Abstract

Based on experiment analytical report arranged by WHO that tetanus was still getting the cause of mortality and maternal, neonatal pain in Indonesia in 2000-2010. There were 137 per 100.000 childbirth who have got death because of tetanus. In 2010 it was reported that 147 cases with 84 were dead. In 2012 there were 495 the pregnant who had TT immunization were only 307 the pregnant (60, 2%) at Baturaden II social center clinic service.This research is analytical observation located at Baturaden II social center clinic service in April 2014. The number of sample is 60 the pregnant which using random techniques. The used instrument of research is closing questionnaires meanwhile for analytical data used unvaried and bivariate analysis. There are correlation between job to mother knowledge about TT immunization at Baturaden II social center clinic service at Baturaden subdistrict at Banyumas regency in 2014 with correlation coefficient of chi square X2 count is 8,085 and 0,018 of p-value;   there are correlation between economic statuses to mother knowledge about toxoids tetanus (TT) immunization at Baturaden II social center clinic service at Baturaden subdistrict at Banyumas regency in 2014 with correlation coefficient of chi square X2 count is 14,447 and 0,001 of p-value;  there are correlation between education to mother knowledge about toxoids tetanus (TT) immunization at Baturaden II social center clinic service at Baturaden subdistrict at Banyumas regency in 2014 with correlation coefficient of chi square X2 count is 25,031 and 0,0008 of p-value. The pregnant are expected to be more active participating counseling   immunization of TTto increase knowledge of TT immunization through electronica or paper media. Abstract

Centella asiatica leaves has been used as diuretic, antihypertensi, anti leprae, skin infection, burning skin and celloid. Cells suspenson culture of mesophyll of Centella asiatica leaves were carried out in three steps: isolation, purification, and cell culture. The aim of this research is to investigate the influences of leaves position (age of leaves) and concentration of macerozyme R-10 for cells number and viability, and the influence of sucrose concentration for cells growth and biosynthesis of asiaticoside. Isolation and purification of mesophyll cell has been performed using factorial completely randomized design. The first factor was leaf position (age of leaves), and the second factor was concentration of macerozyme. Collected data were analysed using Anova and Duncan’s test at 0.0 – 2.5 – 5.0 and 7.5 per cent respectively. Asiaticoside produced by cell biomass were analyzed qualitatively using thin layer chro- matography. The result revealed that mesophyll cells could be used as explant for suspension culture. The highest biomass produced with highest viability were found in cells isolated from second leaves treated with 0.1% macerozyme (1.32 . 107 cells/ml). Addition of sucrose to suspension culture medium produced higher packed cells volume (pcv) percentage than no sucrose was added, (0% sucrose gave pcv 8.5%, 2.5% sucrose pcv 22%, sucrose 5% pcv 21.5%, and 7.5% sucrose pcv 15.75%). The asiaticoside production not only depended on sucrose, because without any sucrose was added, the asiaticoside was also available..Key Words: Cell suspension culture, mesophyll, Centella asiatica (L.) Abstract

Tawangmangu tangerine (Citrus reticulate Blanco subsp. tawangmangu) is one of prime local fruit from Tawangmangu region, Karanganyar, Central Java. This tangerine have good appearance, easy to peeled, sweet flavored fruit, and high production rate. However at 1984 Tawangmangu tangerine run into depreciation of population caused by CVPD (Citrus Vein Phloem Degeneration). As a result, this variety listed as endangeredThe effort of consevation by culture tissue technique can order to obtain prime seeds that free from virus or disease. This study was aimed to understand the effect of IBA and BAP in culture medium on growth of apical and lateral shoot of Tawangmangu tangerine. The treatment provided were IBA (0 ppm, 0.5 ppm, and 1 ppm) and BAP (0 ppm, 1 ppm and 2 ppm). The treatment in this research was using complete randomized factorial design in 9 different treatment. This research resulted that added combination of IBA and BAP affecting growth of Tawangmangu tangerine but the interaction of IBA and BAP did not happen. The highest shoot length occurs on treatment IBA 1 ppm+BAP 2 ppm while when the fastest shoot appears occurs on treatment IBA 0 ppm+ BAP 2 ppm and the highest shoot formed on treatment IBA 0.5 ppm+BAP 2 ppm. Benefit of the research Efforts to conserve Tawangmangu orange plants need to be done to maintain the diversity of germplasm and support the cultivation in a manner. One technique that can support the conservation of plants is by tissue culture techniques.

Abstract

Plant Growth Regulator (PGR) had been widely utilized in improving plant quality and productivity. The mechanism includes the manipulation of plant adaptibility over environmental stress. This research aimed to study the efect of PGR on the growth of C. tagal seedling and to formulate the applicable treatment to improve its growth performance. The research was conducted through laboratory experiment, including tthe combination of NAA and GA3 with the designed concentration of 0 ppm, 150 ppm and 300 ppm. The experiment lasted for one month and involved weekly observation. The plants were treated by spraying once every three days. Observations were conducted to the survival rate, height increment and leaf number development. ANOVA was conducted as statistical analysis method. The result showed that there were no significant effect of PGRs on the height increment of C. tagal seedling. The survival rate was lowest in the control and 150 ppm NAA treatments, while treatment with 300 ppm GA3was highest. The application of 300 ppm GA3was the best treatment for the livelihood of C. tagal seedling, while the application of 150 ppm GA3 was considerable alternative due to its growth performance. This research provide a novel finding concerning the application of PGRs on the seeding process of C. tagal. Thus, it could be applied in the seeding activities by the mangrove farmers to improve the productivity and quality of mangrove seedling.

Abstract

The Kanayatn Dayak in Tapakng Village has a local culture of using balimo plants for traditional medicine. Balimo is a plant, it has an important meaning in traditional medicine by the Kanayatn Dayak community in Tapakng Village. This study aimed to examine the ethnobotanical knowledge of balimo plants and their advantage among the Kanayatn Dayak  community in Tapakng Village. The research method with observation and deep interview and also semi-structured interview. Deep interviews was conducted with four key informants and the semi-structured interview was conducted with 60 respondents. The results showed that, the local people had a traditional treatment system to solve the disease problems. Balimo plants are used as a prevention of alcohol intoxication due to drinking arak and treatment of coughs (dry cough and bloody cough or (hemoptysis). The use of balimo as a prevention of intoxication has an important role because it is related to the Dayak Indigenous rituals. The level of ethnobotanical knowledge of local people showed that, 100% have heard about the balimo, 92% have seen balimo, and 55% have used it. This research provided information to the public about balimo that can be used in medication. Information on the utilization of balimo gives awareness to the people, the population of balimo really needs to be maintained in nature from extinction.

Abstract

Caisim (Brassica juncea L.) is one of vegetable commodities that has a high commercial value. Many people like it because it has a good taste and it contains several kinds of vitamin and mineral that good for health. This research aims to know the influence of liquid bio organic fertilizer and inorganic fertilizer addition to post harvest product caisim leaf and know the best combination of fertilizer for post harvest product caisim leafquality. Data analysis used is Analysis of Variance (ANOVA) and then continued by test significant different used Duncan Multiple Range Test (DMRT) 95%. The caisim plant that have been harvested then being observed on its fiber content, water content, and physical appearance. The result showed thatthe addition of liquid bio organic fertilizer and inorganic fertilizer has not influence for water content and physical appearance of caisim leaf, but it has an influence on fiber content. Treatment with basic media with the addition of 1 mL + 2,17 g NPK is the best treatment to increase fiber content of caisim leaf Tosakan variety.   Keywords: liquid bio organic fertilizer, caisim, quality of post harvest product

Abstract

Upaya yang perlu dilakukan untuk menjaga kuantitas dan kualitas produk hasil panen Gracilaria verrucosa agar tetap baik diantaranya dengan memberlakukan manajemen budidaya dan pengolahan pascapanen yang baik. Salah satu daerah yang berpotensi dan sudah mulai dikembangkan usaha budidaya Gracilaria verrucosa yaitu lahan tambak desa Mororejo, kecamatan Kaliwungu, Kabupaten Kendal.Penelitan ini bertujuan untuk mengkaji manajemen budidaya dan pasca panen serta kualitas Gracilaria verrucosa secara polikultur di Mororejo, Kaliwungu, Kendal.Penelitian ini bersifat observasi, aspek yang dikaji adalah faktor biotik, abiotik dan pengelolaan pascapanen serta kualitas Gracilaria verrucosa. Pengambilan data dilakukan melalui pengukuran secara kuantitatif  yang dilengkapi dengan wawancara kepada petani. Hasil pengamatan di lapangan menunjukkan manajemen budidaya dan pascapanen Gracilaria verrucosa secara polikultur di Mororejo Kendal sudah cukup baik dari aspek biotik, abiotik dan pascapanennya. Kualitas Gracilaria verrucosa dari Mororejo Kendal untuk kandungan agar (0,054 % - 0,064% dari berat kering total), protein (9,28 % - 11,93 %), lemak (0,12 % - 0,15 %) dan serat (11,44 % – 12, 78 %). Kualitas yang demikian termasuk belum memenuhi standar jika dibandingkan dengan jenis Gracilaria yang ada di lokasi lain.

Abstract

Jatropha (Jatropha curcas L.) is one of the important crop for biofuel industry development and have the nature of drought tolerance that can be cultivated in marginal areas. One of cultivation technology that determines the success of planting jatropha seedlings in the field is the provision of plant nurseries in general use media that is around, such as soil and organic fertilizer. This study aims to observe the growth of jatropha seedlings on soil type and the addition of compost that is different from studies on the growth of leaves and roots. The results showed jatropha plant seedlings grown on soil rendzina, grumosol and regosol given compost or not for two months was able to grow and show the same increase in the number of leaves, wet weight and length of plant roots.

Abstract

The increasing demand of tomatoes corresponds to the economic and population growth. To maintain the quality of tomatoes, an effort to holdup the tomato ripeness was done using several kinds of plastic packaging. This research was intended to assess the effects of using plastic packaging and the most effective kind of plastic packaging to extend tomato storage period. The research was done at BSFT Biology Laboratory Diponegoro University and Food Technology Laboratory Soegijapranata Catholic University.    The design used in the research was Completely Randomized Design (CRD) with some treatments using different  plastic packaging type (P 0 = control / without packaging, P1 = Low Density Polyethylene (LDPE), P2 = High Density Polyethylene (HDPE) and P3 = Polypropylene (PP), each treatment was repeated three times. The parameter observed was the percentage of weight loss, the change in color (using chromameter method); the hardness level (texture analyzer method) and total content of sugar. Data were analyzed by Analysis of Variance (ANOVA) then continued with Duncan's Multiple Range Test (DMRT) at the 95% significance level to find out the difference. The results show that the use of plastic as packaging materials is able to delay tomato ripeness and the effective type plastic-based packaging to weight loss and change color is HDPE and PP.

Abstract

The Objective of this research was to analyse glucose, carotenoid, and biomass of Alang-alang (Imperata cylindrical L. Beauv) that grew at open field area on the both Recidence of Blora and Ungaran. Completely Randomized Design was applied in this research that using single factor ( different level of place ) with 3 times replication. Data was analysed by T test with 99 % and 95%  significant level. The parameter was observed include environmental factors, content of glucose and carotenoid, biomass of Alang-alang. The Result showed that Blora Recidence have higher temperature and light intensity. However, the moisture and rainfall intensity is low. Alang-alang that grow in Blora recidence have higher content of glucose and carotenoid, but the its biomass is low.   Keywords : environmental factors, glucose, carotenoid, the biomass of Alang-alang
