Abstract

Some herbal formulation containing α-mangostin as a marker compound in the pericarp of the mangosteen (Garcinia mangostana L.) are marketed now in Indonesia. The determination of the component of preparation both qualitatively and quantitatively is essential to ensure quality of the product. This research is aimed to develop a TLC (Thin Layer Chromatography)-densitometry method to analyze α-mangostin in the herbal drinks of mangosteen rind. The analysis was performed with TLC Scanner using Camag TLC Scanner 4 with Wincats software. Silica gel 60 F254 (Merck) plates (20x20 cm: 200 μm thickness) were used as stationary phase and mixture of chloroform and ethyl acetate (9:1,v/v) as a mobile phase. The developed chromatogram was scanned at 316 nm, the wavelength of maximum absorption for α-mangostin. At this condition, the Rf value of α-mangostin is 0.48. Regression analysis of the calibration data showed an excellent linear relationship between peak-area vs drug concentration. Linearity was found to be in the range of 50-400 µg/mL. The suitability of developed TLC densitometry method for estimation of α-mangostin was established by validating it as per the ICH guidelines. The limits of detection (LOD) and quantification (LOQ) for α-mangostin were found to be 54.38 μg/mL and 181.28 μg/mL. The developed method was found to be linear (r=0,99415), precise (RSD <1.28 % and < 2.39 % for intra-day and inter-day precision) and accurate (mean recovery of 90.04%). Results of the samples analysis showed that sample A does not contain α-mangostin, whereas sample B and C contain α-mangostin. The average concentrations obtained for samples B and C were 0.040% w/v and 0.118% w/v, respectively. The developed method has been successfully applied for the determination of α-mangostin in the herbal drinks of mangosteen rind.

Abstract

Kulit Jeruk nipis (Citrus aurantifolia Swingle) merupakan bagian tanaman yang banyak digunakan untuk pengobatan dengan berbagai aktivitas farmakologis. Namun masih sedikit diketahui kajian tentang bakteri endofit yang terkait dengan kulit buah tanaman ini. Penelitian ini bertujuan untuk mengisolasi bakteri endofit dari kulit buah tanaman ini serta menguji aktivitas dalam mengambat pertumbuhan bakteri Staphylococcus aureus, Escherichia coli dan Methicilin-Resistant Staphylococcus aureus (MRSA). Sebanyak enam Isolat bakteri berhasil diisolasi lalu difermentasi dalam media Nutrient Broth (NB).  Hasil fermentasi kemudian diekstraksi menggunakan pelarut etil asetat. Ekstrak kental masing-masing isolat diujikan aktivitas antibakterinya menggunakan metode difusi Kirby-Bauer. Berdasarkan hasil pengukuran zona hambat menunjukkan bahwa ekstrak dari isolat 3 dan isolat 5 memiliki diameter hambat kuat >10 mm. Hasil identifikasi fitokimia secara kualitatif menunjukkan bahwa ekstrak dari isolat 3 menujukkan adanya kandungan alkaloid dan flavonoid sedangkan isolat 5 teridentifikasi senyawa alkaloid, flavonoid dan polifenol. Hasil identifikasi penamaan bakteri endofit menunjukkan bahwa isolat 3 merupakan Bacillus velezensis strain JS25R dan isolat 5 merupakan Staphylococcus sp. Ekstrak dari bakteri endofit ini terbukti memiliki aktivitas terhadap bakteri patogen dan potensial untuk dilakukan isolasi dan pengembangan terhadap senyawa metabolit sekunder yang memiliki aktivitas tersebut.

Abstract

Indonesia is an important country that produces essential oils that are widely used in fragrance cosmetic products. The use of local essential oils for cosmetics needs to be introduced to the youth generation with the aim that they do not just become consumers. Youth organisations in Kerinci have many activities to improve their skills and abilities for a better future. Collaboration between the university and the youth organisation was achieved by providing training on doing cosmetic product training on making cosmetic products. The product uses essential oils called body mist. Nowadays, the youth still know very little about the manufacturing process of cosmetics products. To provide further benefits, this community service activity collaborated with BBPOM in Padang, West Sumatra, to explain cosmetic safety and the registration process of cosmetic products. With this series topic in this training and outreach-related cosmetics, it is hoped that it can increase business ideas for young people who are members of the Sakti Alam Kerinci Organisation.

Abstract

This research is a continuation study in the inventory of lichens of the genus Stereocaulon which is currently focused on the Stereocaulon massartianum Hue collected in the rocks of Diatas Lake, West Sumatera, Indonesia. Reports on phytochemical studies and pharmacological activities of this species are still limited, based on literature studies that have been carried out. This research was conducted to determine their secondary metabolites and potential antibacterial activity. The air-dried thallus of lichen S. massartianum was macerated successively using n-hexane, ethyl acetate and methanol solvents. The compounds were separated by chromatography and recrystallization methods, then analyzed by spectroscopy (UV-Vis, FTIR, 1H and 13C-NMR). Furthermore, the antibacterial activity assay was performed by agar diffusion method on ethyl acetate extract, and TLC-Bioautography for the isolated compound. Three compounds have been isolated from ethyl acetate extract, i.e. atranorin (1), stictic acid (2) and norstictic acid (3). The results of antibacterial assay from the extract showed antagonistic activity against pathogenic bacteria S. aureus, E. faecalis, E. coli, and P. aeruginosa at concentrations of 10 and 20%, while TLC-bioautography of compound 3 exhibited growth inhibition area in all test bacteria.

Abstract

Hyperlipidemia is a major risk factor for cardiovascular diseases. While conventional treatments exist, there is a growing interest in natural remedies with fewer side effects. Ficus elastica has promising medicinal properties, yet its potential as an anti-hyperlipidemic agent remains unexplored. This study aimed to investigate the anti-hyperlipidemic effects of F. elastica using an integrated approach of LC-HRMS-based chemical bioinformatics and in vitro/in vivo experimental validation. The anti-hyperlipidemic potential of F. elastica and its mechanism of action were screened using integrative computational network pharmacology followed by in vitro HMG-CoA reductase inhibition and in vivo lipid-lowering activity in a hyperlipidemia rat model. Network pharmacology analysis identified STAT3, HSP90AA1, and TLR4 as potential core targets involved in lipid and atherosclerosis-related KEGG pathways. Molecular docking simulations revealed high-affinity interactions between F. elastica compounds and the identified targets, notably compound 41 and compound 61. In vitro assay demonstrated that ethanolic extract of F. elastica inhibited HMG-CoA reductase with an IC50 of 297.73 µg/mL. In vivo experiment using a hyperlipidemic rat model showed significant reductions in total cholesterol, triglycerides, and increased HDL levels. The reduction of triglycerides and elevation of HDL level after F. elastica ethanolic extract supplementation is similar to the effect from supplementation of simvastatin. These findings suggest that F. elastica ethanolic extract possesses notable anti-hyperlipidemic properties, likely mediated through multiple molecular targets and pathways. The study highlights the potential of F. elastica ethanolic extract as a promising candidate for anti-hyperlipidemic therapy and underscores the efficacy of integrating computational and experimental approaches in natural product research.

Abstract

Teloschistes flavicans (Sw.) Norman is a lichen known as the golden-haired lichen. This lichen has been recognized and used in herbal medicine mixtures as an antimicrobial and bioindicator of air pollution that plays a role in ecological systems. The aim of this study was to explore the potential of its secondary metabolites as antibacterial and anticancer agents, particularly against bacterial pneumonia. Two main compounds (vicacinin and parietin) were isolated with chromatography and identified by spectrometry and single-crystal X-ray diffraction. The crystallographic data of vicanicin are reported for the first time. Chromatography and recrystallization methods were used to obtain both compounds with orange (parietin) and white (vicanicin) crystals. Furthermore, these compounds were evaluated for cytotoxicity on keratinocytes (HaCaT) cells and antibacterial activity against pneumonia pathogens (Klebsiella pneumoniae ATCC 1706, Streptococcus pneumoniae ATCC 49619, Moraxella catarhalis ATCC 25240, and Staphylococcus pyogenes ATCC 19615). The cytotoxic activity of these compounds was moderate at the concentration of 50-100 µM. The antibacterial pneumonia activity was relatively weak compared to chloramphenicol. Between the two compounds, vicanicin showed stronger activity than parietin against all strains. Vicanicin was more active against Klebsiella pneumoniae and Staphylococcus pyogenes with minimum inhibitory concentrations of 156±0.77 µM and 156±0.91 µM, respectively. In this study, comprehensive molecular structures of parietin and vicanicin have been successfully elucidated, and their antibacterial and cytotoxic activities have been provided.

Abstract

The increasing number of diabetes mellitus cases every year is alarming for every country. More than 90% of the cases are type-2 diabetes mellitus. The range of diabetes mellitus patients is at a productive age, including in Indonesia. Type-2 diabetes mellitus can be managed by reducing postprandial glucose levels. The approach inhibits &alpha;-amylase and &alpha;-glucosidase enzymes. A preliminary study on the local edible plants in Solok district, Etlingera elatior, showed potential against the &alpha;-glucosidase enzyme by TLC-bioautography screening. The essential oils from the inflorescence of E. elatior were extracted by hydro-distillation. The analysis of components from the essential oils identified 20 compounds. GC-MS data showed that the highest abundance percentages were 1-dodecanol (39.79%), dodecanal (28.20%), &alpha;-pinene (13.11%), decanal (4.16%), and dodecanol acetate (2.75%). The inhibition against the &alpha;-glucosidase enzyme obtains the IC50 value of 73.95 and 67.55 ?g/mL for the essential oils and acarbose, respectively. The inhibition against the &alpha;-amylase enzyme obtains the IC50 value of 70.87 and 12.28 ?g/mL for the essential oils and acarbose, respectively. The essential oils from the inflorescence of E. elatior could inhibit &alpha;-amylase and &alpha;-glucosidase enzymes, even though the potential was lower than that of acarbose as a positive control.

Abstract

Indonesia's diabetes cases were in the top ten list globally, with 90% of the patients being type-2 diabetes mellitus (T2DM).  An approach for screening the local edible plants is made for managing or treating T2DM by inhibiting α-glucosidase and α-amylase enzymes. Results showed that inflorescence of Etlingera elatior inhibition toward both enzymes. TLC-autography and IC50 value of fractions from E. elatior were used to identify the potential fractions and possible compounds for the activity. The non-polar fraction was spotted in the active substances based on TLC-autography. Then, the targeted compounds were separated by column chromatography to obtain stigmasterol as one of the active compounds. The IC50 values of total extract, n-hexane, EtOAc, BuOH, and isolated compound against α-glucosidase were 16.0, 7.5, 13.5, 9.7, and 2.0 ppm, respectively. The IC50 values for α-amylase inhibition were respectively 88.6, 48.6, 23.2, 29.1, and 27.5 ppm. The positive control (acarbose) against α-glucosidase and amylase exhibited IC50 values of 153.2 and 12.3 ppm. The inhibition of E. elatior against the two enzymes could be an alternative to delay carbohydrate absorption.
