Abstract

Abstract

Nata de coco is an alternative food source that uses Acetobacter xylinum in the fermentation process. However, the high rate of nata de coco product failures is still commonly encountered due to the inadequate concentration of the starter and the fermentation duration. This study aims to determine whether there is an effect of starter Acetobacter xylinum and duration of fermentation on the quality of nata de coco. The research is of a quantitative experimental type. The research method employs a Completely Randomized Design (CRD) with two factors, namely the concentration of starter Acetobacter xylinum (10%, 15%, and 20%) and fermentation time (6 days, 10 days, and 14 days) with 9 treatment combinations and 3 replications. Data analysis used ANOVA followed by DMRT. The results showed that the combination of starter and fermentation time did not have a significant effect on the thickness and yield of nata de coco, but had a significant effect if tested based on a single factor. The best organoleptic results vary according to their parameters (color, aroma, texture, and taste), depending on the specific combination of Acetobacter xylinum starter concentration and fermentation duration treatment that has a significant effect. Keywords— Acetobacter xylinum, fermentation time, nata de coco

Abstract

Abstract

The increasing of bacterial resistance to antibiotics, which encourages the use of herbal medicines in treating infectious diseases. Acacia auriculiformis leaves are known to have metabolites that can act as an antibacterial agent. This study aimed to determine the antibacterial activity of A. auriculiformis leaves extract against the growth of Bacillus sp., Staphylococcus aureus and Escherichia coli bacteria in vitro. This research was conducted by extracting the leaves of A. auriculiformis using ethanol as a solvent. The inhibition test of A. auriculiformis leaf extract was carried out using the well diffusion method. Antibacterial activity was indicated by the presence of a clear zone around the well. The results showed that the leaf extract of A. auriculiformis had the ability to inhibit the growth of Bacillus sp. and Staphylococcus aureus with the category of strong inhibition. Leaf extract of A. auriculiformis was not able to inhibit the growth of Escherichia coli. Phytochemicals screening test showed that A. auriculiformis leaf contains flavonoids and tannins compounds as antibacterial. Therefore, A. auriculiformis leaves extract had the ability to inhibit the growth of Gram positive bacteria but not Gram negative bacteria.

Abstract

Pests are one of the major challenges faced by farmers, particularly those cultivating horticultural crops, due to the difficulty in eliminating these pests, which also increases production costs. Therefore, the development of a practical and effective biopesticide is essential. This study aims to develop a nanoemulsion biopesticide from two readily available raw materials, namely the jongi plant and cigarette butts. The research employed an in vitro laboratory testing method to evaluate the effectiveness of five formulas (F0, F1, F2, F3, and F4) against two types of pests, Fusarium sp. and whiteflies. The effectiveness was measured by the inhibition zone in the Fusarium sp. test and the mortality rate of whiteflies. Additionally, a direct toxicity test was conducted on tomato plants to assess the safety of the formula. The results showed that the developed biopesticide was effective in inhibiting the growth of Fusarium sp. compared to the positive control and increased the mortality rate of whiteflies. Among the four formulas tested, three of the best-performing formulas were selected based on their inhibition and pest mortality rates, followed by toxicity testing. Formula 3, composed of 0% tobacco, 70% jongi, 20% starter, and 10% palm sugar, with a final pH of 4.2, was found to be safe for use without causing side effects on the plants.
