Abstract

Memories are storage in cholinoceptive cells, the cells which are enriched with microtubule-associated protein 2 (MAP-2) that localized in the neuronal dendrite and the cell bodies. Phosphorylation of MAP-2 may increase memory with reduce stability of dendrite by altered dendrite length and lead new side-branches of neuronal as a neuronal plasticity processes in cerebral cortex. The aim of this research is to study the distribution of MAP-2 phosphorylation neurons in cerebral cortex of long-tailed macaques in the third semester of gestationalimmunohistochemically using avidin biotin conjugated complex method. Neurons MAP-2 phosphorylation immunoreactive were located in dendrites and cell bodies, mostly in pyramidal neurons of cerebral cortex. Intensity of MAP-2 phosphorylation immunoreactivity in layer V were stronger than another layer and the neurons that very intensely stained were the pyramidal cells in frontal and parietal lobes, that was suggested that neurons in this areas more responsive to neuroplasticity. From the results we concluded that MAP-2 phosphorylation already distributed in the cerebral cortex of long-tailed macaque fetuses at the last trimester of gestation, mostly in the pyramidal cells of layer V that is suggested plays a role for preparation of memoryformation.Keywords: fetus, long-tailed monkey, cerebral cortex, memory, MAP-2 phosphorylation

Abstract

Paraquat dichloride has been used by farmers as a herbicide to kill the grass. On the other hand, paraquatdichloride is harmful if enters to the body, causing Parkinson’s disease, since it is disrupting dopamineproduction in the substantia nigra pars compacta or dopamine pathways Nigro striatal pathway. The studywas done to fi nd out the histological changes of catecholaminergic neurons and Nigro striatal pathway causedby paraquat dichloride treatment in Wistar rats as a model of Parkinson’s disease.Twenty-two Wistar rats 3,5 months old were divided into 4 groups, 5 rats each. Group I (control group)were injected with aquabidest, while groups II, III, and IV were injected intraperitoneally with paraquatdichloride in aquabidest, with the dosage 5 , 10 and 15 mg/kg bw respectively. The rats were injected onceper week for 6 weeks. Three days after the last injection, the rats were anesthetized using xylasin (2 mg/kg)and ketamine (20 mg/kg) intramuscularly, and then were intracardiac perfused using physiological saline asprerinse solution, followed by 10% buffered formalin solution as a fi xative. After animals were fi xed, the brainswere removed and embedded in paraffi n block and cut in 12 μm thickness for immunohistochemistry stainingusing tyrosine hydroxylase antibody. The results of staining then were observed under light microscope andanalyzed descriptively.The results showed that the catecholaminergic neurons were distributed in the substantia nigrapars compacta in all treatment groups, however, the cell density were found decreased only in group IV.Catecholaminergic neurons appear in the bipolar and multipolar form, while dopamine ‘Nigro striatal pathway’was found exist in all treatment groups. From our study, histologycally the decreased of catecholaminergicneurons is only found in rats that received paraquat dichloride in dose 15 mg/kg bw for 6 weeks.

Abstract

Kelelawar merupakan salah satu keanekaragaman dan kekayaan satwa di Indonesia. Adanya keanekaragaman jenis makanan maupun habitat kelelawar menyebabkan terjadinya perbedaan dalam mencari dan cara makan dari berbagai jenis kelelawar. Keterkaitan antara struktur indra penciuman dengan jenis dan cara makan kelelawar sangat menarik untuk dieksplorasi, karena sampai saat ini sangat sedikit data mengenai hal tersebut. Penelitian ini menggunakan 3 ekor kalong betina dewasa dan 3 ekor lasiwen betina dewasa, sebagai sampel. Hidung sebagai sampel, kemudian dibuat preparat histologi dengan metode parafin dan  pewarnaan imunohistokimia terhadap ekspresi NSE. Hasil pewarnaan imunohistokimia menunjukkan bahwa imunoreaktivitas NSE pada lasiwen terindentifikasi dalam epitel olfaktorius di kedua sisi septum nasalis, konka dorsalis dan beberapa fila olfaktoria. Pada kalong tidak terindentifikasi adanya imunoreaktivitas NSE.Kata kunci: saraf, epitel olfaktorius, kalong, lasiwen, Neuron-Specific Enolase(NSE)

Abstract

rea postrema merupakan sepasang penonjolan ke dorsal pada bagian kaudal medula oblongata yang berbatasan dengan ventrikel IV. Area ini berperan sebagai chemoreceptor trigger zone (CTZ) pada proses muntah yang juga melibatkan neuron katekolaminergik (neuron KA). Pada kemoterapi penderita kanker, fungsi AP sebagai CTZ diupayakan untuk ditekan agar tidal( ada refleks muntah pada pasien. Dalam rangka lebih memahami neuron KA tersebut, dalam penelitian ini dilakukan pengamatan perkembangan bentuk neuron KA di area postrema (AP) monyet ekor panjang (MEP) mulai fetus (F) umur 40 sampai anak (P) umur 105 hari secara imunohistokimia menggunakan antibodi terhadap enzim tirosin hidroksilase (TH). Hasil penelitian memperlihatkan neuron KA di medula oblongata belwn terlihat pada F40, dan bare dijumpai pada F55 di daerah bakal AP, bentuk bulat dengan inti besar dengan sitoplasma sedikit yang merupakan ciri perkembangan awal bentuk neuron. Prosesus sitoplasma yang pendek pada mulai ditemukan pada F85, dan neuron KA di AP berubah menjadi bipolar pada F100 yang merupakan tanda perkembangan menengah bentuk neuron. Dengan bertambahnya umur, prosesus sitoplasma neuron KA bipolar bertambah panjang yang merupakan ciri tingkat perkembangan bentuk akhir dan ditemukan dominan di AP pada P105. Dari hasil penelitian ini dapat disimpulkan bahwa neuron KA di AP pada MEP berada dalam stadium perkembangan bentuk awal dan menengah selama masa prenatal, dan stadium perkembangan lanjut menuju ke perkembangan akhir terjadi pada masa postnatal.

Abstract

Long tailed macaque was one of animal models in biomedical research because it has  many similarities with humans, both anatomical and physiological properties. There were many research about cerebellum associated with its role in the coordination of muscle activity. Understanding of normal development of cerebellum long tailed macaque may help to understand about the development in human cerebellum and its abnormalities. Embryonic and fetal brain samples were obtained through caesarean section and were  then made for histological preparation stained with cresyl violet. Staining results were observed using a microscope with a digital camera. Images obtained are processed by graphics software Adobe Photoshop CS 8.0. Cerebellum Macaca fascicularis Ed40 showed the isthmus and rhombic lip that were composed of ventricular layer, mantle layer, and marginal layer. Cerebellum Macaca fascicularis Fd55 showed future lobes and future  fissures, but the cortex and medulla are not bounded clear. The cortex consisted of the external granular layer, neuroblast basket, and neuroblast stellate, while the  medulla consisted of neuroblast deep cerebellar nuclei. From this research, we concluded that neurons were on stage of proliferation and migration in the embryo aged 40 days, then differentiated and migrated to form cortex  cerebellum and deep cerebellar nuclei at the age of 55 days, but the development of the cerebellum was not fully completed yet.
