Abstract

Papaya (Carica papaya L.) fruit was proven scientifically in having medicinal effects: such as gastroprotective, laxative, galactagogues, and antibacterial effect. Unripe papaya fruit contains saponin, alkaloid, tannin, flavonoid, triterpenoid, and quinone which may reveal toxicities as adverse effect when it consumed in high dose. The aim of this study is to determine the acute toxicity degree of unripe Carica papaya L. fruit aqueous extract in mice using proposed new recommended method. The acute toxicity degree conducted by determination of lethal dose50 (LD50) using new recommended method to 12 female mice. The subjects were divided into three stages and each mouse was given different dose of the extract. LD50 calculated by mean of mortality lowest dose and non mortality highest dose then the result classified into the degree of acute toxicity. The study showed there was no mortality in all groups of the experimental subject. LD50 of unripe Carica papaya L. aqueous extract was found >5000 mg/kg BW and classified as Practically Non Toxic (PNT) substance.

Abstract

Abstract. Cigarette smoke exposure is a major risk factor for acute lung injury (ALI), characterized by increased oxidative stress and inflammatory responses in lung tissue. Arabica coffee (Coffea arabica) contains bioactive compounds, particularly chlorogenic acid, which possess antioxidant and anti-inflammatory properties. This study aimed to evaluate the effectiveness of Arabica coffee infusion in reducing the degree of lung tissue inflammation in mice exposed to cigarette smoke. This experimental study involved 30 mice divided into six groups: normal control, cigarette smoke control, dexamethasone control, and three groups receiving different doses of Arabica coffee infusion. Histopathological assessment was performed by evaluating inflammatory cell infiltration, alveolar edema, and alveolar damage. Data were analyzed using the Kruskal-Wallis test. The results demonstrated a significant association between Arabica coffee administration and reductions in all inflammatory parameters (p<0.001). The treatment groups exhibited lower inflammation scores than the cigarette smoke group and showed effects comparable to dexamethasone. These findings suggest that Arabica coffee has potential as a natural protective agent against cigarette smoke-induced lung injury. Abstrak. Paparan asap rokok merupakan faktor risiko utama terjadinya cedera paru akut (acute lung injury/ALI) yang ditandai oleh peningkatan stres oksidatif dan respons inflamasi pada jaringan paru. Kopi arabika (Coffea arabica) mengandung berbagai senyawa bioaktif, terutama asam klorogenat, yang memiliki aktivitas antioksidan dan antiinflamasi. Penelitian ini bertujuan menganalisis efektivitas pemberian air kopi arabika dalam menurunkan derajat inflamasi jaringan paru mencit yang terpapar asap rokok. Penelitian eksperimental ini menggunakan 30 ekor mencit yang dibagi menjadi enam kelompok, yaitu kontrol normal, kontrol asap rokok, kontrol positif deksametason, dan tiga kelompok perlakuan air kopi arabika dengan dosis berbeda. Derajat inflamasi dievaluasi secara histopatologis berdasarkan infiltrasi sel radang, edema alveolar, dan kerusakan alveolar. Analisis data dilakukan menggunakan uji Kruskal-Wallis. Hasil penelitian menunjukkan adanya hubungan yang signifikan antara pemberian air kopi arabika dan penurunan seluruh parameter inflamasi (p<0,001). Kelompok perlakuan menunjukkan skor inflamasi yang lebih rendah dibanding kelompok asap rokok dan sebanding dengan kelompok deksametason. Air kopi arabika berpotensi sebagai agen protektif alami terhadap kerusakan paru akibat paparan asap rokok.

Abstract

BACKGROUND: Diabetes mellitus often exacerbated by cigarette smoke, which disrupts glucose homeostasis and lipid metabolism. Mango (Mangifera indica L.) and lime (Citrus amblycarpa) peels contain various phytochemical compounds, including those are reported to have metabolic and cytoprotective properties, but their combined nanoparticle formulation in cigarette smoke-exposed diabetes cases remains unexplored. Therefore, this study was conducted to evaluate the synergistic effect of mango-lime peel nanoparticles (NanoMC) on glycemic control, lipid profiles, and pancreatic cell damage in cigarette smoke-exposed diabetic rats.METHODS: Mango and lime peel ethanol extract was prepared into nanoparticles using emulsion-solvent evaporation method and followed by sonication and homogenization, and subsequently divided into three concentration groups. Rats were diabetic-induced and smoke-exposed, before being treated with/without Simvastatin + Glibenclamide, 20, 40, or 80 mg/kgBW NanoMC. Parameters including blood glucose, insulin, homeostatic model assessment of insulin resistance (HOMA-IR), homeostatic model assessment of beta-cell function (HOMA-β), hemoglobin A1c (HbA1c), and lipid profiles were then measured. To evaluate pancreatic cell damage, the pancreatic tissue was collected, fixed, and then evaluated by using Hematoxylin and Eosin (H&E) staining.RESULTS: NanoMC administration significantly reduced blood glucose, HbA1c, triglycerides, cholesterol, and low-density lipoprotein (LDL) levels (p<0.05). NanoMC treatment also increased insulin levels, HOMA-β, and high-density lipoprotein (HDL). After the treatment with NanoMC, particularly with 80 mg/kgBW concentration, fewer degenerative and necrotic cells were found compared to the other groups, showing its ability to attenuate pancreatic cell damage.CONCLUSION: The combination of NanoMC could improve glycemic status, modulate lipid profiles, and improved pancreatic cell damage in diabetic rats exposed to cigarette smoke.KEYWORDS: Mangifera indica, Citrus amblycarpa, nanoparticles, diabetes mellitus, cigarette smoke, lipid profiles, pancreatic histopathology
