Abstract

Coccidiosis is one of the important diseases in poultry industry. In Indonesia the morbidity of thedisease is between 80 to 90%. A rapid and prompt diagnosis would be one of the essential steps ineradication and control of the disease. The objective of this study is to determine the protein profile ofsporozoite of Eimeria tenella isolated in Yogyakarta using sodium dodecyl sulphate polyacrilamide gelelectrophoresis (SDS-PAGE) protein analysis. Protein analysis was performed in 12% polyacrilamide geland further electrophoresis at 100 volts and over-staining with Coomasie brilliant blue. The resultsshowed that the sporozoite of E. tenella isolated in Yogyakarta contained five proteins with molecularweights of 15, 26, 32, 80, and 91 kDa, respectively.

Abstract

Heart worm (Dirofilaria immitis) is the causative agent of a serious parasitic disease in dogs.Dirofilariasis is generally diagnosed by microfilariae examination and specific antigen testing. Microfilariaeexamination has low sensitivity due to occult infections. The available antigen test at this time is able todetect circulating antigens secreted by adult female worms only. The aim of the present study was toidentify male (MES) and female (FES) heart worms excretory-secretory antigens which have the potentialas a diagnostic targets. Identification of antigen was done by sodium dodecyl sulphate polyacrylamide gelelectrophoresis (SDS-PAGE) and Western Blotting analysis. The results of this study indicated that therewere differences between the MES and the FES profiles. The results showed 12 bands in MES (14–118kDa) and 18 bands in FES (10–205 kDa). Protein with a molecular weight of 59 kDa has the potential asdiagnostic markers of dirofilariasis.

Abstract

Study of histopathological changes of domestic cat organs which were serologically positive toxoplasmosis and laboratory infected which Toxoplasma have been undertaken. Histological section is prepared from organs including brain, liver, lung, kidney, duodenum, jejunum, ileum, and spleen then stained using hematoxylin and eosin (HE) and observed under microscope for histopathological changes. The results showed that in the serologically positive animals cell proliferation, infiltration of leucocyte and macrophage cells were observed in the ileum, whilst infiltration of eosinophil and leucocyte was seen in the kidney and liver. However, in other organ such as duodenum, jejunum, and spleen there were no changes observed. In cat experimentally infected with Toxoplasma, the infiltration of eosinophil cells were observed in the ileum and lung, while other organs such as kidney, liver, brain, jejunum, duodenum, and spleen showed no infiltration of inflammation cells. In conclusion, based on the results seropositive cat, showed proliferation of epithelial cells, leucocyte cells, and macrophage cells in the ileum, while in the lung, kidney, and liver showed infiltration of eosinophil and leucocyte. No infiltration of inflammation cells were observed in the brain, jejunum, duodenum, and spleen. ABSTRAK Penelitian mengenai histopatologi beberapa organ kucing peliharaan yang positif Toxoplasma baik secara serologi maupun yang diinfeksikan telah dilakukan. Tujuan penelitian adalah untuk mengetahui perubahan histopatologi pada organ kucing yang positif Toxoplasma. Data hasil pemeriksaan histopatologi yang terdapat pada preparat jaringan masing-masing organ dianalisis secara deskriptif dengan melihat gambaran perubahan histopatologi pada organ otak, hati, paru, ginjal, duodenum, jejenum, ileum, dan limpa. Hasil penelitian menunjukkan bahwa dengan menggunakan metode histopatologi organ kucing yang positif toksoplasmosis secara serologi teramati adanya proliferasi sel epitel, infiltrasi sel-sel leukosit dan makrofag pada ileum, ginjal, dan hati terlihat adanya infiltrasi eosinofil dan juga infiltrasi leukosit, sedangkan organ yang lain seperti jejenum, duodenum, dan limpa tidak teramati perubahan pada jaringan yang diperiksa. Sementara pada kucing yang dinfeksikan Toxoplasma, ileum dan paru teramati adanya infiltrasi sel-sel eosinofil, sedangkan organ lainnya seperti ginjal, hati, otak, jejenum, duodenum, dan limpa tidak teramati adanya infiltrasi sel-sel radang. Simpulan yang dapat ditarik adalah pada organ kucing yang positif toksoplasmosis teramati adanya proliferasi sel epitel, infiltrasi sel-sel leukosit, dan makrofag pada ileum, paru, ginjal dan hati teramati adanya infiltrasi eosinofil dan juga infiltrasi leukosit, sedangkan organ-organ lainnya seperti otak, jejenum, duodenum dan limpa tidak terlihat adanya infiltrasi sel-sel radang.

Abstract

Lemaea spp. is very important fish ectoparasites because it spreads all over the world and its economic importance has increased due to numerous epizootics occurrence among the most important farmed fish; and has wide host range. Lemaea sp. has been infected not only the family Cyprinidae and Characidae but also in Osteoglossidae family, such as Red Arwana [Scleropagesformosus) in Malaysia and Arwana fish {Scleropagesjardinii (Saville-Kent, 1892)) in Irian.The objective of this study was to identity Lernaea sp. often attacked Arwana irian fish as a precautionary measure the spread of infection, especially on Arwana irian fish. Arwana irian fish were collected from Merauke, Jakarta, Bogor and Depok, 10 infected fish per area. Found Lernaea sp. fixed in glutaraldehyde for morphology study. Based on morphological charactheristic, there were six different Lemaea sp. attacked Arwana irian fish. They are Lemaea cyprinacea, L. devastatrix, L. lophiara, L.oryzophila, L. papuensis and Lemaea n sp. Lemaea n sp. was considered as a new species due to different variation of holdfast organ.

Abstract

Tujuan penelitian adalah menentukan kemungkinan faktor-faktor risiko terhadap kemunculan toksoplasmosis. Sebanyak 132 sampel serum darah diambil pada kucing lokal di Yogyakarta. Data-data epidemiologis seperti asal usul kucing, ras kucing, jenis kelamin, umur, lokasi sistem pemeliharaan, jenis pakan, dan frekuensi diare diberi kode untuk mempermudah analisis, kemudian dimasukkan, disimpan, dan dianalisis dengan program Statistix Versi 7 (Analytical Sofware inc). Analisis data dilakukan secara bivariat (Chi-square (2), dan kekuatan asosiasi (OR), dan multivariat (regresi logistik). Hasil penelitian menunjukkan bahwa faktor-faktor yang berpengaruh terhadap adanya toksoplasmosis pada kucing berdasarkan analisis bivariat adalah pembersihan kotak pasir 1 kali sehari dan mandi 2-3 kali seminggu sedangkan faktor-faktor yang memiliki peluang meningkatkan seropositif toksoplasmosis berdasarkan analisis multivariat adalah pemeliharaan kucing yang bebas di dalam rumah, dimandikan lebih dari 1 kali seminggu, dan dimandikan lebih besar dari 1 bulan sekali.

Abstract

The purpose of this research was to compare the use of PCR method and histological examination to diagnose toxoplasmosis in tissues of Balb/c mice infected with sporulated oocysts through drinking water. A total of 20 male Balb/c mice aged approximately 2 months were used in this experiment. Each mouse was infected with 1x103 Toxoplasma gondii tachyzoites intraperitoneally. Tissue samples (liver, lung, heart, kidney, and brain) were collected from 5 mice on day 1, day 5, day 7, and day 9 after infection. Samples were then examined by PCR and histological methods. The data collected were analyzed descriptively. The results showed that PCR method was more sensitive than histological examination. PCR examination using primer invitrogen gen can amplify DNA T. gondii at 436 bp of the samples from liver, lung, heart and brain on Day 7 and Day 9 after infection. The histological examination showed that the cyst of toxoplasma was found in the brain while mononuclear cells infiltration was found in other internal organs.

Abstract

Jagung (Zea mays) dan Singkong atau ubi kayu (Manihot utillisima) merupakan beberapa komoditas pertanian mayoritas Desa Cabeyan, Bendosari, Sukoharjo. Data Badan Pusat Statistik menyebutkan bahwa produksi palawija di Kecamatan Bendosari sebesar 1.362 ton dari 138 ha luas panen yang 109 ton merupakan hasil panen dari Desa Cabeyan.  Akan tetapi produktivitas yang tinggi ini tidak diikuti oleh peningkatan pendapatan petani. Hal ini disebabkan karena masyarakat tidak memiliki mempunyai teknologi, keahlian dan pengetahuan tentang pengolahanjagung singkong menjadi produk yang lebih bernilai secara ekonomi. Alternatif pengolahan jagung dan singkong yang mudah dan potensial saat ini adalah pengolahan tepung. Solusi dalam program ini adalah dengan pemanfaatan dan pelatihan penggunaan teknologi alat pembuat tepung dan pemipil serta peningkatan kemampuan petani dalampemanfaatan teknologi. Kegiatan yang dilakukan meliputi: 1) Praktek dan ppenyuluhan penggunaan alat pemipil, 2) Praktek dan penyuluhan alat penepung, 3) Praktek dan penyuluhan pengemasan dengan menggunaan alat sealer plastik kemasan. Penerapan dan pelatihan alat berupa alat pemipil, penepung dan pengemas ini dapat mempermudah dan mempercepat proses penanganan pasca panen jagung dan singkong menjadi tepung jagung dan singkong sehingga dapat menambah nilai jual serta pendapatan kelompok wanita tani. Kata Kunci : Jagung, Singkong, Pemipil, Penepung, Pengemas
