Abstract

Cervical cancer is one of the most prevalent cancer types, making the development of effective anticancer agents critical. Cisplatin (CIS) is a drug that has been used to treat cancer, but it also affects normal cells. Hibiscus sabdariffa L. extract (HSE), which has the potential as an anticancer agent, can be developed as a co-chemotherapy with CIS. This study aimed to determine the potential of HSE as a co-chemotherapy with CIS against HeLa cervical cancer cells and determine specific and non-specific parameters of the studied extract. Hibiscus sabdariffa L. (HS) simplicia powder was macerated with 96% ethanol. An in vitro evaluation was carried out on antiproliferative activity by calculating the doubling time. Additionally, a selectivity test was conducted to calculate combination index (CI) values based on the microtetrazolium (MTT) method. Apoptosis mechanisms were explored based on immunocytochemical methods using p53, caspase, and Bax antibodies, followed by an observation of apoptotic induction using a flow cytometer. The HSE and CIS selectivity index values obtained for Vero cells and HeLa cells were 209 and 278, respectively. The antiproliferation test results showed that the combination of HSE and CIS could better extend the doubling time of cells compared to the negative control. This combination also demonstrated a strong synergistic effect, with a CI value of 0.001. The extract as a co-chemotherapy with CIS was capable of increasing the expression of p53, caspase-3, and Bax. The flow cytometry analysis results indicated that HSE could induce apoptosis. Based on the results on the IC50 and CI value of HSE, as well as on the doubling time and apoptosis induction of HSE-influenced HeLa cells, it is concluded that HSE has the potential as a co-chemotherapy against cervical cancer.

Abstract

Kanker leher rahim merupakan penyebab kematian wanita akibat kanker terbesar di negara-negara berkembang. Pertumbuhan sel kanker bersifat tak terbatas karena mempunyai kemampuan untuk menghindari mekanisme apoptosis sehingga penemuan agen-agen pemacu apoptosis kini terus dikembangkan. Kemangi (Ocimum sanctum L.) dilaporkan memiliki potensi penghambatan perkembangan sel kanker melalui aktivitas sitotoksik, apoptosis, dan penghambatan angiogenesis pada sel kanker paru dan payudara. Penelitian ini bertujuan untuk mengetahui efek ekstrak etanol daun kemangi terhadap aktivitas sitotoksik dan proses pemacuan  apoptosis pada sel kanker leher rahim HeLa. Evaluasi viabilitas sel melalui nilai IC50 ditetapkan dengan MTT assay. Efek pemacuan apoptosis oleh ekstrak etanol daun kemangi terhadap sel kanker HeLa diamati dengan metode pengecatan DNA menggunakan etidium bromida-akridin oranye. Hasil penelitian ini menunjukkan ekstrak etanol daun kemangi memiliki potensi sebagai agen kemopreventif pada kanker leher rahim HeLa dengan IC50 209µg/mL. Berdasarkan hasil tersebut dapat disimpulkan penghambatan pertumbuhan sel HeLa oleh ekstrak etanol daun kemangi terjadi dengan menginduksi terjadinya apoptosis. Kata kunci : kanker leher rahim, sel HeLa, daun  kemangi, sitotoksik, apoptosis

Abstract

The effectiveness of simple rental apartments (rusunawa) as a housing solution for low-income communities (MBR) is examined using the case study of the Cepu Rusunawa in Blora Regency. The housing backlog in Indonesia, which will reach 12.7 million units by 2023, with 84% of the population remaining homeless, is the background to the urgency of this research. The rapid rate of urbanization of 3% per year, with a projection that 66.6% of Indonesia's population will live in urban areas by 2035, further emphasizes the importance of vertical housing solutions. The research method employed a normative legal study approach. Data analysis was conducted through qualitative methods, utilizing legal, conceptual, and comparative perspectives for policy analysis related to the implementation of Law No. 20 of 2011 concerning Flats. The results indicate that the Cepu Rusunawa, built with an investment of IDR 15.8 billion from the state budget and inaugurated in February 2017 with 98 residential units, faces significant challenges in its management. Despite initial interest, occupancy has declined, with 30 of the 99 units vacant by 2023. Several inhibiting factors exist, including unclear management status between the central and regional governments, and the need for a comprehensive evaluation of service quality and the physical condition of the buildings. A holistic and collaborative approach from all stakeholders is needed to optimize the function of the low-cost apartments as a sustainable, affordable housing solution.
