Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh dosis sperma untuk Inseminasi Buatan (IB) pada ayam kampung terhadap fertilitas telurnya. Dosis IB ini berguna untuk mengetahui perbandingan jantan dan betina yang paling efisien dalam suatu pemeliharaan. Sperma ditampung dan dicampur dari 5 ekor ayam Pelung jantan yang berumur sekitar 52 minggu. Sperma yang telah diketahui konsentrasinya kemudian dibagi menjadi 3 bagian dan masing-masing diencerkan dengan NaCl fisiologis sehingga diperoleh dosis akhir yaitu 25x106/0,1 ml (dosis 1); 75x106/0,1 ml (dosis 2); dan 150x106/0,1 ml (dosis 3). Sembilan ekor ayam kampung betina diinseminasi untuk mengetahui pengaruh dosis terhadap fertilitas telur. Ayam betina yang digunakan adalah ayam kampung yang berumur sekitar 30 minggu. Hasil penelitian menunjukkan bahwa perlakuan dosis sperma tidak berpengaruh pada fertilitas telur ayam kampung. Fertilitas telur untuk masing-masing perlakuan dosis 1, 2, dan 3 adalah 44,13±20,84; 66,67±23,57; dan 58,75±14,36%. Kesimpulan dari penelitian ini adalah dosis sperma tidak berpengaruh pada fertilitas telur ayam kampung.(Kata kunci: Ayam kampung, Dosis sperma, Inseminasi buatan)

Abstract

Penelitian ini memanfaatkan hasil samping rumah potong hewan (RPH) sebagai sumber oosit untuk in vitro fertilization (IVF). Untuk meningkatkan keberhasilan IVF dilakukan suplementasi fetal calf serum (FCS) pada medium maturasi in vitro. Ovarium sapi dari RPH dibawa ke laboratorium dalam medium NaCl 0,9% pada suhu 31-34ºC. Selanjutnya oosit diaspirasi menggunakan syringe 3 ml dan jarum 23 G yang berisi Dulbeco’s-Phosphate Buffer Saline (DPBS), kemudian dimaturasikan pada inkubator CO2 modifikasi dalam medium Tissue Culture Medium-199 (TCM-199) (CO2 5%, kelembaban 99% dan suhu 37-39ºC). Oosit dibagi menjadi 2 kelompok yaitu kelompok kontrol (TCM-199) dan kelompok perlakuan (TCM-199 + 10% FCS). Angka maturasi dianalisa dengan Chi-Square, sedangkan kualitas oosit dianalisis secara deskriptif. Maturasi oosit sapi pada kelompok perlakuan berbeda nyata (P≤0,05) dibandingkan kelompok kontrol (55,22% vs 40,09%). Penggunaan 10% FCS pada medium maturasi dapat menghasilkan kualitas oosit matur yang lebih baik dibandingkan kelompok kontrol. Kesimpulan penelitian ini adalah penggunaan 10% FCS suplementasi dapat meningkatkan kemampuan maturasi oosit sapi in vitro.(Kata kunci: Fetal calf serum, Kultur oosit, Maturasi in vitro, Oosit sapi)

Abstract

The study was carried out to investigate the effect of gonadotropin hormone supplementation into in vitro maturation medium on maturation, fertilization and embryo development of Bligon goats. This research steps consist of oocyte collection, in vitro maturation, in vitro fertilization, and in vitro embryo development. At the maturation stage the oocyte that had been collected and divided into two groups based on the maturation medium, that was tissue culture medium (TCM) with supplementation of GnRH 0 IU/mL and GnRH 25 IU/mL. Oocyte and embryo morphology data were analyzed descriptively. Maturation rate and embryo development data were analyzed by using independent sample t-test. Fertilization data was analyzed descriptively. The result showed the percentages of mature oocytes from gonadotropin supplementation of 0 IU/mL and 25 IU/mL were 54.10±25.97 and 54.89±26.44%, respectively. Expansion cumulus cells surrounding the oocytes might indicated the mature oocytes. Cleavage rate of the 2 cells stage were 13,02±11,09 and 27,01±16,65%; respectively, and for the 4 cells stage were 10,16±10,01% and 16,67±14.91%. Embryos obtained from the treatment, indicated uniform of blastomeres in the size, tight, compact, intact, and round-spherical shape. It could be concluded that supplementation of gonadotropin hormone into in vitro maturation medium could not increase the rate of oocyte maturation and 4 cell embryo development, but it could increase 2 cell embryo development of Bligon goats. Hormone supplementation could improved the maturation and embryo quality.

Abstract

The study was carried out to investigate the effect of follicle stimulating hormon (FSH) into in vitro maturation medium to increase oocytes maturation and 4 cell stadium embryo development of Bligon goat. Goat ovaries were obtained from a slaughterhouse and transported to the laboratory in a flask of NaCl at temperature of 31 – 34°C. Oocytes were aspirated from 2 – 6 mm of follicles into a 3 mL syringe (23G needle) that contained Dulbecco’s Phosphate-Buffered Saline. Oocytes were divided into three groups, i.e tissue culture medium (TCM) with FSH supplementation 0, 50, and 100 IU/mL. Oocytes were put into those medium and incubated on 39°C, 5% CO2, and 95% humidity for 24 hours. Matured oocytes were fertilized with capacitated frozen thawed-semen and incubated on 39°C, 5% CO2, and 95% humidity for 5 hours. Fertilized oocytes were washed for 3 times in TCM and incubated in the same condition for embryo culture. The data of FSH supplementation and embryo development were analyzed using randomized completely one way classification. The results showed that the percentages of mature oocytes from FSH supplementation 0, 50, and 100 IU/mL were 70,48±23,22, 78,48±15,80, and 80,29±12,86%, respectively. Cleavage rate of the two cells stage were 36,00±14,22, 44,00±33,94, and 57,45±31,78%, respectively, and for the 4 cells stage were 27,33±22,04, 35,33±40,73, and 39,45±20,38%. It is concluded that supplementation of FSH in the maturation medium could not increase the percentages of in vitro maturation and embryo development.

Abstract

This experiment was conducted to determine the correlation of progesterone and cortisol levels in plasma between ovulated and non-ovulated Crossbreds Ettawa does. Eight does were used in this experiment and they were divided equally into 2 groups, i.e. group I consisting of 4 normal ovulated does with the average body weight of 37.5 ± 3.109 kg and group II consisting of 4 non-ovulated does with the average body weight of 28 ± 2.160 kg as group II. The estrus cycles of all does were synchronized using intravaginal device of controlled internal drug release (CIDR) accompanied by intra-muscular injection of prostaglandin F2 (PGF2) alpha. Immediately after the onset of estrus, blood samples were collected from jugular every 3 and 6 hours for 72 hours after onset of estrus. The concentration of cortisol and progesterone in plasma was assayed by enzyme linked immunosorbent assay (ELISA). The results showed that the average concentrations of cortisol in ovulated does (90.89±26.22 ng/mL) was higher than in non-ovulated does (42.70±37.18 ng/mL). Similarly, the concentrations of progesterone in ovulated does (0.098±0.0423 ng/mL) was higher than in non-ovulated does (0.093±0.056 ng/mL). It was evident that the change in progesterone level was closely associated with the change in cortisol level in plasma.

Abstract

Kegiatan pengabdian masyarakat ini bertujuan untuk membantu peternak meningkatkan pengetahuan dan mempersiapkan pakan sapi perah pada masa transisi, yaitu tiga minggu sebelum dan setelah beranak. Pengabdian masyarakat berlangsung dari bulan April sampai November 2019 di Kelompok Peternak Sapi Perah Ploso Kerep, Umbulharjo, Cangkringan, Sleman. Pelaksanaan kegiatan terdiri dari pengamatan kondisi awal kelompok, penyuluhan, pelatihan pembuatan konsentrat dan mineral blok, serta pemanfaatan produk hasil pelatihan pada sapi perah. Hasil kegiatan menunjukkan peningkatan pengetahuan peternak tentang pakan sapi perah pada periode transisi, ciri-ciri konsentrat yang berkualitas, cara pembuatan konsentrat dan mineral blok masing-masing sebesar 50%; 42,9%; dan 50%. Peternak berhasil membuat konsentrat dan mineral blok. Konsentrat hasil pelatihan memiliki kadar protein kasar 15,77% dan energi dalam bentuk  total digestible nutrient  65,98%. Palatabilitas konsentrat dan mineral blok baik. Sebagai kesimpulan, kegiatan pengabdian masyarakat ini meningkatkan pengetahuan peternak dalam pengelolaan pakan selama periode transisi. Pelatihan memberikan ketrampilan membuat konsentrat dan mineral blok untuk mencegah gangguan kesehatan sapi perah selama periode transisi.

Abstract

This research was aims to determine the effect of formaldehyde using as a protective material in the soy groats and lemuru fish oil mixtured to escape of the protein and poly unsaturated fatty acid (PUFA) by in vitro. The rumen fluid was derived from donor of the Ongole crosbred fistulated as much as 3 heads. The experimental design used completely randomized design factorial 2x3 pattern consists of two factors:the first factor (soybean groats:lemuru fish oil in the ratio 2:1 and 4:1) and the second factor (the use of formaldehyde at levels of 0%, 2% and 4% base on the dry matter) and was repeated 5 times. The parameters are consist of the nutrient content, the substrates soybean groats and lemuru fish oil the rumen fluid fermented PUFA content by in vitro. Results of the study are levels of protein, fatty acids  oleic, linoleic and arachidonic much available on soybean groats:lemuru fish oil protected formaldehyde at levels of 2% and 4%. The difference between the ratio of soybean groats with lemuru fish oil had no significant (P≥0,05) but significantly different at different levels of formaldehyde (P≤0,05). The conclusion are formaldehyde content of 37% with a level of 2% can be used for protection the soybean groats and lemuru fish oil with a ratio of 4:1 and is able to provide protein as well PUFA. Keywords: soy groats, lemuru fish oil, in vitro, PUFA, formaldehyde protection

Abstract

This research aimed to identify the effect of the length of thawing time on the quality  of  Bali  cattle postthaw semen and  to  investigate  the resulting pregnancy  rate. This  research  was  conducted  at  Wanasaba  Village,  specifically  Tanaq  Mira  Village, Wanasaba  District,  East  Lombok  Regency.  Five  samples  of  Bali  cattle  semen  were  collected  for  insemination  from  each  of  three  different  farmer  groups.  The  semen sample  from  the  remaining  frozen  semen  inseminated  by  the  inseminator  was  used  in this research. Tanaq Mira Village's inseminator performed thawing during the trip from the  artificial  insemination  (AI)  station  to  the  farmer  group's  location.  This  research observed  the  microscopic  quality  of  the postthaw frozen  semen  covering  motility, viability, and abnormality. Moreover, the pregnancy rate on AI acceptors using the non return  rate (NRR)  parameter  or  the  number  of  female parents who returned to  estrus after being inseminated were observed. The observation was conducted on three farmer groups  with  different  distances  and  lengths  of  thawing  time.  Each  farmer  group  was observed five times. The collected data were then analysed by using one-way analysis of variance(ANOVA). The research indicated that the distance of the farmer group’s location significantly affected (P<0,01) the sperm motility and viability. However, it did not significantly affect (P>0,05) the sperm abnormalities. The Sapeng farmer group had the  highest NRR, as  much  as 80%. According  to the  NRR  score,  it  can  be  concluded that the thawing process  using  the  thermos  within  less  than 10 minutes  resulted  in  the best yield.
