Abstract

The objectives of study was to determine free radical scavenging and singlet oxygen quenching (SOQ)activity in turmeric leaves extract. Analyses of phytochemicals were based on total phenolic, flavonoid andcondensed tannins. The radical scavenging activities of extracts were determined by 1.1-diphenyl-2-picrylhydrazyl (DPPH) assay. Turmeric leaves powder was extracted with methanol 80%, ethanol 80% danacetone 80% at room temperature for 24 hours, respectively. Singlet oxygen quenching (SOQ) activity ofeach tumiric leaves extracts at 500-1500 ppm level. SOQ activity test was conducted using linoleic acid assubstrates each containing 5 ppm erythrosine as a photosensitizer. The highest total phenolic flavonoidcontent was detected in methanol extract, followed by acetone extract and ethanol extract. Contrary,ethanol extract contained higher contents of condensed tannins than methanol extract and aceton extract.The addition of metanol extracts of turmeric leave in the reaction mixture showed the highest scavengingactivity in DPPH free radical followed by acetone extract and ethanol extract. The singlet oxygenquenching activities of turmeric extracts were greatly different in different solvents. The methanol extractexhibited the highest singlet oxygen quenching activities followed by acetone extract and ethanol extractunder fluorescent light for up to 5 hours. Abstract

The objectives of this study was to synthesized 4,6-diallyl-2-methoxyphenol (DMP) from allyl eugenol by Claisen rearrangement. The reaction was carried out at temperature 170-200oC and the reaction time was 30 minutes. The synthesis of DMP by heating at 170-200oC for a certain period of time appears to be a fairly good and simple method. The synthesis compounds were elucidated by infrared (IR), nuclear magnetic resonance (1H-NMR), and mass spectrometry (MS) techniques. Antioxidant activity evaluated the based on the heating of coconut oil. At higher temperatures of reaction the yield will be higher and at the reaction time 30 minutes will produce will produce higher yield which was 59,19%. The gas chromatographic analysis showed that the purity of DMP was 96,45% The structure of 4,6-diallyl-2-methoxyphenol was identified by IR spectrometry which shows strong absorption at 3527 cm-1 indicating hydroxyl group from the phenolic compound and 1H NMR spectra showed that the number of protons of aromatic ring was two hydrogen while the mass spectrometry showed the molecular ion peak at m/z 204 as base peak and absence of peak at (M-41)+ as the indicator of ally eugenol. The addition of DMP at concentration 200 ppm in coconut oil exhibited excellent inhibition of oxidation reaction more than BHT as control positive. The result concluded that 4,6-diallyl-2-methoxyphenol had antioxidant activity on heating of coconut oil at 80oC for 30 day.

Abstract

Tujuan penelitian ini adalah untuk menentukan potensi ekstrak pisang goroho (Musa spp) dalam menurunkankadar gula darah tikus putih. Penelitian ini dilaksanakan menggunakan metode percobaan laboratorium, yangmenganalisis penurunan gula darah dan kadar MDA tikus putih. Penelitian dilakukan menggunakan rancanganacak lengkap (RAL), kemudian data yang diperoleh, dianalisis perbedaan. Beda nyata antar perlakuan diujimenggunakan ANOVA deng p<0,05. Jika terdapat beda nyata antar perlakuan, data diuji menggunakanDuncan’s multiple range test (DMRT). Hasil penelitian menunjukkan bahwa pisang goroho mengandung kadarair pisang goroho segar sebesar 69,53%. Fitokima antioksidan yang terbesar yaitu total fenolik, diikuti tanninterkondensasi kemudian terendah senyawa flavonoid. Data menunjukkan bahwa injeksi aloksan menaikkankadar gula darah tikus putih (K, EPS, ETP, EK, G). Pada perlakuan EPS, ETP dan EK mempunyai potensidalam menurunkan kadar gula darah sedangkan EK dan GB tidak berbeda nyata dan dapat dijelaskan bahwakeduanya mempunyai potensi yang sama dalam menurunkan kadar gula darah. Kesimpulan penelitian ini yaitukadar Kadar gula darah tikus yang diberikan Ekstrak pisang goroho yang mengandung fitokimia antioksidanseperti senyawa fenolik, flavonoid dan tannin yang dapat menurunkan gula darah tikus paling tinggi adalahEkstrak Pisang Segar sebesar 67,6 mg/dl (61,19%).The objectives of this research are to determine the potential of goroho banana extract (Musa spp.) todecrease blood glucose. This research using laboratory experiment methods to analyze the decreasing ofblood glucose and malonaldehyde (MDA) on mice. This research using complete random design, thusobtained data were analyzed by regression and differences. Significant difference between groups wereanalyzed using ANOVA with p<0,05. If there is significant difference between groups, data were analyzedusing Duncan’s multiple range test (DMRT).The results shows that goroho banana contain water content phenolic total and condensed tannins, the lowestis flavonoids total. Alloxan increase blood glucose of treatment mice (K, EPS, ETP, EK, G). Statistical analyseson EPS, ETP and EK are not significantly different each other, it because EPS, ETP and EK possessphytochemical antioxidant. Although EPS and EK shows potency as like GB (p>0,05).. As conclusions of thisresearch, goroho banana extract which contains antioxidant phenolics, flavonoids and tannins can decreaseblood glucose of white male wistar.

Abstract

ABSTRAK Penelitian ini bertujuan untuk mempelajari aktivitas antioksidan fraksi pelarut Sagu baruk. Penelitian dimulai dengan mengekstraksi serbuk Sagu baruk menggunakan cara maserasi selama 4 hari dengan pelarut etanol 80%. Ekstrak kemudian dipartisi berturut-turut menggunakan pelarut n-heksan, etil asetat, butanol, dan aquades. Kemudian ditentukan kandungan total fenolik, total aktivitas. Hasilnya menunjukkan bahwa fraksi etil asetat memiliki kandungan total fenolik tertinggi diikuti fraksi butanol, n-heksan dan aquades. Kandungan total fenolik berturut-turut adalah 81,22; 41,43; 20,20; dan 14,39 mg/kg. Fraksi etil asetat juga memiliki total aktivitas antioksidan tertinggi dibandingkan dengan fraksi lain. Berdasarkan penelitian ini, dapat disimpulkan bahwa fraksi etil asetat adalah fraksi terbaik yang dapat berperan sebagai antioksidan.ABSTRACT To evaluate the antioxidant from the solvent fraction of Sago Baruk (Arenga microcarpha) was the purpose of this study. These study initiated by extracting the powder of Sago Banrk by way of maceration for 4 days using 80% ethanol. Extract then partitioned using a series of solvent such as n-hexane ethyl acetate, buthanol, and distilled water, respectively. Then continued to determined the total phenolic content, total antioxidant. The results showed that the ethyl acetate fraction had the highest content of total phenolic followed by the fraction of buthanol, n-hexane and distilled water, with the value of total phenolic content as follow 81.22 (ethyl acetate);41.43 (buthanol);20.20 (n-hexane); and 14.39 mg I kg (aquades). Ethyl acetate fraction also have the highest total antioxidant activity compared to the other fractions. Based on this study, the ethyl acetate fraction was the best fraction with the ability of antioxidant.

Abstract

Singlet oxygen chemistry and photooxidation reaction, in general, often require one or more criticalreaction steps that involve a sensitizer and light for photochemistry production of singlet oxygen from atriplet state. Singlet oxygen can be formed chemically, enzimatically, and photochemically. However, themost well documented formation mechanism for singlet oxygen is photochemistry using sensitizers.Chlorophyll, riboflavin, myoglobin, porphyrins, erythrosine, rose bengal, methylene blue, bilirubin andretinal are well-known photosensitizer and can absorb energy from light and transfer it to triplet oxygen toform singlet oxygen. Photooxidation provides an important way to produced hydroperoxides fromunsaturated fatty acids in presence of oxygen, light energy and photosensitizer. Hydroperoxide are themajor initiation products of lipid oxidation chain reactions. Singlet oxygen participated in the initiation stepof oil oxidation and the reaction rate of singlet oxygen with oleic, linoleic acid and linolenic are about30.000, 1450 and 900 times greater than that of triplet oxygen. Abstract

The objectives of this study was to determine phytochemicals content and free radical scavengingactivities of bread fruit leaves. Bread fruit leaves were finely ground in a laboratory mill and samples wasextracted with methanol, ethanol and acetone under room temperature, respectively. Analyses ofphytochemicals based on total phenolic, flavonoid and condensed tannins. The radical scavengingactivities of extracts were determined by 1.1-diphenyl-2-picrylhydrazyl (DPPH) and the ferric reducingability plasma assay. The highest total phenolic content was in methanol extract (EM), followed byethanol extract (EE) and acetone extract are 179.89 ± 3.17; 152.55 ± 3.17 and 62.46 ± 1.31 mg/Kgrespectively. EM possessed highest contents of total flavonoid and condensed tannins compared with EEand EA. The addition of methanol extracts of in the reaction mixture showed the highest scavengingactivity in 1.1-diphenyl-2-picrylhydrazyl (DPPH) radical 74.80% followed by EE 71.80% and 38.80%.These result suggested that contains phenolic compounds groups having free radical scavengingactivities.

Abstract

This research aims to study the anti UV-B activity from solvent fractions of corncobs. The study initiating by extracting the powder corn cobs using way reflux for two hours with 80% ethanol. Extract then partitioned using a series of solvent; n-hexane, ethyl acetate, butanol and distilled water. Then was determined the effect of fractions against lipid oxidation induced by UV-B light. The results showed that the ethyl acetate fraction showed the lowest content of conjugated diene followed fraction of butanol, n-hexane and distilled water. The content of conjugated diene was 21.89; 25.11; 42.94; and 46.08 mmol/kg, respectively. Based on this study, the fraction of ethyl acetate fraction could act as an antioxidant and anti UV-B better than the other fractionsPenelitian ini bertujuan untuk mempelajari aktivitas anti UV-B dari fraksi pelarut tongkol jagung. Penelitian dimulai dengan mengekstraksi serbuk tongkol jagung menggunakan cara refluks selama dua jam dengan pelarut etanol 80%. Kemudian ditentukan pengaruh fraksi terhadap oksidasi lipid yang diinduksi dengan cahaya UV-B. Hasilnya menunjukkan bahwa fraksi etil asetat memiliki kandungan diena terkonjugasi terendahdiikuti fraksi butanol, n-heksan dan aquades. Kandungan diena terkonjugasi berturut-turut adalah 21,89; 25,11; 42,94; dan 46,08 mmol/kg. Berdasarkan penelitian ini, dapat disimpulkan bahwa fraksi etil asetat dapat berperan sebagai antioksidan dan anti UV-B lebih baik dibandingkan dengan fraksi lainnya.

Abstract

ABSTRACTMongan et al., 2011. Production and fractionation of liquid smoke from corn cob to inhibit lipid peroxidation oflaying fish (Decapterus russelli).Corncob is a cellulose material and can be made to liquid smoke by pyrolysis. Liquid smoke from pyrolysis canbe purified using adsorbent. Chitin is an adsorbent produced by crustacea shell. Liquid smoke purified by chitincan act as antimicroba and antioxidant. Layang fish is very abundantly in Gorontalo Province, so preservativetechniques were needed to prevent lipids peroxidation in fish. The aims of this research was to determineantioxidative activity of liquid smoke from corncop to prevent lipids peroxidation on layang fish.This research using laboratory experiment methods to analyze total phenolic content, free radical scavengingactivity, total antioxidant and lipids peroxidation on layang fish by spectrophotometer UV-vis. This researchusing complete random design (rancangan acak lengkap, RAL), thus obtained data were analyzed usingstatistic. Significant difference between groups were analyzed using ANOVA with p<0,05. If there is significantdifference between groups, data were analyzed using Duncan’s multiple range test (DMRT).The results showed that liquid smoke can be produced using corncop by pyrolysis is 31,7 %. Fractioned liquidsmoke using chitin obtained 4 fractions were fraction I, II, III and IV. Liquid smoke and fractioned liquid smokecontain total phenolic content are 280, 273, 280, 286 and 301 mg/kg gallic acid, respectively. Free radicalscavenging activity from liquid smoke and fractioned liquid smoke are 87,39, 86,08, 89,29, 89,90 and 93,73%,respectively. Although, total antioxidant from liquid smoke and fractioned liquid smoke are 59, 35, 51,32, 51,57,52,74 and 53,58 ppm, respectively. Fraction IV of liquid smoke fractioned with chitin was used in antioxidativeactivity of laying fish test. Fraction IV with concentration 25% can inhibit lipid peroxidation on fresh layang fishby 19 mikromolar for two days. Although layang fish without liquid smoke shows high lipid peroxidation by 24,96mikromolar. Fraction IV with concentration 25% can inhibit lipid peroxidation on layang fish by 18,28 mikromolarfor two days. Antioxidative activity from liquid smoke caused by it phenolics compound that act as antioxidant.As conclusions of this research, liquid smoke can be produced from corncob and fractioned with chitin. Liquidsmoke contain phenolics and act as antioxidant. Liquid smoke can inhibit lipids peroxidation in layang fish. Thisresearch suggested that liquid smoke can be used to inhibit lipid peroxidation on layang fish..Keywords : liquid smoke, corn cob, peroxidation, layang fish

Abstract

ABSTRAK Penelitian ini bertujuan untuk mengisolasi dan mengkarakterisasi fraksi flavonoid dari ekstrak etil asetat tongkol jagung serta menentukan aktivitas singlet oxygen quenching. Tongkol jagung diekstraksi dengan cara refluk menggunakan etanol 80% selama 2 jam. Setelah itu disaring dan filtratnya diuapkan dengan rotary evaporator. Ekstrak etanol disuspensikan dalam air dan diekstraksi berturut turut dengan petroleum eter, etil asetat, butanol dan air. Fraksi pelarut terbaik difraksinasi dengan kromatografi kolom menggunakan silika gel 60 dan eluen n-heksana: etil asetat (4:6). Fraksi dievaluasi dalam singlet oxygen quenching dengan sistem reaksi fotooksidasi asam linoleat (0,03 M) dalam sistem emulsi yang mengandung 5,68 x 10-3 mM eritrosin sebagai sensitiser dan campuran reaksi disinari cahaya fluoresen (4000 lux) selama 5 jam. Hasil karakteristik senyawa hasil isolasi (F2) berdasarkan hasil analisa UV-Vis terdapat 2 pita serapan pada panjang gelombang 289 nm (pita I) dan panjang gelombang 313 nm (II) yang mengindikasikan bahwa senyawa hasil isolasi termasuk golongan flavonoid jenis flavanon atau dihidroflavon. Spektra UV senyawa terisolasi dengan pereaksi geser kemungkinan memiliki gugus hidroksi yang terletak pada atom C-5 atau C-7. Analisis spektrofotometer infra merah (IR) menunjukkan adannya bilangan gelombang 3375 cm-1 (OH, fenol), 2920 cm-1 (CH), 1462 cm-1 (C=C aromatik), 1604 cm-1 (C=0) dan 1031 cm-1 dan 1167 cm-1 (C-O-C). Isolat (F2) memiliki aktivitas singlet oxygen quenching (antiphotooxidation) terhadap fotooksidasi asam linoleat yang mengandung eritrosin sebagai sensitizer.ABSTRACT The objective of this research was to isolate and characterized flavonoid fractions from corn cob ethyl acetate extract and to determine the singlet oxygen quenching activity. Corn cob was extracted with a reflux method using 80% ethanol solvent for two hours. After the extraction, filttering was occurred and the filtrate was evaporated with rotary evaporator. The ethanol extract was suspended in water and continually extracted with petroleum ether, ethyl acetate, buthanol and water. The best solvent fraction was fractionated with column chromatography using silica gel 60 and n-hexane and ethyl acetate (4:6) as eluent. The fraction was evaluated in singlet oxygen quenching with the photooxidation of linoleic acid (0.03 M) reaction system inside an emultion system that contains 5,68 x 10-3 mM of eritrosin as sensitizer and the mixed reaction was lit by a fluorescent light (4000 lux) for 5 hours. The characteristic result of the isolated compound (F2) based on the UV-Vis analysis result there are two absorptopn band at 289 nm (band I) and 313 nm wavelength (band II) that indicate that the isolated compound belongs to the flavonoid group as a flavanon or a dihydroflavon type. The UV spectra of isolated compound with shifting reagent showed having a hydroxyl group located at the C-5 or C-7 atom. The infrared (IR) spectrophotometer analysis showed the wave number 3375 cm-1 (OH, phenol), 2920 cm-1 (CH), 1462 cm-1 (C=C aromatic), 1604 cm-1 (C=O) and 1031 cm-1 and 1167 cm-1 (C-O-C). The isolate (F2) has the singlet oxygen quenching activity (antiphotooxidation) on the photooxidation of linoleic acid that contain erythrosine as sensitizer.
