Abstract

ABSTRACTTransformation Strategy for Indonesian Indica Rice in Attempt to Discover Drought-TolerantRelated Genes Using of Transposon Ac/Ds. Attempt to identify, isolate the gene, and study forgene function for several agronomical traits have been done including some drought toleranttraits. Japonica rice cultivars have been used due to its higher efficiencies compared withindica cultivars. Two plasmids namely pNU400 and pUR224 were used to generate mutants ofthese cultivars (Batutegi dan Kasalath cultivars). Those plasmids contain an element calledActivator (Ac) and Dissociator (Ds) respectively. The pNU400 contains GFP (green flourescensprotein) as a selectable marker, whereas the pUR224 contains hygromycine resistant gene andgusA as a reporter gene. Each plasmid was transformed into rice genome of Batutegi andKasalath cultivars by Agrobacterium mediated transformation using three methods oftransformation (A, B and C). The transformation method A was not suitable for both cultivars,where none of plantlets were produced from pNU400 and pUR224 plasmids. The transformationmethod B produced some plantlets from the Kasalath cultivar only using pUR224 plasmid.The transformation method C was the best method to produce transgenic plants from bothcultivars (Batutegi and Kasalath), using both plasmids (pNU400 and pUR224). The PCR analysisshowed that 19 and 9 plants of Batutegi and Kasalath contained both gusA and hpt genesrespectively. None of those plants contained of gusA gene. Southern blot analysis revealed 3independent lines from Batutegi dan 7 independent lines from Kasalath. The integration of Actransposon was analyzed based on expression gfp gene when observed under UV dark reader.This research has proved that indica rice cultivars, especially the Batutegi cultivar of Indonesianorigin, could be transformed. The cultivar could be used as plant model for the indicatransformation.Key words: transformation, drought tolerant, indica rice, Ac/Ds transposons, Agrobacterium.

Abstract

All living cells consist of phospholipids. Phospholipids are known as biosurfactants. Here we report our study of the physico-chemical characteristics of phospholipids derived from coconut endosperm as biosurfactants. We studied the aggregation and emulsification characters. The aggregation proccess was examined by optical microscope. The stability of emulsi formed was montitored at different temperature and pH. Where as emulsification capability was investigated by constructing a ternary diagram. Depending on the amount of phospholipids and additive substances the emulsification capability changes. The coconut phospholipids will form vesicles in various size (polydisperse). We noticed that coconut phospholipids emulsification stability was affected by temperature, pH, and phospholipids composition. It can be concluded that the prospect of phospholipids from coconut endosperm as biosurfactants is promising.   Keywords: coconut phospholipid, emulsification capability, aggregation, ternary diagram, vesicle.

Abstract

The economic disparity that occurred in Badung Regency, Bali was caused by the rapid development of tourism that does not align with the development of the agricultural sector in the north. This then pushed the Pelaga Tourism Village millennial to urbanize into the southern regions to continue education and to obtain employment. This research aims to analyze the role that millennials can do as cross-cutting interpreters in the development of Pelaga Tourism Village. This research uses qualitative data with two data sources: primary data and secondary data. The data collection techniques used in this research are library studies, observations, interviews, and documentation. In determining the informants is used as a purposive sampling technique. In this study, the data analysis technique used is descriptive-qualitative. The results showed that the role of the millennial generation in the development and management of tourism villages has been very minimal. This is very unfortunate considering the millennial has the ability as cross-cutting interpreters and as well as rich in innovation and much-needed creativity in the development of tourism villages. The role that can be performed by millennials in developing tourist villages is by establishing BUMDes as an organization for tourism village management. Millennials should also conduct mapping and development of several tourism potentials in each Banjar by prioritizing the aspect of diversification. At the marketing and promotion stage, millennials who are technologically literate can utilize digital media as a means to introduce and offer tourism products. Abstract

Many villas in Ubud is located around agricultural land owned by farmers who are members of Subak. There is a deal that the villas are required to provide material contributions to local institutions, including Subak. Such contributions produce caritative framework which not in line towards the sustainability of tourism development. This paper addresses to offer the guidelines of sustainable tourism practice in Ubud, Bali. To fulfill this purpose, data has been collected from observations and interviews with selected informants and then analyzed descriptively. The analysis indicated that there is dualism perspective against agriculture, which has implications towards exclusivity in-group, especially farmers and villas. Each group attempts to maintain the dualism perspective by applying various tactics, which can lead to disharmonies relations between groups. It requires social guidance in order to reduce caritative framework by creating common tourism sphere through agrotourism.

Abstract

The potential of such unique nature and culture in the village Penglipuran it is proper to be developed and introduced to the tourists so that rural communities receive economic benefits, such as the availability of additional jobs and increase income by providing services as a local tour guide, provision culinary, providing soulvenir typical village Penglipuran and receiving income in the form of receipts village onggos admission and parking. Businesses that can be taken is the form of making the tour packages of innovative rural to attract the number of tourists visiting the village Penglipuran and improving the quality of human resources supporting. To develop further Penglipuran village, Pengabdi team has managed to make the packaging countryside tour package entitled "Village Tracking Tour" and Penglipuran Rural Cycling tour "second package is already manifested in the form of brochures. Realization of two packets of agrotourism has been preceded by a series of activities such as: 1) justify trajectory tracking path and cycling (something to see, something to do and something to learn) that will be bypassed by tourists in the countryside Penglipuran village and surrounding areas, 2) how to make tour event (itinerary), 3) how to determine the cost and the price of travel and 4) creating rural tourism package packing in the form of brochures.

Abstract

Bali Tourism has grown for the hundred years, but there has never been a mapping of its developmental stages and its future predictions. This article examines the development of tourism in Bali in the past, present, and future predictions, as well as discourse in each stage. Data were collected by literature study, observation, and in-depth interviews with diachronic informants. The main theory used is critical evolution and genealogy. This article offers a model of tourismemorphosis as a stage of tourism development along with its paradigms including the introduction stage, reaction stage, institutional stage, and compromise stage. The prediction of future Bali tourism development is to remain at the compromise stage with the transtourism paradigm that is paradigm with the effort of adjusting all tourism components to respond to the big and rapid changes of tourism globally. Always compromise, Bali tourism stakeholders in the future are still demanded to be creative in the future, customary government, market dynamics, IT development, customs, and religious rituals to survive or even move forward.

Abstract

This article examines the local community participation in the Candidasa tourism area, Karangasem, Bali. The objectives are to identify the capital exchange practice of local communities with the tourism industry and then determine the type of local communities’ participation in the development of Candidasa tourism area. The community based tourism approach adopted to understand the phenomenon of local community participation. Based on observations and interviews can be addressed that the communities surrounding the Candidasa relatively have an exhaustive capital. However, the comprehensiveness of such capital is not accompanied by the proper capital exchange strategies by indigenous village institution as a representation of local communities. The local communities only get relief materials from tourism as a result of the capital exchange. Accordance with the practice of capital exchange, it can be said that this type of local community participation is included in the participation for material incentives. This type of community participation is still relatively limited, so it necessary to improve the their involvement into tourism development in Candidasa.

Abstract

This article is aimed to reinterpret the ritual of Ketupat War based on practical consciousness into reflexive consciousness to preserve agricultural tradition in Kapal Village, Badung Regency. This consciousness could be created by deconstructing the pragmatic materialistic perspective by promoting local knowledge Tri Hita Karana, philosophy on the harmonious relationship among human being, human and nature, and God. This research applied a descriptive qualitative method and structuration approach from Giddens to explain the ritual Perang Ketupat as reflexive consciousness. The research shows that the local people at Kapal Village interpret this ritual as practical consciousness without knowing the function and the meaning of the ritual. It means that ritual is only performed annually as a routine tradition. Therefore, reflexive consciousness needs to be nurtured through constructing the community`s awareness of this ritual as a tourist attraction, so that they can get the economic benefit (touristic meanings). In other words, tourism becomes a melting-pot producing space of the triple sections among nature, culture and tourism which is called Eco-Cultural Tourism Landscape (ECTL).

Abstract

Tenganan Pegringsingan Village Tourism is located in Karangasem Regency, which has many ancestral cultures and traditions that are still held today. One of the famous traditions of the Tenganan Pegringsingan Village Tourism is the tradition perang pandan (Mekare - kare). Perang pandan has its own uniqueness that can only be found in Tenganan Village, from this uniqueness that makes many tourists who visit the Tenganan Pegringsingan Village Tourism just to witness the uniqueness of the perang pandan ritual. This study sought to find diachronic developments in the implementation of the perang pandan ritual, before and after the establishment of Tenganan Pegringsingan Village into a Village Tourism, using the over-time comparative method. The results of this study indicate, first seen from the indicator of time, participants who followed the tradition of the perang pandan ritual experienced a change. Secondly seen from the room indicator, the implementation of the perang pandan ritual is still permanent, cannot be changed. Third, judging from the level indicator, the perang pandan ritual has begun to move to a more complex level than before. and the last seen from the composite indicator, time, space and level, the perang pandan ritual has begun to be packaged into a tourist attraction, in the form of a festival tenganan that is routinely carried out every year.
