Abstract

Penelitian ini bertujuan mengetahui struktur histologi sistem urinaria ikan gabus (Channa striata). Sampel yang diambil adalah ginjal dan vesika urinaria yang berasal dari enam ekor ikan gabus. Sampel kemudian dibuat menjadi preparat histologi dengan pewarnaan hematoksilin-eosin (HE) dan diteliti strukturnya menggunakan metode histologi eksplorasi. Hasil penelitian menunjukkan bahwa ginjal ikan gabus terdiri dari head kidney dan kidney. Kidney terdiri atas glomerulus, tubulus dan pembuluh darah. Glomerulus ikan gabus berbentuk bulat dan berukuran besar. Kapsula Bowman terbagi menjadi stratum visceral dan stratum parietal. Tubulus terdiri dari tubulus kontortus proksimal dan tubulus kontortus distal. Tubulus kontortus distal tidak mempunyai brush border. Kedua tubulus ini disusun oleh epitel kuboid selapis. Vesika urinaria pada ikan gabus mempunyai tiga lapisan yaitu tunika serosa, tunika muskularis dan tunika mukosa yang ditemukan adanya epitel transisional. The study aims to determine the histological urinary system in snakehead(Channa striata). The samples were kidney and urinary bladder came fromof six snakehead. Samples were made be histological preparation withstained haematoxylin-eosin (HE) then observed histologically. The result showed that kidney consisted head kidney and kidney. The kidney consistedof glomerulus, Bowman’s capsule, tubules and vein. Glomerulus of snakeheadwas round and was large in size. Bowman’s capsule divided into stratumviscerale and stratum parietale. Tubules consisted of proximal tubule anddistal tubule. Distal tubule do not  have brush border. Both of this tubulesstructured by  cuboidal epithelia. The urinary bladder of snakehead have three layers namely tunica serous, tunica muscularis muscularis and tunicamucosa  which has transitional epithelium.

Abstract

ABSTRAK            Jantung adalah organ sirkulasi utama yang memiliki peranan penting dalam menyuplai darah keseluruh tubuh. Jantung unggas mempunyai empat ruang yang terdiri dari dua atrium dan dua ventrikel, setiap ruang memiliki tiga lapisan dinding yang terdiri dari; epikardium, miokardium dan endokardium. Penelitian ini bertujuan untuk mendapatkan informasi tentang struktur histologi dan histomorfometri jantung kalkun pada tingkat umur yang berbeda. Sampel penelitian dibagi menjadi tiga kelompok umur (8, 16 dan 24 minggu) dan masing-masing kelompok berjumlah enam jantung kalkun. Jantung kemudian diproses hingga menjadi sediaan histologi dengan ketebalan 5 µm, selanjutnya diwarnai menggunakan pewarnaan Haematoxylin-Eosin (HE). Kemudian dilakukan pengukuran ketebalan dinding epikardium ventrikel kanan dan kiri, miokardium ventrikel kanan dan kiri serta endokardium ventrikel kanan dan kiri. Hasil penelitian didapatkan bahwa struktur histologi jantung kalkun tidak berbeda antar kelompok perlakuan. Rataan ketebalan epikardium kanan pada minggu ke-8, 16 dan 24 secara berturut-turut adalah 177,91±40.57 µm, 838,59±223,05 µm, 1306,96±91,37 µm. ketebalan miokardium kanan pada minggu ke-8, 16 dan 24 secara berturut-turut adalah 1245,55±78,93 µm, 1866,21±257,13 µm, 2465,98±285,82 µm. ketebalan endokardium kanan pada minggu ke-8, 16 dan 24 secara berturut-turut adalah 19,01±1,58 µm, 28,16±5,33 µm, 26,57±2,19 µm. ketebalan epikardium kiri pada minggu ke-8, 16 dan 24 secara berturut-turut adalah 636,31±423,61 µm, 737,19±244,58 µm, 927,43±321,2 µm. ketebalan miokardium kiri pada minggu ke-8, 16 dan 24 secara berturut-turut adalah 5304,99±764,52 µm, 6429,6±685,49 µm, 8334,3±1166,86 µm. ketebalan endokardium kiri pada minggu ke-8, 16 dan 24 secara berturut-turut adalah 31,92±17,36 µm, 37,06±12,53 µm, 49,78±5,16 µm. Data histomorfometri yang telah di analisis menunjukkan bahwa hasil pengukuran ketebalan lapisan dinding jantung pada setiap kelompok umur ialah berbeda nyata (P0,05). Dapat disimpulkan bahwa semakin bertambah umur hewan maka ketebalan dinding akan semakin bertambah. Kata Kunci : Jantung, kalkun (Meleagris gallopavo), histologi, histomorfometri ABSTRACT            The heart is the main circulatory organ that has an important role in supplying blood throughout the body. The avian heart has four chambers consisting of two atria and two ventricles, each chamber has three layers of walls consisting of; epicardium, myocardium and endocardium. This study aimed to obtain information about the histological structure and histomorphometry of the turkey heart at different age levels. The study sample was divided into three age groups (8, 16 and 24 weeks) and each group consisted of six turkey hearts. The heart was then processed into histological preparations with a thickness of 5 µm, then stained using Haematoxylin-Eosin (HE) staining. Then the thickness of the epicardium of the right and left ventricles, the myocardium of the right and left ventricles and the endocardium of the right and left ventricles were measured. The results showed that the histological structure of the turkey heart did not differ between the treatment groups. The mean thickness of the right epicardium at weeks 8, 16 and 24, respectively, was 177.91±40.57 µm, 838.59±223.05 µm, 1306.96±91.37 µm. the thickness of the right myocardium at weeks 8, 16 and 24 respectively were 1245.55±78.93 µm, 1866.21±257.13 µm, 2465.98±285.82 µm. The thickness of the right endocardium at weeks 8, 16 and 24 were 19.01±1.58 µm, 28.16±5.33 µm, 26.57±2.19 µm, respectively. The thickness of the left epicardium at weeks 8, 16 and 24 were 636.31±423.61 µm, 737.19±244.58 µm, 927.43±321.2 µm, respectively. The thickness of the left myocardium at weeks 8, 16 and 24 were 5304.99±764.52 µm, 6429.6±685.49 µm, 8334.3±1166.86 µm, respectively. The thickness of the left endocardium at weeks 8, 16 and 24 were 31.92±17.36 µm, 37.06±12.53 µm, 49.78±5.16 µm, respectively. Histomorphometric data that has been analyzed shows that the results of measuring the thickness of the heart wall layer in each age group are significantly different (P0.05). Abstract

Otot merupakan bagian utama yang penting pada tubuh yang berfungsi sebagai alat gerak aktif, termoregulasi, dan membentuk tubuh. Penelitian ini bertujuan untuk mengetahui gambaran histologi, histomorfometri, dan histokimia sebaran glikogen pada otot dada dan paha ayam kampung periode sebelum dan setelah menetas dengan menggunakan pewarnaan Haematoksilin-Eosin (HE) dan Periodic acid-Schiff (PAS). Sampel penelitian dibagi menjadi empat kelompok tingkat umur berbeda dan masing-masing kelompok berjumlah enam otot dada dan paha ayam. Hasil penelitian menunjukkan perkembangan jaringan yang diawali oleh sel mioblas kemudian menjadi serabut otot yang berkumpul membentuk fasikulus. Hasil pengukuran diameter fasikulus otot dada dan paha serta serabut otot dada dan paha ayam kampung hari ke-14, hari ke-20 sebelum menetas dan hari ke-7 setelah menetas berturut-turut adalah 182,06 ± 4,71µm dan 122,16 ± 5,01µm; 371,43 ± 1,77 µmdan 173,25 ± 5,58 µm; serta 587,47 ± 20,25 µmdan 192,89 ± 5,60µm, serta untuk serabut otot 30,59 ± 1,57 µmdan  23 ± 0,23; 74,23 µm ± 0,52dan 42,47  ± 0,46 µm; 149,11 ± 0,95 µmdan 59,23 ± 0,77 µm. Hasil histokimia sebaran glikogen pada jaringan otot ayam kampung hari ke-14, hari ke-20 sebelum menetas dan hari ke-7 setelah menetas pada otot berturut-turut adalah (+), (++), (+++). Dapat disimpulkan bahwa pertumbuhan otot dada dan paha ayam kampung terdapat perbedaan nyata pada setiap kelompok umur.

Abstract

ABSTRAKGinjal adalah organ ekskresi utama yang memiliki peran penting dalam proses pengeluaran sisa metabolisme. Organ ginjal memiliki perkembangan seiring dengan pertambahan umur. Penelitian ini bertujuan untuk mengetahui gambaran histologi dan histomorfometri ginjal kalkun (Meleagris gallopavo) pada tingkatan umur yang berbeda. Jumlah total sampel yang diamati adalah 18 sampel ginjal kalkun jantan yang dibagi ke dalam tiga kelompok umur yaitu 8, 16, dan 24 minggu dengan masing-masing kelompok umur terdapat enam ekor kalkun. Ginjal kemudian diproses hingga menjadi sediaan histologi dengan ketebalan 3µm, selanjutnya diwarnai menggunakan pewarnaan Hematoksilin-Eosin (HE). Berdasarkan hasil penelitian ditemukan bahwa terdapat perbedaan struktur histologi ginjal pada tiap kelompok umur. Organ pada sistem urinaria unggas terutama ginjal sudah terbentuk dan sudah menjalankan fungsinya dengan sempurna setelah menetas. Data histomorfometri ginjal kalkun yang telah dianalisis menunjukan hasil berbeda nyata (P0,05) antara ukuran diameter glomerulus dan tubulus-tubulus ginjal dari berbagai tingkatan umur. Dapat disimpulkan bahwa terdapat perbedaan perkembangan ginjal kalkun baik secara histologi maupun secara histomorfometri antara umur 8, 16 dan 24 minggu.Kata kunci: Ginjal, histologi, histomorfometri, kalkun (Meleagris gallopavo)ABSTRACTKidneys are the main excretory organs that have an important role in the process of eliminating metabolic waste. Kidneys develop along with the age. This study aimesd to observed the histological and histomorphometric features of the kidney of turkey (Meleagris gallopavo) at different age levels. Total of samples observed were 18 samples of male turkey kidneys which were divided into three age groups, namely 8, 16, and 24 weeks with six turkeys in each age group. The kidneys were then processed into a histology preparation with a thickness of 3µm, then stained using Hematoxylin-Eosin (HE) staining. Based on the results of the study it was found that there were differences in the histological structure of the kidneys in each age group. The organs in the poultry urinary system, especially the kidneys, have been formed and have performed their functions perfectly after hatching. The histomorphometric data of turkey kidneys that have been analyzed showed significantly different results (P0.05) between the diameters of the glomeruli and kidney tubules from various age levels. It can be concluded that there are differences in the development of turkey kidneys both histologically and histomorphometrically between the ages of 8, 16 and 24 weeks.Keyword: Histological, histomorfometry, kidneys, turkey (Meleagris gallopavo)

Abstract

The research aims to find out the  incidence of gastrointestinal parasite in wild long tail macaque in wildlife  reserve  park, Weh  Island, Sabang. For  that purpose,  fecal  specimens  from 25 wild  long  tail macaques  in Weh  Island-  Sabang were  collected.  The  fresh  faeces  samples were  then  preserved  using AFA  (alcohol-formal-acetic)  liquid  prior  to  be  examined  in  Parasitology  Laboratorium,  Faculty  of Veterinary Medicine, Syiah Kuala University.   The  presence of gastrointestinal  parasite was  observed using centrifuge method as well as Boray  sedimentation method. The obtained data were tabulated and analyzed  descriptively.  The  result  revealed  that  two  parasite  species  were  found  in  gastrointestine; Eimeria  spp.    (12%)  and  Strongloides  spp.  (8%).  It  could  be  concluded  that  the  prevalence  of gastrointestinal  parasite  in wild  long  tail macaque  in wildlife  reserve  park, Weh  Island-  Sabang was considered mild.

Abstract

The  research  has  been  conducted  to  determine  hepatoprotective  effect  of  ethanolic  extract  of Acalypha  indica  L.  leaves  on  rats  (Rattus  novergicus)  induced  with  paracetamol.  The  study  was performed  following  a  completely  randomized  design, using  45 male  rats  (Rattus  novergicus)  divided into  5  groups.  Rats  in  group  1  (K1)  were  given  CMC  1  %  for  7  days  and  followed  by  aquadest administration 8 hours post given CMC on day 7. Group 2 was given CMC 1 % for 7 days and treated with paracetamol at dose 2,5 g/kg BW 8 hours post given CMC on day 7. Groups 3 (K3), 4 (K4), and 5 (K5) were pretreated with ethanolic extract of Acalypha indica L.  leaves at the dose of 50, 100, and 200 mg/kg BW for 7 days respectively, followed by paracetamol administration at the dose of 2.5 g/kg BW in the  next  8  hours  on  day  7. Paracetamol  hepatotoxicity  and  extracts  activity were measured  based  on SGPT and SGOT activity level on day 0, 8 hours after  last administration of  extract on day 7, and 24 hours post paracetamol administration. Data were analyzed using one way ANOVA. The result showed that  the  administration  of  ethanolic  extract  of Acalypha  indica  leaves  at  the  dose  of  50,  100,  and  200 mg/kg BW was  significantly  reduced SGPT  and SGOT  level  (P<0,05)  in  rats  that were  induced with paracetamol. Based on the results of SGPT and SGOT serum activity, it can be concluded that ethanolic extract of Acalypha indica L. potential as hepatoprotective agent.

Abstract

This study aimed to verify the effect of ethanol extract of jaloh leaves at various doses on parasitemia percentage of in mice (Mus musculus), wich inoculated Plasmodium berghei. This study used 12 male mice (Mus musculus) strains Balb / C with average body weight of 30.2 grams of 2-month-old. This research using Completely Randomized Design (CRD) with four treatments and three replications. All the mice were infected with Plasmodium berghei prior to treatment on three days post infected the mice in group 2,3 and 4 were given with extract ethanol jaloh leaves at dose 50, 100, 150 mg/kgbw inspectively. The mice in group 1 (K0) were not given anything and consident as control.Observations parasitemia percentage using microscopy on preparations stained thin blood smear Giemsa. The parasitemia percentage data were analyzed using one way ANOVA. On the average (±SD) percentage of parasitemia during the three days in four treatments (K0, K1, K2, and K3) on the first day were 24.87±1.86; 24.33±1.53; 24.20±1.06; and 24.93±1.01. On the second day were 22.67±2.52; 23.00±1.00; 22.80±1.20; and 24.13±2.01; and on the third day were 24.77±1.66; 22.97±3.76; 21.13±1.90; and 24.27±1.42. Statistical analysis showed that ethanol extract of the jaloh leaves for KO, KI, KII, and KIV respectively treatment did not affect the percentage of parasitemia in mice (Mus musculus) inoculated Plasmodium berghei.

Abstract

The study was done to investigate the glycogen distribution in the stomach of long tailed macaques (Macaca fascicularis). The study used eight individuals of long tailed macaques consist of 6 fetuses (55, 70, 85, 100, 120, and 150 days pregnancy) and 2 infants (10 and 105 days). The gastrict tract were histologically stained using Hematoxyllin-Eosin (HE), and histochemically stained using Periodic Acid Schiff (PAS), to detect glycogen. The results revealed that the histogenesis pattern of stomach glandullar cells are developed in line with the age of the fetuses and infant. We also found that glycogen existance are increase in line with the age in the glandullar stomach. From the present study we concluded that the glycogen distribution in gastrict glandular cells were influenced by the age in the fetal period, while in the infant was under influence of the digestion activities, and the activities of the gastrict glandular cells was already seen at 55 days gestation.

Abstract

Penelitian ini bertujuan mengetahui efek injeksi inhibin B terhadap penurunan konsentrasi follicle stimulating hormone (FSH) di dalamserum pada tikus putih (Rattus norvegicus). Dalam penelitian ini digunakan 24 ekor tikus putih berjenis kelamin jantan dengan strain Wistar berumur 4 bulan dengan bobot badan 150-200 g. Tikus-tikus dikelompokkan secara acak ke dalam 4 kelompok, yaitu KK0, KP1, KP2, dan KP3, masing-masing kelompok terdiri atas 6 ekor. Kelompok KK0 merupakan kelompok kontrol hanya diinjeksi dengan phosphate buffer saline (PBS), sedangkan kelompok KP1, KP2, dan KP3 diinjeksi dengan inhibin B dengan dosis berturut-turut 25, 50, dan 100 pg/ekor. Injeksi inhibin B dilakukan secara intraperitoneum sebanyak 5 kali selama 48 hari dengan interval waktu 12 hari. Injeksi pertama inhibin B dilarutkan dengan0,05 ml PBS dan 0,05 ml Freud’s complete adjuvant (FCA). Injeksi kedua sampai kelima, inhibin B dilarutkan dengan 0,05 ml PBS dan 0,05 ml Freud’s incomplete adjuvant (FICA). Pada hari ke-6 setelah injeksi inhibin B terakhir, tikus dikorbankan secara dislocatio cervicalis,lalu darah dikoleksi langsung dari jantung dan didiamkan hingga didapatkan serum untuk pemeriksaan konsentrasi FSH dengan menggunakan metode enzyme-linked immunosorbent assay (ELISA). Hasil penelitian menunjukkan bahwa injeksi inhibin B dengan dosis 100 pg/ekor menurunkan konsentrasi FSH secara nyata (P0,05) bila dibandingkan dengan kelompok kontrol. Berdasarkan hal tersebut, inhibin B berpeluang untuk dikembangkan sebagai kandidat kontrasepsi pria hormon berbasis peptida.

Abstract

The study was conducted to investigate the effects of caffeine treatment during organogenesis period to the fetal birth weight, using rat (Rattus norvegicus) as the animal model. Thirty-six primipararat obtained from Unit Pengembangan Hewan Percobaan, Gadjah Mada University (UPHP-GMU), 3 month old, 165-200 g body weight, were divided into 6 groups, consisted of 6 rats each. Six of the ratshave been selected based on the estrous cycles, and only rat with regular estrous were use for theexperiment. The rat then were mated, and during day 6-14 of the pregnancies were treated orally withcaffeine diluted in aquadest in dosage: placebo (1 cc aquadest) for group I (control), and 5.4, 10.8, 16.2,21.6, and 27 g/200 g body weight/day for treatment groups II-VI respectively. The pregnant rat bodyweights were determined at day 6 of pregnancies for calculating the caffeine treatment dosages. At day 20thof the pregnancies all of the pregnant rats were caesarotomized, and all of the fetuses were removed and weighed. The results showed that all of the treatment groups have significantly lower birth weightcompare to the groups control group. More over, fetal obtained from the treatment groups also showedserious subcutaneous hemorrhagic.Keywords: organogenesis, Rattus norvegicus, birth weight
