Abstract

This study aimed to evaluate the quality of frozen semen of Bali bull resulted from sexing procedure on calf or offspring production with desired sex. The tested sperm of Bali bull were collected from Bali bull raised at Regional Artificial Insemination Center of Riau Province (BIBD Riau). The study was carried out in 2 stages. The first stage was X and Y chromosome separation by albumin method. The extender used in the sexing procedure is trice citrate fructose and egg yolk. The second stage was mainly testing the sexed sperm collected in 60 Bali cow in Langkat Village, Bengkalis Regency. To determine the quality of post thawing frozen semen collected from the sexing procedure, the study evaluated motility, viability, mortality, abnormality and plasma membrane integrity of the spermatozoa. The pregnancy rate, calving rate, and birth accuracy of inseminated sexed sperm to offspring’ sex were also evaluated. The evaluation resulted in motility (66.3-75.3%), viability (70-78.5%), plasma membrane integrity (60-65.8%), abnormality (6.05-8.05%), mortality (20.05-30.05%), and pregnancy rate (83.33-90%). The calving rate on this study was 100% with the birth accuracy of 81.8% for male offspring and 40% for female offspring. As conclusion, the sexed sperm evaluated on this study have fairly good fertility.

Abstract

Artificial Insemination (AI) success depends on the quality of the frozen semen. The quality of the frozen semen of swamp buffalo in Indonesia is still low. The study was conducted to determine the quality of buffalo sperm following freezing using three different diluents and three different doses. The study used buffalo semen from the Tuah Sakato Artificial Insemination Center, Payakumbuh (n = 3). The semen collecting was carried out once a week for 10 weeks (replication). The research method used was 3x3 factorial  randomized block design. The first factor was diluent (Triladyl®, Andromed® and Tris egg- yolk) and the second factor was the dose of spermatozoa (10 and 15 and 20 x 106 sperm/ml). Data were analyzed using variant analysis, while the differences between treatments were tested by Duncan Multiple Range Test. The results showed that the plasma membrane integrity of buffalo sperm was found in Andromed® diluent, while tris egg-yolk diluent gave better motility, viability, plasma membrane integrity and recovery rate at a sperm concentration of 20 x106 sperm/mL compared to triladyl® diluent and a sperm concentration of 10 and 15 x106 sperm/mL. It was concluded that andromed® diluent and tris egg-yolk gave better motility, viability, plasma membrane integrity and recovery rate at a sperm concentration of 20 x106 sperm/mL compared to triladyl® diluent and a sperm concentration of 10 and 15 x106 sperm/mL. of Buffalo of sperm abnormalities not sicnificantly by the type of diluent but are influenced by sperm concentration.

Abstract

The purpose of this study was to obtain the optimal equilibration time to produce quality frozen semen of sexed Bali bulls in Tris-yolk diluent. Semen was collected from two male Bali cows from artificial insemination center, Tenayan Raya. Semen was collected for 8 wk and used as a replicate of the study. The motility of the semen used was 70%. Sperm X and Y were separated with 10% and 30% bovine serum albumin (BSA). The diluent used was tris egg yolk. The research treatment was equilibration time (2, 3, 4, and 5 h). Parameters were measured after sperm separation, dilution and thawing after freezing. Parameters consisted of motility, abnormality and intact plasma membrane. Data analysis was performed using a group randomized design. The results showed that the equilibration times of 2, 3, 4 and 5 h were significantly different on motility, abnormality and intact plasma membrane. The value of motility and intact plasma membrane is significantly different from after separation, dilution and freezing of Bali sperm. The values of motility, abnormality and intact plasma membrane of sperm X and sperm Y were not significantly different. Conclusion, 2 h equilibration time can maintain motility, abnormality and intact plasma membrane of sperm X and sperm Y of Bali cattle.
