Abstract

Tomatoes (Lycopersicon esculentum Mill.) are a source of carotenoids they are easy to find. These compounds function as precursors of vitamin A, antioxidant, and prevent cancer. The extraction of carotenoid compounds for commercial products usually uses fresh plants, which are less efficient and require a lot of raw materials. The supply of these raw materials can be done through tissue culture. The frequency of subculture or supply of nutrients in tissue culture is very influential on the content of callus carotenoids produced. This study aimed to determine the effect of subculture frequency on growth, development and callus carotenoid content and to find out the right frequency of subculture to produce callus with optimal growth, development and carotenoid content. The design this study was a single Completely Randomized Design (CRD) with 4 treatments of subculture frequency and 5 replications. The data obtained were analyzed by ANOVA at the 95% test level followed by DMRT in case a significant different was found The results showed subculture frequency affected growth, development and carotenoid content of callus Lycopersicon esculentum. Mill. The most optimal treatment to induce growth and production of carotenoids in this study was treatment of thrice subculture while the most optimal treatment in inducing development was the twice subculture treatment.

Abstract

TSS (True Shallot Seed) need to be developed to address the quality and quality of shallot seeds. The seeds, however, still has constraints on its viability and germination. The objective of this study was to investigate the effect of corona discharge plasma radiation on the viability of true shallot seeds. The research was conducted in March - April 2023 at the Plasma Laboratory, Faculty of Science and Mathematics, Diponegoro University, and the germination test experiments at the the Agricultural Technology Research and Assessment Installation (IP2TP Ungaran). The experiment was designed completely randomized with 6 treatments of radiation time consisting of P0 (without radiation), P1 (5 min), P2 (10 min), P3 (15 min), P4 (20 min), and P5 (25 min). All treatments were carried out with 5 replications. The collected data were processed using ANOVA and then continued with the DMRT test. The results showed that the corona plasma radiation treatment for 15-25 min affected the parameters of germination, germination rate, seed growth rate, vigor index, seed uniformity, and sprout length.  Keywords: Plasm, Radiation, Shallots, TSS.

Abstract

Plant nutrient absorptions depend on their environment and plants’ stage of development. Beside from soil, nutrient absorption also proceed through resorption in leaves. The resorption efficiency varies based on types of habitat. Drought stress also influences the efficiency of nutrient resorption, growth rate, and plant development. The purpose of this research was to study the change of nitrogen (N) and phosphorus (P) content and leaf resorption value of N and P, and specific leaf area of 6-years old cacao grown under drought stress. Research was carried out in a cacao plantation with 7-years old Gliricidia sepium Jacq. as shading trees. The results showed that drought stress with Troughfall Displacement Experiment (TDE) system did not affect the content and resorption level of N and P of cacao leaves. However, N resorption of cacao leaves tended to increase while P resorption decreased. Cacao plants experienced drought stress have the ability to resorp leaf N for 46.64-50.63%, leaf P for 47.98-58.40%, whereas, the control plants had the leaf N resorption of 45.05-52.97%, and leaf P resorption of 36.64-44.10%. Drought stress for 13 months on 6-years old cacao did not affect specifi c leaf area.

Abstract

Shallots are bulb-shaped commodities that are difficult to maintain in new conditions because they are easily damaged. An effective effort to prevent deterioration of food quality during storage is by coating it using nano chitosan. The effectiveness of nano chitosan is influenced by particle size which related to the ratio of chitosan and STPP (sodium tripolyphosphate). This study aimed to determine the effect of nano chitosan coating with the addition of different ratios of chitosan and STPP and the appropriate ratio of chitosan and STPP on nano chitosan as a coating to maintain the quality of shallot bulbs (Allium ascalonicum L. var. Bauji). This study used a completely randomized design (CRD) method with four treatments and five replications. The treatments of this research were P0= Control, PI= Nano chitosan ratio of chitosan: STPP 1:3, P2= Nano chitosan ratio of chitosan:STPP 1:4, and P3= Nano chitosan ratio of chitosan:STPP 1:5. The variables of this study were the percentage of damage, the percentage of diameter shrinkage, weight loss, color, hardness, and moisture content. Data were analyzed using the ANOVA test and continued with the DMRT test. The application of nano chitosan coating on shallot bulbs could reduce damage, shrinkage of tuber diameter, weight loss, color, hardness, and decrease in water content better than the control treatment. The best results were shown by treatment P3 (1:5) with a percentage of damage of 8%, diameter shrinkage of 20.20%, weight loss of 18.40%, total color change of 54.45, hardness of 226.23 N, and a decrease in water content of 4.65% at the final water content of 79.09%.
