Abstract

Aim This study explored the expression of HIF-1α in hypoxic cardiac muscle in mice, and observed the evidence of apoptosis in hypoxia induced cardiomyocyte.Methods Male Sprague-Dawley rats, were randomized into 7 groups (n = 4 per group): control normoxia group that was exposed to atmospheric oxygen and hypoxia groups that were housed in hypoxic chambers (O2 level 8%) for 1, 3, 7, 14, 21, and 28 days respectively. Animals were sacrificed, hearts were rapidly excised, total RNA was extracted with an mRNA isolation kit and the expression of HIF-1α mRNA was then detected by real-time RT-PCR. Apoptosis was assessed by TUNEL method.Results For rat in hypoxia group, the expression of HIF-1α mRNA in cardiac myocytes was clearly up-regulated compared to the control normoxia group. Further, HIF-1α expression level elevated gradually and reached a peak at 21 days of hypoxia. No cell labeled by the TUNEL method was detected in the control group. Compared with the control group, the apoptotic index was significantly increased in the hypoxia group (P < 0.05). There was no significant correlation between the elevation of HIF-1α mRNA and the elevation of apoptotic index.Conclusion Systemic chronic hypoxia caused the elevation of HIF-1α mRNA and apoptosis in cardiac myocytes. Abstract

Pre-diabetes and type 2 diabetes mellitus are two conditions associated with impaired glucose metabolism in the body, in which blood sugar levels become higher than normal. Pre-diabetes is a condition that often doesn't cause symptoms, so it's often not diagnosed early. However, pre-diabetes can be a risk factor for type 2 diabetes mellitus, heart disease, and stroke. Community outreach, education, and early detection of prediabetes and type 2 diabetes mellitus have an important role in efforts to prevent and control these diseases. Community outreach and education can increase awareness about prediabetes and type 2 diabetes mellitus and the risk factors that influence the occurrence of these conditions. This community service activity was carried out in May 2023 by involving 52 productive age respondents. All respondents attended a series of counseling, physical examinations, and supporting examinations related to diabetes mellitus and its complications. The results of this community service revealed that 8 (15.4%) respondents had a GDP ≥ 126 mg/dL and 22 (42.3%) respondents had a GDP 100 - 125 mg/dL. Checking blood sugar levels regularly can also help in early detection of disease. By raising awareness and promoting early detection, it can help reduce the burden of disease and the costs incurred by individuals and society as a whole. In addition, preventing prediabetes and type 2 diabetes mellitus can also help prevent more serious complications such as heart disease, stroke and kidney failure. By preventing or controlling prediabetes and type 2 diabetes mellitus, people can maintain a better quality of life and avoid complications that can affect quality of life. This can help increase the productivity and welfare of society as a whole.

Abstract

Diabetes melitus adalah gangguan metabolik dengan hiperglikemia kronis sebagai ciri utama. Lebih dari 530 juta orang di dunia didiagnosis dengan diabetes, dan sebagian besar adalah diabetes tipe 2. Penyakit ini membawa beban global dengan komplikasi serius, seperti penyakit kardiovaskular dan mikrovaskular. Mengontrol glukosa darah dan mencegah terjadinya diabetes melitus dapat melalui gaya hidup sehat seperti mengatur pola makan yang seimbang, rutin berolahraga, menghindari merokok, dan memperhatikan berat badan. Selain itu, perlu melakukan pemeriksaan kesehatan secara rutin untuk mendeteksi dini adanya masalah kesehatan. Deteksi dini diabetes melitus yang seringkali muncul pada orang di atas 45 tahun adalah penting sehingga dapat mencegah terjadinya kondisi yang lebih serius. Salah satu pemeriksaan yang dilakukan yaitu skrining Glycated Hemoglobin (HbA1c), yang mencerminkan kadar glukosa darah rata-rata jangka panjang. Pengabdian kesehatan di Panti Lanjut Usia Santa Anna yang diikuti sebanyak 50 peserta dengan rerata usia 75,92 tahun. Berdasarkan pemeriksaan gula darah puasa, sebanyak 11 peserta (22%) tergolong prediabetes dan 3 peserta (6%) terdiagnosis diabetes. Sementara itu berdasarkan HbA1c, sebanyak 11 peserta (22%) terdiagnosis diabetes dan 18 peserta (36%) tergolong prediabetes. Melalui partisipasi dalam kegiatan ini, peserta dapat mendapatkan pemahaman tentang faktor risiko, upaya pencegahan, dan dampak jangka panjang dari diabetes. Selain itu, monitoring secara rutin juga perlu dilakukan. Harapannya, dengan meningkatnya pengetahuan dan pemahaman mereka, peserta dapat menerapkan metode perawatan yang sesuai dan memberikan kontribusi yang signifikan terhadap kesehatan secara keseluruhan dan kualitas hidup.

Abstract

Oxidative stress leads to numerous clinical problems, such as autoimmune, vascular and respiratory impairment, as well as many other degenerative diseases. While daily exposure to UV light, air pollution, and smoke largely contribute to oxidative stress, endogenous antioxidants mostly fail to neutralize these high amounts of free radicals. Stevia rebaudiana or “candyleaf” is largely found in Indonesia, known for its application as a natural sweetener, owing to its content of stevioside compound. This study is experimental research which aims to assess the potential of stevia extract as an antioxidant supplement, which has not been widely applied. In vitro study was performed by extracting stevia leaves in methanol through simple maceration and percolation methods. Subsequently, the stevia extract was subjected to biochemical analysis, i.e. phytochemical screening, total phenolic content assay, antioxidant capacity assay, and toxicity assay. Herein, we found that stevia extract contains flavonoid, saponin, tannin, terpenoid, and phenolic compounds, with total phenolic content of 86.30±1.14 mg GAE/g DW. Moreover, antioxidant capacity assay through 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid (ABTS) method showed that IC50 of stevia extract is 20.13 µg/mL, slightly higher than Trolox (vitamin E analog) with IC50 of 16.50 µg/mL, which indicated that stevia extract is a very strong antioxidant. As for toxicity bioassay, Brine Shrimp Lethality Test indicated that stevia extract is non-toxic with LC50 value of 3388.44 µg/mL.

Abstract

Triglycerides are a type of fat found in the blood and serve as a source of energy. When levels rise beyond normal limits, triglycerides can become a risk factor for various health disorders. Hypertriglyceridemia requires prevention and early detection as essential steps in reducing the risk of metabolic disorders. This community service program utilized the Plan-Do-Check-Act (PDCA) approach to educate adults in the Krendang area on the importance of triglyceride screening and adopting a healthy lifestyle. The health education and routine screenings provided successfully increased public awareness of the risks associated with high triglyceride levels and encouraged healthier lifestyle changes. Program results showed that the community became more proactive in monitoring triglyceride levels and adopting healthy behaviors such as balanced diets and regular physical activity. Triglyceride screening and health education play a vital role in preventing metabolic disorders caused by hypertriglyceridemia, supporting preventive efforts to improve public health

Abstract

Stress oksidatif berperan dalam pathogenesis dari berbagai penyakit degeneratif. Hal ini terjadi apabila adanya ketidakseimbangan antara produksi ROS dan pertahanan dari antioksidan. Antioksidan terdapat yang enzimatik dan non-enzimatik. Antioksidan non-enzimatik dapat diperoleh secara eksogen, salah satu nya pada Bunga Turi ( Sesbania Grandiflora ) yang terdapat banyak senyawa bioaktif yang menjadi dasar dilakukannya penelitian ini sebagai upaya pencegahan penyakit degeneratif. Tujuan penelitian ini untuk Mengetahui kapasitas antioksidan, uji fitokimia, uji antimitotik dan kadar total alkaloid dan fenolik pada Bunga Turi (Sesbania grandiflora L.). Penelitian ini bersifat eksperimental secara in-vitro dan bioassay. Sampel diekstraksi menggunakan teknik maserasi dan di evaporasi dengan rotatory evaporator hingga diperoleh ekstrak.  Uji in-vitro terdiri dari uji kapasitas antioksidan dengan uji DPPH, uji fitokimia kualitatif (Harborne), uji kadar fenolik (Singelton dan Rossi), uji kadar alkaloid (Trivedi et al) dan secara bioassay uji antimitotik dengan uji BSLT (Meyer). Pada uji fitokimia kualitatif diperoleh ekstrak Bunga Turi ( Sesbania Grandiflora ) mengandung fenolik, flavonoid, koumarin, saponin, steroid, terpenoid, kuinon, tanin, alkaloid, kardioglikosida, antosianin dan betasianin. Ekstrak Bunga Turi memiliki kapasitas antioksidan ( IC50 = 3708,77 µg/mL ), kadar fenolik (452,95 µg/mL ), kadar alkaloid (7,78 ?g/mL ) dan uji antimitotik (138,09 ?g/mL ). Ekstrak Bunga Turi ( Sesbania Grandiflora ) memiliki antioksidan yang tergolong lemah dan memiliki potensi untuk digunakan sebagai antimitotik.

Abstract

Latar Belakang: Infeksi SARS CoV-2 pertama kali dilaporkan pada tahun 2019, dan menjadi pandemic hingga saat ini. Pemeriksaan penunjang RT-PCR dengan CT value menjadi standar baku emas dalam penegakan diagnose. Keterbatasan pada pemeriksaan RT-PCR membuat tes cepat antigen menjadi alternatif pemeriksaan masal COVID-19. Secara umum, tes antigen cepat diperkirakan dapat bekerja baik pada pasien dengan kadar viral load yang tinggi (CT value <25), namum perlu adanya studi lebih lanjut mengenai nilai prediksi CT value terhadap antigen seiring dengan banyaknya produk komersial tes antigen cepat yang beredar. Hal ini membuat peneliti tertarik untuk meneliti hal tersebut. Metode: Desain penelitian ini adalah potong lintang. Pengambilan data dilaksanakan di Klinik Citra Semanan dan Klinik Sukma pada peride Januari 2021 hingga November 2022. Sampel penelitian berupa nilai CT value pada ketiga alat jenis antigen yang disamarkan (Antigen A, B dan C) terhadap RT-PCR. Analisa statistik menggunakan independent t-test dan Mann Whitney, uji ROC Curve. Sebelum dilakukan pengujian statistic, dilakuka uji normalitas data menggunakan uji Kolmogorov-Smirnoc dan Shapiro Wilk serta pengujian vairan antar kelompok dengan uji Levene. Hasil: Ketiga alat antigen memiliki sensitifitas yang berbeda-beda. Antigen A memiliki sensitifitas sebesar 68,9%, Antigen B memiliki sensitifitas sebesar 88,9% dan Antigen C memiliki sensitifitas sebesar 82.4%. Didapatkan CT value gen ORF pada ketiga jenis antigen 24,00 dan CT value gen ORF sebesar 22,68. Nilai median CT value gen ORF produk A 20,20 (11,01-30,20), B 22,09 (12,00-30,71), dan C 21,51 (10,20-29,58) dengan p-value 0,005. Nilai median dari CT value gen N pada produk A 18,89 (7,72-28,50), B 21,18 (10-50-30,56), dan C 20,59 (9,23-29,35) dengan p-value 0,004. Kesimpulan: Nilai sensifitas ketiga alat tersebut sangatlah bervariasi dengan rentang 68,9% hingga 88,9%. Penelitian ini didapatkan bahwa ketiga antigen ini mampu mendeteksi dengan nilai CT value pada angka 24 untuk gen ORF dan 22,68 pada gen N. Hal ini dapat membantu tenaga kesehatan untuk menjelaskan kepada pasien yang mendapatkan hasil positif, memperkirakan lama hari paska infeksi, dan besarnya pasien untuk menularkan virus tersebut kepada orang lain.

Abstract

Coronavirus Disease (COVID-19) first reported in Wuhan, China On December 31, 2019. In January 2020, World Health Organization (WHO) declared COVID-19 as a global health emergency. Confirmed cases of COVID-19 in Indonesia until September 21, 2021, amounted to 4 million cases, with death rate of 140 thousand people. This study determine the correlation between the use of PPE with COVID-19 infection in midwives. Analytical research with a cross-sectional design. subjects of this study are midwives who actively treated patients and is an active members of Tigaraksa IBI branch who met inclusion and exclusion criteria totaling 153 respondents, data were collected on March 4, 2022. Average age of respondents was 29 years old. 69 respondents (45.1%) have an education level of D3 and 84 respondents (54.9%) have an education level of S1. The average work experience of each respondent is 6 years, the longest work experience is 32 years and the most recent is 1 year. majority of midwives implemented PPE. 129 out of all respondents (84.3%) implemented PPE and only 24 respondents (15.7%) did not. There were 99 respondents (64.7%) infected with COVID-19 and 54 respondents (35.3%) who were not infected. There were 13 respondents (54.2%) who did not use PPE and were not infected with COVID – 19 and 11 respondents (45.8%) who did not use PPE and were infected with COVID-19. 41 respondents (31.8%) use PPE and are not infected with COVID-19, and there were 88 respondents (68.2%) who use PPE and are infected with COVID-19. The relationship between the use of PPE with COVID-19 infection obtained a significant value of 0.061, there no relationship between the implementation of PPE and COVID-19 infection in midwives at the IBI Branch Tigaraksa secretariat

Abstract

Coronavirus Disease 2019 (COVID-19) is an infectious disease caused by the Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2), which first appeared in Wuhan, China and was reported in December 2019. In early October 2021, it was reported According to the World Health Organization (WHO), there are approximately 233 million people in the world who have confirmed COVID-19, while the number of patients who died has been recorded at approximately 4 million people. This study was to describe the knowledge, attitudes, and behavior of the importance of PPE for health workers in hospitals. This research is a descriptive cross-sectional design. In this study, all health workers at Sunan Kudus Islamic Hospital who met the inclusion and exclusion criteria were 169 respondents. In the study, the average age of respondents was 35 years, 135 respondents (79.9%) were women and 34 respondents (20.1%) were men. The average length of work of respondents is 10 years, the longest working respondent is 30 years and the new one is 1 year. The majority who worked in the inpatient room were 109 respondents (64.5%), outpatient room 31 respondents (18%), intensive care room 8 respondents (4.7%), emergency room 21 respondents (12.4%). The picture of the majority of good knowledge on the importance of PPE 168 respondents (99.4%), less good one respondent (0.6%), good attitude towards the importance of PPE 167 respondents (98.8%), bad attitude 2 respondents (1,2 %), good behavior on the importance of PPE 167 respondents (98.8%), bad behavior 2 respondents (1.2%). Abstract

The number of cases of kidney disorders continues to rise, often going unnoticed due to the absence of symptoms in the early stages. Examination of creatinine levels and estimation of glomerular filtration rate (eGFR) can identify impaired kidney function before permanent damage occurs. The habit of drinking enough water is one simple way to maintain kidney function. The activity was carried out using the Plan-Do-Check-Act (PDCA) approach. Officers took venous blood samples aseptically for analysis in the laboratory to assess creatinine levels and calculate eGFR. Participants received the results of the examination along with education about the role of hydration in supporting kidney excretion. The examination involved 62 participants aged 23 to 76 years. The median eGFR value was recorded as 97.0 mL/minute, and creatinine levels ranged from 0.60 to 3.77 mg/dL. Kidney function screening identified participants at risk of decreased excretory capacity. Education about the importance of drinking water regularly needs to be provided to prevent further kidney damage and maintain optimal filtration function.
