Abstract

This study examined the correlation between the concentration of blood metabolites (triglyceride, albumin, phosphorous) on the reproductive performances (S/C and PPM) of lactating Holstein Friesian crossbred (HFC) cows. Blood samples were collected through the caudal vein 8 hours after feeding and the plasma was analyzed at the Integrated Research and Testing Laboratory of Universitas Gadjah Mada (LPPT UGM). Samples were stored at 5° C. Triglyceride, albumin, and phosphorous were measured using glucose oxidase-phenol 4-aminoantipyrine (GOD-PAP) method, photometric bromocresol green method, and cypress diagnostic, respectively. Blood metabolites data were analyzed using Pearson correlation model. The results showed that the correlation coefficients of triglycerides, albumin, and phosphorus levels on PPM and S/C were -0.521 and -0.650; -0.447 and -0.612; -0.513 and -0.700, respectively. In conclusion, there are significant negative correlation between triglyceride, albumin and phosphorus levels on the PPM, and very significant negative correlation between triglyceride, albumin and phosphorus levels on the S/C of lactating HFC cows.

Abstract

Mouse and vole embryos were transferred into pseudopregnant CD-1 and scid female mice. Cellular changes involved in the formation of decidua in the pregnant mouse uterus up to day 8 of pregnancy were examined by histological, electron microscopic, and histochemical techniques. On day 6 of pregnancy, the vole embryos were laid in interstitium of antimesometrial side of the uterus, as well as in intraspecific pregnancy. Compared with intraspesific pregnant mouse, blood vessels were numerous in the decidua around the vole embryos in interspecific pregnancy. Both distribution and dilation of the blood vessels were increased on day 8. A part of cells in the inner cell mass had not nuclei, suggesting damaged vole embryos on day 8. At the implantation site, the uterine decidua was invaded by extravillous trophoblast (EVT) cells whose function is to destroy the walls of the uterine spiral. Moreover, this decidua was infiltrated by a population of natural killer (NK) cells and macrophage. These cells were particularly numerous in the decidua basalis at the implantation site where they come into close contact with invading EVT cells. These results suggest that interaction between NK, macrophage, and EVT provides the controlling relationship of embryo-maternal in intraspecific and interspecific pregnancy.

Abstract

This research was conducted to identify the estrogen hormone profile and estrus response in the thin tailed ewes synchronized with controlled internal drug release (CIDR) implant. This research was carried out by employing 8 thin tailed ewes, CIDR, and estrogen kit. The ewes were distributed into: 3 ewes as control group and 5 ewes as treated group (with 12 days-CIDR implantation). Ewes were raised in the Sidomukti farm group, Sleman Regency, Yogyakarta. The data obtained were estrogen hormone profile during estrus examined by using ELISA method and estrus response marked with the reddening and oedema of vulva, mucus vaginal discharge, behavioral changes, vaginal pH, and superficial cells population. The data were analyzed by applying independent sample T-test. The result did not show any significant difference (P0.05) in the normal range between the control and CIDR group in estrogen hormone profile, estrus response, and pH level, but showed significant differences with positive correlation (P0.05) in population of superficial cells. In conclusion, CIDR was an effective tool for estrus synchronization at the farmer group, and gave positive estrus response towards thin tailed ewes with estrogen hormone profile, pH level, and superficial cell percentage on the normal range.

Abstract

Penelitian ini bertujuan mengetahui respon estrus pada kambing Peranakan Ettawa (PE) dengan Body Condition Score (BCS) 2 dan 3 terhadap implant Controlled Internal Drug Release (CIDR) jangka pendek dikombinasikan dengan injeksi prostaglandin F alpha (PGF a). Sebanyak 10 ekor kambing PE betina dibagi 2 2 dalam 2 kelompok perlakuan berdasarkan BCS, yaitu kelompok I dengan BCS 2 (kondisi kurus), berat badan antara 25-30 kg (n=5) dan kelompok II dengan BCS 3 (kondisi ideal) berat badan antara 35-40 kg (n=5). Semua kelompok perlakuan disinkronisasi dengan implant CIDR (berisi 1,3 g progesteron) secara intravaginal jangka pendek selama 10 hari dan 48 jam sebelum pencabutan CIDR, kambing diijeksi dengan PGF a. Deteksi estrus dilakukan setiap hari dengan interval 6 jam dimulai dari pencabutan CIDR sampai 60 2 jam setelah pencabutan CIDR. Data onset dan durasi estrus dianalisis menggunakan independent sample T-test sedangkan tanda-tanda visual estrus dianalisis secara deskriptif. Respon estrus semua kelompok perlakuan adalah 100%. Onset dan durasi estrus antara BCS 2 dan 3 masing-masing adalah 21,60±1,47 vs 13,20±1,20 jam dan 27,60±3,06 vs 32,40±1,47 jam. Dari hasil penelitian dapat disimpulkan bahwa kombinasi implant CIDR jangka pendek dan injeksi PGF a efektif untuk sinkronisasi estrus pada kambing PE dengan 2 BCS 2 dan 3.

Abstract

Penelitian ini bertujuan mengetahui pengaruh medium maturasi terhadap kompetensi perkembangan in vitro oosit kambing bligon. Ovarium kambing diperoleh dari Rumah Potong Hewan, dibawa ke laboratorium dalam 0,9% NaCl pada suhu 31-34 C. Oosit diaspirasi dari folikel berdiameter 2-6 mm dengan menggunakan syringe 3 ml dan jarum 23 G. Oosit dengan minimal 2 lapisan sel-sel kumulus diproses untuk maturasi in vitro. Oosit dicuci 3 kali dengan dulbecco’s phosphate buffered saline (DPBS) dan tissue culture medium 199 (TCM) 199, kemudian didistribusikan ke dalam 50 μl medium maturasi yang berbeda, yaitu TCM 199 (tanpa suplementasi),

Abstract

Penelitian ini bertujuan mengetahui pengaruh injeksi media kultur embrio supernatan ke dalam uterus mencit 2 hari sebelum transfer embrio (TE) terhadap angka implantasi, yang diindikasikan oleh adanya embrio dan leukemia inhibitory factor (LIF) pada uterus. Mencit jenis Swiss Albino dibagi menjadi 2 kelompok yakni kelompok perlakuan dan kelompok kontrol, masing-masing terdiri atas 30 mencit. Kelompok perlakuan mendapat injeksi media kultur embrio supernatan (MKES) dalam uterus 2 hari sebelum TE sedangkan kelompok kontrol mendapat injeksi media kultur embrio (MKE) dalam uterus 2 hari sebelum TE. Dua hari setelah TE (h6 kebuntingan), mencit diperfusi dengan menggunakan larutan para formaldehid 4% dan diambil uterusnya. Uterus diproses untuk blok parafin dan dipotong dengan mikrotom setebal 5 µm. Preparat dideparafinisasi dan diproses imunohistokimia dengan kit ABC dan antibodi LIF. Preparat diperiksa di bawah mikroskop untuk melihat implantasi embrio, distribusi LIF, dan jumlah LIF di desidua dan non-desidua. Angka implantasi antara kelompok perlakuan dan kelompok kontrol masing-masing adalah 52,77 dan 40,88% (P0,05). Jumlah LIF desidua pada kelompok perlakuan dan kelompok kontrol masing-masing adalah 12,83 dan 8,83 (P0,05) sedangkan jumlah LIF di non-desidua antara kelompok perlakuan dan kelompok kontrol masing-masing adalah 8,00 dan 4,50. Dapat disimpulkan bahwa injeksi media kultur embrio supernatan 2 hari sebelum TE dapat meningkatkan angka implantasi pada mencit.

Abstract

This study was conducted to evaluate fertility grade of Friesian Holstein cows based on milk urea levels. A number of eight cows which fulfill the criteria of not pregnant and in second to sixth lactation, produced milk of 2.5 liters/day, and body weight ranging from 400-450 kg were used in this research. The cows were given fodder that consisted of 30 kg of forage and 10 kg of a mixture of concentrate (pollard, pulp out, and molasses), in the morning and afternoon. Milk and blood samples were taken once a week for five weeks in the morning before fed. Milk urea levels were measured using the urea FS (DiaSys) and blood samples were tested using enzyme linked immuno-sorbent assay (ELISA) with the progesterone hormone kit (DRG, Germany). The variables measured were pregnancy rate, service per conception (S/C), days open, and progesterone levels. Reproduction and milk urea levels data were analyzed by linear regression to determine the relationship between two variables. Progesterone data were analyzed descriptively. The results showed that the average calving interval, days open, and S/C were 607.5±170.27 days; 341.25±187.65 days; and 2.41±2.32 days, respectively. The average of milk urea levels was 14.16±2.55 mg/dL. Regression analysis showed a significant effect (P0.05) and negative correlation between milk urea level and S/C. The average of non-pregnant progesterone hormone level was 12.04±7.13 ng/mL. In conclusion, the fertility grade of Friesian Holstein cows decreased when the milk urea level at 14.16 mg/dL and non-pregnant progesterone at 12.04±7.13 ng/mL.

Abstract

The season impacts livestock productivity and cattle kept in grazing production systems in tropical conditions. This research aims to determine the effect of season on the weight changes of Bali cows and calves on oil palm plantations. This research used 20 Bali cattle kept semi-intensively in oil palm plantation areas without feed supplementation in 2016–2018. Cows and calves were weighed at calving and every month until weaning (150 days). Season grouping was based on calving time: dry season (DS) May–October and rainy season (RS) November–April. Data were analyzed by independent T-test statistics using the SPSS 20.0 application. The results of observations showed that cows' body weight decreased gradually until the fourth month of lactation in both seasons. Cows' weight changes were only significantly different in the second month, in which cows at DS were better than RS (P<0.05). The pre-weaning average daily gain (ADG) of calves each month was not significantly different between the two seasons, and they could reach a body weight of 60.60 kg (RS) and 61.47 kg (DS) at weaning. Season differences do not impact weight changes in the Bali cows and calves in the cattle-oil palm plantation production system.
