Abstract

Excessive glutamate may give neurotoxic effects and contribute to Autism spectrum disorder(ASD). In this study, we investigated prolonged exposure effects of 10 µg/mL Monosodium Glutamate (MSG) on intracellular calcium level, bax, bcl-2, ratio of bax/bcl-2 genes expression, caspase-3, apoptosis of brain cells and stereotypic behavior of Zebrafish (Danio rerio) larvae at early developmental stages. Genes expression were determined by real time PCR, caspase-3 using ELISA, intracellular Ca2+ and apoptotic cells of brain using confocal microscopy, locomotor activity by using crossing lines assay whereas stereotypic behavior by circle swimming. The results indicated that MSG exposure increased brain bax and bcl-2; and caspase-3; intracellular Ca2+; and apoptosis; stereotypic behavior; and decreased locomotor activity. Termination of MSG treatments resulted in recovery of bax, bcl-2, caspase-3 basal levels and stereotypic behavior. In conclusion, MSG exposure at early embryonic stage increased brain cell damage and risk of behavior changes.

Abstract

Objective: To determine the effect of Bryophyllum pinnatum treatment in modulating immune response and the pregnancy outcomes of pregnant pristane-induced lupus mice model.Methods: Sixteen Balb/c mice were intraperitoneally injected with single 0.5 cc pristane to induce lupus manifestations. After 12 weeks of injection, mice were mated and considered as gestational day 0 (GD0). Mice were divided into 4 groups based on the dosages of Bryophyllum pinnatum: control (no treatment), B1 (10.5 mg/kg), B2 (21 mg/kg), and B3 (42 mg/kg). The treatment was given orally every day started from GD9 until 9 days. At the end of the study, blood pressure and fetal size were measured. Serum anti-dsDNA and urine albumin levels were measured by ELISA. Spleen T helper (Th) and mature B cells percentages were measured by flow cytometry.Results: Administration of Bryophyllum pinnatum reduced the percentages of Th1 (p=0.006), Th2 (p=0.005), Th17 (p=0.000), and mature B cells (p=0.007) in dose-dependent manner. B1 and B2 had significantly lower of systolic blood pressure compared to control (p=0.026 and p=0.022 respectively). Significantly lower of anti-dsDNA levels were found in B1 group compared to control (p=0.014). However, no significantly different of urine albumin levels were found between groups. Bryophyllum pinnatum also significantly increased the fetus body weight in dose-dependent manner (p=0.000).Conclusion: Treatment of Bryophyllum pinnatum could improve the pregnancy outcome and modulate the immune response in pregnant pristane-induced lupus mice. Therefore, Bryophyllum pinnatum is a potential herb which can be developed as an immunosuppressive agent in the future.

Abstract

Sebagian besar masyarakat masih belum mengetahui manfaat yang terkandung pada susu kambing, masyarakat masih menganggap bahwa susu sapi merupakan satu-satunya sumber nutrisi hewani yang memiliki kandungan protein, gizi, dan manfaat yang sangat besar bagi kesehatan. Untuk itu perlu dilakukan sosialisasi tentang pemanfaatan susu kambing karena kurangnya pengetahuan masyarakat sehingga menyebabkan rendahnya daya beli masyarakat. Tujuan dari kegiatan ini adalah untuk menjalin rintisan kerjasama dengan sekolah sebagai salah satu tempat pemasaran produk susu kambing serta orang tua siswa-siswi dalam meningkatkan daya konsumen masyarakat terhadap susu kambing dan memberikan informasi yang berlandaskan data ilmiah tentang manfaat susu kambing. Sosialisasi dilaksanakan melalui pemaparan pemateri tentang manfaat dan pentingnya mengonsumsi susu kambing dalam kehidupan sehari-hari serta pemanfaatan susu kambing menjadi produk nutrisi sehat (susu kambing pasteurisasi). Hasil dari kegiatan sosialisasi adalah para guru dan siswa-siswi bertambah pengetahuannya tentang pentingnya mengonsumsi susu kambing sebagai pangan sehat bernutrisi dan antusias dalam membuat produk-produk sehat berbahan dasar susu kambing.

Abstract

Background : Preeclampsia is a condition where the blood pressure is increasing (≥140/90 mmHg) followed by proteinuria (≥300 mg/24 hour or ≥1+ in stick test) in pregnant women after 20 weeks of pregnancy. Released placenta factor cause the damage in endothelial cells which lead to the excessive increase on pro inflammation factor interleukin 6 (IL-6). Objective: This study aimed to investigate whether level IL-6 on normal HUVECs with normal HUVECs induced by plasma from preeclamptic patients. Research Method: True experimental research using posttest only control group approach, conducted in the laboratory with invitro. HUVECs culture was divided into 5, that were normal HUVECs without treatment (K-); HUVECs preeclampsia model (K+). Measurement on the levels of IL-6 used ELISA (Enzyme-Linked Immunosorbent Assay). Result: Independent sample t-test showed that there were significant differences (p=0.000<α) on the average of IL-6 levels between the negative control group (1.64±0.12 pg/ml) and the positive control group (31.64±11.69 pg/ml). Conclusion: Adding the patient’s preeclampsia plasma into normal HUVECs can increase levels of IL-6. Keyword: Patient’s preeclampsia plasma, HUVECs, and IL-6.

Abstract

Stroke is a cerebrovascular disease that is often found in developed countries and currently also presents in developing countries, and one of them is in Indonesia. The high prevalence of stroke in Indonesia is affected by several risk factors such as obesity, lack of physical activity, unhealthy diet, smoking, high blood pressure, elevated blood sugar, and elevated blood lipid profile. The purpose of this study was to analyze the relationship between predictors of hemorrhagic stroke such as obesity, blood pressure, blood sugar and blood lipid profile with mortality in hemorrhagic stroke patients in RSD dr. Soebandi Jember. This research was a quantitative research with a retrospective approach. The number of samples was 101 medical record data. Univariate analysis was used to identify the description of each variable, a chi-square test was applied to analyze the correlation between variables, and the logistic regression analysis was applied to identify the most dominant factor influencing mortality of hemorrhagic stroke. The result of bivariate test showed p-value of obesity= 0,039, p-value of blood pressure= 0,478, p-value of blood sugar= 0,04, p-value of blood lipid profile= 0,026. Logistic regression obtained p-value for obesity= 0.043, OR = 2.689; p-value of blood sugar= 0,042, OR = 2,656; p-value of blood lipid profile= 0.069, OR = 3,749. There was a significant association between obesity, blood sugar, and blood lipid profile with mortality in hemorrhagic stroke patients. Obesity and blood sugar are predictors that can be used to predict mortality in hemorrhagic stroke patients, where obesity is the most dominant factor affecting mortality in hemorrhagic stroke patients.

Abstract

The increase prevalence of speech and language disorders, which greatly affects the quality of life of a child. The role of educational institutions, especially teachers, is needed in early detection of these disorders. Early detection abilities must be based on the ability to understand a child's speech and language development. Community Service Activities are carried out at the Idayu 2 Malang Special School, by providing training to teachers on speech and language development and early detection methods. The method used is the provision of material followed by an examination of the students status growth ad development. There was an increase in the teacher's ability after the activity was carried out, which was marked by an increase in the average pretest and postest results: 61.15 to 86.54. On examination of the student's growth status, the nutritional status of the majority of students was found to be good nutrition (61%), but the short stature rate was also quite large (49%). The condition of children with special needs with physical limitations greatly affects their growth and development, so that the role of the environment is needed for optimal development, including parents and educational institutions.

Abstract

Lupus is a systemic autoimmune rheumatic disease with mild to severe clinical features and can lead to hopelessness if not treated properly. The purpose of this activity is to give a beauty class and see how it affects the hopelessness of lupus patients in honor of World Lupus Day. Methods: Organizing World Lupus Day activities such as experts seminars on lupus at a glance, being friendly with lupus, and staying beautiful with lupus; a musical drama and dance performance by a lupus support group with the theme “You Are Beautiful”; and a lupus beauty class. The activity was completed in one day in May 2018 by collecting pre-test and post-test data on the incidence of hopelessness in lupus patients using the Beck Hopelessness Scale (BHS). A total of 100 lupus patients participated in the activity, and all of them underwent pre- and post-testing. The Wilcoxon Signed Rank Test with α=0.05 was used to analyze the collected data. Results: All participants expressed delight and satisfaction with their participation in this activity. The mean BHS scores before and after the beauty class were 8.62+2.68 and 6.36+1.59, respectively, with a significant difference (p=0.000). Conclusion: Beauty class is a fun way for lupus patients to feel less hopeless about their illness. Every year, an activity to commemorate World Lupus Day is required, which is beneficial for lupus patients.

Abstract

ABSTRAKHiperlipidemia merupakan penyebab dari berbagai penyakit degeneratif. Mengonsumsi air perasan lemon menjadi tren di Indonesia beberapa tahun ini. Pada penelitian ini, dilakukan perbandingan efektivitas air perasan lemon dan jeruk nipis. Tujuan dari penelitian ini adalah untuk menganalisis perbedaan profil lemak darah: kolesterol total, low density lipid (LDL), very low density lipoprotein (VLDL), dan trigliserida (TG) pada tikus yang diberi diet tinggi lemak, air perasan lemon, dan jeruk nipis. Penelitian ini menggunakan 32 tikus jantan galur Wistar yang dibagi menjadi 8 kelompok dosis perlakuan air perasan lemon dan jeruk nipis. Didapatkan hasil bahwa tidak ada perbedaan kadar LDL (P0.151; p<0.05), VLDL (P0.187; p<0.05), TG (P0.216; p<0.05), namun ditemukan perbedaan signifikan pada kadar kolesterol total (P0.003; p<0.05). Hewan coba yang diberi perlakuan jeruk nipis memiliki sebagian besar rerata kadar lemak yang tidak berbeda signifikan dengan kelompok perlakuan lemon. Meskipun demikian, kolesterol total pada kelompok jeruk nipis lebih rendah dibandingkan kelompok lemon. Konsumsi jeruk nipis tampaknya memiliki kemungkinan menjadi alternatif dari lemon yang lebih murah dan terjangkau untuk mencegah profil lemak yang buruk.   Kata kunci: lemon, jeruk nipis, kolesterol total, LDL, VLDL, TGABSTRACTHyperlipidemia causes various degenerative diseases. Recently, consuming lemon juice has become a trend in Indonesia to improve blood lipid profiles. This study compared the effectiveness of lemon juice and lime juice. This study aimed to determine differences in blood lipid profile that contains total cholesterol (TC), low-density lipoprotein (LDL), very-low-density lipoprotein (VLDL), and triglycerides (TG) in rats given High Fat Diet (HFD), lemon juice, and lime juice. This study used 32 white male Wistar rats (Rattus norvegicus) divided into eight treatment groups of lemon and lime juice doses. The results showed no significant differences in LDL (P0.151; p<0.05), VLDL (P0.187; p<0.05), and TG (P0.216; p<0.05), but there were significant differences in total cholesterol levels (P0.003; p<0.05). The experimental animals receiving lime juice had mean scores of lipid levels that were not significantly different from those with lemon treatment. However, the total cholesterol levels among groups given lime juice showed lower levels than the control and lemon treatment groups. Lime consumption seems possible to be a cheaper and affordable alternative to lemon to prevent lipid profile aberrations.Keywords: lemon, lime, TC, LDL, VLDL, TG

Abstract

Introduction: Vulvovaginal candidiasis (VVC) is an infection of the vaginal mucosa that extends to the vulva due to excessive growth of Candida albicans, causing a progressive decrease of Lactobacillus spp., as the main of normal flora. Decrease in the number of Lactobacillus spp. causes a reduction of lactic acid production that it was causing increasing vaginal pH. The increase in pH is due to the progressive growth of C. albicans colonies in the vaginal mucosa. In addition, the mechanism of candidiasis is also influenced by the secretion of virulence factors by C. albicans. Secreted Aspartyl Protease-5 (SAP5) is the dominant virulence factor that plays a role in changing the morphology of yeast cells into hyphae. This study aims to identify the correlation between SAP5 levels and vaginal pH. Material and Methods: This research was conducted in vivo using a sample of Rattus norvegicus with a posttest randomized experimental group design approach. This study has two groups, i.e., Negative Control (NC) and Positive Control group (PC). SAP5 and pH samples were taken from vaginal fluids and then measured using ELISA and a digital pH meter. The statistical analysis used was Pearson Correlation to analyze the correlation between SAP5 and pH levels. Results: There was a strong correlation (R=0.846) that means the increase of SAP5 is related to the increase of vaginal acidity in the PC group. Abstract

The electric cigarettes are said safe for health, in fact there are toxic substances, carcinogens, and unstable nicotine, it is suspected to cause low birth weight in pregnant women who are exposed to electric cigarette. Nicotine in electric cigarette is free radicals, so to ward off free radicals is required antioxidant. Infused lemon contains antioxidants that are higher than lime and are believed to resist free radicals from electric cigarettes. The purpose of research to determine the influence of infused lemon water in preventing low birth weight in pregnant rats (Rattus novergicus) exposed by electric cigarettes. The study used 30 pregnant rats, divided into 5 groups; Negative control (KN), positive control (KP), Treatment (P1), Treatment (P2), Treatment (P3). Using electric cigarette liquid 9 mg, and the difference in the weight of lemon in the making of infused water (P (1): 25g, P (2): 50g, P (3): 100g lemon + 100ml water). The treatment is given on day 1 ovulation to day 18, sacrificed on the 19th day ovulation then calculated the average weight of the baby. The average weight baby KN: 5.502g, KP: 2.821g, P (1): 4.301g, P (2): 4.888g, P (3): 5.435g. The result of One-way ANOVA indicates there is a significant difference in the treatment group with the value P=0,000 (P<0.05). The KP group differs significantly from KN. Group P (1,2,3) is significantly different from KP. The KN group is significantly different from P (1), but does not differ significantly from P (2, 3). The group P (1) differs significantly with P (3). Infused lemon juice can prevent the occurrence of low birth weight in the child of the pregnant rats that’s exposed by electric cigarettes especially in group P (3).
