Abstract

This experiment was performed to examine the effects of black cumin (Nigella sativa) seed oil extract onreproduction performance and fetus development if given during organogenesis period. Forty pregnantmice were randomly divided into four groups. Extract was treated orally by gavage with doses 0%(control), 0.1 ml, 0.2 ml, and 0.3 ml since gestation day 6 to 15. Caesarean section were performed topregnant mice on day 18 to remove fetuses. Observation covered reproduction performance i.e. the foetusnumber (reabsorptions, dead/ alive foetus), and morphological of the foetus (weight and length of thelitter, and malformations). Statistical analysis was performed using Anova and Duncans Multiple RangeTest. Teratogenic effects of black cumin oil extract caused decreasing of reproductive performance of themother (alive foetus decreased, dead and resorbed foetus increased) and an increased incidence of birthdefects (fetal weight and length and fetal hemorrhage).Keywords: Nigella sativa, congenital malformations

Abstract

Anjing kintamani bali (AKB) adalah kelompok anjing lokal jenis pegunungan yang hidup di sekitar Desa Sukawana, Kecamatan Kintamani, Kabupaten Bangli, Bali. Anjing kintamani bali merupakan plasma nutfah asli Indonesia yang sangat perlu dijaga kelestarian dan kemurniannya. Peripheral blood mononuclear cell (PBMC) sebagai komponen penting sistem kekebalan tubuh yang terlibat dalam imunitas humoral dan seluler, dapat digunakan untuk memonitor perkembangan penyakit atau respon terhadap pengobatan serta menemukan biomarker yang valid dalam kompartemen sel darah. Penelitian ini bertujuan untuk mengetahui ukuran sel limfosit dan sel monosit pada AKB sehat yang dievaluasi berdasarkan pemeriksaan cytomorphometry. Penelitian ini merupakan penelitian eksploratif menggunakan Rancangan Acak Lengkap (RAL) pola faktorial AxB. Data dianalisis dengan analisis ragam dan disajikan dalam bentuk tabel, grafik, dan gambar. Berdasarkan hasil penelitian, AKB memiliki nilai cytomorphometry yaitu diameter sel limfosit (7,312-8,761µm), diameter nukleus limfosit (5,786-7,138 µm), diameter sel monosit (7,785-8,883 µm), diameter nukleus monosit (6,099-7,457 µm), circumference sel limfosit (23,131-27,737 µm), circumference nukleus limfosit (18,160-22,402 µm), circumference sel monosit (24,514-27,966 µm), circumference nukleus monosit (19,093-23,442 µm), surface area sel limfosit (42,318-60,140 µm2), surface area nukleus limfosit (27,331-40,308 µm2), surface area sel monosit (48,313-62,334 µm2), surface area nukleus monosit(28,979-43,736 µm2), luas sitoplasma sel limfosit (8,863-27,509 µm2), dan luas sitoplasma sel monosit (12,230-25,953 µm2).

Abstract

Calliandra calothyrsus Meissn. leaf extract is potential as phytoestrogens. It influenced male mice reproduction, rat estrous cycle, and ovarian histology in previous study. This research aimed to prove the C. calothyrsus leaf extract potential as phytoestrogen source and the effect on endometrial thickness where the embryo implantation take place in early pregnancy. This study used a Completely Randomized Design used 54 days old female rats (Rattus norvegicus). Rats were divided into K as control group (treated with 0.5% Na-CMC as placebo) and P1, P2, and P3 as three groups with C. calothyrsus leaf extract administration with doses of 17.5; 35; and 70 mg/ kg bw respectively. Treatments were given 1 ml/rat/day orally for 20 days. At day 21st, animals were euthanized to collect blood samples for estrogen hormone analysis. After the dissection, all uterus were collected and weighed. Histological preparation was done with paraffin method and Hematoxylin-Eosin staining. The effective dose was 70 mg/ kg bw that did not decrease the weight of the uterus and the body. This dose even maintained the normal diameter and thickness of uterine walls (endometrium, myometrium, and perimetrium layers) like control rats. The extract in this study could increase estrogen concentration in female rats. This research novelty is that C. calothyrsus leaf extract (70 mg/ kg bw) can be used as an alternative herbal suplement to maintain uterine wall thickness and estrogen concentration in productive women. With further clinical research, this extract is a good candidate as potential estrogen source to overcome women infertility or pregnancy difficulties due to problem of endometrial thickness and lack of endogenous estrogen.

Abstract

This research was conducted to determine the effect of Leucaena leucocephala leaf extract on the development of ovarian follicle cells of mice (Mus musculus). Twenty four female mice, 10 weeks old, were divided into 4 groups. The control group (P0) were given 0.9% NaCl and the treatment group P1, P2 and P3 were given 0.5, 1 and 1.5 g/ kg bw of Leucaena leaf extract, respectively. The treatments were administered daily for 15 days by gavage. After the treatment ended, all mice were dissected to collect the ovaries. Histological preparation of the ovaries used paraffin method and Hematoxylin-Eosin staining. Observations were made on the number of primary, secondary, and tertiary oocytes, and also De Graaf follicles and corpus luteum. Data were statistically analyzed using One Way ANOVA method. The results showed there were no significantly differences (P>0.05) on the number of primary, secondary, and tertiary oocytes, as well as De Graaf follicles and corpus luteum among the control and treatment groups. The mimosine content of Leucaena leaf extract was 0.87% (less than 1%) so it could not inhibit the secretion of Follicle Stimulating hormone by the pituitary gland. The doses of Leucaena leaf extract used in this research did not significantly affect the follicles development of mice ovaries. In this study, the mimosine content of Leucaena leaf extract decreased 91.8%.

Abstract

Pineapple peel in animal feed might contain bromelain enzyme which could distrupt pregnancy in animal laboratory. This experiment used a complete randomized design with 16 pregnant rats which were divided into four treatment groups. The treatments are feed containing 0 (K); 4,35 (P1); 8,70 (P2); and 13,05 (P3) g/rat/day of pineapple peel that were given during pregnancy. Parameters of morphological fetus observed were alive fetuses, weight, length, and fetal abnormalities and skeleton development i.e. axial skeleton (costal, sternebral, and vertebral bones) and appendicular skeleton (vertebra caudal, metacarpal and metatarsal bones). Data were analyzed by SPSS version 22, using One Way Anova or Kruskal Wallis. The result showed that fetuses from pregnant rats which administered with feed containing pineapple peel showed teratogenic effect. Fetal morphological abnormalities of the fetus is subcutaneous hemorrhage in were found at dose of 8,70 (P2) and dose of 13,05 (P3) g/rat/day. Feed containing pineapple peel could inhibit ossification of apendicular skeleton in vertebra caudal, metacarpal, and metatarsal.

Abstract

Research on the reproductive biology of fish can provide information on spawning frequency, spawning success, spawning time, and size at first maturity. This research aimed to determine the reproductive biology of common dolphinfish (Coryphaena hippurus Linnaeus, 1758) landed at the fish landing site (PPI) Kedonganan, Badung, Bali. Sampling was conducted in May - September 2018. A total of 42 female common dolphinfish with length ranged between 41-110 cm FL were obtained from hand-line fishery based on FADs. Ovaries were prepared using the paraffin method and Harris Haematoxylin-Eosin staining for histological observation. The results showed that the gonad maturity level in adult females was: spawning (38.10%), spawning capable (35.71%), regressing-potentially reproductive (4.76%), regressed 1 (2.38%), regressed 2 (2.38%), and regenerating (2.38%). Immature female: immature (9.52%) and developing (4.76%). Oocyte diameter ranged between 43,20 – 864,66 µm with an average of 430,99 µm. Fecundity in fish size 49-75 cm FL ranged from 30,720-433,080 eggs / fish with an average of 129,594 eggs / fish. The difference in the level of oocytes in the ovaries at the same time showed that female common dolphinfish in the southern Bali waters has an asynchronous ovary development and indeterminant fecundity with multiple spawning strategies and size at first maturity (Lm50) was 43.2 cm FL. Abstract

This study aims to determine the side effects of high-dose injections of vitamin C in a long term against liver histology. This study used a completely randomized design (CRD) with different long treatments of vitamin C injection in various doses of P0 (control), P1 (30 days injection), P2 (50 days injection), P3 (70 days injection) and P4 (90 days injection). Parameters measured were plasma levels of SGPT and SGOT, the percentage of hydropic degeneration,  fatty degeneration, pyknotik nuclei, and necrosis of liver cells, and also inflammatory cell infiltration and sinusoid congestion. SGOT levels showed a significant differences (P=0,009) between the treatment and control group by ANOVA test. The Kruskal Wallis test showed that ALT levels were not significantly different (P=0,86), however, the levels of creatinine showed significant differences (P=0,002). The hydropic degeneration were not significantly different (P=0,054) by Kruskal Wallis test, whereas the fatty degeneration were significantly different (P=0,000) between the treatment and control groups by ANOVA test. Pyknotik nuclei and necrotic cells showed significant differences between the control and treatment with the P value of 0,016 and 0,01 respetively. The injected white vitamin C in high doses for long periods decreased the physiological functions of female mice liver.

Abstract

The pituitary gland of broiler chickens (Gallus sp.) is empirically known to have gonadotropins to accelerate ovulation in fish. This research was conducted to determine the reproductive performance and survival of sangkuriang catfish (Clarias sp.) Larvae of adult male and female mature gonads by 20 pairs. This study used a Completely Randomized Design consisting of 4 treatments and 5 replications to obtain 20 experimental units. Doses of pituitary extract used is without injection of pituitary chicken pituitary extract (P0), injection of pituitary chicken pituitary extract with a dose of 600 mg/kgbb (P1), 900 mg/kgbb (P2) and 1200 mg/kgbb sangkuriang catfish (P3). Intramuscular injection of extracts in the ventral dorsal fin with a volume of 0.5 mL/kgbb. Descriptive data analysis for gonad maturity level, ovulation response, egg performance. Statistical analysis of data for morphometric aspects using the Wilcoxon test, for latency time, relative fecundity, fertilization success, hatching success, and larval survival aspects using One Way Anova using SPSS version 25. The results of statistical analysis showed significant differences in reproductive performance (relative latency and fecundity time), successful fertilization and successful hatching of eggs, but there was no significant difference in survival (number of live larvae, length and weight) larvae of sangkuriang catfish (Clarias sp.). Broiler chicken pituitary extract (Gallus sp.) can improve reproductive performance, fertilization and hatching success but does not affect egg performance and survival of sangkuriang catfish (Clarias sp.) larvae. Abstract

The aimed of this study was to determine the effect of feed containing various concentrations of Calliandra calothyrsus Meissn. leaf meal mixed with pineapple peels and concentrate feed on histopathology and the activity of liver of local rabbit (Lepus sp). This study used a Post-Test Only Control Group Design, using 24 local male rabbits divided into 4 groups of feed treatment namely P0 = concentrate feed as control; P1 = mixture of 15% of calliandra leaf meal (CLM), 30% of pineapple peels (PP) and 55% of commercial feed (CF); P2 = 30% of CLM, 30% of PP and 40% of CF; and P3 = 45% of CLM, 30% of PP and 25% of CF). The treatments were given for 60 days and used 6 replicates for each group. The data were analyzed statistically by SPSS program version 22. The research results showed the increase of calliandra leaf meal levels in concentrated feed which were added 30% of pineapple peels still caused liver damage namely fatty degeneration, necrosis and congestion. It increased the ALT level, but did not affect the AST level in local rabbit serum.

Abstract

Sodium nitrite (NaNO2) is a food preservative that is often added to processed meat products. Excessive consumption of NaNO2 can be detrimental to health. One effort to minimize the negative effects caused by NaNO2 is by consuming antioxidants. This study aimed to determine the effectiveness of Eucheuma cotonii seaweed extract on improving blood profile, liver and kidney function in male white rats (Rattus norvegicus) induced by NaNO2. This research used a Completely Randomized Design (CRD) consisting of two control groups and three treatment groups. Each group consists of six replications. Negative control (K-) receive aquades and CMC NA 0,5%. Positive control (K +) was only received 22.5 mg/kgBW NaNO2 for 35 days then on the day 36 until the day 45 only given 0,5% CMC Na. The treatment groups P1, P2, and P3 respectively on day 1 to day 15, only received 22.5 mg/kgBW NaNO2 in the morning, and then day 16 to day 35 received 22.5 mg/kgBW NaNO2 in the morning and received 150 mg/kgBW (P1), 300 mg/kgBW (P2), 450 mg/kgBW (P3) Eucheuma cottoni seaweed extract in the evening. Day 30 to day 45, the treatment groups only received Eucheuma cottoni seaweed extracts at each dose. Blood and liver were collected on day 46. Data analysis of serum alanine transaminase (ALT) and aspartate aminotransferase (AST) enzyme levels and liver histology were performed with Anova statistical test and continued with Duncan post hoc test. The results showed Eucheuma cottonii extract dose of 150 mg / kgBB significantly reduced the level of liver damage seen from the levels of ALT and AST and liver histology (percentage inflammatory cell infiltration, congestion, fatty degeneration and necrosis) in white rats induced by sodium nitrite (NaNO2).
