Abstract

Saat ini, jus manggis menjadi “minuman herbal” yang populer. Beberapa produsen mengklaim bahwa jus manggis dapat mengobati diare, masalah menstruasi, infeksi saluran kemih, tuberkulosis, dan berbagai kondisi lainnya. Namun, tidak ada bukti ilmiah yang dapat diandalkan untuk mendukung klaim ini. Penelitian ini bertujuan untuk menganalisis α-mangostin dalam minuman herbal kulit buah manggis yang terdapat di pasaran dengan metode kromatografi lapis tipis densitometri. Analisis dilakukan dengan TLC Scanner menggunakan Camag TLC Scanner 4 dengan software Wincats, panjang gelombang maksimum α-mangostin adalah 316 nm, dilihat menggunakan lampu UV Camag 254 nm dan 366 nm, Camag Nanomat 4, Camag pipet kapiler ukuran 2 µL, Camag Twin Chamber, plat silika gel 60 F254 (Merck) ukuran 20 x 20 cm. Fase gerak yang digunakan adalah kloroform dan etil asetat (9:1). Setelah dilihat di bawah lampu UV 254 nm, standar α-mangostin, sampel B dan C memiliki nilai Rf yang sama yaitu 0,48. Deret konsentrasi yang digunakan 50, 100, 150, 300 dan 400 ppm menghasilkan persamaan regresi y = 1326,1658 + 19,7647x dengan koefisien korelasi (r) = 0,99415. Koefisien korelasi ini menunjukkan hasil yang linear. Batas deteksi (LOD) dan batas kuantifikasi (LOQ) yang diperoleh 54,38 µg/mL dan 181,28 µg/mL. Persentase Recovery yang diperoleh dari analisis sebesar 90,04 %, metode validasi ini memberikan hasil yang akurat dan tepat (nilai intraday presisi RSD/Relatif Standar Deviasi adalah  1,28 %, sedangkan uji presisi interday adalah  2,39 %). Hasil kadar rata-rata yang diperoleh untuk sampel B dan C berturut-turut adalah 0,040% b/v dan 0,118% b/v. Penelitian ini murah, akurat, tepat, linier dan cepat. Metode KLT densitometri memenuhi kriteria validasi untuk penetapan kuantitatif α-mangostin dalam minuman herbal kulit buah manggis.

Abstract

Sumatran lichen has the potential as antiviral, pure isolates that have been isolated and developed as prospective compounds for COVID-19 treatment. Computational methods were used to accelerate the discovery and screening of potential new compounds. The molecular structures of the isolated compounds such as Lobarin, Atranorin, Methyl 2,4-dihydroxy-3,6-dimethylbenzoate, Methyl 3‐formyl‐2,4‐dihydroxy‐6‐methylbenzoate, Ethyl 3-formyl-2,4-dihydroxy-6-methylbenzoate, and Lobaric acid were drawn, then their activities were analyzed, processed by docking with ACE2 protein, and tested for Druglikeness. The activities and druglikeness were determined in the Swiss ADME program, while the ACE2 docking was processed by Blind Docking in Arguslab, AutoDock Vina, Open Babel, and Discovery Studio Visualizer programs. All compounds bound to the ACE2 protein, as apparent from the number of hydrogen bonds between the two. The Gibbs free energy was in the range of -5.6 to -7.0, and the best one was obtained from atranorin. As for lobarin, this compound was found to be non-drug-like.

Abstract

Ripe banana peroxidase activities with Glomus type 1. Ripe banana is very susceptible to blood disease caused by Blood disease bacterium (BDB) and Fusarium wilt caused by Fusarium oxysporum f.sp. cubense. Arbuscular mycorrhizal fungi (AMF) Glomus tipe 1 increased resistance of ripe banana seedlings to both wilt diseases. The resistance mechanism related with the change of hydrolytic enzyme activities: peroxidase (PO), phenylalanin amonialyase (PAL) and polyphenoloksidase (PPO). The green house and laboratorium experiment were conducted to study the effect of different colonization time 4, 12, 24, 36, 48, 72, 96 h after application (haa) and control (without AMF) with 3 replicates. AMF fresh inoculants source is a mixture of sand planting medium that containing spores, hyphae and corn root colonized AMF. Ripe banana seedlings (60 days old) were inoculated with 50 g fresh AMF inoculants and incubated with the treatment. The results showed that the application of Glomus tipe 1 caused changes in the enzyme activity of peroxidase in the roots and leaves. Root peroxidase enzyme activity slightly increased 5.84% (0.326U) at the beginning of colonization (4 haa) while peroxidase enzyme activity in leaves sharply declined (85.83–87.37%).

Abstract

In continuation of our study of Sumatran plants and the bioactivity of their constituents, Stereocaulon halei Lamb (Stereocaulaceae) was extracted and isolated to obtain atranorin (1), lobaric acid (6), and methyl-β-orcinol carboxylate (9). We also prepared methanolic extracts of some Sumatran medicinal plants, i.e. Alpinia denticulata, Alpinia malaccensis, Alpinia submutica, Curcuma zedoaria, Centella asiatia, Mangifera indica, Piper crocatum, Mussaenda frondosa, together with previously isolated compounds, i.e. crocatin A (2) and crocatin B (3) from P. crocatum, curcumin (4) from Curcuma longa, demethoxymatteucinol (5) from Sphaerostephanos polycarpus, mangiferin (7) from M. indica, methyl caffeate (8) from Balanophora elongata, tiliroside (10) from Guazuma ulmifolia, and usnic acid from Usnea sp (11). These extracts and compounds were tested for their cytotoxicity against MCF-7 and HSC-3 cell lines by MTT assay (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide). It was found that the IC50 value of atranorin (1), lobaric acid (6), methyl-β-orcinol carboxylate (9), and methanolic extract of A. submutica towards the MCF-7 cell line was 208.20 µM, 172.05 µM, 382.60 µM and 70.95 µg/mL, respectively. The IC50 value of lobaric acid (6) and methyl-β-orcinol carboxylate (9) against the HSC-3 cell line was 88.92 µM and 260.09 µM, respectively. The other extracts did not show significant cytotoxicity.

Abstract

Investigasi fitokimia dari Lichen Sumatera (Stereocalon halei Lamb) yang dikoleksi di Gunung Singgalang, Sumatera Barat telah dilakukan. Depsid, atranorin dan depsidon, asam lobarat dapat diisolasi dan dipurifikasi secara singkat dari ekstrak etil asetat (15,88 g) masing-masing sebanyak 4,6 g dan 1,18 g. Penentuan struktur dari dua senyawa hasil isolasi diidentifikasi menggunakan 1H dan 13C RMI. Kemudian dilanjutkan dengan pengujian anti mikroba dari setiap ekstrak dan isolat hasil isolasi menggunakan metode difusi agar terhadap bakteri Gram positif (S. aureus, E. faecalis) dan bakteri gram negatif (E. coli, S. thyphosa, S. typhimurium dan P. aureginosa). Pada pengujian anti tuberkulosis, masing-masing ekstrak dan senyawa hasil isolasi memperlihatkan hambatan terhadap pertumbuhan bakteri M.tuberculosis H37Rv menggunakan media Lowenstein Jensen. Asam lobarat dan atranorin memberikan aktivitas yang besar dalam menghambat bakteri S. aureus dengan konsentrasi hambat minimum (KHM) 0,0125%, sedangkan terhadap M. tuberculosis H37Rv keduanya memiliki KHM 5 μg/ mL.

Abstract

Bacillus sp. 3 is one of symbiotic bacteria of marine sponge Haliclona fascigera obtained from Mandeh Island, Pesisir Selatan, West Sumatra that can produce antibacterial compounds. This study aims to isolate the antibacterial compounds produced by Bacillus sp.3 (A1). The methods that has been used were bacterial cultivation on Nutrient Broth medium using Shaker Incubator at 150 RPM at 37ºC for 48 hours and isolation of antibacterial compound. The isolation method of the compound was carried out by column chromatography method. The testing of antibacterial activity was done by using agar diffusion method. Characterization of isolated compounds were performed using UV and IR spectrophotometer. From this research, two compounds RJ1 and RJ2 were obtained. The compound RJ1 was yellow oily (2.8 mg). The compound RJ1 can inhibit Staphylococcus aureus ATCC 25923 at a 1% of Minimum Inhibition Concentration (KHM). RJ2 compound was yellow oily (3.1 mg). The RJ2 compound can inhibit Escherichia coli ATCC 25922 at a 1% of Minimum Inhibition Concentration (MIC).

Abstract

Amorphous solid dispersions of a poorly water-soluble drug genistein in PVP K-30 were prepared by solvent co-evaporation technique using organic solvent methanol. Solid dispersions system was prepared with several variations of drug to polymer 2:1, 1:1 dan 1:2 w/w. Solid state properties of solid dispersion system were evaluated by powder X-ray diffraction, Fourier transform infrared spectroscopy, and differential scanning calorimetry, and microscopic SEM. Dissolution rate profile were conducted in distilled water medium by using dissolution tester apparatus type II USP. Base on X-ray diffractometry analysis, differential scanning calorimetry and microscopic SEM, crystalline phase of genistein decreased in crystallinity index and formation of amophous state. Dissolution rate profile showed that genistein in amorphous solid dispersion had a faster dissolution rate in comparison to intact genistein. This study proved that preparation of solid dispersion of genistein in PVP K-30 is an effective approach to improve dissolution rate of genistein.

Abstract

Sub-chronic toxicity test of the deffated ethanolic extract of  tali putri (Cassytha filiformis L.) on the rat renal function has been carried out. A number of 16 male rats aged 2-3 months, weighing ±250 g were used as experimental animals. The rats were divided into 4 groups which consisted of 1 control group and 3 extract-treated groups given 1.25, 2.5, and 5 mg/kg extract administrated intraperitoneally once a day for 14 days. The creatinine clearance, renal function, and the ratio of renal were calculated and analyzed by two-way ANOVA followed by Duncan’s test. The study showed that the creatinine clearance and renal function decreased significantly during treatment (p<0.05). The kidney weight ratio of groups treated with extract, especially with greater doses, showed a very significant increase as compared with control (p<0,01). These indicated that the ethanolic extract of  C. filiformis on the doses of 1.25 – 5 mg/kg might decrease the renal function, althought it seemed to be relatively safe when used for 14 days.

Abstract

This research is a continuation study in the inventory of lichens of the genus Stereocaulon which is currently focused on the Stereocaulon massartianum Hue collected in the rocks of Diatas Lake, West Sumatera, Indonesia. Reports on phytochemical studies and pharmacological activities of this species are still limited, based on literature studies that have been carried out. This research was conducted to determine their secondary metabolites and potential antibacterial activity. The air-dried thallus of lichen S. massartianum was macerated successively using n-hexane, ethyl acetate and methanol solvents. The compounds were separated by chromatography and recrystallization methods, then analyzed by spectroscopy (UV-Vis, FTIR, 1H and 13C-NMR). Furthermore, the antibacterial activity assay was performed by agar diffusion method on ethyl acetate extract, and TLC-Bioautography for the isolated compound. Three compounds have been isolated from ethyl acetate extract, i.e. atranorin (1), stictic acid (2) and norstictic acid (3). The results of antibacterial assay from the extract showed antagonistic activity against pathogenic bacteria S. aureus, E. faecalis, E. coli, and P. aeruginosa at concentrations of 10 and 20%, while TLC-bioautography of compound 3 exhibited growth inhibition area in all test bacteria.

Abstract

Tujuan penelitian ini adalah untuk meningkatkan laju disolusi piperin dengan pembentukan multikomponen kristal piperin dan asam nikotinat (1:1) dengan metode pelarutan menggunakan pelarut etanol. Multikomponen kristal dikarakterisasi sifat padatannya dengan Difraksi sinar-X, analisa termal Differential Scaning Calorimetry (DSC), spektroskopi FT-IR, analisa mikroskopik Scanning Electron Microscopy (SEM), uji kelarutan dan  profil laju disolusi menggunakan  Aparatus USP 2. Dari penelitian serbuk multikomponen kristal piperin-asam nikotinat diperoleh hasil  pola difraksi sinar-X menunjukkan puncak difraksi yang baru, analisa termal termogram DSC menunjukkan puncak endotermik pada 126,117 ºC, spektrum FTIR terdapat bilangan gelombang 3349,94 cm-1, analisa mikroskopik SEM menunjukkan sudah terbentuk agregat, uji kelarutan menunjukkan peningkatan 1,5 kali lipat dibandingkan piperin murni, profil laju disolusi piperin menunjukkan peningkatan secara signifikan dibandingkan piperin murni yaitu sekitar 2,5 kali lipat. Secara umum preparasi multikomponen kristal piperin-asam nikotinat dapat meningkatkan laju disolusi piperin
