Abstract

This study aims to analyze the kinship of butterflies (Lepidoptera) based on morphological characters in Pombewe Village, Central Sulawesi. The research method used was the roaming method with free collection technique. Morphological characters observed included antenna shape, wing color and pattern, and body size. Kinship analysis was conducted using the Unweighted Pair Group Method with Arithmetic Mean (UPGMA) with the help of Paleontological Statistics software (PAST 4.03). The results identified 13 butterfly species from the Nymphalidae family, namely Euploea redtenbacheri, Euploea algea, Euploea hewitsonii, Danaus genutia leucoglene, Cyrestis strigata, Danaus genutia genutia, Euploea eunice, Vindula erota, Doleschallia polibete, Acraea terpsicore, Ideopsis juventa, Parantica cleona, and Cyrestis paulinus. The relationship dendrogram showed two main clusters with four relationship categories, namely very close relationship (Cyrestis strigata and Cyrestis paulinus, IS = 100%), close relationship (Euploea hewitsonii and Danaus genutia leucoglene, IS = 74.07%), not close relationship (Cyrestis strigata and Euploea eunice, IS = 32.25%).

Abstract

This study aims to identify the presence of Escherichia coli and Salmonella sp. in "ayam geprek" products sold at the canteens of Tadulako University. The type of research used is descriptive-exploratory with quantitative examination methods using the Most Probable Number (MPN), Total Plate Count (TPC), and further identification using the Microbact System and Microbact System 2000 Software. Samples were taken from three different faculty canteens using a stratified random sampling technique. The results showed that (1) Test using the Most Probable Number (MPN) method obtained the highest value of 150x102 CFU / ml came from the FMIPA canteen sample, 28x102 CFU / ml samples from the FKIP canteen and the lowest value of 27x102 CFU / ml samples from the FKIP canteen. (2) Test using the Total PlateCount (TPC) method obtained the highest value of 105x103 CFU / ml came from the FMIPA canteen sample, 217x102 CFU / ml FKIP canteen sample, and the lowest value of 120x102 CFU / ml FKIP canteen sample. (3) Validation identification of the percentage of samples showing contamination by Escherichia coli and Salmonella sp., with a validation level above 90%. Thus, all samples in this study were positive for the presence of Escherichia coli and Salmonella sp. bacteria.

Abstract

This research aimed to isolate and identify the types of bacteria present on the surface of biology education students' mobile phone screens. This was an exploratory study using a bacterial isolation method.Bacteria found on the mobile phone screens were cultivated using MacConkey agar, a selective medium specifically designed to detect and identify Gram-negative bacteria, particularly Enterobacteriaceae like Escherichia coli. The accuracy of the Escherichia coli data from the phone screen samples, after processing with the Microbact System, was 96.39%.Malt Extract Agar (MEA) was also used as a selective medium to detect and identify Gram-positive fungi, such as Candida albicans.Based on these findings, the developed module was deemed highly feasible. A learning medium is considered highly feasible for use if it achieves a presentation rate between 60-100%. With an average score of 82.39%, the module meets this criterion and can be effectively used in learning.

Abstract

This study aims to analyze the phylogenetic relationships among six species of fiddler crabs (Uca spp.) found in Lalombi Village, Banawa Selatan Subdistrict, Donggala Regency, based on morphological characters. A descriptive-exploratory approach was employed, with purposive sampling to select representative specimens. Eighteen morphometric characters were measured from three individuals of each species: U. triangularis, U. perplexa, U. dussumieri, U. annulipes, U. tetragonon, and U. bellator. Data were analyzed using Principal Component Analysis (PCA) and UPGMA clustering with the aid of PAST software version 4.03. The PCA results revealed that carapace length (CL), anterior carapace width (ACW), and posterior carapace width (PCW) contributed significantly to total morphological variation. Cluster analysis indicated a close phylogenetic relationship between U. tetragonon and U. triangularis, as well as between U. perplexa and U. bellator. U. annulipes appeared intermediate between the two groups, suggesting a moderate level of relatedness. In contrast, U. dussumieri formed a distinct branch, likely due to its markedly larger body size compared to the other species. These findings affirm that morphometric characters are valuable in elucidating phylogenetic relationships within the genus Uca and support taxonomy based on morphological traits.

Abstract

This study aims to analyze the relationship between diet and physical activity with the nutritional status of students of SMP Negeri 19 Palu. This study used a cross sectional design with a quantitative approach and analytic observational type. The subjects were 77 students of class VIII (aged 13-15 years) who were selected using simple random sampling technique. Data were collected using FFQ (Food frequency Questionnaire) to assess diet and APARQ (Adolescent physical Activity Recall Questionnaire) for physical activity, both validated instruments from previous studies. As well as anthropometric measurements (height and weight) using the Body Mass Index (BMI) to determine nutritional status. The results of the analysis using the chi-square test showed that there was a significant relationship between diet and nutritional status (χ²(3) = 8.772; p = 0.032), and between physical activity and nutritional status (χ²(4) = 9.919; p = 0.042) Respondents with a good diet and moderate to vigorous physical activity tended to have normal nutritional status, while poor diet and light physical activity were more common among students with abnormal nutritional status. These findings suggest that diet and physical activity play an important role in determining the nutritional status of adolescents. The implications of this study emphasize the importance of educational intervention programs in the school environment to support healthy lifestyles and prevent nutritional problems among students.

Abstract

This study aims to describe the influence of the model Learning Cycle 7E learning assisted by powtoon media on the activities and learning outcomes of class X students of SMA Negeri 4 Palu. The population in this study were all students of class X of SMA Negeri 4 Palu totaling 400 students spread across class X by taking a sample of 25% of the total, namely 70 students. Sampling used a purposive sampling technique, and the research sample was 2 classes consisting of class XE and XG. Data collection techniques used learning outcome tests and observation sheets. The results showed that there was a significant influence of student learning activities between the experimental class given the Learning Cycle 7E learning model treatment and the control class given the conventional learning model treatment. This is shown by the calculation of the observation sheet of student learning activities, student psychomotor activities and student affective activities in the experimental class getting a score of 83.54 percentage 81-100% very good category while in the control class getting a score of 77.64 percentage 61-80% good category and the average value of significant learning outcomes in class XE as an experimental class 82.20 and class XG as a control class 77.34. In addition, it is proven by the results of the hypothesis test (t-test), obtained a sig value in class X which is 0.00 <0.05. Thus, it can be concluded that the Learning Cycle 7E learning model assisted by Powtoon media has a significant effect on the activities and learning outcomes of class X students of SMA Negeri 4 Palu.

Abstract

Metacognitive knowledge refers to what is known about memory function and the strengths and weaknesses of one's own cognition. This research aims to describe the influence of the problem-based learning model on the metacognitive knowledge and learning outcomes of class IX junior high school students. The research was carried out on Class IX students of Palu City Middle School, the entire population of Class IX consisted of three classes with a total of 45 students. Two classes were used as samples determined based on purposive sampling. Metacognitive knowledge data was obtained through questionnaires and learning outcome data through essay and multiple choice tests. Data analysis used an independent t-test with the help of SPPS version 25.0. The results showed that metacognitive knowledge with conventional learning was significantly lower than the PBL class with a gain of t(25) = 9.289, p < 0.000 and learning outcomes in the conventional class were significantly lower than the PBL class with a gain of t(25) = 4.520, p < 0.000. The conclusion of this research is that there is an influence of the problem-based learning model on the metacognitive knowledge and learning outcomes of class IX SMP students

Abstract

This study aimed to determine the phylogenetic relationship of coral trout groupers (Plectropomus spp.) in the waters of Liang Village, Banggai Kepulauan Regency, based on morphological characters. The research employed a descriptive exploratory method with quantitative and qualitative approaches. Samples were collected using purposive sampling, obtaining 15 individuals representing five species: Plectropomus leopardus, Plectropomus oligacanthus, Plectropomus maculatus, Plectropomus areolatus, and Plectropomus leavis. Morphological observations included 25 morphometric characters, 11 meristic characters, and 4 qualitative characters. Data were analyzed using Principal Component Analysis (PCA) to identify the main morphological characters contributing to variation among species, followed by cluster analysis using the Unweighted Pair Group Method with Arithmetic Mean (UPGMA) to assess phylogenetic relationships. The results indicated that the physicochemical conditions of the waters in Liang Village were suitable to support coral trout grouper populations. PCA results showed that the first two principal components (PC1 and PC2) explained 73.31% of the total morphological variation, with interorbital length, dorsal fin base length, and body depth contributing most significantly to species differentiation. The UPGMA dendrogram revealed that Plectropomus leopardus exhibited the closest phylogenetic relationship with Plectropomus areolatus and Plectropomus oligacanthus, while Plectropomus maculatus showed the greatest morphological divergence and formed a separate cluster. These findings demonstrate that morphological characters are effective in revealing phylogenetic relationships among Plectropomus species in the studied waters.

Abstract

Honey is a valuable natural product with nutritional, medicinal, and economic importance, but its high commercial value makes it prone to adulteration, which threatens consumer trust and product quality. This study aimed to evaluate the effectiveness of simple purity tests in identifying adulteration in honey samples collected from several regions in Central Sulawesi Province. A total of 12 honey samples from different bee species and floral sources were analyzed using seven tests, namely solubility, cloudiness and foam, heating, hexagonal pattern, seepage, raw fish, and onion tests. The results showed that most tests successfully distinguished pure honey from adulterated samples. Specifically, the solubility, cloudiness and foam tests, as well as heating, and raw fish tests indicated that 66.67% of the samples were pure, while the hexagonal pattern, seepage, and onion tests confirmed purity in all samples (100%). These findings demonstrate that simple, low-cost methods can provide reliable initial screening of honey purity and are suitable for routine use in local communities where advanced laboratory facilities are limited. The application of these tests is expected to support honey quality assurance, protect consumer trust, and promote the sustainable development of the beekeeping industry in Central Sulawesi.

Abstract

Indonesia is recognized as one of the world’s biodiversity hotspots, supporting a wide range of pollinating insects, including honey bee species of the genus Apis. Among these, Apis cerana is the most widely managed species and contributes significantly to national honey production, particularly in Sulawesi. However, the increasing economic value of honey has led to frequent cases of entomological origin fraud, in which honey is falsely labeled as being produced by a different bee species. This study aimed to determine the entomological origin of honey sold in Central Sulawesi using the Major Royal Jelly Protein 2 (mrjp2) gene as a molecular marker. Seven honey samples claimed as “forest honey” were analyzed through DNA extraction, PCR amplification using species-specific primers (CF-CR), and sequencing followed by BLAST-n analysis. All samples yielded high-quality DNA with purity ratios appropriate for PCR amplification. PCR successfully amplified the mrjp2 gene, producing fragments of approximately 201-208 bp, consistent with the expected size for A. cerana. Sequence analysis showed percent identity values ranging from 99.20% to 100% when compared with reference sequences of A. cerana. Based on molecular verification, three samples (H3, H5, and H7) did not match the seller’s claims and were confirmed to originate from A. cerana rather than A. dorsata binghami. These findings demonstrate that sequencing of the mrjp2 gene is an effective and reliable approach to authenticate the entomological origin of honey and to detect fraudulent labeling in commercial honey products.
