Abstract

Penelitian ini dilakukan bersesuaian dengan program penelitian prioritas Universitas Negeri Manado terutama untuk menjawab berbagai tantangan yang dihadapi bangsa Indonesia dalam mengatasi berbagai problem kesehatan antara lain dalam hal pemenuhan obat-obatan antikanker, telah didisain suatu riset dasar untuk mengeskplorasi potensi molekuler yang terkandung dalam asosiasi mikroalga dan inangnya (Ascidiaceae) dengan pendekatan bioteknologi. Tujuan utamanya mengisolasi dan karakterisasi gen-gen pada mikroalga yang berasosiasi dengan Ascidiciae yang memiliki potensi sebagai sediaan farmasetika, mendapatkan biomassa mikroalga dengan kultivasi, dan mengembangkan metode scalling up perbanyakan mikroalga di luar inang. Tahun pertama penelitian ini isolasi dan kultivasi mikroalga yang berasosiasi dengan Ascidiaceae. Untuk itu telah diadakan pengambilan sampel Ascidian di Pantai Malalayang teluk Manado pada koordinat 1°27?39?LU dan 124°17?31?BT, isolasi mikroalga dan upaya kultivasi di medium Hirata; isolasi DNA mikroalga dan konfirmasi molekuler spesies mikroalga dengan gen penanda filogeni. Walaupun tidak ditargetkan dalam pelaksanaan riset tahun I, namun mendahului mendapatkan gen-gen yang menyandi produk yang memiliki potensi untuk dikembangkan sebagai sediaan farmasi, telah pula dilakukan deteksi metabolomik terhadap sampel mikroalga yang diambil dari alam maupun yang dari hasil kultivasi. Sebagai metabolit dengan berat molekul kecil, komponen metabolomik dapat dimaknai sebagai produk akhir ekspresi gen, sehingga penentuan secara kuantitatif maupun kualitatif dapat menyediakan informasi status biokimia mikroalga karena itu dapat dipakai untuk memantau fungsi gen. Temuan yang diperoleh sampai tahap I berupa hasil kultivasi mikroba simbion, jenis-jenis mebolit dan analisis DNA awal. Luaran penelitian yaitu (1) draft paper untuk publikasi ilmiah di jurnal internasional bereputasi (terindeks Scopus); (2) pemakalah dalam temu ilmiah nasional dan/atau internasional.

Abstract

Penelitian ini dilatar belakangi masalah dengan adanya penggunaan zat pewarna sintetis pada ikan cakalang asap , adanya penggunaan bahan pengawet pada ikan cakalang asap dan ikan cakalang fufu merupakan salah satu produk Sulawesi Utara yang banyak dikonsumsi masyarakat. Penelitian ini dilakukan untuk mengetahui pengaruh zat pewarna alami dari kulit buah naga merah terhadap produk ikan cakalang asap dan mengetahui apakah konsumen tertarik dengan produk ikan cakalang asap yang sudah diberikan zat pewarna alami kulit buah naga merah.  Metode penelitian ini adalah eksperimen, dengan rancangan percobaan rancangan acak lengkap. Dimana dalam rancangan acak lengkap ini digunakan 5 taraf perlakuan yang diulang sebanyak 3 kali, sehingga total unit perlakuan sebanyak 15 sampel. Konsentrasi uji kontrol, 5%, 10%, 15% dan 20% pemberian zat pewarna alami kulit buah naga merah. uji toksisitas metode BSLT, uji fisik organoleptik metode hedonik. Hasil penelitian menunjukkan uji toksisitas ekstrak kulit buah naga merah menunjukkan bahwa ekstrak kulit buah naga merah tidak toksik dengan nilai LC50 1.1529,042 mg/L, hasil uji organoleptik menunjukkan bahwa pemberian ekstrak kulit buah naga merah pada ikan cakalang asap untuk tingkat kesukaan konsumen pada parameter kenampakan, warna dan rasa yang paling berpengaruh nyata adalah konsentrasi 5% dan 20%.

Abstract

Ranowangko II beach is famous for its beautiful beaches and is a natural habitat for macroalgae, including Caulerpa racemosa and Sargassum polycystum. Despite its great potential and abundance, information about the nutritional content of both types of algae, especially in the Ranowangko II beach area, is still limited. This study aimed to determine the nutritional content (water content, ash content, protein, lipid, crude fiber, carbohydrate) of green algae Caulerpa racemosa and brown algae Sargassum polycystum in Ranowangko II beach. The type of research used is descriptive quantitative to determine the proximate nutrient content of two types of algae tested by proximate analysis in the laboratory. Based on the study results, there were variations in the nutritional content of the two types of algae tested. Green algae Caulerpa racemosa has a moisture content of 71.29%, ash content of 6.90%, protein of 2.78%, fat of 1.34%, crude fiber of 3.53%, and carbohydrates of 17.69%. Brown algae Sargassum polycystum has a water content of 83.23%, ash content of 1.82%, protein of 9.93%, fat of 1.72%, crude fiber of 11.18%, and carbohydrates of 3.30%. The conclusion obtained after conducting this research is that Caulerpa racemosa (green algae) and Sargassum polycystum (brown algae) have different nutritional content.

Abstract

This research is motivated by the lack of studies and absence of research data on the diversity of Echinoderms on the Beach of Ranowangko II Village, Kombi District, Minahasa Regency. The aim of this study is to determine the diversity of the phylum Echinodermata found on the Beach of Ranowangko II Village, Kombi District, Minahasa Regency. Sampling in this research used the quadrat transect method with 3 transect lines, each line being 100 m long, and 10 quadrats, each measuring 1 x 1 m². The results of the study found 4 classes and 5 species including the class Asteroidea (Protoreaster nodosus), class Ophiuroidea (Ophiocoma scolopendrina), class Echinoidea (Tripneustes gratilla), class Holothuroidea (Holothuria atra & Holothuria vagabunda), and there were 21 individuals, and the dominant species is Holothuria atra from the Holothuroidea class of which there are 11 individuals, indicating that the diversity index (H’) of the phylum Echinodermata on the location is moderate with a value of 1.204. These findings offer initial insights into the condition of the local ecosystem, classified as moderate, and may serve as a foundational reference for long-term ecological monitoring.

Abstract

This research aims to provide information on the types, and diversity of macroalgae in seagrass ecosystems and water conditions in the waters of Ranowangko Dua Village Beach, Kombi District, Minahasa Regency. Observations of macroalgae in seagrass ecosystems were carried out using the quadrant transect method (1x1m) along 100 meters with an observation distance of every 10 meters per quadrant using 3 transect lines with a distance of 10 meters. The results found 12 species of macroalgae namely Halimeda Macroloba, Ulva intestinalis, Caulerpa Lentillifera, Neomeris Annulata, Halimeda Incrassata, Codium Arabicum, Turbinaria Ornata, Padina Minor, Acanthophora Spicifera, Amphiroa Fragillisima, Galaxaura Rugosa, and Mastophora Rosea. The diversity values of transects 1, 2, and 3 are 1.745; 1.378, and 1.294, respectively, included in the medium category. The absolute density value was highest in transect 2 (12.1), and lowest in transect 3 (7.5). The uniformity values of transects 1, 2, and 3 were 0.702; 0.555, and 0.521, respectively, including moderate (0.50 and less than 0.75). The dominance values of transects 1, 2, and 3 are 0.228; 0.348, and 0.362 respectively, which fall into the stable category or there is no dominating species. The results of physical and chemical parameters show the temperature value of 31.14°C, pH 8.99 is stable, and Dissolved Oxygen 7.62 mg/l is not polluted. In contrast, the salinity value of 35.14 ppt is not optimal, the brightness is in the low category of 0.38 m, and the turbidity value of 103 NTU is in the high category. Abstrak Penelitian ini bertujuan untuk memberikan informasi tentang jenis, keanekaragaman makroalga pada ekosistem padang lamun dan kondisi air di perairan Pantai Desa Ranowangko Dua Kecamatan Kombi Kabupaten Minahasa. Pengamatan makroalga pada ekosistem lamun dilakukan dengan metode transek kuadran (1x1m) sepanjang 100 meter dengan jarak pengamatan setiap 10 meter per kuadran menggunakan 3 line transek dengan jarak 10 meter. Hasil yang ditemukan 12 species makroalga yaitu Halimeda Macroloba, Ulva Intestinalis, Caulerpa Lentillifera, Neomeris Annulata, Halimeda Incrassata, Codium Arabicum, Turbinaria Ornata, Padina Minor, Acanthophora Spicifera, Amphiroa Fragillisima, Galaxaura Rugosa, dan Mastophora Rosea. Nilai keanekaragaman transek 1, 2 dan 3 masing-masing 1,745; 1,378 dan 1,294 termasuk dalam kategori sedang. Nilai Kepadatan mutlak tertinggi pada transek 2 (12,1), dan terendah pada transek 3 (7,5). Nilai keseragaman transek 1, 2 dan 3 masing-masing 0,702; 0,555 dan 0,521 termasuk sedang (0,50 dan kurang 0,75). Nilai dominansi transek 1, 2 dan 3 masing-masing 0,228; 0,348 dan 0,362 dimana masuk dalam kategori stabil atau tidak ada jenis yang mendominasi. Hasil parameter fisik dan kimia menunjukkan nilai suhu 31,14°C, pH 8,99 tergolong stabil, Oksigen Terlarut 7,62 mg/l tidak tercemar, sedangkan nilai salinitas 35,14 ppt tidak optimal, kecerahan dalam kategori rendah 0,38 m, dan nilai kekeruhan 103 NTU dalam kategori tinggi. Abstract

Suatu kegiatan Pemberdayaan Kemitraan Masyarakat (PKM) telah dilaksanakan untuk memberdyakan masyarakat berwirausaha tani terpadu guna menunjang pariwisata pantai desa Marinsow Kecamatan Likupang Timur Minahasa Utara. Metode yang digunakan adalah kombinasi metode kombinasi metode Andragogik dan Pilot Project aplikasi teknlogi tepat guna usaha tani terintegrasi dengan mengoptimalkan lahan dan usaha tani Mitra berada persis di tepian jalan utama menuju kawasan wisata pantai Paal desa Marinsow. Berdasaran kegiatan sampai saat ini diperoleh hasil yaitu, (1) terfasilitasi upaya menghadirkan satu unit cultivator untuk revitalisasi pengolahan lahan pertanian, (2) terfasilitasi pengadaan mulsa plastic hitam perak (MPHP) dan peralatan pertanian lainnya untuk menunjang usaha budidaya tanaman pangan, (3) terfasiitasi pemasangan tower dan pengadaan satu unit tangki air untuk pemenuhan irigasi lahan pertanian saat musim kemarau, (4) terfasilitasi peningkatan pengetahuan dan keterampilan mitra dalam pengolahan lahan menggunakan cultivator, (5) terfasilitasi peningkatan pengetahuan dan keterampilan mitra memproduksi dan mengemas aneka produk kacang tanah menggunakan vacuum sealer dengan plastic vacuum emboose. The application of the conversion results from Eco-Enzyme into dishwashing soap, the fermentation process lasts for one months, where the resulting liquid is dark brown in color and has a very strong sweet and sour fermented aroma. There is a lack of public knowledge about the very profitable benefits of Eco-Enzyme, such as as a multi-purpose cleaner as a multi-functional liquid which can be converted, one of the ways, into dishwashing soap. This research aims to determine the application of the results of converting Eco-Enzyme into dishwashing soap. This research used experimental methods and a research period of 3 months. This Eco-Enzyme is made with a ratio of organic ingredients: sugar: water 3:1:10. The Eco-Enzyme harvest is converted into dishwashing soap by adding lerak and Methyl Ester Sulfonate (MES). Eco-Enzyme conversion soap was then applied to several treatment groups: plastic plates, cans and glass. The results of the application show that the dishes washed with Eco-Enzyme using lerak have not completely removed dirt, oil and odors in each treatment, whereas Eco-Enzyme using Methyl Ester Sulfonate (MES) when compared with Sunlight brand liquid soap sold on the market shows that there are similarities both in terms of cleanliness, tapestry and smell. The research conclusion is that the application of the Eco-Enzyme conversion results has significant potential to be developed into an active ingredient for environmentally friendly dishwashing soap.

Abstract

Suatu kegiatan pengabdian kepada masyarakat pendanaan skema Pemberdayaan Kemitraan Masyarakat (PKM) telah dilaksanakan di desa Libas Pulau Bangka Kecamatan Likupang Timur Minahasa Utara dalam upaya mengatasi masalah rendahnya nilai ekonomi jambu mete karena produk utamanya hanya kacang mete kering dengan kemasan seadanya serta pemasaran langsung diantarkan kepada pedagang pengumpul di daratan Likupang. Pengemasan yang hanya menggunakan kantung pastik direkatkan dengan panas api lilin tentunya tidak dapat menjamin kualitas produk dan dapat bertahan lama. Melalui kegiatan PKM telah diaplikasi teknologi pengolahan jambu (kacang) mete dengan kemasan standar dan beragam produk olahan kacang mete serta dengan pemasaran digital. Metode pendekatan yang diterapkan yaitu diaplikasi Teknologi Tepat Guna dengan teknik pendekatan pembelajaran orang dewasa (Andragogik). Hasil kegiatan yang dicapai yaitu (1) terfasilitasi upaya peningkatan pengetahuan dan keterampilan mitra mengolah aneka produk kacang mete; (2) terfasilitasi peningkatan pengetahuan dan keterampilan mitra memproduksi dan mengemas aneka produk kacang mete menggunakan vacuum sealer dengan plastic vacuum emboos; (3) Dihasilkan empat produk olahan kacang mete meliputi kacang mete kering, mete goreng mentega, mete goreng asam manis, dan mete goreng pedas manis, (4) Terfasilitasi upaya meningkatkan pengetahuan dan keterampilan pemasaran digital bagi Mitra; (5) Terfasilitasi satu aplikasi pemasaran digital.

Abstract

Indonesia is a highly biodiverse country, second only to Brazil. This gives Indonesia enormous potential in ethnopharmacology. Previous studies have shown that the leaves of the Brugmansia suaveolens plant, also known as Angel trumpet, have high antioxidant activity. The aim of this study is to test the antioxidant activity of B. suaveolens flowers and conduct quantitative screening to determine which compounds play the greatest role in their antioxidant activity. This research used the DPPH test and Gas Chromatography-Mass Spectrometry (GC-MS) screening. Ascorbic acid was used as the comparator in this study. Before the DPPH and GC-MS testing, the samples were extracted using the cold maceration technique with a 95% ethanol solvent. Then, the samples were concentrated using a rotary evaporator. The IC₅₀ value of the ethanol extract of B. suaveolens flowers is 196.16 µg/mL, while the IC₅₀ value of ascorbic acid, the comparator, is 3,814 µg/mL. This result shows that the ethanol extract of B. suaveolens flowers has moderate antioxidant activity. Screening bioactive compounds with GC-MS showed that Hexadecenoic acid and Octadecanoic acid play the biggest role in antioxidant activity. However, the compound with the highest content in the ethanol extract of B. suaveolens flowers is scopolamine.
