Abstract

There are many methods for development  of mammae cancer animal model, one of which is chemical induction using carcinogenic  agent, DMBA. This research aimed to explore the influence of dose and time regimens of DMBA on development of mammae carcinogenesis on Sprague dawley female rats. The first study was 50 rats treated with 20 mg/kg bw of DMBA orally for eleven times at twice a week. Morphological evaluations were conducted with mammae palpation for 15 weeks and then all of  rats were sacrificed for collecting mammae organs for histological analysis using hematoxylin-eosin staining. The results showed that the first and the latest nodules appeared at the fourth-week and the fourteenth-week after ending DMBA induction, respectively, in which the  most often nodule appearances were at the seventh-week. The number of nodule incidence and multiplicity were by 74% and 2 noduls/rat, respectively. Histological analysis of mammae glands determined that they fell under in Ductal Carcinoma Invasive (DCIV) category. The second study was 25 rats gavaged orally with DMBA at dose 20 mg/kg bw for five times every three days. After palpating for 15 weeks, the results showed that no nodule was observed but the histological analysis demonstrated developing of mammae gland carcinogenesis reaching about 60%  Ductal Carcinoma Insitu (DCIS) and 40% Ductal Carcinoma Invasive (DCIV) stages. Based on the results of this study can be concluded that the dose and frequency of DMBA will affect the successful development of mammary gland carcinogenesis. In DMBA induction with low frequency, no data showed the incidence and multiplicity of tumor, but histopathologic level  carcinogenesis can be distinguished. In DMBA induction with high frequency, incidence and multiplicity of tumor data can be obtained but can not be distinguished histopathologically.

Abstract

Rocaglamide derivatives are the compounds which have featuring cyclopenta[b]tetra-hydrobenzofuran skeleton. Until now it includes more than 50 naturally occurring derivatives. They were chosen to be interesting candidates for possible therapeutic agents primarily in the field of cancer chemotherapy due to their cytotoxic activities data against various cancer cells. A quantitative structure activity relationship (QSAR) studies were done to investigate physicochemical properties of molecule which  contribute to their activities. Series of rocaglamide derivatives have been used and analyzed using linear free energy regression Hansch model for their cytotoxic activities against MONO-MAC-6 leukemia cells, RAJI lymphoma cells and MEL-JUSO melanoma cells. Results showed that the best QSAR equations were revealed involving C Log P and CMR parameters with nonlinear regression relationships in cytotoxic activities of rocaglamide derivatives against cancer cells above.

Abstract

Isolation of the main compound of sponge Axynissa aplysinoides collected from Lombok seawater, has been conducted. Methanol extract of dried sponge was fractionated into nonpolar, semipolar and polar fractions using hexane, ethyl acetate and buthanol. Identification and final purification of the main compound was done using column chromatography and high performance liquid chromatography (HPLC). Structure elucidation of the main compound was performed using mass spectrometry (EIMS), 1H- and 13C-NMR spectroscopies. The main compound was identified as (E)-(4-hydroxystyryl) trimethylammonium.Key words : isolation, structure elucidation, Axynissa aplysinoides

Abstract

ABSTRACTA research about hair fertilizer test has been done by using a combination of bitter melon leaf and carrot extracts. Bitter melon leaf (Momordica charantia) is known for having hair nutritive activity, and carrot (Daucus carota L.) is also known for containing vitamin A to nourish the hair scalp. In this research, 3 months old New Zaeland white male rabbit with 2 kg of weight was tested to find out the hair growth activity of single extract and the combination of bitter melon leaf extract (EDP) and Carrot Extract (EW) with a ratio of 1EDP:1EW ; 3EDP:1EW; 3EDP:2EW. The growth and weights of hair that has been smeared on the rabbit's back were observed for 28 days. The results on combination extract with 3EDP: 1EW showed the best result with an average growth of 5.23 mm per week and a weight of 53.44 mg. The result of continued activity test of hair tonic with  concentration of 1%; 2% and 3% showed that hair tonic with 3% concentration had the best activity that is 11.56 mm for the average of hair growth and 51.16 mg for its weight. The result of physicochemical analysis and irritation showed that the color of hair tonic had brown, aromatic, clear, homogeneous, had a specific gravity of 0.9419 and had not causes irritation. Keywords : Hair Tonic Formula, Growth Activity,   Bitter Melon Leaf Extract,   Carrot Extract, Rabbit, Irritation. ABSTRAK Telah dilakukan penelitian pengujian penyubur rambut dari  kombinasi ekstrak daun pare dan wortel. Daun pare (Momordica charantia ) diketahui memiliki aktivitas sebagai penyubur rambut, dan wortel (Daucus carota L .) mengandung vitamin A untuk menutrisi kulit kepala rambut. Dalam penelitian ini dilakukan pengujian pada kelinci jantan New Zealand White berumur 3 bulan dengan bobot 2kg untuk mengetahui aktivitas penumbuh rambut ekstrak tunggal dan  kombinasi dari Ekstrak Daun Pare (EDP) dan Ekstrak Wortel (EW) dengan perbandingan 1EDP:1EW; 3EDP:1EW; 3EDP:2EW. Pertumbuhan rambut dan bobot rambut yang telah diolesi pada punggung kelinci diamati selama 28 hari. Hasil uji pada  kombinasi ekstrak, perbandingan  3EDP:1EW menunjukkan hasil terbaik dengan rata-rata pertumbuhan 5.23 mm perminggu dan bobot 53.44 mg. Hasil uji aktivitas lanjutan sediaan hair tonic dengan konsentrasi 1%; 2% dan 3%, menunjukkan bahwa sediaan hair tonic dengan konsentrasi 3% memiliki aktivitas paling baik yaitu rata-rata panjang rambut 11.56 mm rambut dan bobot rambut 51.16 mg. Hasil uji analisis fisikokimia dan uji iritasi menunjukkan bahwa sediaan hair tonic berwarna coklat, bau aromatik, jernih, homogen, memiliki berat jenis 0,9419 dan tidak menimbulkan iritasi.Kata kunci : Formula Hair Tonic,  Aktivitas Penyubur Rambut,   Daun Pare,   Wortel,  Kelinci, Iritasi

Abstract

Background: Animal models of Alzheimer's have now been developed in various studies to become potential animal models suitable for clinical conditions. Alzheimer's in clinical conditions is characterized by Amyloid βeta (Aβ) plaques and intracellular tangles of neurofibrillary tangles as well as disorders of the acetylcholinesterase enzyme in the hippocampus. Rodents used include rats and mice. Mice are used because they are efficient and parts of the brain can be penetrated with a needle without requiring surgery. Objectives: Appropriate toxicant induction is needed so that the process of developing Alzheimer's pathology is known, namely the use of Streptozotocin. Intracerebroventricular induction of strepzotocin in mice needs to be further understood. Material and Methods: The research method uses literature review with identification, screening, evaluation and interpretation of all research results collected through online search engines such as; Sciencedirect, Pubmed, GoogleScholar with keywords and meet the inclusion criteria Results: The results of the synthesis obtained selected data from 8 articles. ICV-STZ has been proven from various studies to induce Alzheimer's pathology in both young and old male mice through neuroinflammatory pathways, oxidative stress, increasing Aβ expression and hyperphosphorylation of Tau as Alzheimer's markers which are similar to clinical patients. Abstract

Content Benger plant extract compounds (Lagerstroemia ovalifolia Teijsm. & Binn.) are reported as a new class of COX-2 enzyme inhibitors. Molecular docking analysis of Benger plant extract compounds (Lagerstroemia ovalifolia Teijsm. & Binn.) against COX-2 enzyme is required to determine the affinity and interaction pattern between the above compounds and COX-2 enzyme. A total of 23 Benger metabolite compounds were evaluated against COX-2 protein structure (PDB: 5KIR) using Molegro Virtual Docker (MVD) software. The docking results showed that three compounds, namely Kaempferol-3-O-acetyl-glucoside isomer, Acacetin malonylgalactoside, and Kaempferol-3-Oa-L-rhamnoside, had a lower rerank score (more negative) compared to the ligand (RCX_601), indicating potential as COX-2 inhibitors that affect inflammation regulation in silico. In addition, in silico ADMET analysis using pkCSM showed that the three compounds were classified as non-toxic according to BPOM Regulation no. 10 of 2022. Analisis docking molekuler senyawa Senyawa ekstrak tanaman Benger (Lagerstroemia ovalifolia Teijsm. & Binn.)  terhadap enzim     COX-2 diperlukan untuk mengetahui afinitas dan pola interaksi antara senyawa di atas dengan enzim COX-2. Sebanyak 23 senyawa metabolit Benger dievaluasi terhadap struktur protein COX-2 (PDB: 5KIR) menggunakan perangkat lunak Molegro Virtual Docker (MVD). Hasil docking menunjukkan bahwa tiga senyawa, yaitu Kaempferol-3-O-acetyl-glucoside isomer, Acacetin malonylgalactoside, dan Kaempferol-3-Oa-L-rhamnoside, memiliki rerank score yang lebih rendah (lebih negatif) dibandingkan dengan ligan (RCX_601), menunjukkan potensi sebagai inhibitor COX-2 yang mempengaruhi regulasi peradangan secara in silico. Selain itu, analisis ADMET secara in silico menggunakan pkCSM menunjukkan bahwa Kaempferol-3-O-a-L-arabinoside (Juglalin) lebih mudah diabsorpsi oleh usus dibandingkan dengan kedua senyawa lainnya namun ketiga senyawa tersebut diklasifikasikan tidak bersifat toksik. Abstract

Present investigation shows that the extract of C. aeruginosa attenuates DMBA-induced spleen carcinogenesis in Wistar rats. Three-week female Wistar rats were treated with three different C. aeruginosa extract doses (CA1: 40 mg/200 g body weight, BW; CA2: 80 mg/200 g BW; CA3: 160 mg/200 g BW) and were induced with DMBA after one-week administration of these doses. A commercial immunostimulant, and DMBA only were also given to each group as positive and negative control, respectively. The development of tumors was evaluated by investigating the incidence of tumor and tumor multiplicity during the experiment. Spleen mass index and histological parameters such as white pulp, centrum germinativum, and marginalis zone were also examined. Based on our study, the administration of C. aeruginosa extract during and after carcinogen induction gave several impacts on rat carcinogenesis. At the extract dose of 80 mg/200 g BW, tumor incidence of animals were least (P<0.05). However, all doses did not show any effect to the spleen mass index, though the highest dose (160 mg/200 g BW) was found to cause changes in white pulp and marginalis zone boards. This trend indicates that it takes higher dose to cause an immune response effect reaching the organs.

Abstract

Andaliman (Zanthoxylum acanthopodium D.C) is a plant widely used by the Batak people in North Sumatera. Several studies demonstrated that Andaliman fruit exhibits antioxidant activities and cytotoxic effects on cancer cells. Quality of Andaliman fruit could affected by post-harvest process including storage time. This research aims to analyze the compound, antioxidant activity and proliferation inhibition on MCF-7 cells of Andaliman fruit extracts (AFE) stored at varying storage time. A series of fresh 250 g Andaliman fruit sealed in a closed container and stored in the oven at 30ºC. After storage at 0, 24, 48, 72, 96, 120 and 144 hours, Andaliman fruit macerated using methanol and obtained successive extract yields of 4.12; 3.70; 12.79; 8.17; 8.15; 3.7 and 2.21. These extracts were analyzed for total phenolic, total flavonoids, free radical scavenging activity and proliferation inhibition on MCF-7 cells. The chemical compound analysis of AFE was performed by ultra performance liquid chromatography (UPLC). Results showed that the AFE stored at zero hours had higher total phenol content (47.32 mg gallic acid equivalent (GAE)/g of extract) and free radical scavenging activity (50.38%) at 500 µg/mL significantly compared to other extracts. The total flavonoid content of all extracts showed no difference level except for the extract at 120 hours of storage. The proliferation inhibition test on MCF-7 cells at 100 µg/mL showed that AFE stored at 72, 96 and 144 hours could inhibit MCF-7 cells above 50%. The storage time variation of Andaliman fruit may affect the total phenolic and flavonoid content, and also activity of free radical scavenging and proliferation inhibition on MCF-7. The UPLC analysis founded the major compound of AFE was predicted as α-sanshool. Analysis of chemicals substance in Andaliman fruit with varying of storage time need to be conducted to evaluated alteration of secondary metabolites contained in Andaliman fruit.
