Abstract

Purification of Cephalosporin-C Acylase and Its Optimization of Ion-Exchange ChromatographyABSTRACTCephalosporin-C acylase (CCA) has an important role in the one-step conversion of cephalosporin-C into 7-ACA. Purification process aims to increase specific activity of CCA enzyme. Purification began with cell lysis, ammonium sulphate precipitation, dialysis, ion exchange chromatography (IEC) and size exclusion chromatography. IEC optimization of elution step was also done to compare gradient and isocratic elusion. Purification was capable to increase the enzyme purity upto 33.66 fold, with specific activity of 3.00 U/mg and the yield reached 41.41%. Optimization of elusion during IEC showed that isocratic protein elusion was more efficient (taking shorter time, 3 column volume (CV) only) than that of gradient batch (up to 9 CV). SDS-PAGE analysis demonstrated that the recombinant CCA enzyme existed in two types, active enzyme containing α-subunit (25 kDa) and β-subunit (58 kDa), and inactive enzyme (83 kDa) as precursor. Furthermore, 30% ammonium sulphate saturated precipitation was able to precipitate this inactive CCA.Keywords: 7-ACA,CCA, cephalosporin C, protein purification, specific activity ABSTRAKSefalosporin-C asilase (CCA) merupakan enzim yang berperan penting dalam konversi satu tahap sefalosporin-C menjadi 7-ACA. Hasil optimasi IEC pada proses elusi secara isokratik lebih efisien dari segi waktu (hanya membutuhkan 3 kolom volume (CV) dibandingkan dengan secara gradien (sampai 9 CV). Hasil SDS-PAGE menunjukkan bahwa CCA rekombinan merupakan enzim dengan 2 macam bentuk yaitu enzim aktif, yang terdiri dari subunit α (25 kDa) dan β (58 kDa), dan enzim tidak aktif berupa prekursor (83 kDa). Proses presipitasi menggunakan amonium sulfat 30% tersaturasi dapat mengendapkan prekursor CCA.Kata Kunci: 7-ACA, aktivitas spesifik, CCA, purifikasi protein, sefalosporin C

Abstract

Annonaceous acetogenin was extracted from Annona squamosa (Srikaya) seeds. It has cytotoxic activity against cancer cells and lower toxicity compared to other cancer drugs. Endophyte from Annonaceae is expected to have similar extracted metabolites to the host, thus increasing the economic value. This research is a preliminary study to obtain active compounds with potential as anti-cancer agents from endophytic fungi of Srikaya plants. Four endophytic fungal strains were isolated from Srikaya plants (Annona squamosa) and identified based on 28S rDNA sequence. The isolates are SKY II.3.1, SKY I.1.2, SKY II.3.2, and SKY III.3.1, and have similarity with Fusarium sp Vega760, Fusarium sp NRRL 22354 NRRL223, Nectria rigidiuscula, and Fusarium sp BOL35, respectively. The identified isolates were fermented in liquid media for three weeks. The liquid and mycelium were extracted using ethyl acetate. Whole extract of each fermented isolate was partitioned and evaporated to obtain ethyl acetate extract. Cytotoxicity assay of ethyl acetate extract was carried out at level 100 ppm by Methyl Thyazole Tetrazolium (MTT) viability test towards MCF-7 (breast cancer cell). The result indicated that each ethyl acetate extract could inhibit the viability of cell MCF-7 with 11.34, 99.78, 91.48, and 96.84%, for SKY II.3.1, SKY I.1.2, SKY II.3.2, and SKY III.3.1, respectively. Based on the results of cytotoxicity assay on MCF-7 breast cancer cells, endophytic fungi isolates SKY I.1.2, SKY II.3.2, and SKY III.3.1 are potential as sources of anti-breast cancer compounds.

Abstract

Brebeg Village has great potential for producing Siamese oranges. Large areas of land have the productivity to produce more oranges, oranges can be harvested on average 18 tons per year. The large potential of orange fruit can improve the economy of orange farmers, the government Department of Agriculture provides assistance in the form of training on the care and rejuvenation of orange trees as well as providing training on processing orange fruit products in collaboration with UNAIC. The problems faced by Gapoktan are a lack of knowledge regarding planting oranges, an irregular irrigation system. The problems faced by KWT are lack of knowledge regarding orange-based products, product management and marketing. The method used is coordinating with all partners, providing knowledge and skills in creating orange cultivation facilities and orange-based food processing, implementing and evaluating activities. The results of Kosabangsa's research are able to provide benefits for community empowerment in this case, namely improving the economy through empowering Siamese orange farmer groups to become functional food-based processed products in Brebeg Village, Jeruklegi District, Cilacap Regency.

Abstract

The level of success of collaboration can be achieved when the collaboration involves people with diverse experiences, multi-disciplinary knowledge, different cultures, education and backgrounds. Various parties will present different thoughts and contributions, so that the problem will be handled from various angles in a comprehensive manner. The problem in Cilacap Regency, with the majority of coastal communities working as fishermen and fish processors, is that sales are stagnant because they only rely on traditional marketing, by selling fresh fish at TPI Padanarang or processing fish into crackers, salted fish, handicrafts that are sold directly at around TPI Pandanarang. The aim of this PkM is providing digital marketing training through Tokopedia as an effort to increase sales of groups supported by the Cilacap Fisheries Service. The method chosen was qualitative with the number of respondents being 10 residents representing groups supported by the Cilacap Fisheries Service. The results of the PkM which was at TPI Pandanarang - Teluk Penyu Area, South Cilacap District, Cilacap Regency were the creation of 10 Business Identification Numbers (NIB) and 10 online shop accounts on Tokopedia for 10 people representatives of groups supported by the Cilacap Regency Fisheries Service. Conclusions from field observations, apart from marketing training, it turns out that the community needs a lot of assistance when processing permits such as PIRT, halal certificates, promotional training, packaging design, hygiene and sanitation training, health safety food. The Cilacap District Fisheries Service can collaborate with several companies/agencies to get assistance with equipment grants, training from Corporate Social Responsibility (CSR) funds.

Abstract

When an organization's performance necessitates the consequences of collaboration between individual performance and affected components, it is considered to be successful and effective. Each administrator's potential is taken into account when dividing management duties into different categories, and all members and the leadership place their trust in them. One of the case studies that can be seen is the education system in Indonesia, specifically the universities, where the performance of lecturers and their leadership, communication, and competency are key factors in determining student achievement. The accomplishment of lecturers in carrying out the execution of instruction, research, and community service is demonstrated by a competent graduate student (Tri Dharma of Higher Education). Using SEM PLS, a quantitative research methodology was applied. Saturation sampling was used in a quantitative study that included all 122 lecturers who responded to questionnaires at the Universities of Al-Irsyad Cilacap and Nahdlatul Ulama Al Ghazali Cilacap. The results of the analysis revealed that, with P values less than 0.05, leadership, competence, communication, and organizational commitment have a substantial impact on lecturer performance. Leadership, in the meantime, can affect performance through the supplementary factor of organizational commitment.

Abstract

Ujunggagak Village is an area prone to flooding often caused by high tides. The community has not utilized the potential of existing natural resources optimally due to a lack of knowledge and skills in processing them. This service aims to increase knowledge and skills in processing coconut sap into healthy sugar and processed healthy sea fish in communities affected by disasters. Service method with education and training on using natural preservatives for penderes farmers and fishermen women groups (KWN). The number of group members is 30 penderes and 25 KWN. The service was carried out for three months. The level of knowledge was measured before and after the education was carried out using a questionnaire. Group skills are measured through quality production results after training. The results show an increase in the knowledge of the Penderes group from 20% poor to 76.7% good, while in the KWN it increased from 28% poor to 80% in the good category. The group skill of the penderes increases from 0% to 100% producing healthy sugar in the form of coins, batteries, and healthy palm sugar. KWN's skills have gone from just making salted fish to being skilled at processing seafood in the form of healthy meatballs, sausages, nuggets, and crackers. This activity can be sustainable and continue to be developed so that it can improve the health and economic status of the community. The community can anticipate crop failure as a result of flood disasters.

Abstract

Masyarakat pesisir sebagian besar memiliki pencaharian sebagai penderes dan nelayan. Produksi nira kelapa oleh penderes disinyalir banyak menggunakan pengawet sistetis yang dikenal dengan laru. Nelayan dengan ikan yang melimpah namun karena ketidaktahuan cara pengolahannya sering dibuang sehingga menjadi limbah dan hanya mengolah menjadi ikan asin. Tujuan pengadian adalah meningkatkan pengetahuan dan keterampilan dalam mengolah nira kelapa dan ikan menjadi produk yang sehat dengan menggunakan pengawet alami. Pengabdian dilaksanakan pada petani penderes dan kelompok wanita nelayan Cilacap, dengan tim pengabdi dari dua perguruan tinggi yaitu Universitas Al-Irsyad Cilacap yang berkolaborasi dengan Universitas Jenderal Soedirman Purwokerto Banyumas. Peserta terdiri dari 30 anggota kelompok petani penderes dan 25 anggota kelompok wanita nelayan (KWN). Peserta diberikan pemahaman sekaligus mempraktikan pencampuran nira kelapa dengan teknologi tangkal kimia untuk sehat (TANGKIS) dan ekstrak bunga kecombrang dalam olahan ikan. Pengukuran tingkat pengetahuan dilakukan dengan menggunakan kuesioner pengetahuan pre dan postes. Keterampilan kelompok diukur melalui hasil produksi yang berkualitas setelah pelatihan. Hasil pengabdian menunjukkan bertambahnya pengetahuan penderes tentang olahan nira kelapa yang sehat dari 20% kategori kurang baik menjadi 76.7% baik. Keterampilan pengolahan nira kelapa menjadi gula kelapa yang sehat dikemas dalam bentuk koin, baterai dan gula semut. KWN bertambah pengetahuan dari kategori kurang baik (28%) menjadi kategori baik (80%) dan dari keterampilan telah dapat membuat empat jenis olahan ikan yang sehat dalam bentuk bakso, kerupuk, nugget dan sosis.

Abstract

Dalam era persaingan pendidikan tinggi yang semakin ketat, strategi promosi yang tepat sasaran menjadi sangat penting untuk menarik calon mahasiswa. Penelitian ini bertujuan untuk mengembangkan model prediksi yang dapat digunakan untuk menentukan strategi promosi efektif bagi UNAIC dengan memanfaatkan metode K-Nearest Neighbors (KNN). Metode KNN dipilih karena kemampuannya dalam mengklasifikasikan data berdasarkan kedekatan geografis, yang memungkinkan identifikasi lokasi asal mahasiswa potensial dengan tingkat akurasi yang tinggi(Fitria et al., 2024). Penelitian ini menganalisis data mahasiswa UNAIC dari tahun 2021 hingga 2023, mencakup variabel demografis dan geografis untuk memprediksi pola asal daerah mahasiswa. Dengan hasil analisis ini, UNAIC dapat merumuskan strategi promosi yang lebih efektif dan efisien, menargetkan daerah-daerah dengan peluang tinggi untuk merekrut calon mahasiswa baru. Hasil penelitian di Universitas Al-Irsyad Cilacap tentang metode K-Nearest Neighbors (KNN) menunjukkan akurasi 99%. Model ini tidak salah dalam kategori "tidak layak" dan berhasil memprediksi 1190 mahasiswa "layak" dengan benar. Namun, ada 12 kasus mahasiswa "ditolak" yang ternyata diterima. KNN memiliki class recall 100% untuk "layak" dan 0% untuk "ditolak", mengindikasikan kinerja yang sangat baik dalam mengidentifikasi mahasiswa yang layak, tetapi membutuhkan penyempurnaan untuk kategori "ditolak".

Abstract

Desa Binangun termasuk dalam kategori kemiskinan ekstrem di Indonesia, dengan mayoritas penduduknya bergantung pada sektor pertanian dan usaha mikro. Koperasi Desa Binangun didirikan untuk memberdayakan masyarakat melalui pengelolaan dan pemasaran produk unggulan desa, yaitu keripik pisang dan minyak Sacha Inchi. Namun, koperasi menghadapi tantangan utama dalam akses pasar dan pemanfaatan teknologi digital, yang menghambat daya saing produk. Saat ini, metode pemasaran koperasi masih konvensional, sehingga sulit menjangkau pasar yang lebih luas. Rendahnya literasi digital di kalangan anggota koperasi serta keterbatasan infrastruktur dan modal juga menjadi kendala dalam strategi pemasaran digital. Selain itu, proses operasional koperasi yang masih manual menyebabkan inefisiensi dalam pencatatan transaksi, manajemen stok, dan layanan pelanggan, serta kurangnya transparansi dalam manajemen keuangan. Untuk mengatasi tantangan ini, diperlukan penerapan digital commerce sebagai solusi strategis. Digital commerce dapat membantu koperasi memperluas jaringan distribusi, meningkatkan efisiensi operasional, serta meningkatkan daya saing produk unggulan desa. Melalui digitalisasi, koperasi dapat memanfaatkan strategi pemasaran berbasis teknologi, mengoptimalkan pencatatan transaksi, serta meningkatkan transparansi keuangan. Penelitian ini berfokus pada perancangan platform digital commerce yang sesuai dengan kebutuhan Koperasi Desa Binangun. Tujuannya adalah memperluas akses pasar, meningkatkan daya saing produk, dan mendorong pertumbuhan ekonomi desa secara berkelanjutan. Dengan adopsi teknologi digital, koperasi dapat bertransformasi menjadi entitas bisnis yang lebih modern dan kompetitif, serta memberikan dampak positif bagi masyarakat desa
