2 and P3; and group P2 with P3. Conclusion: The chitosan of flower crab (Portunus pelagicus) has the potential in inhibiting the biofilm formation of  Porphyromonas gingivalis and Candida albicans. The highest antibacterial effect on the biofilm formation is shown in the concentration of 1%.Keywords: Biofilm, Porphyromonas gingivalis, Candida albicans, chitosan, Portunus pelagicus, periodontitis.

Abstract

Background: Smear layer is a debris consisting of organic and inorganic particles of calcified tissue, necrotic tissue, pulp tissue, and dentinoblast and microorganism processes that can close the entrance to the dentin tubuli. Smear layer, will not only inhibit the penetration of disinfection materials and sealers to the dentin tubuli, but will also reduce the attachment of root canal filling material so that root canal irrigation solution is needed to dissolve the smear layer. Red betel leaf (Piper crocatum) infusion, on the other hand, contains saponin characterized as “surfactants” which can dissolve smear layer. Nevertheless, the effective concentration of the red betel leaf infusion has still not been known clearly. Purpose: This study is aimed to determine the effective concentration of the red betel leaf infusion for cleaning root canal walls from smear layer. Methods: Fiveteen extracted human teeth with straight single roots were randomized into 5 groups (n=3). The specimens were then shaped by using rotary instruments up to a size of 25/.07. During instrumentation, each canal was irrigated with 10, 20, 30 and, 40% red betel leaf infusion for treatment groups, while another was irrigated with aquadest for the control group. Root canal cleanliness was observed by using scanning electron microscope (SEM). Results: There were significant differences among treatment groups (p<0.05), except in the treatment groups irrigated with red betel leaf infusion with concentrations of 30% and 40% (p>0.05). Conclusion: It can be concluded that red betel leaf infusion with a concentration of 30% is effective for cleaning the root canal walls from the smear layer.

Abstract

Background: Porphyromonas gingivalis bacteria are found in dental plaques and have the ability to form biofilms. Biofilms are collections of bacterial cells, which are tightly attached to a surface covered by an extracellular polymer matrix released by the bacteria. Periodontitis therapy generally uses tetracycline antibiotics which if used long-term can cause resistance. Chitosan Rajungan crab extract (Portunus pelagicus) has antimicrobial potential which can be used as an alternative therapy. Objective: To determine the antibacterial power of Rajungan crab chitosan extract (Portunus pelagicus) in various concentrations of Porphyromonas gingivalis bacterial biofilms. Method: This research was true experimental laboratorist. This study used Rajungan crab chitosan extract (Portunus pelagicus) to Porphyromonas gingivalis biofilm tested by the test method biofilm. Divided into 5 groups, where each group consisted of 4 samples. The control group consisted of: K- (negative control group), K + (positive control group / tetracycline group), and 3 treatment groups, namely P1 (0.25% Rajungan crab chitosan), P2 (0.5% Rajungan crab chitosan) , P3 (1% Rajungan crab chitosan). Antibacterial power was determined by the value of Optical density in the ELISA Reader. Data analysis using One Way ANOVA followed by LSD test. Result: There were significant differences (p <0.05) seen from the percentage of biofilm inhibition in the negative control group for all groups, the positive control group for the crab crab chitosan extract concentrations of 0.25% and 0.5%. Conclusion: The chitosan crab chitosan extract (Portunus pelagicus) with 1% concentration has the greatest antibacterial effect on the Porphyromonas gingivalis biofilm.

Abstract

Dental and oral health in Indonesia is still a health problem that needs attention. The most concerning thing is that as many as 93% of school-age children experience dental and mouth problems. This can be caused by a lack of knowledge and awareness of dental hygiene. One of the efforts made to increase knowledge and awareness of dental hygiene is by holding an educational activity called the "School Health Program". This activity was carried out in several schools, one of which was TK ITS. This activity began with the Training of Trainer Program given by dentists to teacher cadres and parents of students/students/students about the benefits of good oral hygiene and how to brush your teeth twice a day with fluoride toothpaste. Dentists provide educational materials using power point media and educational videos through the zoom application. There were 5 cadres consisting of 2 teachers and 3 guardians of students who attended this activity. After that, the teacher cadres and parents of the students/students disseminate information to other parents of students/students so that they can become mentors to their children to apply good and correct behavior of brushing their teeth twice a day for 21 days with the method recorded on the given calendar. The hope is that after this program ends, students at ITS Kindergarten have the behavior of maintaining oral hygiene by brushing their teeth twice a day with fluoride toothpaste.
