Abstract

Abstract

The choice of anticoagulants and the methods of homogenization are critical pre-analytical steps that can significantly influence the accuracy of laboratory results. This study aims to assess the differences in erythrocyte count and hemoglobin levels among blood samples processed with conventional anticoagulants and Vacutainer tubes, taking into account variations in secondary homogenization techniques. A laboratory experimental design was employed, utilizing 32 blood samples acquired from eight participants. The analysis was conducted using an automated hematology analyzer. Data were subjected to a Paired T-Test, indicating no significant differences in erythrocyte counts (p=0.617) and hemoglobin levels (p=0.510) between conventional EDTA samples that were homogenized four times and those homogenized eight times. However, a significant difference was noted in erythrocyte counts (p=0.018), while hemoglobin levels remained unaffected (p=0.393) in EDTA vacutainer samples subjected to four versus eight rounds of secondary homogenization. Additionally, a significant difference was found in both erythrocyte counts (p = 0.008) and hemoglobin levels (p < 0.0001) between conventional EDTA samples and those subjected to four rounds of secondary homogenization using vacutainers. Similarly, there was a significant difference in erythrocyte counts (p = 0.006) and hemoglobin levels (p = 0.014) between conventional EDTA samples and Vacutainer samples that underwent eight rounds of homogenization.  Keywords: Blood Specimen Collection; EDTA; Erythrocyte; Hemoglobin; Secondary Homogenization

Abstract

Blood type in saliva can be examined through bitemarks on an object left at the crime scene in a crime case as a screening test of suspected perpetrators. Saliva deposited at the bite contains glycoproteins which can carry blood type ABH antigens expressed in the salivary glands and excreted in saliva. Salivary examination is influenced by various external factors that damage saliva, one of which is the duration of exposure to room temperature in a certain period of time. This study aimed to determine differences in protein levels and blood group agglutination titers in (Pyrus bretschneideri) pear bitemark saliva during room temperature exposure within 30, 60 and 90 minutes. This type of study was experimental laboratories with time series design. The samples used were 18 pear bitemarks from 6 individuals who had blood groups A, B and AB then incubated for 30, 60 and 90 minutes. Analysis of protein content using trizol reagent was determined spectrophotometrically, while blood group examination used inhibition absorption method. Anova test showed significant difference between groups of 30 minutes exposure duration with 60 minutes and 90 minutes exposure group (P<0.05; 95% confidence interval). The Pearson correlation obtained the results of -0.739, indicating correlation that the longer exposure to room temperature, the lower the protein content. The results showed that there was a decrease in salivary protein levels and blood group agglutination titers. Salivary protein levels were still detected in the time range of 30, 60 and 90 minutes, so that blood type can still be examined from saliva in bitemarks on pear fruit.

Abstract

Typhoid fever is a systemic infectious disease characterized by the patient experiencing fever and abdominal pain due to the spread of Salmonella bacteria. Typhoid fever sufferers cause the immune system to decline which can affect changes in the number of white blood cells and in an infection, an inflammatory process occurs that produces cytokines which are the core stimulators of acute phase protein production, including C-reactive protein (C-reactive protein = CRP). This study was conducted in April 2021 with the aim of knowing the relationship between the number of leukocytes and CRP levels in typhoid fever patients at RA Basoeni Mojokerto Hospital. The design carried out in this study was a laboratory experimental, the sample needed in this study was 30 typhoid fever patients at RA Basoeni Hospital. Examination of the leukocyte count uses an automatic method with a Hematology analyzer, while CRP examination uses a semi-quantitative method. In the Spearman correlation test, it was found that there was no significant relationship between the number of leukocytes and the level of CRP p = 0.460.
