Abstract

Telah dilakukan penelitian uji penolakan distilat minyak atsiri pandan wangi (P.amaryllifolisRoxb.) terhadap hama kutu beras (Sitophilus oryzae L). Penelitian ini menggunakan rancanganacak lengkap sebagai rancangan penelitian. Uji penolakan menunjukkan fraksi A (41 – 500C)signifikan menolak hama kutu beras. Fraksi ini memberikan persen penolakan sebesar 32,22;29,44; dan 13,30 % untuk tiap konsentrasinya (10, 15, dan 20%). Hari pertama hingga hari ketigamerupakan hari efektif fraksi ini menolak hama kutu beras, sedangkan pada hari berikutnyapersen penolakan berkurang tajam. Analisa GC-MS, UV-Vis, dan FTIR menunjukkan bahwasenyawa fraksi ini muncul pada waktu retensi 1,39 dan 1,44; memiliki puncak spektra uv padapanjang gelombang 205 nm (A=0,1268), dan bilangan gelombang. 3129, 1400, 1108, 617 cm-1.

Abstract

The research characterization of cytotoxic fraction against P-388 leukemia murine cells from the extract honje (Etlingera elatior) seed have been reported. This research lead to isolated and characterization of cytotoxic compounds against P-388 leukemia murine cells from the extract E. elantior seed. The extract of E. elantior seed was maserated by methanol, n-hexane, and ethyl acetate, respectively and estimated their cytotoxic activity against P-388 leukemia murine cell with 3- (4, 5-dimetiltiazol-2-yl) -2,5-difeniltetrazolium bromide (MTT) assay guided toxicity test against of shrimp Artemia salina Leach. Brine shirmp Lethality Test (BSLT) method. The active extracts will be separated by fractionation using column chromatography, radial chromatography, and for analyzing the purity of isolate will estimate by HPLC. The chemical structure of pure isolate will be identified by spectroscopies data UV Vis, FTIR, NMR and MS. The ethyl acetate extract from honje seed have cytotoxic activity by leukemia P-388 cell  with IC50 19.21 µg/mL. The compound toxic as cytotoxicagainst P-388 leukemia murine cells is flavonoid compouds their is resveratrol, lapachol, apigenin, methylated chrysin, 6,2’-dihydroxyflavanone, 3-hydroxy-3,4’-dymethoxyflavone and 4’-hydroxy-5,7-dimethoxyflavanone.DOI: http://dx.doi.org/10.15408/jkv.v0i0.3640

Abstract

A research on antioxidant activity of kecombrang flower ( Etlingera Elatior) water extract leading tojelly candy formulation was conducted. This experiment aims to elucidate the potential usage ofkecombrang flower as a functional food ingredient. Antioxidant activity of the kecombrang flowerwater extract was determined using Diphenyl Picryl Hidrazyl ( DPPH) method. In order to elucidatethe responsible compound for antioxidant activity, GCMS analysis was undertaken. Water extract ofkecombrang flower have antioxidant activity ( IC50=61,6497 ppm) and based on GCMS analysis itcontained 1-dodekanol ( tR=11,60, area=11,73, similarity at 95 %), 3-metil-1-oxo-2-buten 1-(21,41,51-trihidroxyl phenil) ( tR=13,02, area=3,17, similarity at 57 %) and 1-tetradecene ( tR= 13,26,area=6,03, similarity at 98 %). Formulation of jelly candy using kecombrang flower was designedand based on organoleptic test, C formula ( pink, sample code 763) was the optimum jelly pepermintformula with average score of 3,52.

Abstract

Namnam is known as a plant family of fabacea that contains phenolic compounds with diverse biological activities including antibacterial. The aim of this study was to isolate and characterize the antibacterial activity of compounds isolated from ethyl acetate extract of the leaves namanam (Cynometra cauliflora L.). Extraction was done by maceration, fractionation by column chromatography, antibacterial essay by disc diffusion method, and characterization of compounds with UV-Vis, FTIR spectroscopy and LCMS. Ethyl acetate extracts have antibacterial activity with inhibition zone respectively 12.25 and 6.00 mm to Escherecia coli and Stapilococcus aureus at a concentration of 100,000 ppm. The results of column chromatography fractions 23-30 yielded three isolates with Rf 0.20 cm; 0.33 cm and 0.87 cm. Characteristics of antibacterial active compounds in isolates 2 (Rf 0.33 cm) is based on the analysis results are UV-Vis absorption at λmaks 206.93 nm, 268.40 nm, 328.58 nm, 383.98 nm and 386.98 nm , FTIR (KBr) showed -OH group 3415.68 cm-1, CH 2958.10 cm-1, C = C aromatic 1651.18 cm-1, C-OH cyclic 1019.88 cm-1 and CH aromatic 694.56 cm-1 and LCMS produce three main peak at a retention time of 4.82; 6.87 and 7.64 which is thought to be the compound 2-isopropyl-5-metilsikloheksil 2-hidroksipropanoat, Cuelure, and 2-[(2-Hydroxycyclohexyl) oxy] cyclohexanecarboxylate.   Key word: antibacterial, disc diffusion, Namnam (Cynometracauliflora L.). Abstract

Penelitian penggunaan bunga kecombrang dalam formulasi permen jelly telah dilakukan. Tujuan daripenelitian ini adalah mengetahui formulasi yang paling disukai berdasarkan uji organoleptik danspesifikasinya berdasarkan standar permen jelly dalam SNI 3547.2-2008. Penelitian ini terdiri dari duatahap yaitu pertama penentuan formulasi permen jelly dan kedua analisis produk meliputi sifat fisik,sifat kimia, aktivitas antioksidan, cemaran logam dan total mikroba. Sediaan bunga kecombrang yangdigunakan dalam formulasi permen jelly adalah ekstrak air dan manisan bunga kecombrang. Hasilpenelitian menunjukan bahwa formula kode M2 merupakan produk yang paling disukai oleh panelisberdasarkan uji organoleptik.Spesifikasi permen jellytersebut adalah kekerasan 130,43 g force;kekenyalan 10; pH 3,84; kadar air 4,92% (b/b); kadar abu 0,14% (b/b); kadar protein 7,34% (b/b);kadar lemak 0,15% (b/b); kadar gula pereduksi 6,18% (b/b); aktivitas antioksidan (IC50) 161,82μg/mL; logam Pb 0,52 mg/L dan total mikroba permen jelly sampai akhir penyimpanan pada hari ke-4masih dibawah 5x104 koloni/gram yaitu 8,19x103 koloni/gram produk. Berdasarkan standar permenjelly, spesifikasi permen jellyyang paling disukai telah memenuhi standar yang ditetapkan dalam SNI3547.2-2008.

Abstract

Characterization of antioxidant compounds from the seeds of basil (Ocimum basilicum L.) has been done. Extraction is done by maceration method using ethanol solvent, fractionation by TLC and column chromatography, antioxidants test using DPPH method, and characterization of antioxidant compound using GCMS. Ethanol extract and results of fractionation ethanol extract of basil seeds using n-hexane, ethyl acetate, n-butanol and methanol-water extracts show that n-butanol extract has the highest antioxidant activity with IC50 values ​​of 41.90 ppm. Results of column chromatography n-butanol extract using n-hexane : ethyl acetate (1:9) as mobile phase yielded 5 fractions with fraction 4 (F4 isolate) has dominant stain of active antioxidants after being sprayed DPPH reagent, it had IC50 values ​​of 39,70 ppm and total phenolic content of 0,003 mg/g. Isolate F4 suspected contains two active compounds as antioxidant which is terpenoid and phenolic compound group, namely squalene and 1,4-di-tert-buthyl-phenol identified by GCMS.DOI :http://dx.doi.org/10.15408/jkv.v0i0.3598

Abstract

ABSTRAKTeknik hidroponik adalah salah satu metode alternatif dalam budidaya hijauan pakan. Tanaman sorgum telah dikembangkan di Indonesia sebagai alternatif penyedia hijauan pakan. Badan Tenaga Nuklir Nasional (BATAN) telah menghasilkan tiga varietas sorgum berupa varietas Pahat (P), Samurai 1 (S1) dan Samurai 2 (S2). Ketiga varietas ini dapat dikembangkan sebagai Sorghum Green Fodder (SGF). Tujuan dari penelitian ini adalah untuk mengetahui profil pertumbuhan, nutrien dan kecernaan SGF dari tiga varietas sorgum yang berbeda yang ditanam secara hidroponik. Perlakuan penelitian pada studi profil pertumbuhan dan nutrisi adalah: 1) P sebagai varietas kontrol; 2) S1 dan 3) S2. Perlakuan pada studi kecernaan in vitro adalah: 1) RL (rumput lapangan); 2) P; 3) S1 dan 4) S2. Penelitian ini menggunakan rancangan acak lengkap (RAL) dengan lima ulangan. Hasil penelitian menunjukkan bahwa Samurai 2 menghasilkan SGF dengan tinggi tanaman dan berat panen segar tertinggi (P<0,05). SGF Samurai 1 mengandung protein kasar tertinggi yaitu 19,26%. Kandungan neutral detergent fiber (NDF) SGF ketiga varietas tidak berbeda nyata. SGF varietas Pahat menghasilkan estimasi RFV tertinggi (P<0,05). Nilai kecernaan bahan kering (BK) dan bahan organik (BO) SGF lebih tinggi dibandingkan rumput lapangan (P<0,05). Kesimpulan dari penelitian ini adalah SGF Samurai 2 menghasilkan produksi biomassa tertinggi. Kecernaan in vitro SGF varietas Pahat lebih tinggi dibandingkan kedua varietas lain dan rumput lapangan.Kata kunci: Hidroponik, kecernaan In vitro, profil nutrisi, relative feed value, sorghum green fodder ABSTRACT     Hydroponics technique is one of the alternative methods for forage cultivation. Sorghum has been developed in Indonesia as an alternative forage. Indonesia National Nuclear Energy Agency (BATAN) has produced three varieties of sorghum (Pahat, Samurai 1, and Samurai 2). These three varieties can develop as Sorghum Green Fodder (SGF). The purpose of this study was to investigate the growth, nutrient, and digestibility profile of SGF from three different sorghum varieties. The treatment of the studies was: 1) P (Pahat) as control; 2) S1 (Samurai 1); 3) S2 (Samurai 2). Treatment on In vitro digestibility study: 1) RL (native grass); 2) P; 3) S1 and 4) S2. Complete randomized design with five replications was used in this study. The results showed that Samurai 2 produced the highest height and fresh weight (21.35 cm and 1.20 kg) (P <0.05). Neutral detergent fiber (NDF) of three varieties SGF was not significantly different. The highest relative feed value (RFV) was produced by SGF from Pahat variety. Dry matter (DM) and organic matter (OM) digestibility of SGF is higher than field grass (P<0,05). The conclusion of this research is SGF Samurai 2 produces the highest biomass production compared to Pahat and Samurai 1 varieties. In vitro digestibility of SGF from Pahat variety is better than other varieties and native grass.Keywords: hydroponics, in vitro digestibility, nutrient profile, relative feed value, sorghum green fodder

Abstract

Penelitian ini dilakukan untuk mengetahui aktivitas antidiabetes dari ekstrak etil asetat daun pandanwangi (Pandanus amaryllifolius Roxb.) menggunakan metode α-glukosidase. Ekstrak dibuat dengancara perendaman menggunakan etil asetat. Uji antidiabetes dilakukan dengan menggunakan enzim α-glukosidase. Ekstrak etil asetat daun pandan wangi bersifat antidiabetes dengan aktivitaspenghambatan (IC50) sebesar 94,23 ppm. Hasil analisa GCMS menunjukkan ekstrak etil asetat daunpandan wangi mengandung senyawa aktif asam lemak dan turunannya, terpenoid, dan steroid.
