Abstract

This research aims to study the  influence of water content in the production of biogas from dairy cattle  feces to the quality of sludge. The method used  in  this study  is the experimental method using a completely  randomized design, with  three  treatments  (P1 =  substrate water  content  of dairy  cow  feces 40%, P2 = substrate water content of dairy cow  feces 60%, P3 = substrate water content of dairy cow feces  80%  ) with  six  replications. Variables measured were  levels  of N  total, P2O5, K2O. The  results showed that 1) the treatment did not significantly affect total N concentration, but significantly different levels of P2O5 and K2O, 2) substrate water content 60% (P1) and 80% (P2) generate levels of P2O5 and K2O are highest, ie 1.03% and 1.13% (P2) and 0.84% and 1.00% (P3).

Abstract

Abstract. Hidayati YA, Nurachma S, Badruzzaman DZ, Marlina ET, Harlia E. 2021. Utilization of sheep dung and rice straw with indigenous microbial agent to optimize vermicompost production and quality. Biodiversitas 22: 5445-5451. Sheep dung is a useful by product that can be potentially processed into more useful products with minimal pollution. Vermicomposting is a waste processing method that produces Solid Organic Fertilizer and worm biomass as the raw materials for drugs and cosmetics. This research aimed to utilize the role of indigenous fungi and bacteria to determine the influence of C/N ratio from sheep dung and rice straw mixture on vermicompost quality (N, P, K, Ca, Mg). This experimental research was conducted in a Completely Randomized Design with three treatments (T1: C/N ratio of 25; T2: C/N ratio 30; T3: C/N ratio 35) and six replicates for each treatment. Data so obtained were analyzed with ANOVA and Duncan's Multiple Range Test. The results showed that C/N ratio had a noticeable influence on vermicompost quality. C/N ratio 30 produced the highest vermicompost quality (N: 1.11 ± 023381%; P: 0.56 ± 0.010328 %; K: 0.51 ± 0.021369 %; Ca: 0.33 ± 0.0248 %; Mg: 0.14 ± 0.0228%).

Abstract

Abstract. Marlina ET, Hidayati YA, Harlia E, Badruzzaman DZ, Meynadhea N, Rahayu NA. 2024. Microbial quality and macronutrient of vermicompost produced by Eisenia fetida in dairy wastewater solids. Biodiversitas 25: 2729-2737. Dairy Wastewater Solids (DWS) contain enough nutrients for the growth of earthworms and utilize the nutritional requirements of livestock waste for growth. The growth of earthworms affects the quality of vermicompost or castings that are useful as organic fertilizers. This research aimed to determine the growth of Eisenia fertida using DWS as a growth medium, and its effect on the quality of vermicompost produced. The organic material decomposed in DWS by indigenous microorganisms and earthworms can convert DWS into organic fertilizer through composting. Rice straw was added as a carbon source to obtain the ideal C/N ratio for the growth of decomposing microorganisms. Good growth of decomposer microorganisms can increase the availability of nutrients in DWS as a source of plant nutrition. E. fetida was used in vermicomposting for 14 days after the initial decomposition phase had passed. This research used the experimental method of Completely Randomized Design (CRD) with three stocking density treatments (T1: 6.6 g/L; T2: 13.3 g/L; T3: 20.0 g/L ) and six replications. The results showed that higher stocking density promoted an increase in the biomass weight of earthworms. Various indigenous microorganisms act as decomposers and work with earthworms to convert organic matter into inorganic matter as a plant nutrient. The stocking density treatment had no significant effect on vermicompost production, total nitrogen content, and P2O5 vermicompost but had a significant effect on K2O, Ca, Mg, population of Nitrogen-Fixing Bacteria (NFB) and Phosphate-Solubilizing Bacteria (PSB). Vermicomposting, DWS, and rice straw mixture produced vermicompost with high functional microbial and macronutrient concentrations.

Abstract

Abstract

Abstract. Marlina ET, Hidayati YA, Harlia E, Maulana YP, Puteri GCK. 2026. Isolation and cellulolytic activity of bacteria from beef cattle feces using rice straw as enrichment substrate. Biodiversitas 27 (6): d270617. https://doi.org/10.13057/biodiv/d270617. Beef cattle (Bos indicus/taurus) feces contain diverse microbial communities with potential roles in lignocellulosic biomass transformation. This study aimed to evaluate the initial decomposition of rice straw and beef cattle feces, using this information as a basis for isolating and preliminarily identifying cellulolytic bacteria from beef cattle waste. Rice straw was mixed with beef cattle feces to initiate decomposition for 7 days. Changes in chemical composition and structure were assessed using fiber fraction analysis and Scanning Electron Microscopy (SEM). The addition of cattle feces reduced Acid Detergent Fiber (ADF) from 51.36% to 42.31%, Neutral Detergent Fiber (NDF) from 72.08% to 60.82%, lignin from 12.89% to 7.40%, cellulose from 38.47% to 34.91%, and hemicellulose from 20.72% to 18.51%. SEM showed increased surface roughness and fiber degradation (loosening and fragmentation), with structural changes that can be attributed to microbial activity. Based on this finding, microbial enrichment was carried out with cattle feces and rice straw, with the addition of molasses as a nutrient source for microbial growth. Seven bacterial isolates were obtained, among which three (LS-6, LS-11 and LS-14) exhibited the highest cellulolytic activity on Carboxy-Methyl Cellulose (CMC) agar and  dinitrosalicylic acid (DNS). Molecular level identification of these isolates through 16S rRNA gene sequence revealed that these isolates were closely affiliated with Bacillus safensis and Bacillus pumilus group. The results suggest that the isolates exhibit cellulolytic activity under the test conditions and may contribute to lignocellulose degradation. This study provides a first step for further research on cellulolytic bacteria from beef cattle feces for the effective use of lignocellulosic biomass.
