Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh kolkisin terhadap keragaman fenotipe tanaman Aglaonema. Penelitian ini dilaksanakan di rumah kasa dan Laboratorium Kultur Jaringan, Fakultas Pertanian USU Medan,dari bulan Juni sampai November 2016. Penelitian dilakukan dengan cara perendaman bahan tanam setek secara keseluruhan dengan 4 konsentrasi yakni 0ppm, 2ppm, 4ppm dan 6ppm.Hasil penelitian menunjukkan tanaman Aglaonema hasil perlakuan kolkisin 6ppm berpengaruh sangat nyata pada peubah amatan panjang daun terpanjang. Hasil penelitian menunjukkan tanaman Aglaonema hasil perlakuan kolkisin memiliki karakter morfologis dan kromosom yang berbeda dibandingkan dengan kontrol. Karakter fenotipe paling baik dihasilkan pada dosis kolkisin 6ppm, dan perlakuan konsentrasi kolkisin mampu menghasilkan karakter fenotipe dan kromosom yang aneh dan berbeda.

Abstract

The ojective of the research was to determine the effect of plant growth regulators on the activity of enzyme superoxide dismutase and the physiology of partially Tapping Panel Drynessin (TPD) the clones quick starter (PB260) and a slow starter (RRIM 921) of rubber plant. This research was conducted at the Research Estate and Laboratory of Physiological Research Center, Rubber Research Centre, Sungei Putih, Galang, Deli Serdang - North Sumatra, from March until June 2016. The completely randomized design was used with three factors: the first factor was the clones of rubber plants (PB260 and RRIM 921), the second factor was the plant condition (Healthy and TPD or conditions), and the third factor was the formula. The Parameters observed were superoxide dismutase (SOD), Thiol, Sucrose, and Inorganic Phosphate. The result showed that the clone of rubber plants was significantly affected the thiol’s, sucrose’s and inorganic phospate in rubber. condition plant and  the formula significantly affected the sucrose’s. Interaction between clone and formula significantly affected the sucrose’s. The Interaction between clone, plant variety, and formula significantly affected the sucrose’s and the enzyme superoxide dismutase (SOD).

Abstract

Penelitian ini bertujuan untuk mengetahui karakteristik kalus pada berbagai varietas kedelai dengan pemberian kombinasi 2,4D dan Kinetin pada kondisi hipoksia secara in vitro. Penelitian dilaksanakan di Laboratorium Kultur Jaringan Tanaman Universitas Sumatera Utara pada bulan Maret sampai Oktober 2016 menggunakan rancangan acak lengkap (RAL) dengan tiga faktor perlakuan yaitu varietas yang diuji (Baluran, Gepak Kuning dan Willis), kombinasi ZPT (5 mg/l 2,4D; 10 mg/l 2,4D + 0,5 mg/l Kinetin; 15 mg/l 2,4D + 1 mg/l Kinetin) dan perlakuan penggenangan. Hasil analisis data menunjukkan bahwa varietas berpengaruh nyata terhadap bobot kalus dan kandungan klorofil. Zat pengatur tumbuh berpengaruh nyata terhadap bobot kalus dan enzim SOD. Perlakuan penggenangan berpengaruh nyata terhadap bobot kalus, kandungan klorofil, enzim SOD dan enzim POD. Interaksi antara varietas dan zat pengatur tumbuh berpengaruh nyata terhadap enzim POD. Interaksi antara varietas, zat pengatur tumbuh dan penggenangan tidak berpengaruh nyata terhadap semua peralakuan.   Kata kunci: 2,4-D, hipoksia, in vitro, kedelai, kinetin

Abstract

Penelitian ini bertujuan untuk mengetahui respon pertumbuhan kalus embriogenik pada berbagai varietas kedelai terhadap pengaruh 2,4-D dan penggenangan secara in vitro. Penelitian dilaksanakan di Laboratorium Kultur Jaringan Fakultas Pertanian Universitas Sumatera Utara, Medan dari bulan Maret sampai Oktober 2016, Rancangan yang digunakan adalah rancangan acak lengkap (RAL) dengan 3 faktor perlakuan. Faktor pertama adalah varietas kedelai  terdiri dari Baluran, Gepak kuning, serta Wilis, faktor kedua adalah konsentasi zat pengatur tumbuh 2,4 D yang terdiri dari 5 ppm, dan 10 ppm; faktor ketiga adalah kondisi hipoksia yang terdiri dari tanpa penggenangan dan penggenangan dengan MS cair. Penelitian ini menggunakan 3 ulangan. Parameter yang diamati adalah persentase pertumbuhan kalus, keadaan visual kalus, bobot kalus, uji aktivitas SOD, uji aktivitas POD, uji protein dan uji klorofil. Hasil penelitian menunjukkan bahwa perlakuan varietas berbeda nyata pada parameter bobot kalus dan uji protein. Pemberian auksin 2,4-D berbeda nyata pada parameter bobot kalus, sedangkan perlakuan penggenangan berbeda nyata pada parameter uji klorofil. Terdapat interaksi antara perlakuan varietas dan penggenangan.

Abstract

Penelitian ini bertujuan untuk mengetahui jenis morfologis dan produksi tanaman                 ubi jalar (Ipomoea batatas L.) didataran tinggi dan dataran rendah di Sumatera Utara. Penelitian ini dilakukan di dua Kabupaten yaitu Kabupaten  Deli Serdang mewakili daerah dataran rendah dan Kabupaten Tanah Karo mewakili daerah dataran tinggi. Penelitian ini di mulai bulan Maret 2016 sampai dengan Agustus 2016 menggunakan metode survei deskriptif dengan teknik pengambilan sampel secara purposive sampling. Hasil penelitian ini menunjukkan bahwa Jenis ubi jalar yang diperoleh dari Kabupaten Deli Serdang (dataran rendah) ada 2, yaitu Genotip 1 dari Kecamatan Hamparan Perak, Namo Rambe dan Delitua, Genotip 2 dari Kabupaten Deli Serdang Kecamatan Sunggal dan Pagar Merbau. Jenis ubi jalar yang diperoleh dari Kabupaten Tanah Kato (dataran tinggi) ada 2, yaitu Genotip 3 dari Kecamatan Berastagi dan Genotip 4 dari Kecamatan Merdeka, Simpang Empat dan Kabanjahe.   Kata kunci : hubungan kekerabatan, karakter morfologis, ubi jalar

Abstract

The purpose of this research was to determinate the growth response of embryogenic callus from some varieties of soybean (Glycine max (L.) Merrill) on hypoxic and in vitro condition. The research was conducted at The Tissue Culture Laboratory of Agriculture Faculty in University of Sumatera Utara on April until December 2016. This research had two stages, induction of embryogenic callus and analysis metabolism of callus after hypoxic condition with t-test. This analysis used factorial Completely Randomized Design with two factors. The first factors were varieties of soybean and the second factors were combination of Plant Growth Regulator (PGR) (5 mg/l 2,4-D + 1 mg/l BAP, 10 mg/l 2,4-D + 1,5 mg/l BAP, and 15 mg/l 2,4-D + 2 mg/l BAP). The result showed the varieties, combination of PGR, and interaction of varieties and PGR give significant effect to weight callus. The result of T-test showed that in hypoxic condition, POD enzyme exercise on Gepak Kuning’s callus in 5 mg/l 2,4-D + 1 mg/l BAP was different before and after hypoxic condition. Abstract

Penelitian ini bertujuan untuk memilih tanaman kedelai ( Glycine max L.Merill ) yang dapat tumbuh dan berproduksi secara baik pada generasi F3 di tanah salin. Penelitian dilakukan di Lahan Percobaan Fakultas Pertanian, Universitas Sumatera Utara, Medan dengan ketinggian tempat ± 25 m dpl, yang dilaksanakan pada bulan Agustus 2015 hingga Januari 2016. Menggunakan analisis sidik lintas pada kedelai varietas Anjasmoro. Parameter yang diamati adalah jumlah Cabang prOduktif, umur berbunga, umur panen, jumlah polong, jumlah polong hampa, produksi biji per tanaman, dan berat biji pertanaman. Hasil penelitian  menunjukkan bahwa tanaman yang memiliki produksi tertinggi yaitu pada nomor tanaman P1(61)7,2 g, yang terendah pada nomor tanaman P3(84) 0,1 g dan diperoleh batas seleksi 2,6-7,2 g. Komponen yang memberikan pengaruh langsung tertinggi terhadap produksi biji per tanaman adalah adalah jumlah biji pertanaman yaitu sebesar 0,96 dan komponen yang memberikan pengaruh tidak langsung yang terbesar adalah langsung jumlah polong sebesar 0,84.Kata kunci : Kedelai, Salin, Seleksi, Varietas

Abstract

The objective of the research was to know growth respons of Pak Coy varieties with applied NAA on floating hydroponics medium. This research was conducted at experimental fields on Compost Centre, Faculty of Agriculture, University of Sumatera Utara, from February-April 2017 with factor such as varieties (Green and Red) and applied NAA (0, 10, 20 ppm) using two way randomized design then analyzed using Duncan Multiple Range Test on 5%. The parameters observed were plants length, number of leaf, chlrorophyll values, stems diameters, roots length, fresh weight per sample,fresh weight per plot and fresh apical weight. The results showed that varieties was significantly different on number of leaf 3, 4, and 5 weeks after planted, chlrorophyll values on morning, noon and afternoon, stems diameters, fresh weight per sample,fresh weight per plot and fresh apical weight. Abstract

This research aims to identify morphological character and relation plant of acid gelugur (Garcinia atroviridis Griff. Ex T. Anders) that exist in Regency of Asahan and Regency of Batubara in North Sumatera. This research was conducted in Asahan and Batubara regency in February-April 2017 with survey method using IPGRI gelmatic descriptors guide. Sampling technique is accidental sampling. The results showed that in three regencies of Sumatera Utara there were 20 accessions of gelugur acid that identified Asahan Regency there were 9 accessions and there were 11 accessions of Batubara Regency. Result of morphological characterization for crown shape (pyramid, round, longitudinal and pattern-shaped), stem surface (slippery, coarse, very coarse), leaf (jorong, elongated, lanceolate), flower (female flowers and hermaphrodite flowers) , Evenly, longitude, longitude). The lowest dissimilarity value or the closest kinship relationship in Asahan and Batubara regency is 1,390 with 4 differences from 19 qualitative characters and the highest dissimilarity value or the furthest kinship relationship in Asahan and Batubara regency is 63,636 with 7 differences from 19 qualitative characters   Keywords : garcinia atroviridis, identification, kinship relationship, morphologycal characteristics

Abstract

The aim of the research  was to determine the effect of colchicine on the genotypic and phenotypic diversity of the Aglaonema plant. The research was conducted at the greenhouse of Faculty of Agriculture, North Sumatera University Medan with 32 meters altitude, from January 2017 to May 2017. The plant materials were immersed in 5 ppm and 10 ppm colchicine for 3 and 6 hours. The result showed that the colchicine concentration significantly affected the increased plant height. The plants were immersed in 10 ppm colhicine for 6 hours showed the morphological and chromosomal difference compared with the control and other treatment.   Keywords:  aglaonema, colchicine, duration of immersio, ghenotypic diversity, phenotypic diversity.
