Abstract

Leucaena leucocephala provides the highest amount of proteins compared to other greens. However, its use is restricted due to the content of mimosine, an antinutrition non-protein amino acid known to be toxic to animals. This study aimed to observe the effect of administration of more than 10% of processed Leucaena leucocephala leaf meal to the level of T3, T4 and the thyroid histopathology. In this study, Leucaena leucocephala leaves were soaked in distilled water for 12 hours. The processed of Leucaena leucocephala leaf meal was made into pellets containing 0%, 7.5%, 15%, and 22.5% leaf meal of total feeds respectively, and was fed to the rats during pregnancy and lactation. The level of triiodothyronine (T3) and thyroxine (T4), and the thyroid histological features were the parameters observed. The collected data were statistically analyzed by SPSS 20.0 for Windows using One-way ANOVA followed by Duncan Multiple Range Test to observe any significant difference among the 4 treatment groups (?=5%). Meanwhile, the presence of hyperplastic cells and follicle lumens filled with vesicles and colloids was descriptively analyzed by means of available literatures. The analysis of T3 and T4 level revealed that there was no any significant difference between the control and treatment groups. The epithelial cells of thyroid follicles in the treatment groups of P1, P2 and P3 showed hyperplasia and were detached from their respective basal membranes.

Abstract

This study aimed to determine CD4 count from cryopreservation of Buffy coat (BC) and PeripheralBlood Mononuclear Cell (PBMC) with and without ficoll. Fifteen EDTA Blood sample (2 ml for eachtube) were drawn from one adult healthy subject. The samples were categorized into five group beforemeasuring the CD4 level (which were fresh whole blood [Group(G)-I], BC without ficoll [fresh <GII>and frozen <G-III>] , and PBMC resulted from BC and ficoll isolation [fresh <G-IV> andfrozen <G-V>]. Each group was replicated three times. Blood storage before preparation was less thanfour hours. Two months cryopreservation using liquid nitrogen (in 40% FBS, 10% DMSO, and RPMI)was conducted. The mean value of CD4 count (cell /mu1) were 522 (G-I), 1410 (G-II), 906 (G-III), 807(G-IV), and 733 (G-V). CD4 count, after 2 month preservation in liquid nitrogen, of the BC sample (G-III) was higher (906 cell /mu1) than PBMC (G-IV) sample (733 cell /mu1).

Abstract

Cryopreservation of PBMC (peripheral blood mononuclear cells) was done to preserve and analyze the number of PBMC derived from blood samples which come in at different time. The batch analysis wasperformed at the same time in order to reduce variations in results. The analysis on the cells numbers carried out after 1, 3, 6, 12, and 24 hours. Heparinized whole blood was collected from healthy subject by venipuncture, and stored at room temperature. Blood is processed by centrifugation in Ficoll density gradient following the established method of Balai Besar Veteriner Denpasar. Buffy coat layer was collected and washed twice with HBSS (Hank's balanced salt solution) and was counted in Turk’s solution. The cells were then dissolved in 1 ml of cold freezing medium containing 10% DMSO and 50% FBS (fetal bovine serum) and stored overnight at -80°C before storage in liquid nitrogen vessel for few weeks. The samples rapidly thawed in a water bath at 37°C and washed twice with PBS (phosphate buffered saline). The cells were stored in 4°C PBS and counted in Turk’s solution after 1, 3, 6, 12, and 24 hours. The results obtained were varied with a declining trend.

Abstract

Smoking habits have been around since ancient times, but nowadays this habit is considered to be detrimental, especially to health. The impact that is often felt by smokers is difficulty in breathing because the lungs are exposed to cigarette smoke. Cigarette smoke contains about 1015-1017 oxidants or free radicals, as well as 4700 harmful chemicals, including aldehydes / carbonyls, NO2, and SO2. Herbal cigarettes are tobacco cigarettes with added ingredients from plants. Gurah terapi sin cigarettes are herbal cigarettes that are sold commercially. The aim of this study was to determine the effect of gurah cigarette smoking on the leukocytes and lung histology of mice. This study used a comparative method consisting of 3 groups, namely the control was not exposed to cigarette smoke, treatment 1 was exposed to commercial cigarette smoke and treatment 2 was exposed to cigarette smoke with herbal ingredients and each group consisted of 10 replications. The results showed that there were significant differences (p <0.05) regarding the number of cell necrosis, type II pneumocytes, inflammatory cell infiltration, hemorrhage, and alveolar dilation. While the results of the analysis of the number of leukocytes showed no significant difference where p > 0.05. The results showed that there was no significant difference in the number of leukocytes in the control group, treatment 1 and treatment 2 (p > 0.05). herbs containing various kinds of antioxidants cause a tendency for differences in the number of leukocytes where there is a decrease in the number of lymphocytes and neutrophils and an improvement in the histopathological structure of the lung against type I pneumocyte cell necrosis, hemorrhage, alveolar dilation, type II pneumocyte cell proliferation, and inflammatory cell infiltration in exposed mice. commercial cigarette smoke without herbal ingredients.

Abstract

Abstract Objective: P-glycoprotein (P-gp) overexpression on neoplastic cells can deteriorate the therapeutic outcome on cancer patients. P-gp plays important role on drug efficacy and toxicity. This research aimed to measure P-gp expression on children with Acute Lymphoblastic Leukemia (ALL) on Sanglah Hospital, Denpasar. Method: Flowcytometry method was used to measure P-gp expression level on Bone Marrow samples from pediatric patients (0-12 years old) who were newly diagnosed with ALL in Sanglah Hospital. P-gp overexpression were based on the percentage of cell stained. Ten percent of P-gp expression were considered as the cut-off value of P-gp overexpression. Result: On this study, 11 samples were obtained with the range value of 56-97% on P-gp expression. Conclusion: All 11 patients had P-gp overexpression.

Abstract

Advances in molecular biology has brought a new branch of multidiscipline study named biotechnology. To comprehend the complex nature of concepts in biology, practical and hands-on approaches are often required. This activity was a collaboration between Biology Teachers Working Group (MGMP Biologi) in Klungkung Regency with university as the facilitator, and aimed to train senior high school biology teachers in compiling learning modules as well as supplying them with the appropriate materials required in simple practical sessions, such as those in animal physiology and reproduction, as well as biotechnology. The event was held successfully, attended by total of 20 biology teachers from six senior high schools (SMA) in Klungkung District, Klungkung Regency, started by introductory lectures and followed by practical sessions on how to perform mice vaginal and sperm smear, human blood smear, and DNA extraction of plant tissues. Abstract

Balai Karantina Pertanian Kelas I Denpasar is located on Jl. Raya Sesetan No 312, Denpasar which is a place of seclusion and/or action as an effort to prevent the entry and spread of pests and animal diseases or nuisance organisms from abroad and from one area to another within the country, or exit from territory of the Republic of Indonesia. The people's need for beef is increasing, demanding more production in order to reach many consumers in various regions. This causes beef producers to pay attention to the quality of the meat to be marketed so that the meat becomes safe. The Total Plate Count (TPC) is one of the initial tests to identify the general microbial count in meat. Therefore, this study aims to determine the TPC in samples of beef that will be marketed outside Bali. There were 20 samples of beef, originating from 5 beef companies in Bali that beef outside the island of Bali through testing services from the Class 1 Agricultural Quarantine Center Denpasar. The test results stated that most of the beef samples from the five companies were contaminated with microbes. The feasibility of consuming the beef sample was declared feasible in accoaccord per 2008 SNI standard 1 x 106 CFU/g.
