Abstract

Growth Promoter Antibiotics are used to prevent disease and promote growth and production in poultry. Repeated admnistration of feed can have a microorganic resistance effect, accumulation of antibiotic residues in animal and environmental products and imbalance of normal microflora in the intestine. The antibacterial and carbohydrate content of some natural ingredients can be potential as a replacement candidate for AGP. This study aims to determine the role of a combination of wild ginger, honey, and probiotics (Bacillus subtilis and Lactobacillus acidophilus) as AGP candiate in Vitro. The antibacterial activity of the combination of wild ginger and honey against pathogens (E. coli) and their use against probiotics was tested by disk diffusion method, while the calculation of optical density values to determine the minimum inhibitory concentration and minimum bactericidal concentration was carried out on E. coli. The ability of inhibition of probiotics against pathogens is also done by the disk diffusion method. The disk diffusion test results showed the best combination was extract of 25% wild ginger aquades + 100% Lombok honey with inhibition zone diameter (8,53 ±, 03). Optical density values indicate this combination is able to inhibit and kill E. coli (DO 0,00 ±, 002) and support B. subtilis (DO 0,18 ±, 002) and L. acidophilus (DO 0.25 ±, 005) significantly better than positive control. MIC value of wild ginger aquades extract and honey combination against E. coli is wild ginger aquades extract 3.13% + Lombok honey 25%, while MBC value is wild ginger aquades extract 6,25% + Lombok honey 25%. The combination of B. subtilis and L. acidophilus showed the largest inhibitory zone diameter against E. coli (7,30 ±, 02 mm) compared to individual colonies. The combination of wild ginger and honey, in addition to inhibiting also able to kill pathogens and support the growth of probiotics, so this formula can be used as one of the replacement candidates for AGP.

Abstract

Tujuan penelitian ini mengetahui kualitas mikrobiologi susu kambing mentah yang diambil langsung dari ambing. Dalam penelitian ini digunakan 50 sampel susu kambing peranakan Ettawa (PE). Sampel dianalisis terhadap total plate count (TPC), jumlah Staphylococcus sp., jumlah koliform, Escherichia coli (E. coli), dan Salmonella sp. berdasarkan reaksi biokimia. Rerata untuk TPC; Staphylococcus sp.; total koliform masing-masing adalah 1,65x10 3 ; 5,75x10 3 ; 1,3x10 cfu/ml, sedangkan E. coli dan Salmonella sp. adalah negatif. Berdasarkan Standar Nasional Indonesia (SNI) No 01-6366-2000 tentang persyaratan susu segar, maka TPC, koliform, E. coli, dan Salmonella sp. memenuhi standar, sedangkan Staphylococcus sp. melebihi ambang batas. Hasil penelitian menunjukkan bahwa susu kambing mentah yang diambil langsung dari ambing masih layak konsumsi.

Abstract

Penelitian ini bertujuan mengevaluasi status virulensi 22 isolat Bacillus anthracis (B. anthracis) asal Nusa Tenggara dan Papua  menggunakan metode polymerase chain reaction (PCR) multiplex dengan dua pasang primer nukleotida yang memiliki target amplifikasi gen spesifik pada kedua plasmid. Ektraksi DNA dilakukan dengan metode lisis panas. Pasangan primer PA5 dan PA8 digunakan untuk mengamplifikasi gen pagA pada pXO1, sedangkan pasangan primer 1234 F dan 1301 R mengamplifikasi gen capABC pada pXO2. Hasil reaksi PCR menghasilkan dua pita DNA berukuran sekitar 600 dan 800 bp pada 20 isolat. Namun, dua isolat lain, masing-masing hanya memiliki salah satu dari kedua ukuran pita DNA tersebut. Sebagian besar koleksi isolat asal Nusa Tenggara dan Papua (91%) masih memiliki kedua plasmid secara lengkap (pXO1+/2+) dan karena itu bersifat virulen, sedangkan dua isolat lain (9%) telah kehilangan salah satu plasmid virulennya sehingga bersifat avirulen. Disimpulkan bahwa PCR multiplex dengan dua pasang primer dengan target amplifikasi pada plasmid dapat digunakan untuk evaluasi status virulensi isolat B. anthraci.

Abstract

Penelitian ini bertujuan mengetahui potensi kit deteksi cepat Anigen® rapid test kit rabies Ag dalam mendeteksi virus rabies pada sampel otakanjing yang diperoleh dari lapangan yang meliputi batas deteksi, kecepatan reaksi, uji reaksi silang, uji sensitivitas, dan spesifisitas. Batas deteksi ditentukan dengan pengenceran secara serial kontrol positif virus rabies dan selanjutnya diuji dengan rapid test kit sesuai petunjuk produsen. Uji reaksi silang dilakukan dengan canine parvovirus, Escherichia coli, dan Salmonella sp. Uji sensitivitas dan spesifisitas dilakukan terhadap sampel otak yang telah dikonfirmasi positif rabies dengan uji fluorescent antibody technique. Konfirmasi uji rapid test tersebut dilakukan dengan reverse transcriptase polymerase chain reaction. Berdasarkan data yang diperoleh dalam penelitian ini dapat disimpulkan bahwa Anigen® rapid test kit rabies Ag mampu mendeteksi sampel yang mengandung virus rabies dengan titer 0,5 x log 106,5/0,03 ml, dengan rata-rata kecepatan reaksi 1,8 menit 29,35 detik (kurang dari 2 menit). Di samping itu Anigen® rapid test kit rabies menunjukkan tidak terdapat reaksi silang dengan canine parvovirus, Escherichia coli, dan Salmonella sp. serta mempunyai sensitivitas 92,30% dan spesifisitas 97,90%

Abstract

Antibiotic Growth Promoters (AGPs) are antibiotics that are used commonly in livestock farming to increase animals growth rate. The use of AGP has been prohibited in Indonesia (No.14/PERMENTAN/PK.350/5/2017). AGP banning urges some innovations to find the alternative of AGP and one of them is utilization of natural resources. Papaya leaves, basil leaves, Curcuma aeruginosa rhizomes and honey contain flavonoid that has antibacterial activity. The purpose of this research is to know the effect of Papaya leaves, basil leaves, Curcuma aeruginosa rhizomes and honey against Serratia marcescens growth. Re-identification of Serratia marcescens were done by looking at the colony morphology, cell morphology and biochemical tests. Antibacterial activity of ethanol (100%) and aquades (33.33%) extract of the herbals and the honey (100%) against Serratia marcescens were tested by disc diffusion method. Each test was repeated 2 times. The results showed that Lanceng (Trigona bee) honey from Gunung Kidul, Black honey from Lombok, White honey from Lombok, ethanol and aquades extract of the herbals are not effective to inhibit Serratia marcescens growth. Commercial honey (7.59±.22 mm) has the highest antibacterial acivity to Serratia marcescens, followed by honey from Kupang (6.69±.21 mm). Commercial honey and honey from Kupang have moderate antibacterial activity.

Abstract

Clinical symptoms in the chicken’s respiratory system, such as sores, nasal discharge, gasping, and so on, can be caused by a variety of possible socks. These signs may be caused by a virus, usually followed by economic losses. Causative diagnosis, especially virus detection, is needed to establish the cause of the symptoms. The study aims to detect Newcastle Disease (ND), Avian Influenza (AI), and Infectious Bronchitis (IB) cases of respiratory symptoms in commercial chickens by 2023. The samples studied in this study were organs from commercial chicken broilers and layers with respiratory symptoms. The organ was prepared as a suspension, then centrifuged to take out the supernatants. Supernatants were extracted using commercial extraction kits to produce templates for molecular detection. Molecular detection was performed using the RT-PCR two-step method. Primer pairs were used for the detection of ND, AI subtypes H5 and H9, as well as IB. The results were visualized by electrophoresis with 1.5% agarose. Interpretation of the positive result of RT-PCR based on the appearance and length of amplicons compared with the positive control of each virus. Positive RT-PCR samples with thick amplicon quality were subjected to sequencing and bioinformatics analysis. Results of detection with RT-PCR against 41 samples are positive ND of 1 out of 8 samples (12.5%), AI of 2 out of 17 samples (11.7%), and IB of 5 out of 16 samples (31.3%). Based on the molecular detection with RT-PCR, the symptoms of respiration in commercial chickens are confirmed due to the cause, namely the virus: ND genotype VII-i, AI subtype H5 clade 2.3.2, AI subtype H9, and IB genotype GI-19 (QX-like).
