Abstract

Development Population of Root-Knot Nematode (Meloidogyne spp.) and TheLevel Of Damage to Some Plants Solanaceae Family This research started from November 2018 until March 2019 in the experimental garden Pegok Faculty of Agriculture, University Udayana. It uses acomplete randomized design with 4 treatments and 6 repeats. The study treatmentuses some of the Solanaceae family plants such us tomato (Lycopersiconesculentum), eggplants (Solanum melongena L), large red chili pepper (Capsicumannum), cayenne pepper (Capsicum frutescens), which was given 500 of nematodesMeloidogyne spp. on Each polybag. The observed parameters include thedevelopment of the population of nematode Meloidogyne spp. for 2 life cycles in 1gram of root and 300 grams of soil and measuring the rate of crop damage.Nematodes Meloidogyne spp. is an important parasitic nematodes in the world.Solanaceace plants are widely cultivated in Indonesia because of their high economicvalue and become an important food ingredient. One of the problems in aquacultureSolanaceae is the attack nematodes Meloidogyne spp.. The highest population of theresearch results is in tomato plants with a damage rate of 68.3%. The lowestpopulation amount in chili pepper plants with a damage rate of 11.7%. The less-likedplant Nematoda Meloidogyne spp. is a chili pepper so it can be used as an alternativecrop rotation.

Abstract

Histopatology Siamese Citrus Leaves (Citrus nobilis) Infected by Citrus Vein Phloem Degeneration (CVPD) at mild, moderate and severe attacks. Aims of this study is to identify the presence of the bacteria Liberobacter asiaticum, cause of CPVD (Citrus Vein Phloem Degeneration) disease and to knowthe differences in the histology of Siamese citrus plant tissue that is attacked by mild, moderate and severe attacks. This research was in the Laboratory of Plant Structure and Development of the Faculty of Biology, University of Gajah Mada and conducted at the UPT of the Genetic and Molecular Biology Laboratory of Udayana University. Field observations and sampling were carried out in the Wanasari Banjar, Kintamani Village, Kintamani Sub-District, Bangli District, Bali Province. There are several symptomatic citrus plants and attacked by CVPD with various levels of severity by visual appearance. On the observed land it were found that the attack percentage was 70% and the intensity of the attack was 46.4%. PCR (Poly polymeration chain reaction) identification method using OI1 and OI2 specific primers. The DNA band 1160 bp was found in mild, moderate, and severe chlorosis symptomatic samples. Leaves without symptoms do not change shape in the epidermis, cortex, xylem and phloem, while form changes occur in phloem, xylem and cortex in symptomatic citrus plants.

Abstract

ABSTRACT Effectiveness Test of Some Plant Seeds Extract to Inhibit the Growth of Spotting Leaf Bacteria (Xanthomonas campestris) on Tomato Plant. The X. campestris bacterium is very harmful to tomato plant because the bacteria able to attack the plant seedling up to full grown stage. This bacterium  not only attack the root, but also infect the leaves, fruits and stems. Utilization of botanical pesticides to inhibit the pathogen becomes one alternative option to be recommended. The control method has selective activity and safe for the ecosystem. The purpose of this study is to determine the ability of some plant seeds extract in inhibiting the growth of X. campestris the causal agent of leaf spot on tomato plants. Rose apple seed extract contains phyto-pesticide that can be used to inhibit the growth of leaf spot caused by X. campestris. The minimum inhibitory concentration of the extract was at a concentration of 0.312% with the diameter of inhibition zone of 8 mm. It was expected that rose apple seed extract may be used as an alternative subtance to inhibit the growth of bacterium X. campestris bacterium of tomato plants. Abstract

Types Of Weed Leaf Extract On The Development Of Cabbage Caterpillar (Crocidolomia pavonana F.) In Laboratory The research was conducted in laboratory of Plant Pest and Disease Faculty of Agriculture Udayana University, from October 2016 until February 2017 to test the three types of weed leaf extracts namely Lantana camara leaf, Wedelia trilobata leaf, Chromolaena odorata leaf and to know the biological development of caterpillar Crocidolomia pavonana after three types of weed leaf extracts were applied. This research used Completely Randomized Design (RAL) with 3 treatment of leaf extract at 50% concentration and 1 type without treatment (control), each repetition was done 10 times. The observation is done until the larvae do nothing activity (dead), or until it becomes imago. Testing three types of weed leaf extracts showed by the following results: the biological development of caterpillars C. pavonana with leaf extract application of L. camara gave the influence of the development of larvae into pupa very slow because it takes 16 days to become a pupa, while W. trilobata leaf extract only takes 12 days to become a pupa. However, with the leaf extract of C. odorata larvae did not reach the pupa and imago phase because this extract resulted in 100% death in the larval phase.

Abstract

ABSTRACT Identification of Arbuscular Mycorrhiza through Microscopis Methode in Rhizosfer of Several Grass and Cacao ( Theobroma cacao L.) Arbuscular mycorrhiza fungi that live in a symbiotic mutualism with plant roots and helps the absorption of plant nutrients, and also able to live in various places. The study aimed to identifiy arbuscular mycorrhiza through microscopic methode in rhizosfer of several grass and cacao was conducted from December 2013 to March 2014. The method used in this study were spore isolation by wet sieving and root staining techniques. The results showed that arbuscular mycorrhiza spores and structures found in Imperata cylindrica L. are 2 types spores of genus Glomus (Glomales: Glomeaceae) with vesicular and hyphae structures. The presence of spores and structures in Paspalum notatum are Acaulospora (Glomales: Acaulosporaceae) and genus Gigaspora (Glomales: Gigasporineae) with arbuscular, vesicular and hyphae structure. Spores and structures of mycorrhiza found in the Pennisetum purpureum are 3 types of the genus Glomus with hyphae and arbuscular structure. Spores and structures mycorrhiza in the Cyperus rotundus are 1 type spore of the genus Gigaspora and 2 types of spores Glomus with internal hyphae structure. While spores and mycorrhiza structures in the cacao are 2 types spore of the genus Glomus with hyphae and vesicular structures.   Keywords: Glomus, Gigaspora, Acaulospora, Arbuscular Mycorrhiza Structure

Abstract

Effectiveness Of Some Extract Of Plants Leaf To Cabbage Crop Caterpillar Crocidolomia pavonana F. (Lepidoptera: Pyralidae) In The Field Cabbage crop caterpillar (Crocidolomia pavonana F.) is one of the most important pests in cabbage plants. This caterpillar is capable todecreasing cabbage production. This study was conducted in order to determinethe ability of some extracts of leaf plants such as: Chromolaena odorata L., Lantana camara L, Tithonia diversifoliaHemsl., and Nicotiana tabacum L., to suppress the development of the population, the most effective leaf extract against Crocidolomia pavonana F.and its effect on quality of cabbage leaves. This study used Randomized Block Design (RBD) with 5 treatments and replicated 5 times. This study was done from November 2017 to February 2018. The results showed that from the 4 plants extracts that significantly affect the population ofCrocidolomia pavonana F compare to control treatment. The highest population decline was found in C. odorata leaf extract of 1.8 /plants and T. diversifolia of 1.8 /plant. In the effectivity test of leaf extract proved able to suppress population of C. pavonana with different potency. The result of observation of cabbage crop weight showed that the treatment of plant extract had significantly effect on the weight of cabbage. Treatment with C. odorata leaf extract had higher average cabbage weight compared to other treatment which was 2.1 kg, while the lowest weight of cabbage was 1.4 kg. The average yield of cabbage crop quality with different quality categories. In treatment with C. odorata extract, L. camara, T. diversifolia, and N. tabacum resulted in crop quality with quality category 2.

Abstract

Identification of Fusarium solani Fungi Associated with Stem Rot Disease in Dragon Fruit Plant (Hylocereus sp.) at Bangorejo District, Banyuwangi Regency Dragon fruit plants at Banyuwangi in the maintenance process always experience disorder such as pathogenic attacks. The part of dragon fruit plant whichis often attacked by pathogens is the stem which is caused the stem to rot. One type of photogenic fungus in dragon fruit plants is Fusarium solani. This fungus attacks the stem of the dragon fruit plant, so that it can cause the stem of the plant looks wilted and brown rot.The cause of stem rot disease of dragon fruit plant in Banyuwangi is not yet known exactly, thus identification in macroscopic, microscopic, and molecular need to be done. Recently, the use of molecular methods in fungi identification has increased to solve this problem. One of the molecular approaches to identify fungi is by using a device called Polymerase Chain Reaction (PCR).The result of this research showed mycelium that grown on PDA media (Potato Dextrose Agar) shaped like a thick cotton expands like a cloud, the shape of the edge of the colony is uneven, the surface color of the colony is white, the color of the bottom of the colony is white, the direction of the growth of the mycelium is to the side and the structure of the mycelium is smooth. In microscopic identification there found a long macroconidia, crescent shaped with blunt ends, 1-5 septa, microconidia shaped in round to oval, long hyphae and bersepta. The molecular identification with PCR to determine the presence of Fusarium solani mushroom by using universal primary pairs of ITS4 and ITS5 successfully amplified and produced DNA fragments measuring 560 bp.

Abstract

ABSTRACT Inhibiting Ability of Some Fruit Rinds Extract Against the Growth of Ralstonia solanacearum on Chili Ralstonia solanacearum infect plant root through wound that occured indirectly. The symptoms of the disease are whole plant to wilt, the leaves turn yellow to blackish-brown, and eventually the plant dies. In the culture media contained  Triphenyltetrazolium Chloride (TZC) the bacterium tend to form white colony and irregular round shape. The method used to determine the extract activating was well diffusion method. The results of the study showed that application of coconut rind extract inhibited the bacterium. The minimum concentration was 1,25% with diameter of inhibition zone was 15 mm. The phenolic compounds contained on the coconut rind was possibly inhibit the growth of the bacterial colonies. Further research needs to be conducted in vivo to determine the effectiveness of antibacterial compounds contained in the extract of coconut rind. Abstract

Effectility of some Extract to Larvae of Plutella xylostella Development in the LaboratoryThe research was conducted in November 2017 to Februari 2018 in Udayana University Plant Pest and Disease Laboratory. The aim of this research is to know the ability of some plant leaf extract to larvae of Diamonback Moth (Plutella xylostella) development in the laboratory. Plant extract materials used in this experiment were leaf of Lantana camara, Chromolaena odorata, Tithonia diversifolia, and Nicotiana tabacum. Process of extracting was done in Laboratory of Genetic Resources and Molecular Biology Faculty of Agriculture Udayana University. This research uses Completely Randomized Design (CRD) with 4 treatment of leaf extract at 10% concentration and 1 without treatment (control), each treatment was repeated 10 times. Observations were made until the larvae does not do activity or until become adult. Testing of four types of plant leaf extracts showed the following results: Each of C. odorata leaf extract, L. camara, T. diversifolia and N.tabacum used as research material had different potential in suppressing P.xylostella development. Application of L. camara, T. diversifolia and N. tabacum on larvae can develop through 4 instars before becoming pupa, treatment with plant leaf extract is most effective to suppress the development of larvae biology P.xylostella is a leaf extract of C. odorata that result were 100% mortality on larvae stase and the lowest is on the treatment of N. tabacum extract were 20%.

Abstract

The Influence of The Presence of Weeds on The Population Number of main Pest of Cabbage on The Cabbage Croppings The aim of the research was to find out the development of population number of Plutella xylostella and Crocidolomia pavonana in the precence and the absence of weeds on the cabbage croppings. The results showed that there were 23 species of weeds in association with plant cabbage, which was made up of 7 species of grasses, 3 species of weeds from Cyperaceae family, and 13 species of broadleaved weeds. Main pests of cabbage on cabbage croppings (Plutella xylostella and Crocidolomia pavonana) was found more on cabbage plants in the absence of weeds compared than that of the presence of weeds.
