Abstract

Purification of Cephalosporin-C Acylase and Its Optimization of Ion-Exchange ChromatographyABSTRACTCephalosporin-C acylase (CCA) has an important role in the one-step conversion of cephalosporin-C into 7-ACA. Purification process aims to increase specific activity of CCA enzyme. Purification began with cell lysis, ammonium sulphate precipitation, dialysis, ion exchange chromatography (IEC) and size exclusion chromatography. IEC optimization of elution step was also done to compare gradient and isocratic elusion. Purification was capable to increase the enzyme purity upto 33.66 fold, with specific activity of 3.00 U/mg and the yield reached 41.41%. Optimization of elusion during IEC showed that isocratic protein elusion was more efficient (taking shorter time, 3 column volume (CV) only) than that of gradient batch (up to 9 CV). SDS-PAGE analysis demonstrated that the recombinant CCA enzyme existed in two types, active enzyme containing α-subunit (25 kDa) and β-subunit (58 kDa), and inactive enzyme (83 kDa) as precursor. Furthermore, 30% ammonium sulphate saturated precipitation was able to precipitate this inactive CCA.Keywords: 7-ACA,CCA, cephalosporin C, protein purification, specific activity ABSTRAKSefalosporin-C asilase (CCA) merupakan enzim yang berperan penting dalam konversi satu tahap sefalosporin-C menjadi 7-ACA.

Abstract

The research that has been done aims to analyze the effect of the work environment and employee competence on employee performance at a seafood processing company in Probolinggo City, East Java. Based on the results of the analysis of the calculation of the first hypothesis (H1), which states that the work environment has a positive and significant effect on employee performance, the t-count value of 19,845 is greater than the t-table value, which is 1,677 and the significance value obtained is less than 0.05, which means that the work environment has a positive and significant effect on employee performance, the results of the calculation analysis of the second hypothesis (H2) which states that employee competence has a positive and significant effect on employee performance obtained a t-count value of 14,471 greater than the t-table value of 1,677 and the value the significance obtained is less than 0.05, which means that employee competence has a positive and significant effect on employee performance, while the R-square value obtained is 0.897 which states that work environment variables and employee competency affect employee performance by 89.70% and the remaining 10.30% influenced by other variables.

Abstract

The objective of this research is to examine the relationship between leadership styles and employee motivation in the context of contemporary management. This research employs a literature review approach to analyse various leadership theories, including transformational, transactional, autocratic, democratic, laissez-faire, and servant leadership. Each leadership style is evaluated in relation to employees' intrinsic and extrinsic motivation. The findings indicate that transformational and democratic leadership styles have the most significant influence on employees' intrinsic motivation, whereas transactional and autocratic leadership styles emphasize extrinsic motivation. This study contributes to contemporary management literature by providing insights into the importance of tailoring leadership styles to employees' motivational needs to achieve optimal performance.

Abstract

Social media feed—which is populated with text embedded in numerous forms of multimedia contents—has been a crucial battleground for politicians to situate their policy relevance to the public during election campaign. Narrative building is somewhat played out by politicians in digital space to seek an earlier assurance of their victory. This article analyses how the proponent 2019 Indonesia presidential election candidate, Joko Widodo and his running mate, Ma’ruf Amin assigns a duality of function to the anti-hoax narrative besides renders it as a firm stance they take to differ from their rival. We employ action research at the scene drawing upon the five-month period of observing one of the consultative groups to the candidate on the matter of digital campaign, and at times sharing our perspective on digital literacy and tackling disinformation. Anti-hoax narrative in the context of election turns out to be an inescapable hot topic that arguably serves as a sort of ‘free-to-use political resource’ ready to be availed of by politicians to reiterate an agenda, counterbalance and issue, or even instill a new idea regarding nationhood.

Abstract

This study investigates the relationships among knowledge sharing, cross-functional collaboration, adaptive leadership, and innovation success in organizational settings. Employing Partial Least Squares Structural Equation Modeling (PLS-SEM), the research analyzes data collected from employees across various industries. The findings reveal that knowledge sharing significantly influences innovation success both directly and indirectly through cross-functional collaboration, which also serves as a critical mediator between adaptive leadership and innovation outcomes. Furthermore, adaptive leadership is shown to enhance cross-functional collaboration, emphasizing the importance of leadership style in fostering a collaborative culture conducive to innovation. These results underscore the necessity for organizations to cultivate knowledge-sharing practices, promote cross-functional teamwork, and develop adaptive leadership capabilities to drive innovation success. The study contributes to the literature by elucidating the dynamic interplay among these constructs and providing practical implications for managers seeking to enhance their organization's innovative capacities.

Abstract

This study aims to analyze the effect of job satisfaction and tenure on turnover intention. Using the survey method, data was collected from 30 employees from various industry sectors. Pearson correlation analysis showed that there is a significant negative relationship between job satisfaction and turnover intention (r = -0.527, p < 0.05), and between tenure and turnover intention (r = -0.382, p < 0.05). Multiple regression analysis shows that job satisfaction and tenure significantly affect turnover intention (p < 0.05), with a negative coefficient indicating that an increase in job satisfaction and tenure will decrease turnover intention. The interaction analysis shows that there is a significant interaction between job satisfaction and tenure in influencing turnover intention (p < 0.05). The results of this study provide insights for human resource managers to increase job satisfaction and retain long-term employees to reduce turnover intention. With the right strategies, organizations can improve employee retention and overall productivity.
