Abstract

Abstract

Turmeric (Curcuma longa) is widely used as herbal medicine, not an exception by pregnant women. Turmeric consumption by expectant mothers requires standard dose, because of its antiangiogenic effect could be harmful on placentation process and embryonic development. This experiment was undertaken to determine the effect of different concentrations of turmeric decoctum to the expression of Vascular Endothelial Growth Factor Receptor-2 (VEGFR-2) and Angiopoietin 1 (Ang-1) on the 48-hours-old chicken embryo. In this study, turmeric was extracted using decoction method to mimic the common method as adopted by people. The turmeric decoctum were freeze dried into a powder form and was used in preparing the stock solution for 200 ppm (P1), 300 ppm (P2), and 400 ppm (P3) as experimental treatments. The control group (P0) received 2% DMSO without turmeric decoctum. These were administered on the yolk sack of 16 hours incubation of fertile chicken egg by number of 200 µL. After 48 hours incubation, the expression of VEGFR-2 and Ang-1 on the chicken embryo were counted by ImageJ software. The results revealed that there is no significant effect of turmeric decoctum to the expression of VEGFR-2 and Ang-1. This suggested that turmeric decoctum was safe up to 400 ppm on chicken embryo.

Abstract

Embryonic development forms three layers of germinativum that is endoderm, mesoderm and ectoderm. At the beginning of intrauterine fetus exchange of substances through diffusion, but in line with the development of the embryo, nutrition is not obtained through diffusion alone. Endoderm lining mesoderm cells will then be formed angioblasts as an early sign of vascularity. In the development of flexi cranial can be known from the development of the brain and heart which is characterized by bending the head of the embryo and decreased heart from the cranial down. The development of a normal vascular system will have an effect on organogenesis and morphogesis processes. If there are disturbances in the process of growth and development at that stage can cause congenital abnormalities. Xanthones mangosteen peel is rich in benefits, but in research on the effects of xanthones on embryos is still very limited and has not been able to explain their effects on the condition of the fetus. The study was conducted with the aim to determine the effect of mangosteen peel extract inhibiting the flexi cranial of 48 hours chicken embryos using doses of 100 µg/mL, 150 µg/mL and 200 µg/mL. Mangosteen peel extract is injected into chicken eggs less than 7 days after oviposition then incubated for 48 hours. The results showed that mangosteen peel extract inhibited flexi cranial of chicken embryos with Chi-Square test results (p = 0.002). The conclusion of the study was that exposure to mangosteen peel extract inhibited flexi cranial and gave significant results.

Abstract

The most common cause of patalogis leucorrhoea is infection, one of which is Vulvovaginal Candidiasis (VVC) caused by fungus, while 80% -90% is caused by Candida albicans. Approximately 90% of young women in Indonesia have the potential to experience vaginal discharge because Indonesia is a tropical climate. Alternative treatment for antifungal drugs is needed through the exploration of natural material sources that have the potential as antifungals to deal with this infection safely and easily. The design used in this study was experimental research by in vitro using the dilution method of the tube. Treatment concentrations are 45%; 40%; 35%; 30%; 25%; 20%, 15% and 0% with Candida albicans 104 CFU / ml samples from the microbiology lab of the medical faculty Brawijaya University and repeated 3 times. The result showed that at all concentrations, growth was still obtained in agar media so that it could be denied that the boiled water of avocado seeds had no effect on the growth of Candida albicans fungal colonies in vitro because there were still fungal colonies on the plates.

Abstract

Approximately 70-75% of reproductive women have experienced vulvovaginal candidiasis at least once, and 40-45% will have recurrences. Candida albicans is the most etiology of vulvovaginal candidiasis and is able to form a biofilm that can lead to antifungal resistances and recurrences. One of the natural products that have an anti biofilm effect is propolis. This study expected that propolis from Lawang can be one of the anti-biofilm agent candidates for antifungal resistant cases. This study aimed to know the anti-biofilm effect of propolis ethanol extract on cell adhesion index, morphology, and viability of Candida albicans cell on biofilm formation. The adhesion index was counted on 100 epithelial cells under light microscopy (1000x). Morphology was seen using light microscopy (400x). Cell viability was examined by CFU assay. At 12.5% concentration of propolis ethanol extract, adhesion index decreased (p=0,000), and hyphal growth was inhibited. Colony growth decreased at 2.5% concentration and was not seen at 10% concentration of propolis ethanol extract (p=0,000). These results indicated that propolis ethanol extract could decrease adhesion index, failed the Candida albicans morphology transition from yeast to hyphal, and decreased Candida albicans cell viability on biofilm formation. Propolis ethanol extract is likely to be one of the alternatives to recurrent vulvovaginal candidiasis treatment, especially caused by Candida albicans biofilm formation.

Abstract

Introduction: Preeclampsia has two stages: improper placental and maternal circulation. Abnormal trophoblast invasion causes uteroplacental ischemia and hypoxia. Ischemia in the placenta produces endothelial cell dysfunction, which is defined by a change in endothelial cell activity to a reduced ability to vasodilate (decreased eNOS) and prothrombotic conditions (decreased PECAM-1). Reduced maternal eNOS activity and PECAM-1 can cause preeclampsia. Pravastatin is the statin class's most hydrophilic medication, and it limits placental transfer. Pravastatin can reduce endothelial dysfunction by targeting pleiotropic effects in pregnancy. The aim was to show the effects of pravastatin on the eNOS and PECAM-1 expression in the placenta of preeclampsia rat model. Materials and Methods: This is a true experimental study with only a post-test and a control group design. The sample was biological material in the form of placental tissue from a pravastatin-treated preeclampsia rat model (using L-NAME exposure). This research was divided into five groups, each with five samples. The parameters studied were eNOS and PECAM-1 expression. Results: The Shapiro-Wilk test result was significant (p>0.05). Annova tests on eNOS (p=0.000) and PECAM-1 expression (p=0.000) confirmed the hypothesis. The Tukey test showed significant differences in eNOS (sig. 0.001) and PECAM-1 (sig. 0.000) expression between normal and preeclampsia rats. Positive controls and treatment groups P1, P2, and P3 all showed significant changes in eNOS and PECAM-1 expression. Pravastatin dose considerably increased eNOS (p=0.015; r=0.536) and PECAM-1 (p=0.000; r=0.734) expression.. Conclusion: Pravastatin has been shown to increase eNOS and PECAM-1 expression in the placenta of preeclampsia rat model.
