Abstract

Abstract:  Urinary tract infection (UTI) is caused by the growth of microorganisms in the urinary tract. The gold standard in diagnosing UTI is bacterial count. This study aimed to determine the effects of temperature and storage time for urine of patients with UTI on the number and the species of bacteria, which is important in supporting the diagnosis of UTI. This was a laboratory and experimental study using accidental sampling techniques in accordance with the inclusion criteria. The results showed that at room temperature (20-25oC) the number of bacteria increased faster and was higher than the number of bacteria stored at cold temperature (2-8oC). The number of bacterial colonies was the highest at storage time of 5 hours (5.3x104 CFU/ml) meanwhile the lowest was in fresh urine (3.4 x 104 CFU/ml). The Friedman test showed that there was an effect of temperature and storage time for urine of patients with UTI on the number of bacteria with a sig p-value of 0.001 (p<0.05). In conclusion, the average number of bacteria at room temperature (20-25oC) increased faster than at cold temperature (2-8oC) during a storage time of five hours. Keywords: urinary tract infection; number of bacteria; storage time; urine    Abstrak: Infeksi saluran kemih (ISK) diakibatkan adanya pertumbuhan mikroorganisme di dalam saluran kemih. Abstract

Background : Kidney failure is one of the causes of death in the world. Laboratory tests related to kidney function are very important in the management of patients with kidney failure because they are useful in identifying any decline in kidney function, monitoring treatment and progression of kidney disease. The urine albumin creatinine ratio (UACR): is related to assess the protein filtration function in the glomerulus. The urine osmolality is to assess pre-renal, renal and post-renal hemodynamic status. The estimated glomerular filtration rate (EGFR) is to assess overall glomerular function. EGFR is the gold standard. The aim of this research is to determine 1). The correlation between UACR value to urine osmolality value, 2). The correlation between UACR value to EGFR value. Patient with kidney failure is the independent variable, meanwhile UACR value, urine osmolality value and EGFR value are dependent variables. Methods : This study is a quantitative study with analytical observations through a cross sectional design using 30 sample patients with kidney failure either acute kidney injury (AKI) or chronic kidney disease (CKD). This study was conducted from May to June 2024 at the Central Laboratory Installation of Dr. Saiful Anwar Hospital, East Java Province. UACR and EGFR examination used the principle of photometric test while urine osmolality examination used the principle of electrical conductivity. The data were analyzed using the Shapiro-Wilk normality test and the Spearman non-parametric correlation test through SPSS 27. Results : In the normality test results, it was found that UACR and EGFR values were not normally distributed (p = <0.001) while urine osmolality values were normally distributed (p = 0.523). Spearman's non-parametric correlation test showed that there was no correlation between UACR to urine osmolality (p = 0.342) and EGFR value (p = 0.481). Conclusion : The results of this study showed no correlation between UACR value to urine osmolality value and EGFR value in patients with kidney failure.

Abstract

Specimen homogenization and the addition of anticoagulants are pre-analytical steps that influence the accuracy of the test results. Complete blood count (CBC) is a screening test that supports disease diagnosis and aids in determining the appropriate therapy for patients. Inadequate specimens are inappropriate for subsequent testing, as they can lead to inaccurate results in the CBC examination. This study aims to determine the optimization of complete blood count results with variations in specimen handling and secondary homogenization techniques for whole blood. The type of research used is quantitative analysis using laboratory experimental methods. The study was conducted in May 2024 in the Clinical Pathology Laboratory of the Diploma-4 Medical Laboratory Technology Study Program, Faculty of Health Sciences, Muhammadiyah University of Sidoarjo, using 48 blood samples. The results of the MANOVA test indicate that the type of anticoagulant has a significant effect on the erythrocyte count (p=0.041) but does not have a significant impact on the leukocyte count (p=0.844) and platelet count (p=0.920). Meanwhile, the homogenization technique does not significantly affect the erythrocyte count (p=0.959), leukocyte count (p=0.991), or platelet count (p=0.867). This study concludes that the secondary homogenization technique has no significant effect. In contrast, the type of anticoagulant significantly impacts the results of the Complete Blood Count (CBC) test. This research suggests collecting whole blood specimens using Ethylene Diamine Tetra Acetate (EDTA) vacutainers as the anticoagulant and applying a secondary homogenization technique before performing the CBC analysis.
