Abstract

Background: Monosodium glutamate (MSG) consumption in excess can have an impact on a man's fertility and lead to infertility. MSG has the potential to produce free radicals, which can harm cells. Antioxidants can fight off free radicals. Exogenous antioxidants are required to maintain equilibrium since there will be an imbalance if the body produces more free radicals than endogenous antioxidants. Exogenous antioxidants flavonoids are present in the leaves of the Moringa plant (Moringa oleifera L). It is thought that flavonoids have 4-5 times more antioxidant potential than vitamins. Objectives: To evaluate how moringa leaf extract affects the histopathological profile of MSG-exposed mouse testes. Methods: Only the posttest was used as a control group in this study. 25 male mice were used as the sample, and they were split up into five groups. The treatment group 1 (P1) received 6 g/day of MSG exposure + 300 mg/kg BW/day of moringa leaf extract, the treatment group 2 (P2) received 6 g/day of MSG exposure + 600 mg/kg BW/day of moringa leaf extract, and the treatment group 3 (P3) received 6 g/day of MSG exposure + 1200 mg/kg BW/day of moringa leaf extract. The control group (K-) received only standard feed. The mice were put to death after 30 days. The testes were removed for histological examination using hematoxylin-eosin staining, and the Johnsen scoring criteria were applied to their evaluation. Results: The data were not significant according to the Shapiro-Wilk test in the P3 group (MSG 6 g/day + Moringa Extract 1200 mg/kg BW/day). The Kruskal-Wallis test revealed that there was no significant difference between the seminiferous tubules according to the treatment group (p = 0.117). Conclusion: Moringa oleifera leaf extract protects the histopathological picture of the testes of male mice compared to the group given monosodium glutamate.

Abstract

Background: Histological processing encompasses the steps of acquiring fresh tissue, preserving it through fixation to maintain its natural appearance, slicing it into thin sections, placing the sections on glass slides, and subsequently staining them. This enables the observation of various histological components within the tissue using a microscope. The objective of this study was to ascertain the microscopic characteristics of heparin preparations and the kidneys of mice (Mus musculus) through fixation using a formalin buffer solution.Method: This kind of research encompasses experimental research. The study employed a purposive sample methodology in its research design. This study concludes that preparations containing images provide a representative depic-tion of the cell nucleus, cytoplasm, and exhibit consistent coloration with different cutting thicknesses. The standard thickness for cutting the mice hepatic tissue using a microtome is 5 μm.Result: This thickness yields good microscopic results overall, with 25 field views from 5 preparations. The percentage of high-quality microscopic images obtained at this thickness is 100%. The statistical analysis revealed significant differences (p<0.05) between the 2 μm and 5 μm groups, as well as between the 5 μm and 8 μm groups. There was no statistically significant difference (p>0.05) observed between the 2 μm and 8 μm groups in terms of many aspects including structural, nucleolus, cytoplasm, color uniformity, and total structure scores. Conclusion: The optimum thickness in this research was 5 μm. Additional investigation is required to examine the microscopic characteristics of tissue samples by varying the duration of staining on the samples.

Abstract

Background: Moringa provides extra dietary benefits. The immune system is significantly influenced by the nine necessary amino acids, calcium, iron, potassium, magnesium, zinc, and vitamins A, C, E, and B that are included in this food. Previous studies state that moringa causes liver and kidney damage. Another study found that moringa has an abortive effect. Objectives: This study aimed to determine how exposure to kelor (moringa oleifera) leaf extract affected the hepar and kidney in pregnant mice and found the secondary metabolite of the moringa leaves.Materials and Methods: An experimental laboratory design was conducted in 24 pregnant female Balb/c mice. They were randomized into four groups. Group K was not given anything. Group P1-P3 were given moringa leaf extract during pregnancy at days with a dose dose of 10, 20, and 30 mg in 10, 20, and 18 days, and termination was done to take the organs and make the tissue processing. Results: We found no dead mice and no aggression during the experiment. Significant statistics differed between the experimental group in the microscopic appearance of the kidney and hepar. This study found degeneration and inflammation in the hepar and kidney in the treatment group. Qualitative phytochemical tests on leaves contained alkaloids, flavonoids, phenolics, tannins, and steroids. Conclusion: The ethanol extract of Moringa leaf has shown a mild heparin and kidney effect compared to the control.

Abstract

This study investigated the impact of mobile phone–derived electromagnetic wave (EMW) exposure on the uterus and progeny of pregnant Balb/c mice. Fourteen pregnant mice were randomly assigned to two groups (n=7 each). The control group (CG) was not exposed to EMW. In contrast, the experimental group (EG) was subjected to continuous exposure to EMW at a whole-body specific absorption rate (SAR) of 1.74 W.kg-1 for 24 hours daily from gestational day (GD) 1 to 19. On GD 20, all mice were euthanized, and body weight, oxidative stress biomarkers, and uterine histological parameters were assessed. Maternal body weight at termination showed a significant reduction in EG compared with CG (p < 0.05). However, differences in overall body weight gain, malondialdehyde (MDA), and superoxide dismutase (SOD) levels were not statistically significant (p > 0.05). Histological evaluation revealed marked alterations: endometrial scores were significantly higher in EG (3.32) than in CG (0.56) (p < 0.05), and myometrial scores were elevated in EG (1.72) versus CG (0.64). Endometrial thickness was reduced in EG (0.748 mm) compared with CG (1.470 mm) (p < 0.05), whereas myometrial thickness showed no significant difference. Continuous EMW exposure (24 h.day-1, GD1–GD19; SAR 1.74 W.kg-1) was associated with lower maternal weight at GD20, reduced litter size, higher endometrial/myometrial injury scores, and reduced endometrial thickness, while MDA and SOD differences were not statistically significant (p > 0.05). These findings warrant further studies to clarify the mechanisms involved without overstating the changes in oxidative biomarkers.

Abstract

BACKGROUND: Diabetes mellitus (DM) often leads to chronic, slow-healing wounds due to impaired tissue regeneration and sustained inflammation, which can progress to diabetic ulcers. Opuntia cochenillifera has been reported to possess antioxidant and anti-inflammatory potential, making it a promising candidate for natural wound therapies. However, standardized topical formulations and in vivo evaluations are limited. This study was conducted to develop and assess the wound-healing effects of Opuntia cochenillifera cream in diabetic mice.METHODS: Forty-eight male Balb/C mice were randomly assigned to different groups: healthy control (HC), negative control (C−), positive control (C+), and treatment (T) groups. DM was induced by a single intraperitoneal injection of 180 mg/kgBW streptozotocin. After confirming hyperglycemia, incisions and burn wounds were created and monitored for 7 and 14 days. Wound healing was assessed macroscopically (incision length, width, and burn diameter) and microscopically (fibroblast proliferation and re-epithelialization). The formulated Opuntia cochenillifera cream was evaluated for pH, homogeneity, adhesion, and spreadability using standard topical formulation tests.RESULTS: The formulated cream exhibited good homogeneity, adhesion (4.97 s), spreadability (5.10 cm), and skin-compatible pH (6.44). Phytochemical analysis confirmed the presence of flavonoids, tannins, terpenoids and alkaloids. In vivo, Opuntia cochenillifera cream significantly accelerated wound closure and increased fibroblast proliferation compared to the C− group (p<0.05). Its effects were comparable to those of the standard treatment for burn wounds and superior for incision wounds, demonstrating enhanced fibroblast activity and more organized re-epithelialization.CONCLUSION: Opuntia cochenillifera cream effectively improves wound healing in diabetic mice through the enchantment fibroblast activity and organized re-epithelialization. These findings support the potential of this formulation as a natural topical therapy for diabetic wounds, warranting further mechanistic investigation.KEYWORDS: Opuntia cochenillifera, diabetic wound, wound healing, topical cream, phytochemical

Abstract

Background: Pregnant women often use mobile phones for communication and health-related information. The World Health Organization (WHO) recommends minimizing mobile phone use during pregnancy to reduce maternal and child health risks. Electromagnetic waves (EMW) from cell phones can be absorbed by the brain and may affect neuronal function. Objective: This study aimed to evaluate the size, histology, and number of Purkinje cells in the cerebellum of pregnant mice exposed to cell phone EMW compared to unexposed controls. Methods: Fourteen pregnant Balb/c mice were randomly divided into exposed (EG) and control (CG) groups. The EG was exposed to an electromagnetic wave with a specific absorption rate (SAR) of 1.74 W/kg for 24 hours per day from gestation day 0 to 19. On day 20, cerebellar tissues were harvested and processed with hematoxylin–eosin (HE) staining for histological assessment. Results: The mean Purkinje cell size in the EG (0.069 ± 0.009 mm) was slightly smaller than in the CG (0.070 ± 0.007 mm), but this difference was not statistically significant (p = 0.741). However, the Purkinje cell count was significantly lower in the EG (6.160 ± 0.746) than in the CG (7.800 ± 0.764; p < 0.001), indicating that EMW exposure reduced Purkinje cell numbers. Conclusion: Exposure to cell phone EMW during pregnancy led to a significant reduction in Purkinje cell numbers in the cerebellum, while cell size remained unchanged. These findings suggest that prenatal exposure to EMW may negatively affect fetal cerebellar development. This underscores the need for pregnant women to be cautious with mobile phone use and highlights the importance of further research to clarify the risks to maternal and fetal health.

Abstract

Deparaffinization is a stage before the staining process to remove/dissolve paraffin so that the absorption of color in tissue preparations is maximized. Deparaffinization is usually carried out using xylol and toluol. Xylol has toxic effects including acute neurotoxicity, heart and kidney damage, hepatotoxicity, fatal blood dyscrasias, skin erythema, dry skin, peeling skin, and also has a carcinogenic effect. The toxicity effect of olive oil is lower than that of xylol. Oils that have non-polar properties can remove the remaining paraffin contained in the tissue. The purpose of this study was to determine the microscopic appearance of the kidney tissue preparations of mice deparaffinized with olive oil on hematoxylin eosin (HE) staining. The type of research used is experimental research which is analyzed with a descriptive approach. The results of the assessment of preparations deparaffinized with xylol in 80 visual fields obtained 100% good preparations and preparations deparaffinized with olive oil in 80 visual fields obtained 0% poor preparations, 11.3% poor preparations, and 88.7% good preparation. So it can be said that better results are found in the microscopic picture of the kidney preparations of mice (Mus musculus) deparaffinized with xylol.
